    Notizen: Abstract Hill reaction and noncyclic photophosphorylation of isolated class C chloroplasts of spruce (Picea abies (L.) Karst.), as well as 14CO2 fixation by whole needles at constant laboratory conditions proceeded at high rates during spring and early summer, declined during late summer and autumn by about 60%, remained at this level during winter, and recovered quickly in early spring. During summer, the whole needles proved to be frost labile, since after exposure to-20°C and careful thawing, fast chlorophyll degradation occurred. In addition, only photosynthetically inactive chloroplasts could be isolated from those precooled needles. On the contrary, during winter the photochemical activities of plastids from freshly harvested needles did not differ from those of artificially frozen-thawed needles. When isolated spruce chloroplasts were exposed to the same subfreezing temperatures as the whole needles, no influence of freezing on the photochemical activities was observed, irrespective of whether the plastids were isolated from frost sensitive or frost hardened needles. It is concluded that frost damage to spruce chloroplasts is due to an attack of membrane toxic compounds or lytic enzymes which were liberated upon freezing from more labile compartments. Frost hardening of the chloroplasts, as determined by the stability of chlorophyll after exposure of the needles to low temperatures, as well as by the isolation of photosynthetically active chloroplasts from such precooled needles, appeared to depend at least on 2 processes: (i) an alteration of the composition of the photosynthetically active membranes and (ii) and additional stabilization of these membranes by protecting substances. The first process was indicated by a large increase (decrease) of the capability of isolated chloroplasts for PMS-mediated photophosphorylation which accompanied natural or artificial frost hardening (dehardening). Production of cryoprotecting compounds was suggested by a significant higher stability against NaCl observed with class C chloroplasts isolated from frost hardened needles as compared to that of plastids from frost labile material. The decrease of the capability for both, the ferricyanide dependent photoreactions of the plastids and the CO2 fixation by whole needles, which was observed during the frost hardening phase, cannot be due to freezing injuries; it rather appears to be a consequence of the frost hardening process.

    Notizen: Summary It is concluded from 14C labelling kinetics as well as from enzyme analysis that formation of malate and aspartate from glycerate-3P via carboxylation of P-enolpyruvate occurs in isolated photosynthesizing spinach (Spinacia oleracea L.) chloroplasts. Carbon incorporation into both dicarboxylic acids was about 1% of the total CO2 fixation. Both acids are suggested to act as substrates for counter exchange for glutamate and α-ketoglutarate.

    Notizen: Summary Seasonal changes of ultrastructure were studied by electron microscopy and by determining the chlorophyll and starch content of the plastids. Young plastids of spruce (Picea abies (L.) Karst.) first function as amyloplasts which store reserve material for the growth of the young needles. Then they develop a normal thylakoid system and produce assimilation starch during the day. In autumn, starch synthesis ceases and the plastids group together. In winter they swell and their membrane system becomes disorganized and reduced. With increasing temperatures in spring the chloroplasts recover, but then they accumulate large amounts of starch, which is not broken down during the night or even during a dark period of several days. As in the previous year they now function as amyloplasts providing reserve material for the new shoot. In summer these plastids are again converted into typical chloroplasts. The same seasonal changes of structure and function could be observed in chloroplasts from 2- or 3-year old needles. Thus these changes represent cyclic processes, which repeat each year. Features of slow aging are superimposed on to these cycles.

In the guinea pig heart-lung-preparation (HLP) anaphylactic bronchospasm was not greatly affected by mepyramine (2–40 μg/ml blood), adrenaline (0.1 and 1 μg/ml), noradrenaline (0.1 μg/ml) or papaverine (100 μg/ml) but was completely abolished by combining mepyramine with the lower dose of adrenaline. A partial antagonism was achieved by the combined action of mepyramine and papaverine which became complete when adrenaline was added. 2. Bronchospasm due to histamine was completely prevented by mepyramine, but only partially antagonized by both adrenaline doses used. 3. The increase of pulmonary vascular resistance in anaphylaxis was not inhibited by the highest tolerated amounts of mepyramine or papaverine, no matter whether the drugs were given alone or in combination. Adrenaline, in contrast to noradrenaline, was partially effective at the lower dose (0.1 μg/ml). The higher concentration did not act more strongly. It became fully effective when mepyramine was added either alone or combined with papaverine. 4. The increase of pulmonary vascular resistance induced by histamine was strongly and significantly antagonized by mepyramine alone as well as by the lower concentration of adrenaline (0.1 μg/ml). Noradrenaline had no significant effect in either amount given (0.1 and 1 μg/ml). 5. Antigen increased heart rate in sensitized HLPs, probably due to histamine release. This effect was resistant to mepyramine. 6. The present findings are discussed a) with respect to the refractoriness of the anaphylactic bronchospasm to mepyramine, an effect displayed in isolated lung preparations but not in intact animals. b) with respect to the mechanism of the so-called protracted shock which is mainly due to heart failure.

In the heart lung preparation (HLP) of the guinea pig 2.5 ml rat serum anaphylatoxin (AT) released approximately ten times less histamine than was released by 5 mg ovalbumin in the sensitized HLP. In intact animals the plasma histamine increase caused by 5 ml/kg AT was not significantly different from that caused by 10 mg/kg ovalbumin. It is supposed that in AT shock extrapulmonary sources contribute to plasma histamine more than in anaphylactic shock. 2. Mepyramine maleate (40 μg/ml blood) did not significantly reduce the bronchospasm caused by AT in HLP except when a very high AT dose (0.3 ml/ml) was given. In the intact animal, mepyramine lowered the AT-induced bronchospasm by 50–65%. This effect of mepyramine is completely abolished byβ-adrenoceptive receptor blockade. 3. It is concluded that the prevalence of histamine effects in AT shock in vivo, as documented by the life saving effect of antihistamines, is due to 2 facts: influx of histamine from the organs other than the lung, and preferential antagonism by catecholamines of such shock mechanisms which are not based on histamine release.

    Notizen: Zusammenfassung Eine früher von den Verff. beschriebene Methode, mit der man staubbeladene Membranfilter für die elektronenoptische Untersuchung präparieren kann, wurde für weitere Messungen des Schwebestaubes in Bergwerken verwandt. Der durch das Schießen entwickelte Staub ließ hohe Staubkonzentrationen erkennen und zeigte ein enges Teilchenspektrum. Außerdem wurden mit der Membranfiltermethode Untersuchungen der atmosphärischen Luftverunreinigungen durchgeführt. Die in Düsseldorf entnommenen Staubproben wiesen vor allem Teilchen mit einer Korngröße unter 0,1Μ auf, während in Bochum vor allem Rußpartikeln in den Vordergrund traten.
