The yolks of White Leghorn chicken (Gallus domesticus) eggs were injected prior to incubation with either 3,3′,4,4′,5- pentachlorobiphenyl (PCB 126) at doses ranging from 0.1 to 12.8 μg/kg egg or 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) at doses ranging from 0.04 to 0.64 μg/kg egg. Chicks were subjected to necropsy within 24 h of hatching. The brain, bursa, heart, liver, and spleen were removed and weighed. Assessment of the rate of hatching indicated an LD50±S.E. of 2.3±0.19 μg/kg egg (7.1±0.58 nmol/kg egg) for PCB 126 and 0.15 ± 0.012 μg/kg egg (0.47 ± 0.037 nmol/kg egg) for TCDD. No significant differences in the incidence of developmental abnormalities (structural defects and edema) were observed in TCDD-exposed embryos, while PCB 126 caused significantly more developmental abnormalities at 3.2, 6.4, and 12.8 μg/kg egg than the vehicle control. PCB 126 caused lower hatchling weights and greater relative brain, heart, and liver weights when compared to the vehicle control group at a dose of 3.2 μg/kg egg which is greater than the LD50. TCDD at 0.08 μg/kg egg caused relative bursa weights to be less than those of the vehicle control. A toxic equivalency factor (TEF) of 0.07 was determined for PCB 126 in relation to TCDD based on overt lethality.

    Notizen: Abstract Uptake and depuration kinetics for benzo(a)pyrene (B(a)P) were determined for the midgeChironomus riparius (Diptera) with one and two compartment models. Nonfeeding animals were exposed to nominal 1.0μg·L−1 14CB(a)P for eight hr. Depuration over eight hr was determined in animals with and without substrate. The uptake rate constant was 214+-20 hr−1 (X+-SE, n=3), while elimination rate constants for the first four hr were 0.22 hr−1 (with substrate) and 0.06 hr−1 (without substrate). Biphasic depuration was observed with an initial rapid phase that lasted several hr. Approximately 10% of accumulated14C was associated with exoskeleton. As much as 50% of the accumulated B(a)P was transformed into polar compounds after one hr. Based on steady state14C concentration, an apparent bioconcentration factor of 650 was determined. The bioconcentration value based on B(a)P analysis was 200.

    Notizen: Abstract Carp (Cyprinus carpio) collected from Saginaw Bay, Michigan, containing 8.4 mg total polychlorinated biphenyls (PCBs)/kg and 194 ng of 2,3,7,8-tetrachloro-dibenzo-p-dioxin equivalents (TEQs)/kg, were substituted for marine fish at levels of 0, 10, 20, or 40% in the diets of adult ranch mink (Mustela vison). The diets, containing 0.015, 0.72, 1.53, and 2.56 mg PCBs/kg diet, or 1.03, 19.41, 40.02, and 80.76 ng TEQs/kg diet, respectively, were fed to mink prior to and throughout the reproductive period to evaluate the effects of a naturally-contaminated prey species on their survival and reproductive performance. The total quantities of PCBs ingested by the mink fed 0, 10, 20, or 40% carp over the 85-day treatment period were 0.34, 13.2, 25.3, and 32.3 mg PCBs/mink, respectively. The corresponding quantities of TEQs ingested by the mink over the same treatment period were 23, 356, 661, and 1,019 ng TEQs/mink, respectively. Consumption of feed by mink was inversely proportional to the PCB and TEQ content of the diet. The diets containing Saginaw Bay carp caused impaired reproduction and/or reduced survival of the kits. Compared to controls, body weights of kits at birth were significantly reduced in the 20 and 40% carp groups, and kit body weights and survival in the 10 and 20% carp groups were significantly reduced at three and six weeks of age. The females fed 40% carp whelped the fewest number of kits, all of which were stillborn or died within 24 hours. Lowest observable adverse effect levels (LOAEL) of 0.134 mg PCBs/kg body weight/day or 3.6 ng TEQs/kg body weight/day for adult female mink were determined. The potential effects of exposure of wild mink to contaminated Great Lakes fish were assessed by calculating “maximum allowable daily intakes” and “hazard indices” based on total concentrations of PCB residues in several species of Great Lakes fish and mink toxicity data derived from the study.

    Notizen: Abstract The hypothesis that the concentrations of 2,3,7,8-tetrachlorodibenzo-p-dioxin equivalents (TEqs) contributed by non-ortho- and mono-ortho-substituted (coplanar) PCB congeners have changed relative to total concentrations of PCBs over time was tested by analyzing eggs of red-breasted mergansers collected from the same locality in 1977–78 and in 1990. Twelve eggs from each time period were analyzed for coplanar PCBs, using a porous graphitic carbon HPLC column and GCECD. TEqs were calculated from coplanar PCB concentrations and bioassay-derived toxic equivalency factors. Median total PCB concentrations decreased from 25 to 13 μg/g, fresh weight (fw), and TEqs decreased from 320 pg/g to 160 pg/g, fw. The relative potency of the PCB mixtures, expressed as the ratio of TEqs contributed by the coplanar PCB congeners to total PCBs, did not change, nor did the ratio of concentrations of coplanar congeners to total PCBs. Thus, the relative potency of the mixture of PCBs in the eggs has not changed even as the concentrations declined significantly. The greater relative potency of TEqs relative to that which would be expected from the Aroclor® mixtures released into the environment is due to selective bioaccumulation, biomagnification, and metabolism, rather than as a result of differential weathering as a function of time.

    Notizen: Abstract Recently, there have been discussions of the relative merits of passage of fishes around hydroelectric dams on three rivers (Au Sable, Manistee, and Muskegon) in Michigan. A hazard assessment was conducted to determine the potential for adverse effects on bald eagles that could consume such fishes from above and below dams on the three primary rivers. The hazard assessments were verified by comparing the reproductive productivities of eagles nesting in areas where they ate primarily fish from either above or below dams on the three primary rivers, as well as on two additional rivers in Michigan, the Menominee and Thunder Bay. Concentrations of organochlorine insecticides (OCI), polychlorinated biphenyls (total PCBs), 2,3,7,8-tetrachlorodibenzo-p-dioxin equivalents (TCDD-EQ), and total mercury (Hg) were measured in composite samples of fishes from above and below hydroelectric dams on the Manistee and Muskegon Rivers, which flow into Lake Michigan, and the Au Sable River, which flows into Lake Huron. Mean concentrations of OCI, total PCBs, and TCDD-EQ were all greater in fishes from below the dams than in those from above. The hazard assessment indicated that current concentrations of Hg and OCI other than DDT (DDT + DDE + DDD) in fish from neither above nor below dams would present a significant hazard to bald eagles (Haliaeetus leucocephalus). Both total PCBs and TCDD-EQ in fishes from below the dams currently present a significant hazard to bald eagles, since their mean hazard quotients (HQ) were all greater than one.

    Notizen: Abstract Great Lakes colonial waterbirds have experienced poor reproduction and a greater incidence of birth defects than those in remote areas. An egg was collected from each of 1,000 marked cormorant nests at Spider Island (Lake Michigan). Nine pools comprised of three eggs were randomly selected for instrumental quantification of polychlorinated biphenyls (PCB) congeners, calculation of 2,3,7,8-tetrachlorodibenzo-p-dioxin equivalents (TEq) and measurement of equivalents by bioassay (TCDD-EQ). PCB analysis of the nine samples was semi-automated with high performance liquid chromatography (HPLC) columns including a porous graphitic carbon column. TEqs were calculated from concentrations of PCB congeners and bioassay-derived toxic equivalency factors (TEfs), and TCDD-EQ were measured directly with an H4IIE bioassay. Total PCB concentrations ranged from 9.7 to 38 μg/g, wet weight (ww). Mean concentrations of PCB 77, 126, and 169 were 2, 7, and 1 ng/g, ww. The mean TEqs and TCDD-EQ were 150 and 350 pg/g, ww, respectively. Thus, PCB congeners contributed less than 50% of the total TCDD-EQs as measured by the bioassay.

    Notizen: Abstract The effects of consumption of environmental contaminants contained in carp (Cyprinus carpio) from Saginaw Bay, Michigan on various hematological parameters and liver integrity of adult female mink (Mustela vison) were determined. Mink were fed diets that contained 0 (control), 10, 20, or 40% carp prior to and throughout the reproductive period (182 days). The diets contained 0.015, 0.72, 1.53, and 2.56 mg polychlorinated biphenyls (PCBs)/kg diet and 1.0, 19, 40, and 81 pg 2,3,7,8-tetrachlorodibenzo-p-dioxin equivalents (TEQs)/g diet, respectively. Mink fed the diets containing carp showed a general dose-dependent occurrence of clinical signs commonly associated with chlorinated hydrocarbon toxicity, including listlessness, nervousness when approached, anorexia, and melena. Erythrocyte counts were less in mink exposed to Saginaw Bay carp than in controls, while the number of white blood cells was greater than in controls. Significant differences (p〈0.05) in the concentrations of neutrophils, lymphocytes, monocytes, and eosinophils were also found between the control and carp-fed groups, but are considered to be of limited clinical or biological importance. Hematocrit values for the mink fed the 20 and 40% carp diets were significantly less than those of mink in the control and 10% carp groups. There were no significant differences in hemoglobin concentrations among the groups. Necropsies revealed enlarged yellowish livers in many of the carp-fed mink, especially those fed the 40% carp diet. Liver, spleen, and lung weights of carp-fed mink were significantly greater than those of control mink. Histopathologic examination of the livers revealed various degrees of congestion, hepatocellular fatty changes, and scattered portal lymphocytic infiltration which were most prevalent in mink fed the carp diets. These clinical signs, hematological effects, and histologic alterations are similar to those previously described for chlorinated hydrocarbon toxicoses in mink.

Concentrations of PCBs, DDTs, toxaphene, chlordanes, dieldrin, and mercury were determined in smallmouth bass (Micropterus dolomieui) collected from Fumee Lake, a remote lake in the Upper Peninsula of Michigan. An ecological hazard assessment was conducted to determine potential impacts of contaminants on bald eagles and mink eating fish from this lake. Concentrations of organochlorines, except toxaphene, and mercury in smallmouth bass were similar to those found in fish from Lake Superior, where atmospheric inputs are the primary sources. Bioaccumulation was indicated by a positive correlation between fish weight and contaminant concentrations for organochlorines, while mercury concentrations did not appear to correspond predictably to body weight. Concentrations of mercury and PCBs in smallmouth bass were sufficiently great to be of concern regarding their consumption by eagles or mink.

    Notizen: Abstract Blue mussels (Mytilus edulis) collected from 34 locations along the south and east coast of Korea were analyzed for polychlorinated biphenyls (PCBs) and organochlorine (OC) pesticides. Maximum concentrations of PCBs and total OC pesticides were 98.5 and 20.5 ng/g, wet weight, respectively. Extracts were fractionated by Florisil chromatography and each fraction was screened for dioxin-like activity in vitro, using recombinant rat hepatoma cells (H4IIE-luc). Fraction 2 (F-2), which contained hexachlorocyclohexanes, chlordanes, p,p′-DDD, and p,p′-DDT, generally elicited significant dioxin-like activity compared to control, whereas Fraction 1 (F-1), which contained PCBs, p,p′-DDE, and hexachlorobenzene, did not. The greatest magnitude of dioxin-like response observed was 44% of the maximum response elicited by a 2,000 pM 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) standard. The relatively low magnitudes of dioxin-like response observed for F-1 samples were consistent with the relatively low PCB concentrations. At concentrations equal to the maximum observed in the mussel samples, neither individual OC pesticides nor a mixture of OC pesticides yielded a significant dioxin-like response in the H4IIE-luc assay. Thus, the concentrations of OC pesticides in F-2 did not appear to have accounted for the dioxin-like activity observed. This suggests the presence of unidentified and/or unknown, acid-stable, dioxin-like compounds in F-2. This study suggests that in vitro bioassays are useful in assessing the contamination of mussels collected from coastal marine locations.
