    Notizen: Abstract  We isolated a cDNA clone encoding the human prostaglandin (PG) E receptor EP4 subtype and examined the gene expression in human blood cells. Northern blot analysis revealed that the EP4 gene is expressed at a high level in peripheral blood mononuclear cells, and at lower levels in cultured human blood cell lines, THP-1 and U937 (monocytoid cell lines), MOLT-4 and Jurkat (T-cell lines), and Raji (B-cell line). To examine regulation of the EP4 gene expression in the immune system, we studied the effects of phorbol 12-myristate 13-acetate (PMA) on these cell lines. Gene expression was upregulated in THP-1, U937, and Raji cells by PMA, and was downregulated in MOLT-4 and Jurkat cells. In THP-1 cells the effects of PMA were further analyzed, and the upregulation of the EP4 gene was shown to be followed by an increase in PGE2 binding sites and in PGE2-induced cAMP accumulation. In the striking contrast, other PGE receptor subtypes (EP1, EP2 and EP3) and other prostanoid receptors (IP and DP) were shown not to be upregulated by PMA. Therefore, this is the first demonstration of a highly specific upregulation of the EP4 subtype in THP-1 cells treated with PMA, suggesting the importance of the EP4 subtype in the immune system. In the present study we also clarified that EP4 gene expression is regulated differently among human monocytoid and lymphoid lineage cells, thus leading to the better understanding of the regulatory mechanisms for the human EP4 gene expression in the immune system.

    Notizen: Copolymers of phenylvinylene carbonate with styrene and p-chlorostyrene are readily prepared with radical initiators. The carbonate portion of the copolymers has been hydrolyzed to a 1,2-glycol which was then oxididatively cleaved with periodate-perman-ganate to produce a polymer with carboxyl and keto endgroups. Block copolymers were then prepared by conversion of the carboxyl groups to an acid chloride, followed by coupling with diols or a diamine.

    Notizen: The synthetic details of solution polymerization in benzene and bulk polymerization of vinylferrocene are reported. In benzene solutions, with azobisisobutyronitrile (AIBN) as the initiator, small yields of low-polydispersity low molecular weight (M̄n ≃ 5000) polyvinylferrocene is obtained. However, high yields can be obtained by continuous or multiple AIBN addition. Higher molecular weight polymers and binodal polymers can be obtained as the monomer concentration is increased. In bulk polymerizations, yields of 80% can be obtained. The molecular weight increases as temperature decreases from 80 to 60°C in bulk polymerizations, and an increasing amount of insoluble polymer results. The soluble portion is often binodal, the higher molecular weight node consisting of an increasingly branched structure. Lower molecular weight polymer was readily fractionated into narrow fractions from benzene-methanol systems, but higher molecular weight polymer proved impossible to fractionate into narrow fractions due to branching.

    Notizen: Kinetics of grafting reactions of styrene to preirradiated cellulose acetate have been studied by labeling the active sites with bromine atoms. The dilution of styrene monomer with methanol affected grafting reactions remarkedly: e.g., as the concentration of methanol in monomer solutions increased, the growth rates of individual graft radicals decreased while the average lifetimes increased. The integrated amounts of active sites which participated in grafting reactions were also affected by the constitution of monomer solutions and varied roughly in proportion to the extents of swelling of cellulose acetate. Grafting yields for styrene/methanol = 1/1 were higher than for 3/1 and 1/3 throughout the duration of grafting reactions, which is due not only to the high molecular weight of graft chains but also to the large number of graft chains for the 1/1 system compared to the other systems.
