    Notes: SUMMARY– Investigations were conducted on the effect of three storage temperatures, 2°, 16°, and 37°, on the changes and relationships of certain chemical and physical properties of post-mortem bovine semitendinosus and psoas muscle. Post-mortem muscle shortening was measured with the isometer. Isometric tension development was maximal at 2°, minimal at 16°, and at 37° tension was approximately one-half that developed at 2°. The large tension development at 2° very likely originates from the same events as those in “cold shortening.” Differences in isometric tension parameters were noted between muscles in that psoas muscle developed tension and lost the ability to maintain tension more quickly than did the semitendinosus. Loss of ability to maintain tension was observed only at 2°, and this could correspond to a “resolution” of rigor mortis. Adenosine triphosphate (ATP) degradation was measured by two methods, ammonia production and bioluminescent enzymic method; the bioluminescent method proved to be the more satisfactory. A common relationship was observed between pH and ATP for both muscles and the three temperatures studied. No direct relationship was found between ATP degradation and shear resistance with the possible exception of muscle stored at 37°. Isometric tension parameters and shear resistance were related somewhat at 2° in semitendinosus muscle, but no relationship existed at 16° and 37°. Although considerable tension developed in psoas and semitendinosus muscle at 37°, shear resistance values decreased continuously, indicating that factors other than shortening are more important at high temperature and that these factors are temperature-dependent.The role of ATP degradation in tension development was difficult to interpret, since at 2°, only a small change occurred in ATP level during large tension development, and the level of ATP at 2° did not differ from ATP level in muscle stored at 16° which developed little tension.Differences in post-mortem muscle shortening at 2 and 37 are discussed.

Die Epithel-Bindegewebsgrenze wurde bei 36 Mammacarcinomen, einem Fibroadenom der Mamma und einer proliferierenden Mastopathie systematisch elektronenmikroskopisch untersucht. 2. Während das Epithel bei Fibroadenom und Mastopathie überall durch eine geschlossene, elektronenmikroskopisch eindeutig darstellbare Basalmembran vom umgebenden Bindegewebe abgegrenzt wird, fehlt eine Basalmembran in der Regel beim Mammacarcinom. 3. Die Carcinomzellen liegen dem umgebenden Bindegewebe entweder mit glatter oder unregelmäßiger Zellmembran an. Die Zerklüftung der Zelloberfläche stellt sich als submikroskopisches Äquivalent von zwei verschiedenen biologischen Vorgängen an der Tumor-Stroma-Grenze dar: 1. einer Phagocytose von Fibrin und Erythrocyten und 2. einer Antigen-Antikörperreaktion. 4. Das peritumorale Bindegewebe zeigt ausnahmslos eine hochgradige Aktivitätssteigerung der Fibroblasten mit vermehrter Kollagensynthese. Außerdem treten aktivierte Muskelzellen auf. Elastische Elemente gehen zugrunde. 5. Bei einem Ductuscarcinom einer 38jährigen Frau war eine peritumorale Mastocytose nachweisbar; der wachstumshemmende Effekt des Heparins wird diskutiert. 6. Der (in 2 Fällen beobachtete) exzessive Lymphocytenreichtum des parablastomatösen Stromas spricht für eine lokale immunologische Tumorabwehrreaktion. 7. Dem nach Entdifferenzierung der Zelle und Basalmembranverlust des Parenchyms wirksam werdenden Wachstumsdruck des Tumorgewebes mit Phagocytoseleistung als Ausdruck der Aggressivität des Malignoms begegnet das Stroma mit Stoffwechselaktivierung, Phagocytose, Faserneubildung, Antihyaluronidase-Effekt und Antikörperbildung. Die Prognose eines Tumors ist das Ergebnis komplexer, temporär differenter biologischer Faktoren.
    Notes: Summary An electron microscopic study was made of the Tumor-connectine tissue junction of 36 breast carcinomas, of 1 fibroadenoma, and of 1 proliferating mastopathy. The basement membranes were seen in the fibroadenoma and the proliferating mastopathy but normally were lacking in the mammary gland carcinomas. The malignant epithel cells adjoined the contiguous connective tissue by either smooth or irregular cell membranes. The irregularity of the cell membranes is interpreted as the submicroscopic equivalent of two different biological processes at the junction of tumor and connective tissue: 1) the phagocytosis of fibrin and red blood cells; 2) the reaction between antigen and antibody. The connective tissue surrounding the tumor showed increased activity of fibroblasts. Collagen fibres appeard newly formed. Muscle cells with increased metabolism were found. In two cases the peritumorous tissue showed an excessive abundance of lymphocytes suggesting a local immunological reaction of the organism against neoplastic tissue. In one case mast cells were found increased in the connective tissue. A reaction of heparin against hyaluronidasis is discussed. The prognosis of a malignant tumor is the sum of many complex, temporarily different biological factors.

    Notes: Summary To evaluate the long-term effects of orally administered levodopa, 11 patients with chronic congestive heart failure (NYHA III–IV) were studied during maintenance therapy (30±1 days) and after withdrawal from levodopa. The daily levodopa dose was 4 g in six patients; because of side effects the levodopa dose was reduced to 2–3 g in the remaining patients. After withdrawal of levodopa, mean pulmonary capillary wedge pressure and mean right atrial pressure increased significantly (from 19±2 to 24±3 and from 7±2 to 9±2 mmHg, respectively). Effective renal plasma flow was 329±57 during levodopa therapy and decreased significantly to 252±27 ml/min after withdrawal of levodopa. The number of ventricular premature contractions and couplets increased during levodopa therapy and decreased again significantly after withdrawal of levodopa. No significant differences between on and off levodopa were observed in resting heart rate, arterial blood pressure, cardiac index, stroke work index, systemic vascular resistance, sodium and water excretion, or creatinine clearance. Seven patients improved on levodopa therapy by one NYHA class; four of these seven patients deteriorated again by one NYHA class after withdrawal of levodopa. Regarding both clinical and hemodynamic changes after withdrawal of levodopa, three patients were classified as responders to long-term levodopa therapy. All three responders received 4 g levodopa per day. Average dopamine plasma level was 5.3±0.8 ng/ml in the responder group and 2.0±0.5 ng/ml in the nonresponder group. Long-term administration of oral levodopa is associated with beneficial clinical and hemodynamic response in only a minority of patients with chronic congestive heart failure.
