    Notes: Summary The metastasis of malignant tumors from a primary site to near and distant secondary sites is probably the most important event in the pathogenesis of cancer and it accounts for most cancer deaths1. Whereas advances in the treatment of primary cancer have led to increased patient survival, metastatic cancers are still the most difficult group of diseases to treat successfully2. As organ-characteristic lectins play an important role in the organ manifestation of metastatic islets3,4, it might be possible (e.g. during surgical operations on malignant tumors) to block those organ-characteristic lectins with the appropriate receptor-bearing glycoconjugates in order to inhibit the metastatic spread. Recent experiments have demonstrated that neuraminidase treatment of tumor cells (mouse sarcoma-1) alters in vivo (Balb/c-mice) the organotropic distribution of metastases; instead of being found exclusively in the lung, they are found both in lung and liver. However, pre-injection and regular application of D-galactose — the same holds for arabinogalactan5,6,13 — prevents the settling of metastases in the liver but does not influence the metastatic process to the lung, whereas mannan — as a galactose-free control substance — does not alter the initial pattern of metastasis to lung and liver.

    Description / Table of Contents: Zusammenfassung Es wird über die Spezifität des Lektins aus Soja hispida berichtet. Zunächst werden verschiedene Erythrozyten-Agglutinationstypen beschrieben. Dann wird an Hand von Hemmtesten gezeigt, daß das Agglutinin mit terminalem α- und β-gebundenem N-Acetyl-D-galaktosamin reagiert, nicht jedoch mit subterminalem. Abschließend wird ein Vergleich mit anderen Agglutininen, die ähnliche Spezifität haben, gegeben.
    Notes: Summary A report on the specificity of the lectin from Soja hispida is given. First of all different agglutination types of red blood cells are described and secondly the specificity is elucidated by hemagglutination inhibition tests. It is suggested that the agglutinin can react with terminal α and β linked N-acetyl-D-galactosamine but not with the subterminal hexosamine. A comparison with other antibody-like substances having a similar specificity is given.

    Notes: Summary An anti-A-agglutinin from the albumin gland ofHelix aspersa was purified with the use of Sephadex adsorption and subsequent elution with N-acetyl-D-glucosamine. This purification was studied also in immunoelectrophoresis. The agglutinin has no C′ or hemolytic properties.

    Description / Table of Contents: Summary The serological specificity of some agglutinins from snails is extensively discussed. A new agglutinin is described, which detects a receptor called Mduc, the specificity of which is mainly determined by N-acetyl-neuraminic acid. The receptor occurs on certain red cells, tumor cells and in some glycoproteins.
    Notes: Zusammenfassung Die serologische Spezifität einiger Agglutinine aus Schnecken wird ausführlich diskutiert. Dabei wird ein neues Agglutinin beschrieben, welches einen Rezeptor Mduc erfaßt, dessen Spezifität in der Hauptsache durch N-Acetyl-Neuraminsäure bestimmt wird. Der Rezeptor kommt auf verschiedenen Erythrozyten, Tumorzellen und in einigen Glykoproteinen vor.

    Notes: Summary The granular cell tumourettes of the posterior lobe of the pituitary possess neuraminic acid containing carbohydrate. After removal of neuraminic acid with neuraminidase and exposure to FITC (Fluorescein isothiocyanate) labelled peanut agglutinin (Arachis hypogaea) (PNA), intracellular receptor structures could be demonstrated. The significance of the findings is discussed.

    Notes: Summary Development of liver metastases in 1542 cancer patients was investigated. It was found that in certain liver diseases the incidence of liver metastases was reduced compared to that in cancer patients with otherwise normal livers. We propose that this reduction may be due to a reduced function of the liver-specific lectins.

Three different methods are described for the visualisation of the Thomsen-Friedenreich (TF) antigen on cell suspensions, formalin-fixed and paraffin embedded or frozen tissue sections: a) Rosette formation with chicken and sheep erythrocytes. b) Fluorescence-microscopy with fluorescein labelled peanut agglutinin. c) Autoradiography with3H-labelled peanut agglutinin. 2. The TF antigen was shown, as far as presently investigated, to be exposed on various blood cells, glomerula of the kidney and normal mammary gland after neuraminidase treatment. Mammary gland was also shown to possess TF receptors without prior treatment with neuraminidase. 3. The exposure of this cryptantigen can be brought about by bacterial or viral neuraminidase and is followed by an antigen/antibody reaction, which can lead to possible pathological consequences.
    Notes: Zusammenfassung 1. Drei verschiedene Methoden zur Darstellung von sog. Thomsen-Friedenreich (TF)-Antigenen in Zellsuspensionen, an Formalin-fixiertem und in Paraffin eingebettetem Gewebe sowie in Gefrierschnitten werden beschrieben: a) Rosetten-Bildung mit Hühner- und Schaferythrocyten. b) Fluorescenzmikroskopie mit Fluorescein-markiertem Erdnußagglutinin. c) Autoradiographie mit3H-markiertem Erdnußagglutinin. 2. Nach Neuraminidasebehandlung konnten TF-Antigene — soweit bisher untersucht — auf verschiedenen Blutzellen, in Nierenglomerula und in normalem Brustdrüsengewebe nachgewiesen werden. Ein Teil derartiger Antigene lag im Brustdrüsenparenchym auch bereits als freie Rezeptoren vor, d. h. waren nicht von Neuraminsäure bedeckt. 3. Die Freilegung dieser Kryptantigene kann im Rahmen von Infektionen durch bakterielle und virale Neuraminidase erfolgen. Dabei kommt es zu einer Antigen/Antikörper Reaktion mit unter Umständen klinisch relevanten Folgen.

    Notes: Summary Isoelectric focusing was the method applied to demonstrate the heterogeneity of the erythroagglutinin receptor for the lectin from Phaseolus vulgaris. The receptor activity was found in the fractions containing neuraminic acid and not in the neutral glycoprotein material.

    Description / Table of Contents: Zusammenfassung EC-2 A-se verhindert nicht nur die Umwandlung von PHA-stimulierten menschlichen Lymphocyten zu blasten-ähnlichen Zellen, sondern auch ihre zellzerstörende Wirkung auf HeLa-Zellen in vitro (cytoaggressiver Effekt). Das charakteristische Verhalten der stimulierten Lymphocyten gegenüber den HeLa-Zellen bleibt jedoch unter der Wirkung von EC-2 A-se unbeeinflußt. Daraus leitet sich die Vermutung ab, daß dieses Verhalten allein nicht ausreicht, um HeLa- oder, ganz allgemein, Target-Zellen in vitro zu zerstören. Zusätzliche Faktoren, die durch EC-2 A-se unterdrückt zu werden scheinen, sind offenbar für das Zustandekommen des cytoaggressiven Effektes notwendig.
    Notes: Summary EC-2 A-se not only suppresses the transformation of PHA-stimulated human lymphocytes into blast like cells but also their destructive effect on HeLa cellsin vitro (cytoaggressive effect). The typical behaviour of the stimulated lymphocytes towards HeLa cells, however, remains unaffected by EC-2. It may be concluded thatin vitro this behaviour alone would not suffice to destroy the HeLa cells, or target cells more generally. Additional factors, being most probably suppressed by EC-2-Ase, seem necessary to bring about the cytoaggressive effect.
