    Notizen: Abstract A total of 90 alleles found in white Caucasians from North-West Germany were sequenced and 26 alleles chosen to construct a uniform and reliable allelic ladder for the STR system ACTBP2 (SE33). In addition 3 new aspects concerning the sequence structure were observed. Population studies were carried out on white Caucasians (n = 278) from North-West Germany using the new improved ladder. A total of 24 alleles and 14 “interalleles” were found and reproducible results obtained. No significant deviation from Hardy-Weinberg-equilibrium could be observed.

    Notizen: Summary Forty-seven patients with 48 unoperated acoustic neuromas have been studied by computed tomography (CT) with respect to bone changes. CT and conventional X-ray examination of the skull were compared. The importance of using a small collimator, proper selection of the plane of the CT section and changing of the window level is stressed. CT detects porus changes as accurately as conventional X-ray methods.

    Notizen: Summary The changes of the enhancement of the lesion with time after intravenous administration of contrast medium were measured with computed tomography (CT) in 23 cases of acoustic neuroma. The pattern of high initial enhancement followed by a fairly rapid decrease is confirmed. A more rigorous study of the enhancement pattern using a mathematical description of the enhancement curve of the tumor as well as a measurement of the individual blood enhancement curve is discussed.

    Notizen: Summary Attenuation profiles across the petrous bone covering the whole internal auditory meatus (IAM) were constructed from the printouts obtained by computed tomography (CT) with narrow collimation performed on 12 patients with 13 acoustic neuromas. In healthy patients the attenuation profiles of right and left petrous bone were very similar in shape. The attenuation values of the individual pixels in the pixel columns of the printout located at the site of the porus and the IAM reflected the demineralization of the petrous bone and the widening of the porus and the IAM caused by the acoustic neuroma. A widening deep in the meatus was demonstrated in a patient with an intracanalicular tumor, and therefore it seems possible to make this diagnosis by CT scanning combined with the construction of attenuation profiles across the petrous bone. In the presence of unilateral acoustic neuroma there was a significant and characteristic difference in shape between the attenuation profiles of the two sides with generally lower attenuation values on the tumor side together with signs of widening of the porus and the IAM. In cases of bilateral acoustic neuroma comparison of the attenuation profiles can be made with mean attenuation curves obtained from scanning normal petrous bones. The prevailing physical limitations for demonstrating a narrow bony canal like the IAM with CT was experimentally analyzed using bone-simulating plastic material.

    Beschreibung / Inhaltsverzeichnis: Zusammenfassung 50 verschiedene ACTBP2 Allele und die Zell-Linie K562 wurden sequenziert und analysiert. Die Sequenzierergebnisse zeigen nicht nur einen Längensondern auch einen hoch variablen Sequenzpolymorphismus. 2 verschiedene Sequenzstrukturen wurden charakterisiert. Typ 1 (223–259bp) enthält den regulären 4bp-Repeat AAAG. Im Typ II (265–309 bp) tritt zusätzlich die Hexanukleotideinheit AAAAAG auf, deren Position innerhalb der Repeatregion stark variabel ist. 20 sequenzierte Allele wurden für die Konstruktion einer allelischen Leiter ausgewählt. Denaturierende und hochauflösende, nicht denaturierende Gele wurden ausgewertet und Unterschiede im Wanderungsverhalten der Allele festgestellt. In denaturierenden Gelen ist die elektrische Mobilität abhängig von den Fragmentlängen der Allele. Dagegen zeigen in nicht denaturierenden Gelen einige Allele unterschiedliches Wanderungsverhalten im Vergleich zur sequenzierten allelischen Leiter, welches durch unterschiedliche Fragmentlängen und/oder unterschiedliche Sequenzstrukturen erklärt werden kann.
    Notizen: Summary A total of 50 different ACTBP2 (human betaactin related pseudogene) alleles and the cell line K562 were sequenced and analysed. Sequence data revealed not only length polymorphism but also a highly variable sequence polymorphism. 2 different sequence structures were characterised. Type I (223–259bp) contained the known regular 4bp repeat AAAG. Type II (265–309 bp) revealed a further hexanucleotide unit AAAAAG in addition to the common AAAG which only occurs once in the repeat region. The position of this insertion showed considerable variation. To obtain a regular spaced allelic ladder 20 of the sequenced alleles were selected. Denaturing gels and high resolution/non-denaturing gels were compared and striking differences could be seen between the 2 gel systems. Separation of the alleles on a 6% denaturing gel and analysis on the ABI 373A Sequencer revealed fragment sizes which corresponded to the sequencing data but were in general 6–10bp longer. In contrast, in non-denaturing gels some alleles showed different electrophoretic mobilities compared to the sequenced allelic ladder which could indicate different fragment length and/or different sequence structure.
