    Notes: A new method which should have relatively general applicability for the identification and quantitative analysis of reactive adsorbed molecular intermediates in surface reactions will be described, and the first examples of its application will be presented. When a reactive intermediate is generated on a surface, it often has a tendency to dissociate before desorbing. Since dissociation generally requires additional free sites on the surface, dissociation can be suppressed and desorption correspondingly enhanced if the free sites on the surface can be properly poisoned. We have found that bismuth adatoms are very good inert site blockers, which can be postdosed to the surface of a transition metal containing a reactive adsorbed hydrocarbon without destroying the hydrocarbon. Whereas in the absence of bismuth, the hydrocarbon would completely dehydrogenate during thermal desorption spectroscopy (TDS) and liberate only H2 into the gas phase, after bismuth postdosing the reactive hydrocarbon desorbs intact for mass spectral identification and quantitative analysis. This method has been used to prove that adsorbed benzene is the initial product of the dehydrogenation of cyclohexane on Pt(111) at ∼235 K. In the absence of bismuth, this benzene all dissociates during TDS to liberate only H2, leaving graphitic carbon residue on the surface. When one-third monolayer of Bi is postdosed at 110 K, the dehydrogenation pathway is sterically poisoned and the adsorbed benzene quantitatively desorbs during TDS, where it is unambiguously identified by mass spectroscopy. By briefly heating the reactive adsorbed intermediate to increasing temperatures prior to Bi deposition, the thermal stability limits of the intermediate and the kinetic parameters for its dissociation can be established. This is demonstrated for the dehydrogenation reaction of adsorbed cyclopentene on Pt(111). Bismuth postdosing in thermal desorption mass spectroscopy (BPTDS) should be a very useful but inexpensive addition to surface analytical capabilities.

    Notes: We have investigated the use of frequency variation in ac calorimetry in the measurement of thermal quantities within controllable distances of the sample heater and thermometer. This technique, useful in studies of heterogeneous samples where only a small part of the sample is of interest, e.g., surfaces, thin films, and interfaces, is applied to study the thermal properties of a test sample made from indium (In) sandwiched between layers of bulk gallium arsenide. We measure and analyze the frequency dependence of the thermal response of the sample, as well as the variation with frequency of the relative size of the anomaly observed at the In superconducting transition. The data are consistent with a model assuming a thermal diffusion-limited sampling volume.

Two groups of spontaneously hypertensive rats (SHR) were treated with enalapril (25–30 mg/kg per day): Group I received treatment from 4 to 14 weeks of age to inhibit development of hypertension and Group R received the drug from 14 to 20 weeks of age to reverse established hypertension.2. Systolic blood pressure, ploidy of aortic smooth muscle cells (flow cytometric DNA analysis) and aortic hypertrophy (medial cross-sectional area) were determined at times both during and after enalapril treatment (up to 30 weeks).3. Enalapril treatment normalized blood pressure to that of age-matched Wistar-Kyoto rats in both groups. Blood pressure rose again following cessation of treatment.4. In untreated SHR the incidence of polyploid cells increased concomitantly with increasing pressure throughout the time studied, whereas in Group I the incidence remained low. In Group R, the incidence of polyploidy directly paralleled both the decrease (normalization) and the rise in blood pressure following cessation of treatment.5. Hence, the incidence of vascular smooth muscle cell polyploidy is not simply a result of growth of the vessel with increasing age of the SHR, but parallels inhibition, reversal, and redevelopment of hypertension.

    Notes: Summary Both metabolic rate and protein catabolism are known to increase following severe head trauma, but the etiology of this hypermetabolism is unknown. To further investigate the problem, we studied the metabolism of 17 patients with indirect calorimetry who had severe craniocerebral trauma only and who required ICP monitoring for management. Patients were studied daily and immediately after ICP spikes greater than 20 mmHg, prior to treatment with hyperventilation, osmotic diuretics, or barbiturates. Oxygen consumption (VO2) was correlated with ICP. Two groups of patients were identified. Group I patients were treated with hyperventilation and osmotic diuretics while Group II patients additionally received cerebral metabolic depressants. Group I had a significant correlation coefficent between VO2 and ICP. Significant hypercatabolism early in the post trauma period was demonstrated by increased urine urea nitrogen. Our observations suggest that in patients with craniocerebral trauma, elevated ICP is associated with increased oxygen consumption, protein catabolism and systemic hypermetabolism. Cerebral metabolic depressants blunted increases in VO2 which were seen with elevated ICP.

    Notes: Abstract Twenty-one primary bone tumours of the hand in children from 8 paediatric hospitals are reported. Osteochondromas and enchondromas were not included. Our material consisted of 16 patients withcommon tumours (3 Ewing's sarcoma, 5 aneurysmal bone cyst, 6 osteoid osteoma and 2 epidermoid cyst) and 5 patients withuncommon tumours (osteoma, simple bone cyst, haemangiopericytoma, capillary angiomatous tumour and benign ossifying fibroma or osteoblastoma). The X-ray diagnosis of thecommon tumours should have high concordance with histology, whereas that ofuncommon tumours is much more difficult and uncertain. The characteristic features of Ewing's sarcoma are stressed as all our children with this tumour had a delayed diagnosis and a fatal outcome. Differential diagnosis with other short tubular bone lesions of the hand — specifically osteomyelitis — is discussed and the possibilities of microscopic diagnosis are stressed.

    Notes: Abstract Restriction endonuclease cleavage maps have been constructed for the genome of a canine adenovirus type 1 (CAV-1) vaccine strain (CLL; Connaught Laboratories, Ltd., Willowdale, Ontario). Restriction enzyme analyses were also carried out on CAV-1 (CLL) genomes isolated from viral stocks over 8 serial passages in a dog kidney cell line (DK 6722). The right hand 20% of the genome became more heterogeneous in size with increasing passage in DK 6722 cells due to deletions up to 3–4 kb, whereas the left terminal region was stable throughout these passages. A comparative study of CAV-1(CLL) and a virulent strain of CAV-1, Glaxo, revealed that the genome of CAV-1(CLL) was the shorter, by about 480 bp, within the region covering 0.83–0.91 map units. By virtue of its location within the genome and its dispensable nature for viral growth, this region would appear to encompass a genetic sequence corresponding to the E3 region of human adenoviruses. In terms of viral attenuation, the possible importance of the observed differences between CAV-1(CLL) and CAV-1(Glaxo) is discussed.

    Notes: Abstract Electromanometry and electromyography were used to study gastro-oesophageal motility in two planes of halothane anaesthesia in sheep. Gastro-oesophageal motility when present was greater in light than in deep anaesthesia. The caudal thoracic oesophagus contracted more frequently and for longer than the rostral thoracic oesophagus. In light anaesthesia oesophageal movements were peristaltic in direction with a propagation velocity of 26–29 cm sec-1. Rumen pressures increased throughout anaesthesia and the rate of increase was greatest when the plane of anaesthesia was deep at the start. Gastro-oesophageal reflux (GOR) occurred in both planes of anaesthesia and must occur by passive mechanisms during deep anaesthesia because gastro-oesophageal motility was inhibited. A high pressure zone (HPZ) was demonstrated for a length of 2.9 cm at the gastro-oesophageal junction with a balloon-tipped catheter and a ‘pull through’ technique. Open-tipped catheters could detect the HPZ but were less sensitive. The pressure in the HPZ was not significantly influenced by the depth of anaesthesia used. In 80% of cases of light anaesthesia an increase in HPZ pressure preceeded the contraction of the cranial sac of the rumen. In deep anaesthesia the HPZ continued to have rhythmic changes in tone. Spontaneous GOR coincided with a maximum gastro-oesophageal pressure gradient in 24% of cases. Rumen insufflation with oxygen provoked GOR at a rumen pressure above 33 mmHg compared with 7.2 mmHg during spontaneous reflux. The study demonstrates that a gastro-oesophageal pressure gradient was not primarily responsible for the initiation of GOR during anaesthesia and that the HPZ at the gastro-oesophageal junction of sheep had some of the properties of a lower oesophageal sphincter and played an important role in the initiation of GOR during anaesthesia.
