    Notes: Summary We studied thrombin-induced metabolism of phosphoinositide, protein phosphorylation and platelet aggregation in platelets from 32 NIDDM patients and 12 control subjects. To clarify the effect of diet, sulphonylureas, or insulin treatment, the subjects were divided into three groups based on the type of treatment. Thrombin-induced platelet aggregation was measured with an aggregometer. Low-dose thrombin (0.25 U/ml)-stimulated platelet aggregation in diabetic patients was significantly increased compared with the control subjects. Platelet aggregation in the sulphonylurea and insulin groups was significantly lower than in the diet group. On the other hand, in platelets incubated with [32P]orthophosphate, thrombin-induced incorporation of 32P radioactivity into phosphatidic acid (PA) was significantly lower in the sulphonylurea and insulin groups than in the diet group. Thrombin-induced incorporation of [32P] radioactivity into phosphatidylinositol (PIP) for 10 s was significantly higher in the sulphonylurea group than in the diet group. There were no differences in thrombin-induced 47 kDa protein phosphorylation between platelets from the diet, sulphonylurea, or insulin groups. These results suggest that sulphonylureas and insulin induce suppression of thrombin-induced activation of phospholipase C, which mediates hydrolysis of PIP and PIP2 and production of PA, which leads to inhibition of platelet aggregation.     Notes: Summary The relationship between diabetic nephropathy and an insertion (I)/deletion (D) polymorphism in intron 16 of the angiotensin-converting enzyme (ACE) gene is still under debate. The association of ACE gene polymorphism with nephropathy and retinopathy was therefore examined in 362 Japanese patients with non-insulin-dependent diabetes mellitus (NIDDM) and 105 healthy control subjects. Distribution of the ACE genotype did not differ between healthy control subjects and diabetic patients without complications. However, the frequency of the D allele was significantly higher in the diabetic subjects with nephropathy than in those without (0.32 in normoalbuminuric patients vs 0.44 in albuminuria patients with albuminuria) (χ2=7.7; p=0.006). There was no significant association between ACE genotype and retinopathy. These observations thus demonstrate a significant association of the ACE gene polymorphism with nephropathy, but not with retinopathy, in Japanese patients with NIDDM.

    Notes: Summary While non-enzymatic glycation of long-lived tissue proteins such as collagen has been implicated in chronic complications of diabetes mellitus, its role in the aetiology of diabetic macroangiopathy has not been elucidated. To test the hypothesis that glycation of collagen abolishes the inhibitory effect of native collagen on the proliferation of human smooth muscle cells, we obtained smooth muscle cells from human gastric arteries and cultured them on dishes coated with glycated or non-glycated collagen. The proliferation of human smooth muscle cells in the presence of 10 % fetal calf serum or platelet derived growth factor-BB (10 ng/ml) was inhibited by type 1 collagen coated on the dishes. Glycation of collagen with glucose 6-phosphate for 7 days abolished the growth-inhibitory effect of native collagen. Succinylation of collagen, which like glycation blocked the lysyl residues in collagen, also abolished the growth-inhibitory effect. Adhesion of human smooth muscle cells to collagen-coated dishes was not affected by glycation of collagen. Addition of glycated albumin to the medium did not affect the growth of human smooth muscle cells on plastic dishes. The inhibition of human smooth muscle cell proliferation by collagen was not reversed by the glycation of collagen in the presence of aminoguanidine. Results suggest that early glycation abolishes the inhibitory effect of collagen on human smooth muscle cell proliferation and may thus participate in the progression of macroangiopathy in diabetes.     Description / Table of Contents: Summary The case has been presented of a Japanese aged 22 who was suffering from hereditary non-spherocytic haemolytic anaemia. This was a constant type of anaemia associated with reticulocytosis, jaundice, and glucose-6-phosphate dehydrogenase deficiency of the erythrocytes. Although it was not possible, owing to the absence of enzymic activity, to estimate isoenzyme in the patient's erythrocytes, the isoenzyme was assigned, in the patient's mother and in other members of his family, as well as in controls, to the B type. Cytochemical investigations revealed the presence of a cellular mosaic in the patient's mother who was heterozygous, but in the patient, as well, who himself was hemizygous.
    Notes: Zusammenfassung Es wird über einen 22jährigen Japaner mit einer hereditären, nichtsphärozytären hämolytischen Anämie berichtet. Es bestand eine konstante Anämie mit Retikulozytose, Ikterus und Mangel an Glucose-6-Phosphat-Dehydrogenase in den Erythrozyten. Obwohl der Nachweis von Isoenzym an den Erythrozyten des Patienten wegen fehlender Fermentaktivität nicht gelang, konnte es bei der Mutter, bei den übrigen Familienangehörigen und bei Kontrollfällen dem Typ B zugeordnet werden. Cytochemische Untersuchungen ergaben das Vorliegen eines Zellmosaiks in der heterozygoten Mutter des Patienten, jedoch auch im hemizygoten Patienten selber.

    Notes: Summary Histochemical investigations were carried out on skeletal muscle biopsies from ten patients with chronic progressive external ophthalmoplegia with ragged-red fibers (RRF). In addition to the RRF, mild myopathic change consisting of variation in size of both type 1 and 2 fibers was seen in all patients, as well as neuropathic change in eight. Scattered fibers with absent cytochromec oxidase (CCO) activity (focal deficiency) were seen in all patients. In serial sections, CCO deficiency did not always occupy the entire length of a fiber but was localized segmentally to regions measuring several hundred micrometers in lenghth, suggesting the heterogeneity of CCO activity even in the same fiber.

    Notes: Summary A 43-year-old woman who had four recurrences of acute transverse myelitis (ATM) at the fifth thoracic vertebral level in 7 years is reported. Her haematological and immunological abnormalities included antibodies to DNA and nRNP and markedly reduced serum complement during the course of the disease. These findings suggest systemic lupus erythematosus although she had no symptoms other than those of ATM. Methylprednisolone and prednisolone were effective in treating the ATM.

    Notes: Abstract Fractionation of sulfur isotopes and selenium was measured between coexisting pyrite and chalcopyrite and between coexisting pyrrhotite and chalcopyrite from the Besshi deposit of Kieslager-type, Central Shikoku, Japan. In all the pyrite-chalcopyrite pairs studied, 34S is enriched in pyrite relative to chalcopyrite, while selenium is enriched conversely in chalcopyrite relative to pyrite. The mean △34Spy-cp value is +0.53±0.36 per mil, and the mean value of the distribution coefficient of selenium, Dcp-py, is 2.58±0.64. In all the pyrrhotite-chalcopyrite pairs studied, the two minerals are very close to each other both in sulfur isotope and Se/S ratios. The mean △34Spo-cp value is −0.08±0.16 per mil and the mean Dcp-po value is 0.99±0.05. The results have been discussed in comparison with similar data obtained for the Hitachi deposits of Kieslager-type, Japan (Yamamoto et al. 1983).

    Notes: Abstract  In non-destructive dimensional measurement of high aspect ratio micro structures (HARMS), optical methods cannot offer full three dimensional information due to the lack of observation light. Again, conventional mechanical measurement, such as a surface profiler or a coordinate measurement machine, cannot be applied because their stylus is too large. Furthermore, the AFM, though popular among the semiconductor industry, is also limited in terms of dimensional measurement, because its system is usually designed for planar samples. Thus, we have developed a new sensor-integrated micro resonating cantilever probe and a new dimensional measurement machine, which allows the probe's vertical access to microstructures in a sample. The new probe is made of tungsten carbide super hard alloy and possesses design flexibility according to its intended application. Validity of the system is confirmed through the measurement experiment of EDM drilled and chemically etched micro holes.

    Notes: Abstract The pH and fO 2 dependences of the [Se2−]/[S2−] ratio in chloride solutions at 100°, 200° and 300°C are predicted thermodynamically. Under the high fO 2 conditions where sulfate species are dominant in solution, the [Se2−]/[S2−] ratio always increases with increasing pH and/or fO 2. Under the low fO 2 conditions where sulfide species are dominant in solution, the pH and fO 2 dependences of the [Se2−]/[S2−] ratio are seriously affected by the presence of native selenium. With native selenium present, the [Se2−]/[S2−] ratio decreases with increasing fO 2, but almost independent of pH in geologically important pH regions. When native selenium is absent, the [Se2−]/[S2−] ratio is solely a function of pH and independent of fO 2. Combining the above with the pH and fO 2 dependences of δ34S value of aqueous sulfur species, we discuss the possible influences of the pH and fO 2 of ore-forming solutions on the relationship between the Se/S ratio and δ34S value of hydrothermal sulfide minerals. The results are applied to some Japanese sulfide ore deposits.
