    Notes: Abstract This paper deals with the computational methods for limit analysis of plane strain problems. The finite element mathematical programming formula (FE-MPF) for determining the upper bound load multiplier established by the authors earlier is adopted and modified for plane strain problems. The penalty method is used to impose the incompressibility constraint. The FE-MPF is solved by a direct iteration procedure without the need of a searching process. This algorithm is not sensitive to the volumetric locking effect. And it can be easily extended to the limit analysis of three dimensional problems. The results of numerical examples are satisfactory and show the stable convergency of the present algorithm.

    Notes: Undecaprenyl diphosphate synthase from Micrococcus luteus B-P 26, one of the Z-prenyl chain-elongating enzymes, was crystallized using the sitting-drop vapour-diffusion method with ammonium sulfate and lithium sulfate as precipitants. The crystals belong to the monoclinic space group C2, with unit-cell parameters a = 127.2, b = 60.2, c = 75.7 Å, β = 105.6°. The crystals diffract X-rays to at least 2.2 Å resolution using synchrotron radiation and are suitable for high-resolution crystal structure analysis.

    Notes: Background and Aim:  Tubular atrophy is a major feature of most renal diseases and is closely associated with the loss of renal function. The present study sought to investigate whether Fas/FasL-induced tubular epithelial cell apoptosis was a feature of experimental diabetic nephropathy. The effects of renoprotective therapy with blockade of the renin-angiotensin (RAS) system were also examined.Method:  Six-week-old female Ren-2 rats were injected with streptozotocin and maintained diabetic for 12 weeks. Further groups of diabetic rats were treated with the angiotensin-converting enzyme inhibitor, perindopril, for 12 weeks.Results:  Widespread apoptosis, identified by using mediated Terminal dUTP nick-end labelling (TUNEL) staining was noted in the tubules of diabetic Ren-2 rats. These changes were associated with an increase in both Fas mRNA and Fas L (ligand) within the tubules (P 〈 0.01). Treatment of diabetic Ren-2 rats with perindopril (6 mg/kg per day) reduced the apoptosis to control levels and was associated with a reduction in Fas mRNA and Fas L protein (P 〈 0.05).Conclusion:  In conclusion, Fas/Fas L-induced tubular apoptosis is a feature of diabetic Ren-2 rats and is attenuated by the blockade of the RAS.

    Notes: This study was designed to provide evidence for elucidating the mechanisms of neurokinin-3 receptor (NK3) in spinal pain modulation. First, colocalization of NK3 with the µ-opioid receptor (MOR1) was studied in the spinal dorsal horn of the rat. Confocal microscopy showed that about 44% of NK3-expressing neurons in laminae I and II were immunoreactive for MOR1, which corresponded to about 93% of the total population of MOR1-containing neurons in these laminae. Second, the relationship between NK3/MOR1-coexpressing neurons and those that express nitric oxide synthase (NOS) was examined by using a triple immunofluorescent staining method. About 37% of NK3-immunoreactive neurons were also NOS-immunoreactive, which constituted about 82% of NOS-immunoreacitve neurons in the superficial laminae. However, no triple-labelled neurons were detected. The present results indicate that there are two major distinct subpopulations of NK3-expressing neurons in the superficial dorsal horn, which suggests that the involvement of NK3 receptor in spinal nociception could be mediated by two distinct mechanisms, i.e. opioid and nitric oxide.

    Notes: Diode-laser isotope enrichment of rubidium by magnetic deflection of a polarized atomic beam has been experimentally studied with laser-induced fluorescence detection. The abundance of 85Rb was enhanced from the natural value of 72.8% to 97% and that of 87Rb, from 27.2% to 83%. The results were compared with those with hot-wire detection.

    Notes: Abstract Purpose. We have demonstrated that a heteromolecular complex consisting of cationic lipids and DNA can be prepared and isolated (1). Cationic lipids bind DNA through electrostatic interactions. However, when sufficient lipids are bound to DNA the physical and chemical properties of the complex are governed by hydrophobic effects. Here we describe an approach where this hydrophobic complex is used as an intermediate in the preparation of lipid-DNA particles (LDPs). Methods. The approach relies on the generation of mixed micelles containing the detergent, n-octyl β-D-glucopyranoside (OGP), the cationic lipid, N-N-dioleoyl-N, N-dimethylammonium chloride (DODAC), and selected zwitterionic lipids, 1,2-dioleoyl-sn-glycero-3 -phosphoethanolamine (DOPE) or egg sphingomyelin (SM). Results. When these micelles were prepared at low detergent concentrations (20 mM OGP) and combined with pCMVβ DNA, LDPs spontaneously formed. The mean diameter of these particles as measured by quasielastic light scattering was 55−70 nm, a result that was confirmed by negative stain electron microscopy. Further characterization of these LDPs showed that DNA within the particles was inaccessible to the small fluorochrome TO-PRO-1 and protected against DNase I degradation. LDPs could also be prepared in high concentrations of OGP (100 mM), however particles formed only after removal of OGP by dialysis. Particles formed in this manner were large (〉2000nm) and mediated efficient transfection of Chinese hamster ovary cells. Transfection activity was greater when the lipid composition used consisted of SM/ DODAC. Small particles (〈100nm) prepared of SM/DODAC were, however, inefficient transfecting agents. Conclusions. We believe that LDP formation is a consequence of the molecular forces that promote optimal hydrocarbon-hydrocarbon interactions and elimination of the hydrocarbon-water interface.

    Notes: Abstract In the brain, the γ-aminobutyric acid (GABA) level is primarily controlled by the activity of its synthesizing enzyme,L-glutamate decarboxylase (GAD). At present, mechanisms responsible for regulation of GAD activity remain largely unknown. Here we report that GAD activity is inhibited by conditions favoring protein phosphorylation, and this inhibition can be reversed by phosphatase treatment. Furthermore, this inhibition appears to result from the suppression of a Ca2+-dependent phosphatase. Phosphorylation of GAD is demonstrated by direct incorporation of32P into the GAD protein. These results suggest that GAD activity in the brain is inhibited by phosphorylation and activated by dephosphorylation. A model for regulation of GABA synthesis related to neuronal excitation is discussed.

    Notes: For discussing the mechanism of load-transferring for reinforced steel fiber reinforced concrete (SFRC) double-column combined six-pile caps, the large-scale general finite element software-ABAQUS is used for the tested SFRC caps with computing in the aspects of modeling, cracking load, limit load, load-deformation curve, distribution of stress in caps and cracks and etc. The results of computing is compared with the result of experiments and the comparison is indicated: The results of computing and experiments are coincided well; The destroy pattern of SFRC double-column combined six-pile caps is sheared damage or punched damage and the model of load-transferring accords with spatial strut-and-tie method (STM); The mixture of steel fiber can improve cracking load and limit load for RC caps, delay the crack for caps, block the cracks’ expansion, and enhance the cap’s ductility

    Notes: The interface bonding between the carbides and the solid solution matrix is studied insurfacing welding metal of Fe-Nb-Ti-V-C alloy system. Both the quenched welding droplets and thedeposited metal are investigated by means of SEM and TEM. In the deposited metal, the carbidesare dispersed, and the matrix is low carbon martensite. According to welding metallurgicalcharacteristics, it is deduced that the carbides are formed during the droplets stage and then transmitinto the welding pool. When the welding pool solidify, the carbides serve as nucleation site, so theinterface between the carbides and the matrix is clean and free from other reactants. The selectedarea electron diffraction (SAED) analysis reveals that the mismatch between the two phase is δ=(6.1~8.3)%, and there exist a crystal parallel relationship at the interface: (002)α-Fe // (220)(NbTiV)C.The metallurgical bonding between the carbides and the matrix favors to prevent the carbides todetach from the matrix during wear process, so the surfacing welding metal can achieve goodwear-resistance
