    Notes: Aims : To investigate the value of platelet-derived growth factor receptors (PDGFRs) by immunohistochemistry in discriminating KIT-negative gastrointestinal stromal tumours (GISTs) from other soft-tissue neoplasms of the digestive tract.Methods and results : One-hundred and sixty-seven primary gastrointestinal mesenchymal tumours (125 GISTs, 15 intra-abdominal desmoids, 12 leiomyomas, eight leiomyosarcomas, three schwannomas, two solitary fibrous tumours, and one case each of inflammatory pseudotumour and fibroid polyp) were reclassified based on morphology and on the immunohistochemical panel recommended by the National Institutes of Health consensus on GIST. All cases were then tested with antibodies specific for PDGFR α and β. Of 125 GISTs, 117 were KIT-positive (93.6%) and eight KIT-negative (6.4%). All the KIT-positive GISTs were negative for both PDGFRs, while all the eight KIT-negative GISTs expressed PDGFR-α, with two of them also coexpressing PDGFR-β. Among the 42 non-GIST tumours, only a small percentage (26.6%) of desmoids immunostained for PDGFR-α, two of them coexpressing PDGFR-β.Conclusions : Immunostaining with PDGFR-α is a helpful marker in discriminating between KIT-negative GISTs and other gastrointestinal mesenchymal lesions: all KIT-negative GISTs were positive for PDFGR-α, while none of the other gastrointestinal mesenchymal tumours analysed, except a small subset of desmoids, was reactive with anti-PDGFRs. These preliminary data demonstrate the suitability of commercially available antibodies to detect immunohistochemically the mutually exclusive expression of KIT and PDGFR-α previously reported in GISTs by molecular biological techniques. Since PDGFR exists in the form of a homodimer (αα, ββ) or heterodimer (αβ) and two of the KIT-negative GISTs coexpressed both PDGFR isoforms, further investigations are required to elucidate the role of PDGFR-β in GISTs.

    Notes: Summary The aim of the present study was to investigate whether the ovulation-maintaining effect of melatonin in rats, exposed to continuous light (LL), was also exerted by other pineal indoles which have been reported to influence the reproductive processes of mammals. The effect of 10μg melatonin was compared with that of similar amounts of either N-acetylserotonin, 5-methoxytryptophol, 5-methoxyindole-3-acetic acid, 5-hydroxytryptophol, 5-methoxytryptamine or 5-methoxytryptophan. All these compounds appeared to be significantly less effective than melatonin in preventing the effect of LL, ovulation being preserved in only 20–33 % of the rats investigated, with melatonin this percentage being 60–75%. Investigations were also carried out to assess the effect of these indole derivatives on HIOMT (hydroxyindole-O-methyl transferase) activity in synthesizing different 5-methoxyindoles in the abnormally influenced pineal gland due to LL. Melatonin, the compound the effect of which on ovarian cyclicity is strongest, stimulates 5-methoxytryptophol synthesis; while other less active compounds stimulate the synthesis of melatonin and inhibit that of O-acetyl-5-methoxytryptophol. The possibility that the effect of other indoles than melatonin on ovarian cyclicity might be due to stimulation of melatonin synthesis was considered. A possible functional relationship of the different indoles cannot be excluded.

    Description / Table of Contents: Zusammenfassung Nach Pinealektomie konnte eine Anhäufung von Eisenpigment in den Histiocyten der Pulpastränge und der Follikel in der Rattenmilz festgestellt werden; parallel damit ging eine leichte Erhöhung der enzymatischen Aktivität der Histiocyten und eine Hypertrophie des argyrophilen Reticulums. Die Anhäufung von Eisenpigment und Erhöhung der enzymatischen Aktivität kann weiter gesteigert werden durch subcutane Injektion von Trypan-Blau. Folgende Arbeitshypothese wird zur Erklärung dieser Tatsache vorgeschlagen: Die Pinealektomie führt unmittelbar zu einer Erhöhung der Aktivität der Histiocyten im allgemeinen und des Abbaues der roten Blutkörperchen durch die Milz im besonderen.
    Notes: Summary After pinealectomy we were able to observe an accumulation of iron pigment in the histiocytes of the cords and of the follicles of the spleen in the rat, parallel with a slight increase in the enzymatic activity of the histiocytes and hypertrophy of the argentophil reticulum. The accumulation of iron and the enzymatic activity can be further increased by subcutaneous injection of trypan blue. The working hypothesis is proposed that pinealectomy leads directly to an increase in the histiocytic activity in general and in the red cell destruction by the spleen in particular.
