    Notes: Abstract The central sector of Mühlig-Hofmannfjellet (3°E/71°S) in western Dronning Maud Land (East Antarctic shield) is dominated by large intrusive bodies of predominantly orthopyroxene-bearing quartz syenites (charnockites). Metasedimentary rocks are rare; however, two distinct areas with banded gneiss–marble–quartzite sequences of sedimentary origin were found during the Norwegian Antarctic Research Expedition NARE 1989/90. Cordierite-bearing metapelitic gneisses from two different localities contain the characteristic mineral assemblage: cordierite + garnet + biotite + K-feldspar + plagioclase + quartz ± sillimanite ± spinel. Thermobarometry indicates equilibration conditions of about 650°C and 4 kbar. Associated orthopyroxene–garnet granulites, on the other hand, revealed pressures of about 8 kbar and temperatures of 750°C. The earlier granulite facies metamorphism is not well preserved in the cordierite gneisses as a result of excess K-feldspar combined with interaction with an H2O-rich fluid phase, probably released by the cooling intrusives. These two features allowed the original high-grade K-feldspar + garnet assemblages to recrystallize as cordierite–biotite–sillimanite gneisses, completely re-equilibrating them. Phase relationships indicate that the younger metamorphic event occurred in the presence of a fluid phase that varied in composition between the lithologies.

    Notes: Abstract: Myelins of the PNS were isolated from human motor and sensory nerves of cauda equina, and their ganglioside compositions were compared. The predominant ganglioside in the human PNS myelins, both from motor and sensory nerves, was LM1 (sialosylneolactotetraosylceramide). Sialosyl-nLc6Cer and disialosyl-nLc4Cer, GD3, GM3, and GDlb were detected as common components of the two nerve myelins. Furthermore, it was revealed that the motor nerve myelin contained GM1 (about 15% of total gangliosides), whereas sensory nerve myelin contained only a trace amount of GM1 (less than 5%), by TLC analyses together with TLC immunostaining using anti-GM 1 antibody. As for the disialoganglioside fraction, the content of GD1 a, as well as that of GM1, differed in motor and sensory nerves. Thus, the different contents of the ganglioseries gangliosides in human motor and sensory nerve myelins were demonstrated.

    Notes: Abstract: Compositions of neutral and sulfated glucuronyl glycosphingolipids purified from human motor and sensory nerves and myelins were studied. Higher neutral glycosphingolipids (fraction B), which were separated from GazlCer (fraction A), were analyzed by TLC and TLC-immunostaining. Both nerve myelins contained paragloboside (nLc4Cer) and nLc6Cer dominantly as major higher glycosphingolipids and very little globoside (Gb4Cer), whereas both nerves contained Gb4Cer and nLc4Cer. Besides these major glycosphingolipids, a neutral glycolipid containing asialoGMI (Gg4Cer) epitope and other minor components such as ceramide trihexoside and ceramide dihexoside were detected in both nerves and their myelins. Furthermore, sulfated glucuronyl nLc4Cer and n Lc6Cer, which were monoclonal antibody HNK-1 reactive glycolipids, were detected in both nerves and myelins.

    Notes: The ammo-terminal pro-sequence consisting of 77 amino acid residues is required to guide the folding of secreted subtilisin E, a serine protease, into active, mature enzyme (Ikemura et al., 1987). Furthermore, denatured subtilisin E can be folded to active enzyme in an intermolecular process with the aid of an exogenously added pro-subtilisin E, the active site of which was mutated (Zhu et al., 1989). In this report, we have synthesized the pro-peptide of 77 residues (corresponding to -1 to -77 in the sequence, where residue +1 is the N-terminal amino acid residue of the mature protein), and have found that it could intermolecularly complement the folding of denatured subtilisin E to active enzyme. Furthermore, we have found that the synthetic pro-peptide exhibits specific strong binding to the active mature enzyme by inhibiting it competitively at its active centre with an upper limit to a Ki of 5.4 × 10−7. In contrast, synthetic pro-peptides corresponding to -44 to -77, -1 to 64 and -1 to -43 inhibited the enzyme with Ki values weaker by two orders of magnitude. The results indicate that the sequence extending from -1 to -77 is essential for specificity of interaction, perhaps generating a conformation that accounts for both roles found hitherto, i.e. specific binding to the active centre, and guiding of the refolding to active enzyme. Thus these results suggest that the pro-peptide functions as an intermolecular chaperone.

    Notes: The formation of active subtilisin E from pro-subtilisin E requires the removal of the N-terminal pro-sequence of 77 residues. Pro-subtilisin E produced in Escherichia coli using a pINIII-ompA vector was first extracted with 6M guanidine-HCl and 5M urea and purified to homogeneity in the presence of 5M urea. Upon drop dialysis against 0.2M sodium phosphate buffer (pH 6.2), the purified pro-subtilisin in 5M urea was processed to active subtilisin of which the N-terminal sequence and migration in SDS-polyacrylamide gel electrophoresis were identical to those of authentic active subtilisin E. This process was found to be very sensitive to the ionic strengths and anions used. Under the optimum conditions (dialysis against 0.5 M (NH4)2SO4 and 1 mM CaCl2 in 10mM Tris-HCl buffer (pH 7.0) at 4°C for 1 h), approximately 20% of pro-subtitisin E was converted to active subtilisin E. The activation process was not inhibited by Streptomyces subtilisin inhibitor, and pro-subtiiisin E in which the active site was mutated (Asp32 to Asn) was unable to be processed under the optimum conditions. These results confirmed the previous hypothesis that the processing of pro-subtilisin occurs by an intramolecular, autoprocessing mechanism.

    Notes: As a part of the Norwegian Antarctic Research Expedition 1984/85, geological mapping was performed in Gjelsvikfjella and western Mühlig-Hofmannfjella, Dronning Maud Land. The northern part of Gjelsvikfjella is dominated by the Jutulsessen metasupracrustals which have been intruded by a major gabbroic body and several generations of dykes. To the south the metasupracrustals gradually transform into the Risemedet migmatites. In western Mühlig-Hofmannfjella the bedrock is dominated by the large Svarthamaren Charnockite batholith. The batholith is bordered by the Snøtoa metamorphic complex outcropping to the south and west in Mühlig-Hofmannfjella and it is characterized by a high content of partly assimilated country rock inclusions. Mineral paragenesis and geothermometry/geobarometry suggest a two-stage tectonothermal-igneous history with an initial intermediate pressure, upper amphibolite to granulite facies metamorphism followed by high temperature transformations related to the charnockite intrusion. The age of the initial tectonothermal event is probably about 1,100 Ma. Geochronological work in the present study (Rb/Sr whole rock) gave an age of 500 ± 24 Ma for the Svarthamaren Charnockite, interpreted to record the age of crystallization. Late brittle faulting and undeformed dolerite dykes outcropping in Jutulsessen are believed to be related to Mesozoic crustal stretching in the Jutulstraumen-Pencksøkket Rift Zone to the west.

    Notes: A preliminary U/Pb zircon age determination has been carried out on a grey gneiss of the Eskolabreen Formation, the lowest observable lithostratigraphic unit of Precambrian metamorphic rocks in southern Ny Friesland, NE Spitsbergen. The obtained age, ca. 2, 400 Ma, is considered to be a metamorphic age and suggests an Early Proterozoic tectonothermal event.

    Notes: Abstract The cytotoxicity of arsenic compounds towards BALB/c 3T3 cells in culture was investigated, together with the role of glutathione (GSH) in the induction of the cytotoxic effects. The rank order of cytotoxicity was as follows: arsenite (As3+)〉arsenate (As5+)〉dimethylarsinic acid (DMAA)〉methylarsonic acid (MAA)〉trimethylarsine oxide (TMAO). Arsenobetaine, arsenocholine and the tetramethylarsonium ion were less toxic. Depletion of GSH enhanced the cytotoxic effects of As3+, As5+, MAA and TMAO, while the cytotoxicity of DMAA was markedly reduced by depletion of GSH. These results suggest that GSH plays a role in protecting the cells against the toxic effects of As3+, As5+, MAA and TMAO while it is involved in the induction of the cytotoxic effects of DMAA.

    Notes: Summary Psoriasis is a disease of abnormal proliferation and differentiation of epidermal cells. Several cytokines released by keratinocytes are implicated as factors responsible for this pathological condition of the epidermis. In order to elucidate the role of these cytokines in psoriasis, messenger RNA (mRNA) expression of interleukin-1 (IL-1) and IL-6 in psoriatic epidermis was investigated using biotin-labelled complementary DNA (cDNA) of the cytokines. Messenger RNA of IL-1α was weakly detected in some normal healthy epidermis specimens and more strongly in all the perilesional uninvolved psoriatic epidermis specimens. It was also expressed in the transitional zone between uninvolved and fully developed psoriatic skin, but was not expressed in lesional skin. In contrast, IL-6 mRNA was rarely expressed in normal healthy epidermis, but was expressed in perilesional uninvolved psoriatic epidermis, in the transitional zone and in the fully developed lesional epidermis, with the maximum intensity in the transitional zone. Expression of mRNA of IL-6 receptor showed a similar tendency to that of IL-6. It was expressed in psoriatic epidermis, most strongly in the transitional zone, but not in normal healthy epidermis. IL-6 was demonstrated immunohistochemically in psoriatic epidermis, but IL-6 receptor was demonstrated only in the transitional zone. Thus IL-6 and its receptor expression correlated well with the formation of psoriatic lesions where IL-1 may initiate their expression. IL-6 may play an important role in the pathogenesis of psoriasis.
