    Notizen: Abstract We have recently shown that the angiotensin I converting enzyme (ACE) gene is linked to NaCl-loaded blood pressure in the stroke-prone spontaneously hypertensive rat (SHRSP), and that high-NaCl loading selectively stimulates ACE in the aorta of SHRSP but not in normotensive Wistar-Kyoto (WKY) rats. We therefore investigated the relationship between cardiac ACE and the development of hypertension and left ventricular hypertrophy in response to normal- and high-NaCl diet in these rats. ACE mRNA and ACE activity were measured in left ventricular tissue after completion of hemodynamic characterization of the animals. While SHRSP rats increased blood pressure (P〈0.0001) and heart rate (P〈0.005) in response to high NaCl, blood pressure remained unchanged in WKY. Similarly, relative left ventricular weight increased only in SHRSP after high NaCl (P〈0.002). A significant two- to threefold increase of cardiac ACE mRNA and fourfold stimulation of ACE enzyme activity in response to high NaCl was found in both WKY and SHRSP rats (P〈0.005). The induction of ACE gene expression was significantly more pronounced in SHRSP compared to WKY (P〈0.02), whereas no significant strain differences in left ventricular ACE activity were found after either normal- or high-NaCl diet. Thus, arterial blood pressure and left ventricular weight remained unchanged in the WKY rats despite the activation of left ventricular ACE activity after high-NaCl exposure. These results demonstrate that left ventricular ACE activity is equally upregulated in response to high-NaCl in the normotensive and hypertensive strain, independently from the development of hypertension. We conclude that the pretranslational induction of left ventricular ACE with high-NaCl loading may be important both for the regulation of cardiac angiotensins and kinins and for local therapeutic ACE inhibition in the heart during high-salt status.

    Notizen: Abstract We live on a cold planet where more than 80% of the biosphere is permanently below 5°C, and yet comparatively little is known about the genetics and physiology of the microorganisms inhabiting these environments. Based on molecular probe and sequencing studies, it is clear that Archaea are numerically abundant in diverse low-temperature environments throughout the globe. In addition, non-low-temperature-adapted Archaea are commonly exposed to sudden decreases in temperature, as are other microorganisms, animals, and plants. Considering their ubiquity in nature, it is perhaps surprising to find that there is such a lack of knowledge regarding low-temperature adaptation mechanisms in Archaea, particularly in comparison to what is known about archaeal thermophiles and hyperthermophiles and responses to heat shock. This review covers what is presently known about adaptation to cold shock and growth at low temperature, with a particular focus on Antarctic Archaea. The review highlights the similarities and differences that exist between Archaea and Bacteria and eukaryotes, and addresses the potentially important role that protein synthesis plays in adaptation to the cold. By reviewing the present state of the field, a number of important areas for future research are identified.

    Notizen: Abstract An Atriplex nummularia L. cDNA probe encoding the partial sequence of an isoform of the plasma-membrane H+ -ATPase was isolated, and used to characterize the NaCl regulation of mRNA accumulation in cultured cells of this halophyte. The peptide (447 amino acids) translated from the open reading frame has the highest sequence homology to the Nicotiana plumbaginifolia plasma-membrane H+-ATPase isoform pma4 (greater than 80% identity) and detected a transcript of approximately 3.7 kb on Northern blots of both total and poly(A)+ RNA. The mRNA levels were comparable in unadapted cells, adapted cells (cells adapted to and growing in 342 mM NaCl) and deadapted cells (cells previously adapted to 342 mM NaCl that are now growing without salt). Increased mRNA abundance was detected in deadapted cells within 24 h after exposure to NaCl but not in unadapted cells with similar salt treatments. The NaCl up-regulation of message abundance in deadapted cells was subject to developmental control. Analogous to those reported for glycophytes, the plasma-membrane H+-ATPase are encoded by a multigene family in the halophyte.

    Notizen: The kinetics of ethidium's intercalative binding to DNA packaged in bacteriophage T7 and two T7 deletion mutants have been determined, using enhancement of fluorescence to quantitate binding. At a constant ethidium concentration, the results can be described as first-order binding with two different rate constants, k1* (= k1 + k-1) and k2* (= k2 + k-2). The larger rate constant (k1*) was at least four orders of magnitude smaller than the comparable first-order forward rate constant for binding to DNA released from its capsid. At 25°C values of k1* decreased as the amount of DNA packaged per internal volume increased. This latter observation indicates that the rate of ethidium's binding to packaged T7 DNA is limited by an event that occurs inside of the DNA-containing region of T7, not by the crossing of T7 capsid's outer shell. Arrhenius plots of kM1* are biphasic, indicating a transition for packaged DNA at a temperature of 20°C. The data indicate that k1* s are limited by either sieving of ethidium during its passage through the packaged DNA or subsequent hindered intercalation.

    Notizen: A method is described for formation of hydroxamic acids by direct coupling, in aqueous medium, of hydroxylamine hydrochloride and the sodium salt of a carboxylic acid. The reaction is mediated by a water-soluble carbodiimide, 1-cyclohexyl-3-[2-morpholinyl-(4)-ethyl]-carbodiimide metho-p-toluene sulfonate. Using model compounds, the production of α-, β- and γ-acyl hydroxamic acids was studied. Hydroxamic acid derivatives of α-polyglutamic and γ-polyglutamic acids were also prepared. Dinitro-phenylhydroxamate derivatives were prepared and subjected to Lossen rearrangement, and the reaction mixtures hydrolyzed. Analysis showed that α-polyglutamic acid yielded as many molecules of α,γ-diaminobutyric acid as there had been glutamic acid residues in the form of the dinitrophenylhydroxamate, and gave rise to no ammonia and succinic semialdehyde. In contrast, γ-polyglutamic acid yielded exactly twice the molar quantity of ammonia as there had been glutamic acid residues in the form of dinitrophenylhydroxamate, and also gave rise to significant quantity of succinic semi-aldehyde but no α,γ-diaminobutyric acid. Since production of the latter is characteristic of α-glutamyl hydroxamates and production of the former is associated with γ-glutamyl hydroxamates, the results indicated that the side-chain carboxyl groups of either polymer retained their identities during formation of the hydroxamic acid derivative, and no interchange had occurred between α- and γ-carboxyl groups. These methods were then used to establish that gelatin of ichthyocol contains (per 1000 total residues) at least 20 residues of glutamic acid in γ-peptide linkage. Due to the incomplete dinitrophenylation of the hydroxamic acid derivative of the protein, gelatin of calf skin collagen gave a lower figure of 10 such linkages per 1000 total amino acid residues-a value which must be considered a minimum value. These results show that γ-glutamyl peptide bonds exist in collagen.

    Notizen: Polyphenylquinoxalines (PPQ's) containing various amounts of crosslinkable pendant phenylethynyl groups were prepared from the reaction of 3,3′,4,4′-tetraaminodiphenyl ether, 4,4′-oxydibenzil, and 4,4′-oxybis(4″-phenylethynylbenzil). The distribution of the pendant phenylethynyl groups along the polymer chain was varied in an attempt to alter the properties of the polymers. Preliminary film, adhesive, and laminate properties of PPQ void of pendant crosslinkable groups and containing pendant phenylethynyl groups were determined. The thermally induced reaction of the phenylethynyl group crosslinked the polymer which resulted in better dimensional stability at elevated temperatures.

    Notizen: Molecular weights were studied by gel permeation chromatography of derivatized poly(α-tetrahydrothiophenio para-xylylene) chloride produced by aqueous or methanolic base-induced polymerization of 1,4-bis(tetrahydrothiopheniomethyl) benzene dichloride, both with and without a variety of added polymerization inhibiting agents. Efficient radical scavenging agents such as 2,2,6,6-tetramethylpiperidinoxyl and hydrogen atom donor 2,4,6-tri-tert-butylaniline reduced the degree of polymerization of the reactive α-(tetrahydrothiophenium chloride)-para-xylylene intermediate produced in this chemistry, and in some cases completely suppressed formation of high polymer. These two traps did not affect the equilibrium production of the para-xylylene by UV-Vis spectral analysis; hence they must affect the subsequent polymerization chain propagation steps in the mechanism. Electron spin resonance studies of polymerization in the presence of 0.00025 equiv of TEMPO showed disappearance of the spin label, a result consistent with a radical scavenging process. The results suggest that production of high molecular weight poly(α-tetrahydrothiophenio para-xylylene) chloride proceeds through a radical chain propagation sequence. © 1992 John Wiley & Sons, Inc.

    Notizen: para-Xylylenes are generated by treatment of various 1,4-bis(dialkylsulfoniomethyl)arene dihalides with base in water, methanol, and aqueous acetonitrile, as shown by UV-Vis spectroscopy. This procedure allows the monitoring of the transient xylylene monomers that yield polyelectrolyte precursor polymers for poly(arylene vinylene)s, formed by variations of the chemistry developed originally by Wessling and co-workers. Alkoxy, alkyl, and halogen ring substituents on the sulfonium salt precursors do not greatly affect the ability to generate and observe the growth and decay of steady-state concentrations of the para-xylylene intermediates. Use of strong resonance-acceptor substituents - such as cyano or nitro - on the ring reduces production of a strong para-xylylene absorption, possibly due to reluctance of the ylides initially formed from the bis-sulfonium salts to eliminate to the xylylenes. By variation of UV-Vis conditions, it was found that use of 20% aqueous acetonitrile rather than water allowed formation of low to modest molecular weight polyelectrolytes in cyano-substituted cases (Mw = 8000-37,000). Use of UV-Vis test experiments should be useful for screening of bis sulfonium salt precursors that may be expected to give high molecular weight polyelectrolytes - in cases where para-xylylene formation is easy - as well as for finding reaction conditions that will optimize polymer formation. © 1992 John Wiley & Sons, Inc.

    Notizen: Synthesis of poly(4′-vinyl-2,4,6-tri-tert-butyl-diphenyloxalate) and its 65 : 35 and 19 : 81 copolymers with styrene are described. Rigid phase broad-band photolysis of the homopolymer with a quartz filtered xenon arc at 77 K results in production of up to 25% of the theoretical number of spins/mol expected for quantitative production of pendant phenoxyl radicals, and shows no major loss of radical signal in the ESR at temperatures below 100 K. Curie law analysis of the temperature dependence of the ESR radical signal intensity for the neat photolyzed homopolymer 1 shows curvature consistent with antiferromagnetic pairing of radical spins at low temperatures. Since through-bond conjugation of radical spins is not possible in this system, the antiferromagnetic interaction is interpreted in terms of intrachain and/or interchain through-space exchange interactions.
