    Notizen: Abstract Hybrid rice has contributed significantly to the dramatic increase of rice production in the world. Despite this, little attention has been given to studying the genetic basis of heterosis in rice. In this paper, we report a diallel analysis of heterosis using two classes of molecular markers: restriction fragment length polymorphisms, (RFLPs) and microsatellites. Eight lines, which represent a significant portion of hybrid rice germ plasm, were crossed in all possible pairs, and the F1s were evaluated for yield and yield component traits in a replicated field trial. The parental lines were surveyed for polymorphisms with 117 RFLP probes and ten microsatellites, resulting in a total of 76 polymorphic markers well-spaced in the rice RFLP map. The results indicated that high level heterosis is common among these crosses: more than 100% midparent and 40% better-parent heterosis were observed in many F1s, including some crosses between maintainer lines. Heterosis was found to be much higher for yield than for yield component traits, which fits a multiplicative model almost perfectly. Between 16 and 30 marker loci (positive markers) detected highly significant effects on yield or its component traits. Heterozygosity was significantly correlated with several attributes of performance and heterosis. Correlations based on positive markers (specific heterozygosity) were large for midparent heterosis of yield and seeds/panicle and also for F1 kernel weight. These large correlations may have practical utility for predicting heterosis.

    Notizen: Abstract  Although F2s are the most informative populations for genetic analysis, it has been difficult to use F2 populations directly for QTL analysis because it is usually difficult to assess the reliability of the data, due to an inability to estimate the experimental errors. In this study, we performed a QTL analysis for yield and yield-component traits of an F2 population based on data from replicated field trials over 2 years using vegetative shoots of ratooned plants, making use of the ratooning habit of rice. The objective of this study was to explore the possibility of conducting QTL analyses directly based on an F2 population by means of ratooning plants. The experimental population was from a cross between ’Zhenshan 97’ and ’Minghui 63’, the parents of ’Shanyou 63’, an elite rice hybrid widely grown in China. A genetic linkage map containing 151 molecular markers was constructed for QTL mapping. A total of 20 distinct QTLs were detected; eight of these were detected in both years and remaining 12 in only 1 year. Compared with the results of our previous analysis of the F2:3 families from the same cross, it was shown that most of the QTLs detected in the ratooned F2 population were also detected in the F2:3 population. However, the estimates of both additive and dominant types of genetic effects for many of the QTLs based on F2 ratoons were substantially larger than those based on F2:3 families. The results indicate that vegetatively ratooned F2 populations may have considerable utility in the mapping of QTLs, especially if dominant types of gene actions are of concern, although there were certain technical limitations in making use of such populations in the experiments.

    Notizen: We have investigated the interface formation of Ca with poly(p-phenylene α,α′-diphenyl vinylene) (PPV-DP) and poly(p-phenylene α-phenyl vinylene) (PPV-P) using X-ray photoemission spectroscopy (XPS). Similarly to our earlier findings in metal/PPV interface formation, the O 1s peak shifted toward a lower binding energy as soon as Ca was deposited on to the polymers. This was accompanied by the formation of Ca—O, suggesting a chemical origin for the O 1s shift. By contrast, the C 1s peak shift toward a lower binding energy was observed relatively later, after about 4 Å of Ca deposition. At the same time, a new C 1s component became noticable at about -1.5 eV relative to the initial C 1s peak. This component signifies the possibility of polymer disruption by the Ca atoms to form Ca—C species. The C 1s peak shift is attributed to Ca induced surface band bending and barrier formation as in the case of metal/PPV interface formation. The disruption of the polymer may also induce changes in the interface electronic states and contribute to the C 1s peak shift. From the intensity attenuation analysis, we conclude that the initial 15 Å of Ca overlayer is contaminated by the Ca—O and Ca—C species and the overlayer is pure beyond 15 Å of Ca coverage.

    Notizen: A molecular-dynamics simulation was used to carry out an introductory study of the hydration of a section of a rigid single A- or B-DNA helix with one Na+ counterion per nucleotide. Four Na+ ions and four nucleotides and periodic boundary conditions were used to mimic an infinite helix. The atoms of the helix and the Na+ ions were assumed to be Lennard-Jones spheres that also carried charges. Stillinger four-point charge model water molecules were used. We carried out five calculations, for 26 and 46 water molecules in B-DNA and 20, 32, and 46 in A-DNA fragments. The arrangements of the Na+ ions are found to have some similarities to those obtained by Clementi and Corongiu. In the calculations with 46 water molecules, we found that two Na+ ions can be bridged by about two water molecules and form a hydrated bound pair, which in turn forms a bridge between the guanine N7 and a near phosphate group. These bound pairs may be important in stabilizing the helix structure of DNA molecules.

    Notizen: We report on our recent x-ray photoemission spectroscopy investigations of the interface formation of metals with poly(p-phenylene vinylene) (PPV) prepared under various conditions. We have found that during deposition the metal reacts with residual hydroxyl groups in the polymer. In addition, we have found that Schottky barrier formation and the associated band bending depend strongly on surface preparation. In the case of Al deposition, samples converted in situ, containing 5% surface oxygen, show band bending that depends on the thickness of the metal overlayer, with effects arising after as little as 1 Å of metal. On the other hand, a sample converted ex situ, with 10% surface oxygen, is insensitive to aluminium deposition. We feel that surface impurities and adsorbed species may delay Schottky barrier formation by acting as a buffer layer that prevents the PPV substrate from interacting with the growing layer of Al. By contrast, the Ca/PPV surface exhibits delayed band bending, with strong interactions between surface oxygen and Ca. Our results indicate that band bending at the metal/PPV interface is governed by the metallicity of the metal overlayer, which itself is influenced by the interface reaction of the deposited metal with the PPV substrate or the surface residual impurities. Finally, the degree of band bending observed did not correlate directly with the differences in work functions between the metals and PPV.

    Notizen: Round-robin studies on relative sensitivity factors (RSFs) in secondary ion mass spectrometry (SIMS) were conducted using bulk GaAs samples uniformly doped with various impurity elements. A total of 31 laboratories participated in two round-robins. More than 30 sets of relative ion intensities were obtained for B, Si, Cr, Mn, Fe, Cu, Zn, In and Te in GaAs. The RSFs for both positive and negative ions were derived for several types of SIMS instruments. The effect of primary ion incident angle was examined using quadrupole-based instruments and found to be the determining factor of the instrumental dependence of RSF. © 1998 John Wiley & Sons, Ltd.

    Notizen: The CCl4 flooding technique combined with oxygen ion bombardment has been used during SIMS analysis. The secondary ion yield and the matrix effect have been investigated. The results have been compared with those obtained under oxygen bombardment or under oxygen bombardment plus oxygen flooding. We show that this simple technique allows a significant enhancement of ion yield for all the elements studied and induces a weak matrix effect.
