    Notes: Adult male rats of Simonsen strain were given a dose of 5000 r at 50 peak KV of x-irradiation directed at the lower lip, which was everted through a hole in a lead rubber cylinder shielding the head and body of the animal. Light and electron microscopic observations were made on specimens of lip oral mucosa from animals killed at 2, 6, 26 and 50 h and at 12 days after irradiation. The experimental model met the aim of permitting the study of the sequential effects of a high dose of irradiation without causing ulceration of the mucosa.Widespread degenerative changes were noted in the basal cells as early as 2 h after x-irradiation, increasing in degree up to 50 h. They included inflation of the outer nuclear envelope and rough endoplasmic reticulum (RER) with loss of ribosomes, swelling of mitochondria and disarrangement of cristae evident at 2 h, followed at 6 h by swelling of nucleus and cytoplasm and 26 h by frank membrane breaks. Irreversible degrees of damage were noted in a small though growing minority of cells. Immediate mobilization of the Golgi-lysosomal system was evident as an increase in size of the zone, maximal at 2 h, and subsequent autophagic activity. Signs of recovery, beginning at 26 h, were noted in nuclear envelope, RER, intercellular space, and in mitotic activity by 50 h. Virtually complete recovery was seen at 12 days.It is held that the successful confinement of irradiation to a small tissue volume, the effective activity of the lysosomal system and the short epithelial turnover time were important factors in limiting the damage and in permitting recovery.

    Notes: Abstract The transfer of solute through a membrane separating two aqueous solutions is studied with the time-dependent diffusion equation for composite media. By introducing new independent and dependent variables it is shown that the differential equations and boundary conditions can be transformed into a dimensionless form which does not explicitly depend on the diffusivities of the media. Laplace transforms are used to derive explicit solutions for the solute concentration as a function of position and time. It is shown that at large time the concentration approaches the equilibrium distribution exponentially. Explicit results are given for the decay time as a function of the parameters of the system. In addition, an accurate and simplified expression is derived for the decay time for the case of small membrane permeability. The accuracy of the analytic solutions for the concentration profiles is tested by comparing them with numerical results obtained by solving the diffusion equations by the method of finite differences. Excellent agreement is found.

    Notes: Abstract Past measurement of gastric emptying of solid food in man has depended on external counting of surface-absorbed isotopes without verification that isotopic labels remain attached to solid food in the stomach. In this study chicken liver was isotopically labeled with99mTc incorporated uniformly and intracellularly throughout the liver substance.In vitro studies showed less than 10% loss of99mTc from liver incubated with pepsin HCl. By contrast, up to 90% of51Cr adsorbed to scrambled eggs became detached under similar conditions. In feeding experiments less than 10% of99mTc was liberated from fed99mTc liver, while significantly more51Cr became detached from egg under identical intragastric conditions. We conclude that99mTc-tagged chicken liver is an adequate marker of the rate of emptying of solid food and appears to be more reliable than51Cr-labeled scrambled eggs from which51Cr dissociates in the stomach.

    Notes: Abstract The differing sensitivities of the gallbladder and pancreas to graded doses of CCK have been claimed as important in normal digestive events. As both secretin and CCK are thought to be released in response to normal feeding, we have reexamined the sensitivities of the gallbladder and pancreas to graded increases of CCK with a constant secretin background. Under such conditions the increase in output of pancreatic trypsin and bile salt occurred simultaneously, the dose-response curves being almost superimposable. Thus, under conditions more closely approaching normal postcibal events, the previously described differing sensitivities of the pancreas and gallbladder were not observed.
