    Notes: Summary The expression of specific T-cell receptor gene segments by T lymphocytes appears to be critically important for the induction of several experimental autoimmune diseases mediated by these cells. We examined whether this situation also applied to non-obese diabetic mice by using various T-cell receptor VΒ-specific monoclonal antibodies. No significant age- or sex-related differences were observed in VΒ usage by peripheral and splenic T lymphocytes. CD8+ T lymphocytes among the islet-derived mononuclear cells isolated from 20-week-old female non-obese diabetic mice showed heterogeneity of their VΒ gene usage. In order to examine the role of T lymphocyte subsets expressing specific T-cell receptor VΒ gene segments in the development of diabetes mellitus, T-cell receptor VΒ-specific monoclonal antibodies were administered to 10-week-old male non-obese diabetic mice treated with cyclophosphamide. None of the antibodies used could significantly diminish the incidence of cyclophosphamide-induced diabetes and the severity of insulitis [anti-VΒ3 (11 of 22 mice became diabetic, 50%), anti-VΒ5 (9 of 14, 64%), anti-VΒ8 (9 of 21, 43%), anti-VΒ11 (12 of 23, 52%), anti-VΒ14 (7 of 12, 58%), and anti-VΒ5 + anti-VΒ11 (6 of 12, 50%)] when compared with control mice (12 of 21, 57%). In addition, there were no significant differences in T-cell receptor VΒ usage between diabetic and non-diabetic cyclophosphamide-treated mice. These results suggest that five T-lymphocyte subsets expressing different T-cell receptor VΒ gene segments, considered to be candidates involved in the pathogenesis of autoimmune diabetes, do not individually contribute to the development of cyclophosphamide-induced diabetes in non-obese diabetic mice.

    Notes: A low-duty pulsed Penning source has been developed for the injector of the medical synchrotron facility at NIRS. The repetition rate is 1 Hz, with a duty factor of 4×10−4. The lifetime of the cathodes has been evaluated to be longer than two months, and that of the filament (cw) longer than one week. The intensity of multiply charged ions was increased by a factor of 10 compared with that obtained under dc operation. The reliability and stability were also improved. The results concerning the performance and some improvements of the source are discussed.

    Notes: A low-duty pulsed PIG source has been developed for the injector (e/m= (1)/(7) – 1/4 ) of HIMAC. This source is of an indirectly heated PIG type. The requirement for this source is a high current with a low repetition rate and a low-duty factor. To obtain good stability during pulsed operation, the source is operated in a positive arc impedance range. Pulsed operation also allows the arc voltage to be high (1.4 kV), resulting in a high yield of multiply charged ions. With an arc pulse width of 500 μs (1 Hz and a duty of 0.05%), a useful beam pulse width of 200 μs with a peak intensity of 120 μA for Ar6+ has been obtained at an optimum gas pressure of 10−3 Torr. This intensity is higher than for cw mode by a factor of 5. A 90% emittance has also been measured to be 250π mm mrad in both the X and Y directions.

    Notes: Summary The present investigation was conducted to examine the combination effect of N-butyl-N-(4-hydroxybutyl) nitrosamine (BBN) and N-methyl-N-nitrosourea(MNU) in rat urinary bladder carcinogenesis. Experiment 1 was performed in the groups treated by oral BBN administration in combination with MNU intraversical instillation and their control groups. In the groups given both BBN and MNU, the ratio of rats with the nonpapillary type to carcinoma-bearing rats was significantly higher than in the controls. Since most of the carcinomas were non-invasive, the observation period was prolonged in the groups given both BBN and MNU in experiment 2. However, even after a longer observation period, no invasive carcinoma was observed. These results suggest that high-grade and invasive carcinomas similar to those induced by BBN and MNU in the heterotopically transplanted rat bladder cannot be induced in the natural bladder of the rat.
