    Description / Table of Contents: Summary A rapid method is reported for testing the sensitivity of tumors to cancerostatic agents. The method was developed using solid tumors that were treated with hydroxyurea agents. The method may be performed within a few hours and should prove applicable for testing the sensitivity of human tumors to cancerostatic agents.
    Notes: Zusammenfassung Es wird eine Schnellmethode der Sensibilitätstestung von Tumoren beschrieben, die ihre Brauchbarkeit für solide Tiertumoren unter Einwirkung von Hydroxyharnstoff erwiesen hat. Sie ist innerhalb weniger Stunden durchführbar und dürfte sich auch in der Humanmedizin für die Testung der Empfindlichkeit bösartiger Geschwülste gegenüber Cytostatica eignen.

    Description / Table of Contents: Summary The effect of 1.38×10−5 M adriamycin on tritiated uridine incorporation was studied after 3 hours treatment of suspensions of 25 advanced human lung carcinomas in vitro. The results were correlated with the responses to clinical therapy. All tumours which showed an inhibition of uridine incorporation in vitro of more than 40% were also sensitive to clinical treatment with adriamycin.
    Notes: Zusammenfassung Es wurde die Wirkung von 1,38×10−5 molar Adriamycin auf den 3H-Uridineinbau an Zellsuspensionen von 25 menschlichen Lungentumoren untersucht und mit dem Therapie-Erfolg in der Klinik in Beziehung gesetzt. Alle Tumoren mit einer Uridineinbauhemmung in vitro von mehr als 40% waren auch in der Klinik auf eine Behandlung mit Adriamycin sensibel.

    Description / Table of Contents: Zusammenfassung Mit einer früher beschriebenen in vitro-Schnellmethode (DNS-Synthese) zur Sensibilitätstestung wurden Gewebsproben von 21 operativ entfernten menschlichen Tumoren untersucht. Eine Testung gegenüber Trenimon, Methotrexat und Natulan in 3 Dosierungen zeigte, daß selbst Tumoren gleicher Lokalisation und gleicher Histologie unterschiedlich reagieren. Das bisher vorgelegte Material reicht zwar für eine statistische Sicherung nicht aus, dennoch scheint es, daß die beobachtete Individualität der Tumoren bei weiteren Untersuchungen gesichert werden kann. Dies würde bedeuten, daß eine Chemotherapie auch histologisch gleichartiger maligner Tumoren individualisiert werden muß.
    Notes: Summary Slices from 21 human tumors were obtained immediately after resection. The tissue samples were exposed to Trenimon, Methotrexat, and Natulan. The incorporation of3H-thymidine into the DNA of these slices was evaluated by a rapid and simple method described earlier. It could be demonstrated that all tumors, even those of a same origin and with the same histological features showed individual sensitivity against any one of these cytostatic drugs.—The findings presented here are not yet sufficient for statistical evaluation. If the individuality of every single tumor can be confirmed in further experiments, the chemotherapy of even histologically similar malignant tumors will have to be individual too.

    Description / Table of Contents: Zusammenfassung Mit einer früher beschriebenen Methode zur Sensibilitätstestung (3H-Thymidineinbau, Flüssigkeitsscintillationsmessung) haben wir 30 menschliche Adenocarcinome (10 Rectum-Sigma-Carcinome, 10 Magen-Carcinome, 30 Mamma-Carcinome) gegenüber Trenimon®, Methotrexat® und Natulan® getestet und statistisch ausgewertet. Die in unserer früheren Untersuchung vermutete Individualität der Tumoren konnte bestätigt werden.
    Notes: Summary Using a sensibility testing method previously described by the authors (3H-thymidine incorporation, liquid scintillation counting), the sensitivity of 30 human tumours (10 recto-sigmoid carcinomas, 10 gastric carcinomas, 10 carcinomas of the breast) was tested in vitro against Trenimon®, Methotrexat® and Natulan®. The results obtained were evaluated statistically. Each tumour proved to react specifically to the agents.

    Description / Table of Contents: Zusammenfassung Auf Grund tierexperimenteller Untersuchungen wird belegt, daß Therapieergebnisse bei Tumoren mit in vitro-Testverfahren voraussagbar sind. Die Über-einstimmung zwischen bisherigen Testergebnissen and Therapieergebnissen beim Men schen sind zwar hoffnungsvoll, einen eindeutigen Nachweis können jedoch erst prospektive, statistisch auswertbare klinische Studien bringen. Zwei Erkenntnisse erhält man schon heute aus den Testergebnissen: Hinweise auf die Proliferationsrate, von der die Zytostatika-Wirkung abhängt, und Hinweise auf biochemisch bedingte Resistenzen.
    Notes: Summary On the basis of animal experiments it is shown that the results of tumor chemotherapy can be predicted with the aid of in-vitro test systems. In investigations carried out on human tumors, the correlations obtained between in-vitro test results and response to therapy are promising. However, definitive proof of a correlation can only be provided by planned, statistically supported clinical studies. Two types of information can already be obtained from the in-vitro tests: data concerning the rate of proliferation of the tumor, on which the activity of cytostatic agents depends, and data on certain types of resistance to cytostatics.
