    Notes: The ultrafast time dependence of the energy absorption of covalent solids upon excitation with femtosecond laser pulses is theoretically analyzed. We use a microscopic theory to describe laser induced structural changes and their influence on the electronic properties. We show that from the time evolution of the energy absorbed by the system important information on the electronic and atomic structure during ultrafast phase transitions can be gained. Our results reflect how structural changes affect the capability of the system to absorb external energy. © 2002 American Institute of Physics.

    Notes: Abstract: Myelin and lymphocyte protein (MAL) is a putative tetraspan proteolipid that is highly expressed by Schwann cells and oligodendrocytes as a component of compact myelin. Outside of the nervous system, MAL is found in apical membranes of epithelial cells, mainly in the kidney and stomach. Because MAL is associated with glycosphingolipids, it is thought to be involved in the organization, transport, and maintenance of glycosphingolipid-enriched membrane microdomains. In this report, we describe the generation and analysis of transgenic mice with increased MAL gene dosage. Immunohistochemical analysis revealed that the localization of MAL overexpression in the transgenic animals corresponded closely to the MAL expression pattern observed in wildtype animals, indicating correct spatial regulation of the transgene. Phenotypically, MAL overexpression led to progressive dissociation of unmyelinated axons from bundles in the PNS, a tendency to hypomyelination and aberrant myelin formation in the CNS, and the formation of large cysts in the tubular region of the kidney. Thus, increased expression of MAL appears to be deleterious to membranous structures in the affected tissues, indicating a requirement for tight control of endogenous MAL expression in Schwann cells, oligodendrocytes, and kidney epithelial cells.

    Notes: There is evidence suggesting that nitric oxide (NO) may play an important role in dopamine (DA) cell death. Thus, the aim of this study was to investigate the effects of NO on apoptosis and functionality of DA neurones and glial cells. The experiments were carried out in neuronal-enriched midbrain cultures treated with the NO donor diethylamine–nitric oxide complexed sodium (DEA–NO). DEA–NO, at doses of 25 and 50 µm, exerted neurotrophic effects on dopamine cells, increasing the number of tyrosine hydroxylase positive (TH+) cells, TH+ neurite processes, DA levels and [3H]DA uptake. A dose of 25 µm DEA–NO protected DA cells from apoptosis. In addition, it induced de novo TH synthesis and increased intracellular reduced glutathione (GSH) levels, indicating a possible neuroprotective role for GSH. However, in doses ranging from 200 to 400 µm, DEA–NO decreased TH+ cells, DA levels, [3H]DA uptake and the number of mature oligodendrocytes (O1+ cells). No changes in either the amount or morphology of astrocytes and glial progenitors were detected. A dose- and time-dependent increase in apoptotic cells in the DEA–NO-treated culture was also observed, with a concomitant increase in the proapoptotic Bax protein levels and a reduction in the ratio between Bcl-xL and Bcl-xS proteins. In addition, DEA–NO induced a dose- and time-dependent increase in necrotic cells. 1H-[1,2,4]oxadiazolo[4,3a]quinoxaline-1-one (ODQ, 0.5 µm), a selective guanylate cyclase inhibitor, did not revert the NO-induced effect on [3H]DA uptake. Glia-conditioned medium, obtained from fetal midbrain astrocyte cultures, totally protected neuronal-enriched midbrain cultures from NO-induced apoptosis and rescued [3H]DA uptake and TH+ cell number. In conclusion, our results show that low NO concentrations have neurotrophic effects on DA cells via a cGMP-independent mechanism that may implicate up-regulation of GSH. On the other hand, higher levels of NO induce cell death in both dopamine neurones and mature oligodendrocytes that is totally reverted by soluble factors released from glia.

    Notes: The aim of this study was to investigate the effect of l-DOPA and glia-conditioned medium (GCM) on cell viability, tyrosine hydroxylase (TH) expression, dopamine (DA) metabolism and glutathione (GSH) levels of NB69 cells. l-DOPA (200 µm) induced differentiation of NB69 cells of more than 4 weeks in vitro, as shown by phase-contrast microscopy and TH immunocytochemistry, and decreased replication, as shown by 5-bromodeoxyuridine immunostaining. l-DOPA did not increase the number of necrotic or apoptotic cells, as shown by morphological features, Trypan Blue, lactate dehydrogenase activity, bis-benzimide staining and TUNEL assay. Furthermore, l-DOPA (200 µm) increased Bcl-xL protein expression. Incubation of cells with l-DOPA (50, 100, 200 µm) for 24 h resulted in an increase in TH protein levels (174, 196 and 212% versus control). Neither carbidopa, an inhibitor of l-aromatic amino acid decarboxylase enzyme, nor l-buthionine sulfoximine, which inhibits GSH synthesis, or ascorbic acid, an antioxidant, blocked the l-DOPA-induced effect on TH protein expression. l-DOPA (0, 50, 100 and 200 µm) plus GCM further increased the amount of TH protein (346, 446, 472 and 424%). l-DOPA (200 µm) increased TH protein levels to 132, 191 and 245% of controls after incubation for 24, 48 and 72 h. DA metabolism in NB69 cells was increased in cultures treated with either l-DOPA (200–300 µm) or GCM and these two agents had a synergistic effect on DA metabolism. In addition, l-DOPA (200 µm) or/and GCM-treated cells increased their GSH extracellular levels (223, 257, 300% of controls) after 48 h of treatment. The l-DOPA-induced increase of TH protein expression in NB69 cells was independent of DA production, free radicals and GSH up-regulation.

    Notes: A major aim of the Labour Government elected in May 1997 was to change British social policy away from passive income maintenance towards promoting employment, investment and opportunity. Many of its policy initiatives bear the clear imprint of a social development model. Investment in education and health are seen as paramount, community rebuilding through partnership-based economic renewal and the promotion of social capital through neighbourhood renewal are all explicit priorities in the government's spending plans. Note: this approach to policy will continue in Britain now that Labour has been returned for a second term in the June 2001 general election.

    Notes: A prototype multichannel time-resolved medical optical tomography system is presented, and various instrumental aspects and performance issues are discussed. The instrument has been designed primarily as a continuous bedside monitor for obtaining functional images of premature infants' brains that are at an increased risk of injury due to dysfunction in cerebral oxygenation or hemodynamics. Separate maps of the internal absorption and scattering properties can be reconstructed from purely temporal measurements of photons transmitted diffusely through the tissue, and without recourse to reference or baseline measurements. The instrument employs 32 source fibers that sequentially deliver near-infrared pulsed laser radiation of picosecond duration. Transit time measurements of very high temporal resolution and stability are made between these sources and 32 detector optodes that are located on the surface. The effectiveness of this instrument is demonstrated by successfully imaging a tissue-equivalent phantom. © 2000 American Institute of Physics.

    Notes: : The use of high methylated (60% to 66% methylation) and low methylated (8% methylation) pectin for the continuous production of pectic-oligosaccharides in an ultrafiltration dead-end membrane reactor was investigated. Substrate concentration (1% to 5% w/v), enzyme concentration (90 to 2700 Ul-1), and residence time (40 to 120 min) were the factors studied. Response surface methodology was used to model the processes. Two responses were determined: conversion of pectin(s) into product, and product dextrose equivalent values (DE). Mathematical models were fitted for each response. Conversions to product ranged from 67% to 99.7%. Product DE values increased with increasing residence time from 27 to 70. The average molecular-weight distribution of the products obtained was 3.5 kDa when high methylated pectin was used as substrate and 3.8 kDa for low methylated pectin.

    Notes: This randomised, placebo-controlled, patient and observer blinded trial was conducted to determine whether acupuncture at the acupuncture point P6 is effective in preventing postoperative nausea and vomiting (PONV) compared to placebo acupuncture. Female patients (n = 220) scheduled for gynaecological or breast surgery were randomly assigned to two groups receiving either acupuncture (n = 109) or placebo acupuncture (n = 111). Each group was stratified for type of surgery and included two subgroups receiving intervention either before or after induction of anaesthesia. The incidence of PONV and/or antiemetic rescue medication within 24 h after surgery was the main outcome measure which showed no statistically significant difference between groups (43.7% acupuncture, 50.9% placebo, p = 0.27). The differences were more pronounced for patients having gynaecological surgery (48.9% acupuncture, 67.6% placebo, p = 0.07) than for those having breast surgery (38.7% acupuncture, 40.3% placebo, p = 0.86). The secondary outcome, vomiting, was significantly reduced by acupuncture from 39.6% to 24.8% (p = 0.03). Subgroup analysis showed no difference between applications of acupuncture before compared to after induction of anaesthesia.

    Notes: In 25 cardiac surgical patients, right ventricular ejection fraction was continuously measured with a new pulmonary artery catheter and transoesophageal echocardiography, scanning the ‘fractional area change’ in a standardised transatrial cross section area. Measurements were recorded at three predefined time points (pre-, intra-, and postoperatively). Both methods were compared using the Bland-Altman analysis. Comparing right ventricular ejection fraction values obtained from the pulmonary artery catheter with those assessed by transoesophageal echocardiography, bias was −3.7%, with a precision of 30.9%. Bias and precision significantly improved when the heart rate was less than 100 beats.min−1, pulmonary artery pressures were low and cardiac performance adequate. In conclusion, the new continuous pulmonary artery catheter system appears to be a valid and useful bedside monitoring device in the haemodynamic management of critically ill patients.

    Notes: Peri-operative myocardial ischaemia is the single most important risk factor for an adverse cardiac outcome after non-cardiac surgery. The present study examines whether intermittent 12-lead ECG recordings can be used as an early warning tool to identify patients suffering from peri-operative myocardial ischaemia and subsequent myocardial cell damage. Fifty-five vascular surgery patients at risk for or with a history of coronary artery disease were monitored for peri-operative myocardial ischaemia using intermittent 12-lead ECG recordings taken pre-operatively and at 15 min, 20 h, 48 h, 72 h and 84 h postoperatively. The effectiveness of the 12-lead ECG was gauged by examining concordance with continuous 3-channel Holter monitoring and capturing peri-operative myocardial ischaemia by serial analyses of creatine kinase myocardial band isoenzyme and cardiac troponin T and I. The incidence of peri-operative myocardial ischaemia detected by 12-lead ECG was 44% and was identifiable in most patients (88%) 15 min after surgery. The incidence of peri-operative myocardial ischaemia detected by continuous monitoring was 53%, with the most severe episodes occurring intra-operatively and during emergence from anaesthesia. The concordance of the 12-lead method with continuous monitoring was 72%. The concordance of creatine kinase myocardial band isoenzyme activity with the 12-lead method was 71% and with Holter monitoring 57%. The concordance of mass concentration of creatine kinase myocardial band with 12-lead ECG recordings was 75%, and the corresponding value for Holter monitoring was 68%. The concordance of cardiac troponin T and I levels with the 12-lead method was 85% and 87%, respectively, and concordance with Holter monitoring was 72% and 66%, respectively. The postoperative 12-lead ECG identified peri-operative myocardial ischaemia associated with subsequent myocardial cell damage in most patients undergoing vascular surgery.
