    Description / Table of Contents: Zusammenfassung Trotz vorheriger Gabe von Propranolol bewirkt Oxyfedrin beim Hund einen signifikanten Anstieg der Frequenz, des arteriellen Spiegels der freien Fettsäuren, der arteriellen 3-Hydroxybutyrat-Konzentration und der Aufnahme von freien Fettsäuren in die Leber sowie eine ebenfalls statistisch gesicherte Verminderung des coronaren Widerstandes. Ein Vergleich der gefundenen Wirkungen mit bereits publizierten Ergebnissen zeigt dennoch eine, wenn auch nur partielle Blockierbarkeit der Oxyfedrineffekte durch Propranolol. Die anderen, z. T. gegenüber der alleinigen Gabe von Oxyfedrin sogar verstärkten Wirkungen wie die signifikante Abnahme des arteriellen Spiegels und der myokardialen Bilanz von Pyruvat werden durch gleichsinnige, additive Wirkungen von Propranolol und Oxyfedrin erklärt.
    Notes: Summary Following beta-adrenergic blockade with propranolol, infusions with oxyfedrin caused significant increases in heart rate, arterial free fatty acids and 3-hydroxy-butyrate levels, uptake of free fatty acids into the heart and liver, and a significant decrease in coronary vascular resistance. A comparison with data from previous experiments without beta-adrenergic blockade shows at least a partial blockade of the hemodynamic and metabolic effects caused by oxyfedrin. The other metabolic parameters, showing identical changes with oxyfedrin and propranolol, could not be taken for a comparison of the action of oxyfedrin before and after betaadrenergic blockade.

    Notes: Summary The intravenous infusion of fructose (0.7 and 2.4 g/kg/30 min) increases the incorporation of14C-glycine into rat liver adenine nucleotides significantly as compared to controls treated with 0.9% NaCl. A decrease of the total adenine nucleotide content of the livers occurs concomitantly. In contrast to its effect upon liver tissue fructose neither effects the content of adenine nucleotides nor the incorporation of glycine into these nucleotides in skeletal muscle of rats. — The results support the hypothesis that large quantities of fructose cause an increasedde novo synthesis of purine nucleotides in the liver. However, they do not as yet present clear cut evidence as to whether this is a direct effect of fructose or is secondary to the depletion of adenine nucleotides.

    Description / Table of Contents: Zusammenfassung In den Erythrocyten eines Falles von Trisomie G-21 konnte im Gegensatz zu bisher vorliegenden Berichten eine Verminderung der Aktivität von Phosphofructokinase (PFK) gefunden werden. Daneben wurde eine hohe Aktivität der Glycerophosphatdehydrogenase und in geringerem Ausmaß auch anderer Enzyme nachgewiesen. Die mit dem PFK-Mangel in Zusammenhang stehenden Stoffwechselstörungen sowie die Frage der genetischen Lokalisation dieses Enzyms werden diskutiert.
    Notes: Abstract A deficiency of erythrocyte-phosphofructokinase has been demonstrated in a case of Trisomy-G-21. This is in contrast to findings of other authors who reported an increase of erythrocyte-PFK in cases of Down's syndrome. The metabolic changes caused by PFK deficiency and the problem of genetic localization of this enzyme are discussed. In addition an increased activity of glycerophosphate-dehydrogenase and, to a lesser degree, some other enzyme was found in the patients red blood cells.

    Description / Table of Contents: Zusammenfassung Die Arbeitsgruppen um Spiegelman und unserer Laboratorien haben nachgewiesen, daß das Enzym reverse Transcriptase auch extrazellulär bei malignen Erkrankungen aufgefunden werden kann. Ein solches Enzym wurde von Spiegelman aus der Muttermilch, von uns aus dem Plasma von Patientinnen mit Mammacarcinom und leukämischen Erkrankungen isoliert. Das von uns isolierte Enzym wurde zur elektrophoretischen Homogenität angereichert und kann 1) synthetische Templates (Primers) mit der der viralen Transcriptase inhärenten Spezifität ablesen; 2) heteropolymere RNA-Anteile einer einsträngigen Phagen-RNA in eine homologe DNA-Sequenz übersetzen und 3) weist chromatographisch ähnliches Verhalten wie die reverse Transcriptase onkogener RNA-Tumorviren auf. Vorläufige Untersuchungen zum Nachweis eines analogen Enzyms im Plasma bei anderen malignen Erkrankungen haben bisher kein positives Ergebnis gebracht.
    Notes: Summary Work from Spiegelman and from our laboratories documented extracellular occurrence of the enzyme reverse transcriptase in malignant states. This enzyme was shown to be present in particles of human milk by Spiegelman and in patients with breast cancer or leukemias by us. The isolated enzyme was purified to electrophoretic homogenity and 1) Utilizes synthetic template—primers with a specificity like the viral reverse transcriptase. 2) Is capable of copying heteropolymeric regions of natural RNA-template-primer complexes. 3) Shows the chromatographic behaviour of the viral enzyme. These preliminary investigations documented the presence of the above mentioned enzyme only in these two forms of malignancies, while the other patients looked treated for the probably viralrelated enzyme in the plasma were negative.
