    Notes: Bullhead catfish, Ictalurus nebulosus, and rainbow trout, Salmo gairdneri, were subjected to two coal-derived substances to evaluate each material for potential carcinogenic activity. Substances tested included a solvent-refined coal heavy distillate and a sediment extract from an industrially polluted river in Ohio. Two subfractions, one containing the polycyclic aromatic hydrocarbons and the other containing the nitrogen heterocycles of each material were also tested. Following a 16-h exposure period, the gill and liver tissues were removed and examined by standard light and electron microscopy techniques. Hyperplasia of the gill tissue and some engorgement with blood of the secondary lamellae were observed with the light microscope. At the electron microscope level, hepatocyte mitochondria were swollen with a loss of cristae, and rough endoplasmic reticulum fragmented. Results suggest that most of the treatments studied induced pathological responses in fish. Of the two fish tested, the rainbow trout was the most sensitive indicator of environmental pollution.

    Notes: Special electronic scanning circuits have been built to increase the resolution time of an intensified photodiode array assembly used for detecting EUV emission. The system can operate at 1-MHz sampling rate to provide data readout on two parallel output lines. The scanning time of a 1000-element array can be reduced to 500 μs. The system is programmable and can function sequentially at three different frequencies within a single scan. The device is CAMAC compatible and operated remotely by computer. It has been used to record spatial as well as temporal development of EUV radiation from the TEXTOR tokamak plasma.

    Notes: A method is described for finding the magnetotelluric transfer function that has the least amount of curvature consistent with most of the data and with a 1-D conductivity interpretation over the widest possible frequency range. This could be called an ‘Occam’ transfer function. It is represented by the transfer function for the best fitting 1-D conductivity model times a distortion function. The latter permits smooth departures of the transfer function from the 1-D case if the data are inconsistent with a 1-D interpretation. The transfer function, for single-station or remote reference magnetotelluric data, is found by a method of successive interations that is found to converge within six to eight iterations. The estimate of the transfer functions is made robust by using frequency and time weights that remove the effects of outliers in the time and frequency domain. If the weighted residuals for remote reference data satisfy certain necessary conditions for uncorrelated noise then the contribution to the noise by the electric and magnetic data can be estimated and used to evaluate the least-squares and remote reference estimates.Examples illustrate the application of this method to artificial and real data. The latter consist of hourly cable voltage data from the Florida Straits, 1/256-s remote reference magnetotelluric survey data from the Phillippines and daily magnetic data from Tucson and Honolulu.

    Notes: An effective method for specific depletion of mature T lymphocytes from human bone marrow mononuclear cells (BMMC) with preservation of prethymic T cells and natural killer (NK) cells is presented. The BMMC were incubated with F101.01, a monoclonal antibody recognizing an epitope of the T-cell receptor CD3 complex, and subsequently with immunomagnetic beads. Flow cytometric analysis demonstrated that mature T cells were efficiently depleted and that NK cells and prethymic T cells were preserved in the BMMC. Furthermore, T cell-mediated immune reaction as measured by thymidine incorporation after stimulation with phytohaemagglutinin was abolished, whereas NK activity as measured by 51Cr release using K562 as target cells was preserved. Recovery of colony-forming units of granulocyte macrophages was 60–70%.

    Notes: A murine monoclonal antibody (MoAb) F101 01 reacting with the T cell receptor (TCR)-T3 complex is presented. Immunohistological studies showed that FH101.01 specifically stains T-zone lymphocytes in lymph nodes, tonsils, and splenic tissue. Two-colour immunofluorescence and flow cytometry demonstrated co-expression of the antigen defined by F101.01 and the pan-T cell antigens defined by CD2, CD3, CD5 and CD7 antibodies. Cells Stained with CD4 and CDS antibodies were both included in the F101.01-positive population, whereas CD16-positive natural killer cells (NK). B cells (CD19 and CD20), and myeloid cells (CD13 and CD33) were excluded. The target antigen of F101.01 comodulated with the CD3-defined antigen (T3) and the TCR recognized by the MoAb WT-31. CD3 antibody and WT-31 both blocked binding or F101.01. F101.10 precipitated the TCR-T3 complex from lysates of 125-labelled peripheral blood mononuclear cells (PBMC) and HPB-ALL, when the lysate was prepared with a detergent (digitonin) that conserves the TCR-T3 complex. FACS analysis of T cells from a patient with a T cell immunodeficiency demonstrated that δ-TCS-1-CD3-CD4- and δ-TCS-1-CD3+CD8+ cells were brightly F101.01+, whereas a large subpopulation of δ-TCS-1+CD3+CD4− CD8− cells were weakly F101.01+. We conclude that F101.01 recognizes a conformational epitope of theTCR-T3 complex and that it reacts with the αβ TCR-T3 and the γδ TCR-T3 complexes with different intensities.

    Notes: Two lineages of T lymphocytes bearing the CD3 antigen can be defined on the basis of the nature of the heterodimeric receptor chain (αβ or γδ T cell receptor (TCR)) expressed. Precise identification of αβ and γδ TCR+ cells is essential when studying the tissue distribution and function of these different T ceils. In immunofluorescence studies γδ TCR− cells have been identified as CD3+ WT-31 or CD3+CD4− CD8− cells. However, this may not be the optimal procedure because γδ TCR+ cells are weakly WT-31+, and some are CD8+. The aim of this study was to evaluate a panel of monoclonal antibodies (MoAb) directed against different chains of the TCR-T3 complex for a more precise identification of αβ+ and γδ TCR+ cells in flow cytometric studies. We found that the MoAb anti-Ti-γA and δ-TCS-1, recognizing the TCR-γ and the TCR-δ chain respectively, only reacted with a subpopulation of γδ TCR+ cells, whereas another TCR-γ chain recognizing MoAb anti-TCR-δ1 reacted with all γδ TCR+ cells. All MoAb reported to belong to the CD3 group reacted with both αβ TCR+ and γδ TCR+ cells as expected. Our results indicate that all γδ TCR+ cells can be identified with the MoAb anti-TCR-δ1. Because no MoAb recognizing the TCR-α or TCR-β chains at the cell surface of intact cells are yet available, we suggest that αβ TCR+ cells could be identified as CD3+ anti-TCR-δ1− cells.

    Notes: Summary The winter wheat varieties ‘Starke’ and ‘Cappelle Desprez’ and the spring wheat ‘Chinese Spring’ were analysed for structural chromosome rearrangements that resulted in the formation of multivalents in F1 hybrids. The analyses were carried out using hybrids involving euploids, monosomic and ditelosomic stocks, and double-monotelodisomic constructs. The study confirmed that ‘Cappelle Desprez’ differs from ‘Chinese Spring’ in a reciprocal translocation between chromosomes 5B and 7B (Riley et al. 1967); a translocation involving chromosomes 3B and 3D could not be verified. Furthermore, the analysis showed that ‘Starke’ differs from ‘Chinese Spring’ in a reciprocal translocation between chromosomes 7A and 7D. Both translocations have a coefficient of multivalent realisation of about 0.84. Further multivalents in euploid ‘Starke’, in euploid and some aneuploid stocks of ‘Cappelle Desprez’, and in euploid as well as various types of aneuploid hybrids between all three varieties could nearly all be explained hypothesizing that chromosome 2B of both ‘Starke’ and ‘Cappelle Desprez’ is a duplication-deficiency chromosome. In the hypothesis a part of the long arm of 2B is missing and replaced by a duplicated part of the long arm of chromosome 2D. The multivalents of this rearrangement showed an average coefficient of realisation of about 0.09.
