    Notes: Summary The influence of usual multiple ingestions of dietary caffeine on oral single-dose pharmacokinetics of theophylline has been investigated in 6 healthy male subjects. The subjects consumed 2 to 7 cups of regular instant coffee during the 24 h study period. Their mean serum concentrations of caffeine varied from 1.2 to 3.1 mg/l. After their usual intake of dietary caffeine, the serum concentrations of theophylline from 3 to 24 h after administration were significantly higher than after deprivation of dietary caffeine. The apparent elimination of theophylline half-life was prolonged from 6.3 (0.61) h (mean with (SEM)) to 8.3 (0.47) h (32% increase, P〈0.01) and the total body clearance was reduced from 55.0 (1.31) ml·h−1·kg−1 to 42.5 (2.63) ml·h−1·kg−1 (23% decrase, P〈0.001). Saturation of theophylline metabolism and/or competition between theophylline and caffeine metabolism in addition to theophylline derived from caffeine may be the cause of the delayed elimination of theophylline. The present study has indicated that a significant reduction in theophylline metabolism may be caused by a conventional intake of dietary caffeine. In bronchodilator therapy with theophylline, therefore, the daily consumption of caffeine should be taken into consideration.

    Notes: The characteristics of Ag-(Mg,Ni) and Ag-Mn alloys as substrate materials for Bi2Sr2CaCu2Ox (Bi-2212) tapes and their influence on the superconducting properties were studied. Alloying with Mg+Ni or Mn drastically decreased the grain size of Ag after heat treatment. The additives in Ag-(Mg,Ni) and Ag-Mn alloys were oxidized during heat treatment in air. Both the fine grain structure and the internal oxidation of the additives increased the mechanical strength of the Ag alloys. Both alloys were 3–6 times stronger than pure Ag. The superconducting properties of Bi-2212 prepared on these Ag alloys were slightly degraded; however the critical current densities at 4.2 K in 8 T were still well over 104 A/cm2.

    Notes: Oxide superconductor tape with Ag-Au sheath was studied for current leads. A 100-cm-long current lead of 1000 A with 0.2 W/kA heat leakage was designed using Ag-11 at. % Au alloy sheath with low thermal conductivity. The overall current density of the superconducting tape is required to be 350–770 A/cm2 at 77 K when the superconductor cross-section ratio in the tape is 0.35–0.7. Bi-Pb-Sr-Ca-Cu-O superconducting tape with Ag-11 at. % Au alloy sheath was fabricated and the critical current density Jc was measured. The overall-Jc was 1700 A/cm2 at 77 K under 0 T. The possibility of the current lead of 1000 A under 0.28 T with 0.2 W/kA was made clear.

    Notes: The influence of Ag-Au and Ag-Cu alloy substrates on the microstructure and superconducting properties of Bi2Sr2CaCu2Ox (Bi-2212) was studied. The critical current density (Jc) of Bi-2212/(Ag-Au) was not degraded by increasing the Au content of the Ag-Au alloy, while the Jc of Bi-2212/(Ag-Cu) was drastically degraded by increasing the Cu content of the Ag-Cu alloy. The microstructure of Bi-2212 on the Ag-Au alloy was uniform and no precipitates were observed. The Jc degradation of Bi-2212 in contact with the Ag-Cu alloy was understood by the fact that Cu absorption of Bi-2212 from the Ag-Cu alloys changed the composition of the oxide layer. Ag-Au alloys are superior substrate materials for Bi-2212 for power current leads for superconducting magnets, taking into account their low thermal conductivity.

    Notes: The localization of fibroblast growth factor-1 (FGF-1) and FGF-2 in human oral squamous cell carcinoma (SCC) was examined by immunohistochemical techniques using anli-FGF-1 and anti-FGF-2 monoclonal antibodies. Immunofluorescence staining of two oral SCC cell lines revealed that growing cancel-cells were intensely positive for both FGF-1 and FGF-2, but confluent cells showed a faint immunostaining. In addition, two molecular mass species of FGF-1(16 and 18 kDa) and one of FGF-2 (18 kDa) were identified by Western blot in cell extracts derived from growing SCC cells, but not from confluent SCC cells. The growing cell extracts significantly stimulated the proliferation of human umbilical vein endothelial cells. Immunoperoxidase staining of 13 oral SCC cases showed that both well-differentiated and poorly-differentiated cancer cells were positive for FGF-1 and FGF-2 with high frequency and intensity as compared to normal oral epithelium. These results indicate that SCC cells express high levels of endogenous FGF-1 and FGF-2, and suggest that these growth factors may contribute to cancer cell growth.

    Notes: Abstract Forty two male patients with borderline and mild hypertension were followed up for 3.9 years without medication. During this period, all subjects had nonpharmacological interventions including salt restriction, weight reduction, anti-smoking or anti-alcohol instruction, and physical exercise after the established protocol. Comparisons of biomedical data between pre- and post-observation revealed no significant improvements. Although the smoking habit decreased during this period, the drinking habit rarely changed and even significant increase of the total amount of alcohol ingestion was observed. These data indicate great limitation of nonpharmacological intervention in the care of these asymptomatic hypertensive patients for a lifelong base.

    Notes: Summary Effects of exogenous prostaglandin (PG) E2 and PGI2 on testicular polymodal receptor activities were compared in in vitro recordings of single- or multi-fiber discharges from canine testis-spermatic nerve preparations. PGI2 up to 1.4×10−6 mol/l (cumulative method) or 1.0×10−5 mol/l (non-cumulative method) excited only weakly some of the receptors, and similar observations were made with PGE2. Both PGs applied cumulatively or non-cumulatively at concentrations above 1.4×10−8 mol/l augmented the response to bradykinin (9.4×10−8 mol/l) in more than half of the cases tested. The augmenting effect of PGE2 lasted longer than that of PGI2 both with the cumulative and the non-cumulative method. The degree of augmentation tended to increase dependent on concentration, but some cases showed no further increase or rather a decrease in augmentation by PGs at a ten times higher concentration, especially when PGs were applied cumulatively. A second challenge by PG after a short interval (2 min) did not induce augmentation. These phenomena were considered to be tachyphylaxis to PGs. Cross-tachyphylaxis to PGE2 and PGI2 was also observed. There was not much difference in excitatory and augmenting potencies between these two PGs, but there was a clear difference in the concentrations of the PGs necessary to induce excitation of polymodal receptors and to facilitate their response to bradykinin.

    Notes: Summary The concept of ex vivo expansion of the human progenitor cell population was tested in this study. In the presence of interleukin-3 and -6 we compared the abilities of various liquid culture conditions of human blood-derived hematopoietic progenitor cells to yield a suitable condition for the expansion of blood progenitor cells. Although the best result was obtained in the gas-permeable bag culture system, the enhancement effect (maximum of 3.8-fold for granulocyte/macrophage colony-forming units and 3.0-fold for erythroid burst-forming units) lasted for only a short period (〈 6 d). The results of this study are disappointing for clinical use, and attempts with currently available technology appear to be limited in their potential.
