Notes: Summary The effect of dibutyryladenosine 3′,5′-cyclic monophosphate (dibutyryl cAMP) on the expression of glutathione S-transferase placental type (GST-P) was examined in rat glioma cell line using an immunohistochemical technique. Cultured T9 glioma cells were negative for GST-P activity under normal conditions. However, treatment with 1 mM dibutyryl cAMP produced GST-P expression in about 50% of the cells, as well as some morphological changes. The expression of GST-P was increased with addition of dibutyryl cAMP together with 1 μg/ml allyl isothiocyanate (AITC) or 0.1 μg/ml benzyl isothiocyanate (BITC). With these combinated treatments, almost all cultured cells showed a strong positive reaction for GST-P, although AITC or BITC alone elicited GST-P in only 5% of the cultured cells. The results of the present study indicate that dibutyryl cAMP causes functional as well as morphological differentiation of T9 glioma cells.

Notes: Summary Lewy body-like hyaline inclusion (LI) in the neuronal soma and swollen cord-like processes is a characteristic feature in the anterior horn cells and neurons in thoracic nucleus (Clarke) of familial amyotrophic lateral sclerosis (ALS) with posterior column involvement. We have studied the LI in the case of two sisters with this disorders. Microscopically the LI consists of an eosinophilic “core” surrounded by a basophilic “halo”. Ultrastructurally the core consists of granule-associated filaments, while the halo consists of normal-looking neurofilament. Immunocytochemistry with anti-ubiquitin antibody shows that these granule-associated filaments in the core are highly ubiquitinated, while the normal-looking neurofilaments in the halo are not recognized by antiubiquitin antibody. Our study proves that LI consists of an aggregation of ubiquitinated filaments among a neurofilamentous accumulation, possibly representing a form of neuronal cytoskeletal disorganization in familial ALS.

Notes: Summary To clarify the structure of Hirano bodies, electron micrographs of Hirano bodies taken at various tilting angles have been studied by digital image analysis. On the electron micrographs, the beaded filaments of Hirano bodies were turned into a pattern of lattice-like arrays by changing the tilting angles. Based on computer-procesed diffraction patterns and filtered images, it is proposed that the filaments of Hirano bodies are helical strands with a pitch of 185 Å. A model for the helical strand drawn by microcomputer at various angles of rotation is in accordance with the filtered images of the tilted filaments. Computer simulation also reveals that the helical strands appear to be lattice-like when they are arranged in parallel.

Notes: Summary The accumulation of amyloid in pituitary adenomas was examined in relation to the types of adenoma and the effect of bromocriptine treatment. Amyloid had accumulated in 34 of 48 adenomas (71%). The occurrence in prolactin-secreting adenomas and growth hormone-secreting adenomas was 79%, respectively, while that in non-functioning adenomas was 50%. Treatment with bromocriptine enhanced the occurrence and extent of the amyloid accumulation in prolactin- or growth hormone-secreting adenomas. Electron microscopy revealed the initial appearance of the amyloid fibrils in the smooth endoplasmic reticulum and a possible sequential process of their release from the cells. The presence of secretory granules in vesicles containing amyloid fibrils and their simultaneous release with amyloid fibrils suggested that degradation of secretory granules was involved in the formation of amyloid.

Notes: Summary The lectin-binding patterns of the cells involved in amelogenesis and dentinogenesis in developing teeth of rats, were studied. Undifferentiated odontogenic epithelia exhibited very slight staining with almost all of the lectins examined. The lectin-staining affinities of secretory ameloblasts could be divided into two categories: Concanavalin-A (Con-A), Wheat germ agglutinin (WGA) and Soybean agglutinin (SBA) binding occurred from the middle to apical cytoplasm, whereas Ricinus communis agglutinin-I (RCA-I) and Ulex europeus I (UEA-I) binding predominated in the basal regions. The cells of the stratum intermedium exhibited relatively stranges lectin staining, which appeared to be dependent on ameloblastic maturation. The basement membranes in undifferentiated epithelia were markedly positive for lectin binding. Odontoblasts showed moderate Con-A staining on the apical side of the cells, as well as slight-to-moderate reactions with WGA and SBA. Pulp cells and dental papillae showed slight-to-moderate lectin staining, and predentin and dentin were also moderately positive for Con-A and RCA-I binding and slightly so for WGA and SBA. The lectin-binding affinities were enhanced during the formation of enamel and dentin, and appeared to be dependent on the degree of cellular differentiation in ameloblasts and odontoblasts.

Notes: Summary We examined rat cells undergoing amelogenesis for the presence of three types of keratin proteins using a polyclonal antibody to keratin (against total keratins (TK) with molecular masses ranging from 41 to 65 kilodaltons (kd) and monoclonal antibodies keratins to KL1 and PKK1 (reactive with keratins with molecular masses of 55–57 and 41–56 kd, respectively). In normal oral epithelia from young rats, the TK, KL1, and PKK1 antibodies bound to all of the epithelial strata. The epithelial cap on the top of incisors and the dental lamina of molar teeth exhibited strong TK staining, moderate staining KL1, and little or no PKK1 staining. In developing molar enamel organs, both the outer and inner enamel epithelia, the stratum intermedium, and stellate reticulum cells were all positively stained by the TK immunoreagent. In developing incisors, TK only bound strongly to stratum-intermedium cells, and no KL1 and PKK1 staining antibodies was observed in ameloblasts or the stratum intermedium.

Notes: Abstract Electrochromic properties of methyl, ethyl, n-hexyl and n-heptyl viologens, and α-, β-, γ-cyclodextrin systems were studied using cyclic voltammetry and absorption spectroscopy. Colourations of red-violet (λ max = 530 or 560 nm), blue (λ max = 600nm) and light purple (λ max = 560 and 600 nm) could be obtained by changing the length of the alkyl chain of viologens or the radius of cyclodextrin. The intermolecular interaction between viologen radicals is weakened when the radicals form inclusion complexes so the blue colour inherent in the monomer of viologen radical appears. The red-violet colour is ascribed to the dimer of the reduced viologen. With the addition of γ-cyclodextrin, viologens were found to be included as dimer. The light purple colour is ascribed to the mixed colour of the monomer and the dimer. Furthermore, since the ratio of monomer and dimer depends upon the concentration of cyclodextrin, any colour between the red-violet of the dimer and the blue of the monomer can be obtained by adjusting the concentration of cyclodextrin. The redox reaction of an inclusion complex was found to depend upon the length of the alkyl viologen and the size of the cavity in cyclodextrin.

Notes: Summary Fifty cases of facial palsy (FP) due to otitis media treated in our hospital during the 10-year period from 1972 to 1981 were analyzed. The averaged percentage of this incidence was 3.1% of all the FP occurring. FP due to otitis media acuta (OMA) was frequently seen in infants. FP due to otitis media chronica (OMC) tended to occur in combination with cholesteatoma or acute exacerbations of infection. Paralyses were generally mild and bacteria from the otorrhea were similar to those of OMC without FP. When present, sites of dehiscence of the facial canal were most frequently seen in its horizontal portion. Electromyographically, the posterior (mastoid) side of the horizontal portion of the facial nerve bundle seemed to be related to the branch to the orbicularis oculi muscle, while the anterior (stapes) side was directed to the orbicularis oris muscle.

Notes: Abstract Resistivity, specific heat, and susceptibility studies on CeGe2 and its pseudobinary system Ce y La1−y Ge2 are reported. The effective moment and the occurrence of ferromagnetic order (0.4≤y≤1.0) indicate the trivalence of Ce in the entirey range. However, the present results suggest that a Kondo effect coexists with the interionic ferromagnetic interaction; the magnetic entropy at the ordering temperatureT c is significantly lower thanR ln 2 (R is the gas constant) and the resistivity of La-diluted samples (y≤0.6) exhibits a Kondo-like minimum at about 10 K. The magnetic entropy atT c is related to the ratioT K/T c by a simple thermodynamic argument, whereT K is the Kondo temperature. TheT K is estimated to be about 2.7 K for 0.4≤y≤1.0, in fair agreement with what the resistivity anomaly suggests.
