    Notes: Background Mast cells (MCs) arise from haematopoietic stem cells. We have recently reported that CD34+ progenitors derived from human bone marrow (BM) develop into tryptase+, chymase+ MCs when cultured in the presence of recombinant human stem cell factor (rhSCF) and recombinant human IL-6 (rhIL-6). In an experiment for the expression of chymase during differentiation, chymase+ cells were detected in human BM, but tryptase+ cells were not found.Objective The purpose of this study was to show the appearance of chymase+ cells in CD34+ cells with an origin different from MC differentiation.Methods CD34+ cells from human BM were sorted with anti-CD117 monoclonal antibody (mAb), and cytospins of CD34+, CD34+CD117+, or CD34+CD117− were prepared. These cells were cultured with rhSCF+rhIL-6 for 12 weeks. Some of the cells were subjected to either histological stain with Wright–Giemsa or immunocytochemistry with anti-chymase mAb. Real-time RT-PCR was also performed to compare the transcriptional level of chymase from each cell preparation.Results Chymase was expressed in CD34+ cells as well as human MCs by immunocytochemistry. Substantial CD34+CD117− cells, but not CD34+CD117+ cells, were stained immunocytochemically with anti-chymase mAb. For 1 week culture with rhSCF+rhIL-6, no cells expressed chymase in any preparation. Real-time RT-PCR revealed positivity for chymase mRNA in CD34+ cells, but it reduced at 1 week of culture, and increased as cells developed into MCs. Chymase mRNA in CD34+CD117+ cells was negligible compared with that in CD34+CD117−. Tryptase mRNA was below the detectable level in CD34+ cells, and increased along with MC differentiation. After 12 weeks of culture, CD34+CD117+ developed predominantly into MCs, whereas CD34+CD117− developed into monocytes/macrophages.Conclusion Our findings suggested that chymase is present not only in MCs but also in CD34+CD117− BM progenitors, but that its origin is different from the MC lineage.

    Notes: Background CD34+ progenitor cells develop into tryptase+, CD117+ mast cells when cultured in the presence of recombinant human stem cell factor (rhSCF). However, spontaneous IgE receptor (FcɛRI) expression during human mast cell development is not well examined.Objective Here, the expression and function of FcɛRI in and on human bone marrow-derived mast cells (HBMMCs) during development were investigated.Methods and Results At 4 weeks of culture, predominant cells expressed high-affinity IgE receptor α chain (FcɛRIα) on the cell surface determined by flow cytometry, but CD117 was less expressed. Immunocytochemistry with antitryptase mAb and anti-FcɛRIα mAb revealed intracellular and surface expression of FcɛRIα at 2 weeks of culture, but tryptase was less expressed. FcɛRIα mRNA transcript preceded that of tryptase mRNA at 2 weeks of culture determined by real-time RT-PCR, and FcɛRIα, FcɛRIβ, FcɛRIγ, and tryptase mRNA increased along with differentiation. FcɛRIα cross-link on HBMMC and 4-week-old mast cells/mast cell precursors induced the release of IL-5 and granulocyte macrophage-colony stimulating factor, which was enhanced by rhSCF.Conclusion These data indicated that HBMMC constitutively and spontaneously expressed functional FcɛRI subunits at the early stage of differentiation, probably because of the differences in the ability and functional property of progenitors.

    Notes: summary  We report here on a patient with bilateral cleft lip and palate (BCLP) and a missing premaxilla, who underwent dentoalveolar reconstruction of the cleft and premaxillary alveolus using endosteal implants after bone grafting. The patient, whose maxillary incisors and premaxilla were missing, had corticocancellous bone grafting from the iliac crest, followed by excellent bone formation at the anterior alveolus. After the placement of the endosteal implants and the completion of the pre-surgical orthodontic alignment, orthognathic surgery was performed for the restoration of a Class III open bite. After post-operative orthodontic preparation, the final fixed prostheses were completed. This treatment procedure offers an option of dentoalveolar reconstruction for BCLP patients with an excised premaxilla.

    Notes: We describe the use of endosseous implants in the autogenous particulate cancellous bone and marrow grafted alveoli after orthognathic surgery for dental rehabilitation of patients with cleft lip and palate. This procedure has been applied to two patients and produced good results functionally and aesthetically. The results are encouraging and indicate that implant placement after orthognathic surgery is useful for patients both with congenital missing teeth and retrognathic maxillae.

    Notes: An n=1 mode global motion on a field-reversed configuration (FRC) plasma is observed by means of a newly developed optical system. The deviation of the FRC from the coil axis reaches 20%–40% of the plasma radius. In order to push back the FRC to the equilibrium position, a multipole field (quadrupole or hexapole field) is applied. The n=1 motion can be easily controlled by the quadrupole field, the critical field strength of which is required to be about 15% of the confinement field. It is found that the n=2 rotational instability can also be stabilized by strength of the same order. The critical strength for the n=1 motion is theoretically obtained from a model such that the driving energy of the motion given at the formation phase balances with the work done by the multipole field. The theoretical estimation agrees within a factor of 2 with the experimental results. © 2002 American Institute of Physics.

    Notes: We applied photoacoustic (PA), photoluminescence (PL), photoluminescence excitation (PLE), and atomic force microscopy (AFM) techniques on porous silicon (PS) layers to study the influence of chemical etching by low-concentration hydrofluoric acid. The chemical etching reveals the formation of PS layers of small dimensions by AFM observations, indicating the possibility of a strong quantum confinement effect. PA spectroscopy is useful to obtain the optical absorption characteristic for strongly scattering media such as PS and it helps to confirm the above speculation by indicating the blueshift of the fundamental absorption edge for the PS layer with chemical etching. PL spectroscopy also confirms the possibility of a quantum confinement effect by revealing the strong intensity and blueshift for the PS layer with chemical etching. PLE measurements suggest that the site for the radiative processes is different from that for the recombination of carriers and the PL of PS layers were dominated only by small crystallites in various size distributions. © 2000 American Institute of Physics.

    Notes: Transconductance oscillations were observed for silicon-on-insulator metal–oxide–semiconductor field-effect transistors with 50 nm channel length and 6 nm Si-layer thickness in the temperature range of 39–50 K. By investigating the temperature dependence of the oscillations it was found that the oscillations were caused by two reasons. One reason is the roughness at the Si/insulator interface responsible for the low-gate-voltage oscillations. The roughness results in different thicknesses of the Si layer along the channel, causing different quantized energy levels, which act as barriers for carriers moving in the channel. The other reason is the tunneling through the potential barrier at the p/n junctions between the contacts and the channel, which is responsible for the high-gate-voltage oscillations. © 2002 American Institute of Physics.

    Notes: Capacitance images responsible for surface depletion were observed on an InAs dot-covered GaAs surface by scanning capacitance microscopy. We performed local capacitance versus bias voltage measurements on quantum dots (QDs) and a wetting layer (WL) as well as conductance versus bias voltage (G–V) measurements. Both results indicate that the surface depletion is more suppressed beneath the QDs than under the WL. In addition, the conventional thermionic equation theory fitted to the measured G–V curves shows that the interface barrier height between the GaAs and the InAs QD increases as the QD size is reduced. We ascribe this result to the influence of the surrounding WL, whose surface Fermi level is strongly pinned at the midgap of the n-GaAs. © 2000 American Institute of Physics.

    Notes: This study investigated the effects of novelty stress on neuroendocrine activities and running performance in Thoroughbred horses. First, to examine the neuroendocrine responses to novelty stress, we exposed horses to two types of novel environmental stimuli (audiovisual or novel field stimuli). After the stimuli, plasma concentrations of vasopressin, catecholamines and adrenocorticotropin (ACTH), as well as heart rates, were significantly increased in each experiment. Second, we investigated neuroendocrine activities during incremental exercise. Plasma concentrations of vasopressin, catecholamines, ACTH and blood lactate increased as the exercise load increased. Finally, we investigated the effects of novelty stimuli on neuroendocrine activities and running performance during supra-maximal exercise (110% VHRmax). When the novelty stimuli were presented to horses, the increases in plasma vasopressin and catecholamines due to exercise load were significantly smaller than those in the control experiments. Blood lactate during supra-maximal exercise was also significantly lower and total run time until exhaustion was prolonged in the novel environmental stimuli compared to the control. These results suggest that novelty stimuli facilitate vasopressin release from the posterior pituitary in addition to activating the sympatho-adrenomedullary and the hypothalamic-pituitary-adrenocortical axes in thoroughbred horses, and increase exercise capacity, resulting in improvement of running performance during supra-maximal exercise.
