    Notes: The angular distribution of lead in films deposited by pulsed laser irradiation of lead–zirconate–titanate and lead targets are studied as a function of ambient gas (argon or oxygen), gas pressure, and substrate temperature. When the substrate is kept in vacuum and at room temperature, a dip in the lead content attributable to the intrinsic resputtering of lead is observed at the position of the target surface normal. In the presence of an ambient gas, the dip disappears and the lead content increases at all angles. These results are attributed to a reduction of resputtering arising from scattering of the ablated species by ambient gas molecules. Under ambient oxygen and at high substrate temperature, the retention of lead content in the deposited films is largely due to the formation of lead oxide. © 1996 American Institute of Physics.

    Notes: Cutaneous melanoma is rare among Chinese people. The clinicopathologic features and prognostic factors of 63 Hong Kong ethnic Chinese cases were analysed. We found that the mean age of onset was 65.5 years with a female : male ratio of 1 : 1.33. Forty-two (66.7%) patients had their tumours on their feet. More than 50% had the acral lentiginous histologic subtype. The mean tumour thickness was 5.73 mm with 92.1% (n = 58) having a Clark's level of III or more. Of the tumours, 57.1% were ulcerated. Stage I and II tumours comprised 73% (n = 46) while stage III and IV tumours made up of 26% (n = 17). The 1-, 3- and 5-year survival rates were 75%, 30% and 17%, respectively. Univariate analysis for overall survival showed that advanced clinical staging (stage III and IV), tumour ulceration and thicker tumour (〉4 mm) had a significantly poorer prognosis. Multivariate analysis demonstrated that advanced clinical staging was the most decisive prognostic factor followed by tumour ulceration. Our study showed that cutaneous melanoma in Chinese people is a disease predominantly of an older age group with the acral lentiginous histologic type located mainly on the feet. The prognosis depends on clinical staging and ulceration status.

    Notes: Abstract: The increased frequency and severity of candidal infections in human immunodeficiency virus (HIV)-infected individuals has prompted the wide use of antifungals, such as amphotericin B, ketoconazole, and fluconazole, resulting in the emergence of drug-resistant strains of Candida albicans. To study this phenomenon in an ethnic Chinese cohort, we isolated multiple colonies of Candida from the oral cavities of 16 HIV-infected patients on single and subsequent sequential visits over a period of 12 months. Ten of the 16 patients had sporadic episodes of oropharyngeal candidiasis (Group A), while the remainder were asymptomatic with respect to this condition (Group B). Oral rinses were collected and immediately processed in the laboratory for the isolation of C. albicans in a standard manner. A total of 433 C. albicans isolates were tested for their susceptibility to amphotericin B, ketoconazole and fluconazole by an agar diffusion method using the commercially available E-test. All tested isolates demonstrated variable susceptibility to amphotericin B, ketoconazole and fluconazole. The minimum inhibitory concentration (MIC) of the isolates for amphotericin B, ketoconazole and fluconazole ranged from 〈0.002–1.5 μg/ml, 〈0.002–4.0 μg/ml and 〈0.016–32 μg/ml, respectively. Sequential isolates of a few patients demonstrated variable susceptibility to all the antifungals, and no discernible MIC pattern emerged either in group A or B over time. Interestingly, significant variation in antifungal susceptibility was also noted in isolates obtained from the same patient on a single visit. Sequential yeast isolates in 9 of 16 patients (56%) demonstrated significant differences in MIC within and between visits for both amphotericin B and ketoconazole, while a lower percentage – 44% (7/16) – exhibited this trait for fluconazole. Our study demonstrates the diversity in antifungal susceptibility in either commensal or “infective” oral strains of C. albicans in HIV disease, and shows the need for vigilance for the emergence of resistant strains, and for frequent antifungal susceptibility studies.

    Notes: Summary Time resolved fluorescence technique was used to demonstrate physically the ionpair aggregation phenomena in the ionomers containing fluorenylsodium as the pendant group along the polymer chains in THF at 25°C. It was found that ionpairs in the ionomer with mole fraction of fluorenylsodium equal to 0.02 did not associate to form stable aggregate. However, the ionpairs would aggregate in those ionomers with higher fluorenylsodium contents. In the homopolymer of vinylfluorenylsodium, extensive aggregation of the ionpairs was observed.

    Notes: Abstract In the present studies, the pharmacology and regulation of the functional muscarinic receptors on HSDM1C1 cells were probed using phosphoinositide (PI) turnover assays. In addition, the receptor binding of the putative M3-selective radioligand, [3H]4-DAMP, to cell homogenates was characterized. Carbachol (EC50=9 μM), (+)muscarine (EC50=4.5 μM) and cis-dioxolane (EC5=0.72 μM) were full agonists which stimulated PI turnover by 13.3±1.0 fold above basal values. The potencies of numerous agonists in this assay system were relatively similar to their affinities in receptor binding assays. Exposure of HSDM1C1 cells to 10 nM–10 μM muscarine during the last 24h of [3H]myo-inositol-labeling resulted in a concentration-dependent reduction in the cisdioxolane affinity and maximal PI response induced by subsequent treatment with cis-dioxolane. pertussis toxin (5–2000 ng/ml) caused a partial reduction in the cis-dioxolane-induced PI turnover. Likewise, exposure of the HSDM1C1 cells to an active phorbol ester (TPA) resulted in a partial inhibition of the cis-dioxolane-induced (100 μM) PI turnover. The half-maximal effect of TPA was produced at 1.8±0.3 nM. [3H]4-DAMP binding to cell homogenates was of high affinity (Kd=0.19±0.04 nM) and moderate capacity (Bmax=201±22 fmol/mg protein). The pharmacological specificity (4-DAMP〉p-FHHSiD〉dicyclomine〉pirenzepine〉methoctramine〉AFDX-116 〉gallamine) resembled that for [3H]NMS binding and correlated well with that observed for inhibition of PI turnover. These studies further support the identification of M3 receptors on HSDM1C1 cells. These receptors have been shown to be influenced by pertussis toxin, an active phorbol ester and to exhibit desensitization.
