    Notizen: Abstract A method is described for the dissociation of guinea pig lung and mesentery into their component cells. The method comprises incubation of the tissues with the enzyme pronase in Ca++- and Mg++-free saline followed by mechanical dispersion and yields on average 3–8% free mast cells. These cells are morphologically intact and viable. They retain an active sensitization or can be passively sensitized. The levels of cAMP and cGMP in the whole cell suspension respond to catecholamine, histamine or cholinergic stimulation in a way similar to that observed in the corresponding undispersed tissue. Although the separation of guinea pig mast cells from other cells proved to be more difficult than the purification of rat mast cells, an inrichment of greater than 50% has been achieved. The mast cells isolated by this method proliferate in vitro and have been cultured successfully for several months.

    Notizen: Summary Complaints of the patient by serous otitis media are considerable. Chronic serous otitis media may lead to irreversible alterations of the middle ear and even deafness. A certain number of our patients did not respond satisfactorily to conventional treatment. Further diagnosis pointed to type I sensitization. Serous otitis media was combined as well with seasonal as with perennial rhinitis. The diagnosis of these allergies — search of allergens and demonstration of pathogenetic relevance — is reported. The indications for hyposensitization and the results of this therapy are described.

    Notizen: Summary Success of hyposensitization is often described as an amelioration of the symptom score subjectively described by patients. Indeed generally accepted criteria for a successful treatment have not yet been agreed. Clear cut indications for hyposensitization, e.g. demonstration of pathogenetic relevance of allergens, and a definite proof for successful hyposensitization of allergic rhinitis are reported.

    Notizen: Summary Moulds cause infections in the upper airways. Additionally they can induce sensitization of type I and III. Sensitizations against moulds are not often diagnosed. Proof of sensitization is not easy, because several ways of invasion of allergen have to be mentioned. The tested spectrum for the diagnosis of these allergies should cover the most important allergenic moulds. The importance of moulds as cause of allergic rhinitis is reported.

    Notizen: Summary With the exception of ragweed antigen E all attempts to purify pollen allergens have been unsuccessful. Quantitations of allergens are conventionally based on parameters devoid of any relevance to allergenicity in vivo (e. g., Noon — units, w/v etc.). RABS (The Reference Allergen Bioassay Standardization) is carried out in isolated guinea pig mast cells and gives relative allergenicities compared to a standard allergen (Human serum albumin). RABS standardized rye pollen extracts were tested in the course of routine diagnostics in patients: the results of RABS standardization and the results of skin tests were found to agree very well.

    Notizen: Summary The enzymic tissue dispersion method yields single mast cells from human adenoids. These cells are light- and electronmicroscopically intact. The mast cells react to the calcium-ionophor A 23187 and to anti-human-immunglobulin E or acetylcholine after passive sensitization with histamine release. After passive sensitization with serum of patients suffering from hay feaver the specific antigen induces a histamine release. Isolated mast cells prove to be a simple system to diagnose allergies and to test antiallergic drugs in human material.

    Notizen: Summary In the literature allergens were reported to produce vestibulocochlear symptoms (review Beikert, 1960). Connections between the suspected allergens, the patients complaints and measurable symptoms are difficult to demonstrate. The modern diagnosis of allergies comprises provocation of clinical symptoms and demonstration of functional alterations — e.g. rhinomanometry, pulmonary function test. We observed occasionally in orally provocated patients vestibulocochlear symptoms. This prompted us to extend our allergy diagnosis also to vestibulocochlear disturbances.
