    Notes: The aim of this study was to develop an assay to assist in the diagnosis of delayed onset of adverse responses to cow milk in children, by measurement of gamma-interferon (GIFN) produced in vitro in response to β-lactoglobulin-stimulated blood mononuclear cells. Diagnostic procedures identified 75 children with immediate reactions who had high total IeE and IgE-isotype responses to cow milk, 17 children who developed reactions after 24 h and had low total IgE and low IgE-isotype response to cow milk and 59 milk-tolerant children. GIFN production was less in children with immediate reactions compared to those with late reactions (P≤0.009) or milk-tolerant children (P=0.022). The results of this study suggest enhanced T-cell reactivity may be involved in the immuno-pathogenesis of non-immediate cow milk allergy, but GIFN production was not a clinically useful diagnostic test.

    Notes: We investigated the response of the Datex Capnomac to alcohol in expired air. The greatest response was seen when the Capnomac was set to monitor halothane. There was an approximately linear correlation with blood alcohol levels. This effect is of sufficient magnitude to introduce inaccuracies in the measured concentration of halothane.

    Notes: Cellular responses to several purified antigens oi Mycobacterium bovis were examined in experimentally infected cattle over a period of 36 months, using in vitro cellular proliferation and interferon-γ assays. These antigens (12, 19, 22a, b. 24, 25.30, 32, 39.65 and 70 kDa) included the majority of M. /lom protein antigens described to date and are highly homologous to those purified from M. tuberculosis. Cellular responses vi'ere examined at 3-month time intervals during the 36-month course of infection. All purified antigens induced cellular immune responses in the infected animals. The onset and magnitude of response to individual antigens varied among the animals. At any specific time during the period of infection one or more antigens appeared to be immunodominant but the immunodominance profile changed as the infection progressed. Humoral immune responses were low or absent in the first half of the infection period, but increased substantially for some of the antigens during the second half. Variation was observed among the different animals as to which antigens they recognized.

    Notes: A Mycobacterium bovis antigen has been purified from culture filtrate by chromatofocusing. This antigen is a major component of culture filtrate and cell extracts and shows a considerable degree of micro-heterogeneity in electric charge and molecular weight. Studies with monoclonal and polyclonal antibodies raised against the purified antigen show that some of its antigenic determinants also occur in higher molecular weight species in culture filtrate and particularly in whole cell preparations. Immunoblotting and ELISA studies, using sera from M. bovis-infected animals, showed that this antigen is one of the most immunoreactive components of M. bovis, recognized by the majority of animals with detectable antibody response to M. bovis. The specificity of the purified antigen is far superior to that of the crude culture filtrate, with very few false positive results. The purified antigen also elicits strong in vivo and in vitro cell-mediated responses The amino acid compositions of two variants of this antigen have been determined and found to be similar to that of MPB-70.

    Notes: Summary Adrenal cells from C57BL/Tb mice produced more steroid than those from DBA/2J mice. Reciprocal differences between both backcross and F1 hybrids showed a persistent maternal effect. Mothers with high output produce offspring with reduced hormone production when adult. Corticosterone output thus depends on maternal phenotype as well as on the genotype of the isolated cells.

    Notes: Abstract Infection withMycobacterium avium, M. intracellulare and M. scrofulaceum (MAIS) organisms in normal children may result in cervical lymphadenopathy. There is a poor response to anti-tuberculous therapy and surgical excision of infected nodes is necessary. The diagnosis therefore requires consideration in children with cervical lymphadenopathy. A simple in vitro assay is described which may be useful for diagnosis prior to excision. Whole blood is cultured withM. avium purified protein derivative. After 24h the plasma is removed and the concentration of gamma interferon (IFN-γ) quantified by a radioimmunometric assay. In a prospective study of 38 children with neck lesions, 16 showed MAIS organisms isolated from cultures of excised tissue. The level of IFN-γ produced by peripheral blood lymphocytes from children in the MAIS group was significantly greater when compared with children having operations for neck lesions that were not due to MAIS organisms (P〈0.001).
