    Notes: Background Atopy is closely associated with the cellular T helper type-2 (Th2) phenotype, that is dominated by the pleiotrophic cytokine IL-4. The cellular source of IL-4 has yet to be determined, although basophils have been proposed. Eosinophils and mast cells are likely contenders investigated here, and the eosinophil-like leukaemia line AML14.3D10 is compared to eosinophils as an in vitro culturable model for eosinophils. Lectins can cross-link-specific surface glycoproteins and are found in the ingested (processed foods) and inhaled (airborne pollen grains) human environment. Therefore it is of interest to determine whether lectins can elicit the release of IL-4 from Th2-associated granulocytes other than basophils.Method This study investigated the ability of eosinophils, AML14.3D10 and mast cells to secrete preformed IL-4 in response to stimulation with lectins, and explored molecular mechanisms underlying the interaction.Results Purified eosinophils and basophils, and cultured mast cells and AML14.3D10 cells were incubated with 1 µm lectin. Agglutination was scored by microscopy. IL-4 secretion was measured by enayme-linked immunosorbent assay. Biotinylated lectins were used to determine binding to cells by flow cytometry and in lectin blots of sodium dodecyl sulphate (SDS) gels.Discussion Purified human eosinophils, AML14.3D10 cells and cultured mast cells secrete IL-4 with a pattern similar to that found in basophils when stimulated with a panel of reactive and unreactive lectins. The lectin SNA induces IL-4 secretion from mast cells and basophils, but not from eosinophils or AML14.3D10. Eosinophils appear to secrete only pre-formed IL-4, whereas mast cells may synthesize IL-4 on ligation with the lectin LCA. Lectins that agglutinate the granulocytes investigated do not necessarily induce secretion of IL-4. Lectins that elicit secretion of IL-4 bind more to eosinophils than unreactive lectins as determined by flow cytometry and lectin blotting of SDS gels.Conclusion As granulocytes with functions related to that of basophils, eosinophils, AML14.3D10 and cultured mast cells respond to stimulation with lectins similarly to basophils. This emphasizes the possibility that eosinophils and mast cells may be linked in their cellular heritage as the cellular partners, and lectins as ligands, may contribute to the maintenance of a Th2-favoured microenvironment that is thought to underlie the allergic march.

    Notes: We have investigated the magnetic properties of flat permalloy cylinders by Lorentz transmission electron microscopy and micromagnetic simulations. The magnetization patterns during in situ magnetizing experiments have been imaged and they revealed that the magnetization reversal of the cylindrically shaped dots investigated is determined by the formation and annihilation of magnetic vortices. Furthermore, the experiments and micromagnetic simulations showed a dependence of the vortex annihilation field not only on the aspect ratio but also on the absolute thickness of the cylinders. The diameter of the cylindrical dots was varied between 150 and 1000 nm, and the thicknesses were 3, 5.5, 8.3, 15, and 20 nm, respectively. The formation of inhomogeneous magnetization patterns prior to vortex evolution was observed and by a comparison of the experimental to simulated Fresnel images these patterns can be identified as S- and C-like states. © 2002 American Institute of Physics.

    Notes: We report on the observation of the Coulomb blockade with Coulomb staircases at room temperature in cobalt nanoparticles, with sizes ranging between 1 and 4 nm. A monolayer of these particles is supported by a thin 1–2 nm thick Al2O3 film, deposited on a smooth Au(111) surface. The local electrical transport on isolated Co clusters was investigated with a scanning tunneling microscope (STM). The tunnel contact of the STM tip allowed us to observe single-electron tunneling in the double barrier system STM-tip/Co/Al2O3/Au. Very high values of the Coulomb blockade of up to 1.0 V were reproducibly measured at room temperature on different particles with this setup. The current–voltage characteristics fit well by simulations based on the orthodox theory of single-electron tunneling. © 2002 American Institute of Physics.

    Notes: Magnetic vortices play an important role in the switching behavior of micron- and submicron-sized ferromagnetic elements. We have prepared submicron permalloy elements by a combination of electron-beam lithography and liftoff technique on electron transparent membrane substrates. The magnetization reversal mechanism and the remanent magnetization configuration were observed by means of Lorentz transmission electron microscopy. In remanence, the investigated structures form a vortex configuration. In situ magnetizing experiments showed the possibility of adjusting the sense of magnetization rotation by introducing a slight geometric asymmetry to the otherwise circular nanostructures. © 2001 American Institute of Physics.

    Notes: Circular permalloy elements were fabricated by a combination of electron beam lithography, thermal evaporation and liftoff technique on electron transparent membrane substrates. The magnetic properties have been studied by Lorentz transmission electron microscopy. In situ magnetizing experiments have been carried out to obtain information about the nucleation and propagation of magnetic domains within the permalloy nanodisks and to determine the nucleation and saturation fields. The diameter of the patterned elements has been varied between 180 and 950 nm, the height was 15 nm. The experiments showed that the vortex configuration is the most favorable state in zero field conditions of all investigated permalloy nanodisks. © 2000 American Institute of Physics.

    Notes: Combining inhaled long-acting β-2 agonist (LABA) and inhaled corticosteroid (ICS) seems to offer asthma control at a lower dose of ICS than achieved by ICS alone. Fine mapping of T-cell surface markers by flow cytometry offers a detailed status of the individual's inflammatory response. The frequency of MT2 (CD4+CD45RA–CD62L+CD11adim) and MT1 (CD4+CD45RA–CD62L–CD11abright) cells in peripheral blood, and their ratio, has been shown to differ predictably in atopics and patients with leprosy, where MT2 correlates with a Th2 phenotype and MT1 with a Th1 phenotype. Stable asthmatics, requiring fluticasone propionate (FP) 750–1000 µg daily or equivalent, were randomized to receive, double-blinded, either Seretide®[salmeterol and fluticasone propionate (SFC, n = 16)] 50 µg/500 µg bd or FP 500 µg bd (n = 17). If asthma was controlled based on lung function and symptoms at clinic visits every 6 weeks, ICS dose was tapered until asthma exacerbated or 0 µg was reached. The frequency and ratio of MT2 and MT1 T cells of the patients was monitored at 6 week intervals. As treatment tapered, the frequency of MT2 cells decreased (P = 0038 from first to final visit), whereas that of MT1 cells increased. The ratio of MT2/MT1 decreased (P = 0049 from first to final visit). In patients receiving LABA + ICS, the fall in MT2/MT1 ratio appeared to be more pronounced than in patients receiving ICS alone. Thus, the MT2 phenotype may be associated with stable asthma, whereas an imminent exacerbation may associate with an increase in the MT1 phenotype. LABA may allow for a greater effect of FP on the MT ratio.

    Notes: The molecular mechanisms underlying protective granuloma formation and control of bacterial growth during infection with Mycobacterium tuberculosis (MTB) are not yet completely understood. MTB-infected mice with natural deficiency in complement component C5 are unable to develop productive granulomatous responses, and are impaired in limiting organism growth within the lung. To address the molecular basis for this histologic dysfunction, congenic complement C5-sufficient (B10.D2-H2d H2-T18c Hcl/nSnJ) and complement C5-deficient strains (B10.D2-H2d H2-T18c Hco/oSnJ) congenic mice were infected with Mycobacterium tuberculosis, and cytokine and chemokine responses were examined. Twelve and 28 days after infection, lungs showed elevated messages for multiple inflammatory cytokines in both congenic strains. Interleukin (IL)-12(p40) mRNA was also induced during infection in C5-deficient mice, although levels were significantly decreased compared to C5-sufficient congenics. C5-deficient mice also demonstrated reduced KC, MIP-2, IP-10, and MCP-1 mRNA. The defect may directly involve C5-mediated effects on macrophage responses; C5-deficient bone marrow derived macrophages had significantly reduced secretion of KC, MIP-1α and MIP-2 compared to C5-sufficient macrophages following in vitro infection. These findings indicate a role for C5 in mediation of chemotactic and activation events that are the basis for granulomatous responses during murine tuberculosis.

    Notes: Mycobacterium tuberculosis (MTB) causes tuberculosis in man, which occurs as an acute, chronic or dormant disease reactivating over several years. The mechanisms of persistence and reactivation are not well understood and there is a need for animal models. Moderate-dose, aerosol infection killed A/J mice earlier than partially resistant C57Bl/6 mice, whereas a low-dose, aerosol-induced chronic infection exacerbated earlier in A/J mice. A/J mice lethally infected with MTB but drug cured of disease underwent reactivation of tuberculosis at least 100 days before similarly infected C57Bl/6 mice. Because A/J mice were C5 deficient, congenic B10 mice sufficient and deficient for C5 were infected intravenously with MTB to define the role of C5. C5-deficient mice again showed enhanced growth of MTB in the lungs. MTB-infected macrophages from C5-deficient mice showed enhanced growth of MTB coinciding with a reduced secretion of both cytokines (TNF-α, IL-1β, IL-6, IL-12) and chemokines (KC, MIP-2 and MIP-1α) in A/J and TNF-α and chemokines in C5-deficient mice. Because C5-deficient macrophages could be activated from extraneous C5 and TNF-α we suggest that both play a role in the macrophage-mediated killing as well as containment mechanisms in tuberculosis.

    Notes: Background:  Mast cells have long been recognized as the principal cell type that initiates the inflammatory response characteristic of acute allergic type 1 reactions. Our goal has been to further characterize maturation of progenitors to mast cells.Methods:  Mast cells were cultured from human cord blood derived CD133+ progenitors. Mast cell function was tested using histamine release. During differentiation mast cells surface marker expression was monitored by flow cytometry.Results:  CD133+ progenitors expressed the early haematopoietic and myeloid lineage markers CD34, CD117, CD13 and CD33. Mature mast cells expressed CD117, CD13 and CD33, and expression of the high affinity immunoglobulin E recpetor FcɛRI increased during culture. Cytokine receptors interleukin (IL)-5R, IL-3R, granulocyte-macrophage-colony stimulating factor (GM-CSF)R and IL-18R were expressed at high levels during maturation. Chemokine receptors CXCR4 and CXCR2 were highly expressed on both newly purified CD133+ cells and mature cells.Conclusion:  Human mast cells can be cultured from a CD34+/CD117+/CD13+/CD33+ progenitor cell population in cord blood that is tryptase and chymase negative. Developing and mature mast cells express a wide range of chemokine and cytokine receptors. We found high levels of expression of CD123, IL-5R and GM-CSF receptors, also found on eosinophils and basophils, and high levels of expression of the receptor for the inflammatory cytokine IL-18.
