Summary After a review ofliterature on the problem of splenomegaly and “hypersplenism”, the authors report on studies with liquid cultures of human bone marrow for the investigation of the actions of albumin-free spleen extracts. In corroboration of earlier results with cover glass cultures, the authors found an inhibition of maturation partly dependent on the dose; this inhibition was expressed by a decrease in the absolute numbers of the polymorphonuclears. The authors present the findings for 4 extracts studied, and, in conclusion, discuss whether the active substances are specific in the actual sence. In addition to the possibility that other parts of the RES, too, can produce such substances or substances with a similar action, it is pointed out that the catabolic or metabolic products of the increased number of cells dying in an enlarged spleen may be involved.

Zusammenfassung Nach einer Literaturübersicht zum Problem Splenomegalie und „Hyperspleinsmus” wird über Untersuchungen mit Flüssigkeitskulturen von Menschlichem Knochenmark berichtet, an denen die Wirkungen eiweißfreier Milzextrakte untersucht wurden. In Bestätigung früherer Ergebnisse mit Deckglaskulturen fand sich eine z. T. dosisabhängige Reifungshemmung, die in einem Absinken der absoluten Zahlen für die Segmentkernigen zum Ausdruck kam. Die Befunde für 4 untersuchte Extrakte werden migeteil und abschließend diskutiert, ob die wirksamen Substanzen spezifisch im eigentlichen Sinne sind. Neben der Möglichkeit, daß auch andere Teile des RES solche oder ähnlich wirksame Stoffe produzieren können, wird darauf hingewiesen, daß es sich vielleicht um Abbau- oder Stoffwechselprodukte der in einer vergrößerten Milz vermehrt zugrunde gehenden Zellen handelt.
