    Notes: The coexistence of antidesmoglein 3 (Dsg3) and antidesmoglein 1 (Dsg1) autoantibodies is well described in patients with pemphigus vulgaris (PV); however, there is little evidence of sequential development of these two autoantibodies. Autoantibody responses to Dsg3 and Dsg1 were studied in seven PV patients over time by enzyme-linked immunosorbent assay, using baculovirus expressed recombinant fusion proteins. All patients had anti-Dsg3 IgG antibodies at presentation. Two patients developed anti-Dsg1 later in the course of the disease. The transition in autoantibody profile was associated with disease progression to generalized PV involving mucous membranes and skin in both patients; one patient initially presented with a predominantly mucosal phenotype, the other with herpetiform pemphigus-like features. These findings demonstrate that there is an extension of autoimmune response from anti-Dsg3 only to both anti-Dsg3 and anti-Dsg1 in some patients, which is associated with an alteration in clinical expression in PV.

    Notes: In nutrient medium, aluminium (Al) accumulation in tobacco cells occurs only in the presence of ferrous ion [Fe(II)]. The localization of Al was examined to elucidate a mechanism of Al accumulation. After the digestion of Al-treated cells with cellulase and pectolyase together, the resulting spheroplasts contained as much Al as the intact cells. However, the cell walls isolated from Al-treated cells also contained as much Al as the intact cells. Comparison of sugar and Al contents in polysaccharide components extracted chemically from cell walls isolated from intact cells and spheroplasts revealed that the enzymes digested most of the cellulose and hemicellulose, but only half of the pectin, and that Al mainly existed in the pectin remaining in the spheroplasts. Gel-permeation chromatography of the pectin fraction (NH4-oxalate extract) from the cell walls of the intact cells indicated that Al was associated with small polysaccharides of approximately 3–7 kDa. These results suggest that a minor part of pectin is a major site of Al accumulation. The content of cell wall pectin increased during Al treatment in nutrient medium. Taken together, we hypothesize that Al may bind to the pectin newly produced during Al treatment.

    Notes: Follicular epithelium of the thyroid gland of the one-humped camel was examined by light and electron microscopy. It consisted of a single type of epithelial cell which varied from flattened to columnar in shape. Follicular epithelial cells were characterized by the presence of markedly dilated cisternae of rough-surfaced endoplasmic reticulum, well developed Golgi apparatus, abundant small vesicles (150 nm to 200 nm in diameter) in the apical cytoplasm, and electron-dense colloid droplets measuring from 250 nm to 1600 nm. Follicular epithelial cells frequently showed apocrine secretion into colloidal lumens. Apocrine protrusions with smooth surface were dome-like or balloon-like structures and contained a fine granular matrix. These findings indicate that the morphological features of the follicular epithelial cells of the thyroid gland of the camel are essentially similar to those of mammals except for the presence of apocrine secretion, which is unique to the camel.

    Notes: Abstract Background. The overall survival rate and contributory factors were examined in patients undergoing hemodialysis in one dialysis program. The clinical profiles of dialysis patients who suffered from cerebro- and cardiovascular diseases (CCVD) were also studied. Methods. Data were available for 1064 patients who had received hemodialysis treatment for more than 3 months in our program (17 centers) in Miyazaki, Japan, between 1971 and 1994. Results. The respective 3-, 5-, and 10-year actuarial survival rates were 85%, 79%, and 66%. The Cox proportional hazard model identified three variables: age at start of hemodialysis (adjusted relative hazard [RH], 1.05), sex (RH, 1.42 for males), and type of underlying renal disease (RH, 1.00 for chronic glomerulonephritis, 2.80 for diabetes mellitus, 1.59 for nephrosclerosis), as being significantly associated with survival. Patients with CCVD had a high incidence of cerebral hemorrhage (8.7 per 1000 person-years). Patients whose first CCVD episode occurred within 36 months of starting hemodialysis were older, and more frequently had diabetes mellitus or nephrosclerosis and suffered from cerebral infarction. Long-term hemodialysis patients with CCVD more frequently had chronic glomerulonephritis as the underlying disease and had a higher incidence of cerebral hemorrhage. Conclusion. Age, sex, and underlying renal disease were independent factors that contributed to the mortality of the hemodialysis patients. There was a high incidence of CCVD among the hemodialysis patients, but the etiology of CCVD may differ depending on the underlying renal disease.

    Notes: Abstract. Bisphosphonates, therapeutic reagents against tumoral bone diseases (Paget's disease or osteoporosis), are potent inhibitors of bone resorption. The mechanisms by which they directly act on mature osteoclasts remain unclear. Using a recently developed technique for isolation of highly purified mammalian mature osteoclasts, we demonstrated that etidronate [ethane-1-hydroxy-1,1-diphosphonate (EHDP), 1-hydroxy-1,1-ethylidenebisphosphonate], inhibited directly osteoclastic bone-resorbing activity by pit assay. In addition, EHDP also directly induced apoptosis and disrupted actin rings in osteoclasts. The data support previous data on non-purified osteoclasts and results in vivo.

    Notes: Abstract Recessive alleles (va, va 1 , va 2 , etc) of the tobacco Va locus confer resistance to potato virus Y (PVY). To elucidate the mechanism underlying this resistance, we attempted to identify randomly amplified polymorphic (RAPD) markers that reveal polymorphism between two nearly isogenic lines (NILs) that differ in their susceptibility to PVY. Using each of 500 primers and 800 pairs of primers, we identified over 100 RAPD fragments that differed between the NILs. We applied these RAPD primers or primer combinations to an F2 population obtained from a cross between the susceptible line BY4 and the resistant va 2 -bearing NIL, F55. It was found that only 10 RAPD markers were polymorphic between resistant and susceptible plants. Unexpectedly, these markers were all linked to Va. All 10 RAPD markers were present in all 8 susceptible varieties tested. At least one RAPD marker was not detected in 8 out of 10 resistant varieties. Southern analysis revealed that the sequences of markers were not present in the genomes of resistant varieties, and the markers were found in individually distinct positions on the chromosomes of susceptible tobacco varieties. These results strongly suggest that the resistance conferred by va is due to deletions at the Va locus governing susceptibility to PVY.

    Notes: 31 P nucleus can be used to represent a quantum bit (‘qubit’) with a relatively long relaxation time. In a CeP crystal lattice, 31P nuclei are periodically situated in three dimensions at distances of about 6 Å. The application of a static magnetic field gradient in one direction causes differences in the Zeemanfrequencies of separate nuclei. This allows thousands of distinct qubits to be individually addressed. Initializations of the qubits can be done efficiently by the Pound–Overhauser double resonance effect on the nuclear spins and the antiferromagnetically ordered 4f electron spins of cerium ions. Logic operations can be performed by simple pulse sequences, and computational results after logic operations can be measured by the nuclear magnetic resonance of neighboring nuclei, or the electron resonance of neighboring 4f electrons of cerium ions.
