Summary Beef serum kininogen yields on incubation with pepsin „pepsitocic“ activity, which is, however, about 100 times weaker than the kinin effects following incubation with crotalus venom or trypsin. Kinin and „pepsitocin“ content of protein fractions run parallel during preparative work with blood serum. The kinin-yielding group of purified kininogen can be extracted with glacial acetic acid following pepsin digestion. Its substrate function for crotalus venom remains stable even for many hours of peptic digestion; the kinin activity which can be released by pancreatic kallikrein, however, is diminished quickly and considerably by pepsin. A chromatographically and electrophoretically homogeneous peptide (PKFL) has been obtained by combined chromatography on carboxymethyl cellulose, paper and phosphorylated cellulose; it consists to 65% of kinin-9. In addition to the amino acids of methionyl lysyl bradykinin, it contains each one serine, valine and glutamic acid residue. A second peptide fraction (PKFS) represents about 1/7 of the kinin-yielding group recovered. A separation of substrate function for trypsin, crotalus venom, pancreatic kallikrein or carboxypeptidase A was never demonstrable. Both kinin-yielding fragments as well as a third fraction separable by electrophoresis or column chromatography can be labelled by the common term „pepsitocin“. They act—like kinins, but much weaker—on rat uterus, guinea pig ileum, rabbit blood pressure or cutaneous vessels; they differ from kinin-9 by the relative activities on blood pressure and ileum. Following peptic digestion of crude kininogen preparations further, relatively very weak pepsitocic and kinin-yielding fractions are demonstrated by chromatography.

Zusammenfassung Setzt man gereinigtes Rinderserum-Kininogen mit Pepsin um, so erhält man „pepsitocische“ Aktivität, die aber ca. 100 mal geringer ist als die durch Einwirkung von Trypsin oder Crotalusgift erzielbaren Kinin-Effekte. Bei der Aufarbeitung von Serum laufen Kinin- und „Pepsitocin“-Gehalt der Proteinfraktionen parallel. Die kininliefernde Gruppierung von gereinigtem Kininogen ist nach Umsatz mit Pepsin mittels Eisessig extrahierbar. Während ihre Substratfunktion für Crotalusgift auch bei mehrstündiger Inkubation weitgehend erhalten bleibt, sinkt die durch Pankreaskallikrein freilegbare Kininaktivität schnell und stark ab. Durch Kombination von Chromatographie an Carboxymethylcellulose, auf Papier und schließlich an phosphorylierter Cellulose wird ein chromatographisch und elektrophoretisch einheitliches, basisches Peptid (PKFL) erhalten, das zu 65% aus Kinin-9 besteht. Neben den in Methionyl-Lysyl-Bradykinin vorkommenden Aminosäuren enthält es noch je einen Serin-, Valin- und Glutaminsäure-Rest. Eine zweite, ebenfalls basische Peptidfraktion (PKFS) enthält rund ein Siebentel der insgesamt wiedergefundenen kininliefernden Gruppierung. Eine Trennbarkeit der Substratfunktionen für Trypsin, Crotalusgift, Pankreaskallikrein oder Carboxypeptidase A deutete sich nirgends an. Beide kininliefernde Fragmente sowie eine dritte, von ihnen elektrophoretisch und säulenchromatographisch abtrennbare Fraktion können unter der Sammelbezeichnung „Pepsitocin“ zusammengefaßt werden. Wie die Kinine, aber sehr viel schwächer, wirken sie auf Rattenuterus, Meerschweinchenileum, Kaninchenblutdruck und Hautgefäße; in ihrer relativen Blutdruck- und Darmwirksamkeit sind sie von Kinin-9 verschieden. Nach Pepsinbehandlung roher Kininogenpräparate (Fraktion IV-6) werden weitere, vergleichsweise sehr schwache Fraktionen mit pepsitocischen und kininliefernden Eigenschaften chromatographisch gekennzeichnet.
