    Notizen: Abstract: Little is known about the histogenesis of the human odontogenic myxoma or the relation between tumour cells and the matrix. In order to attempt to remedy this situation, we established and investigated a cell line derived from a human odontogenic myxoma. To our knowledge this is the first cell line derived from this tumour. The cell line, named Mix 1, preserved features of the tumour cells. Mix 1 cells expressed vimentin, type I collagen, fibronectin, tenascin and hyaluronic acid. Ultrastructural analysis of cells of the tumour and cell line demonstrated similarities, both containing Golgi apparatus, rough endoplasmic reticulum and mitochondria indicative of secretory cells. Ultrastructural analysis showed the matrix to be represented by bundles of collagen fibrils in the tumour, and by irregular filaments in cultures more than 60 days old. The Mix 1 cell line promises to be an excellent model for investigating the biology of the odontogenic myxoma.

Evaluation of the prevalence of skin prick test positivity to Alternaria and Cladosporium in patients with suspected respiratory allergy. A European multicenter study promoted by the Subcommittee on Aerobiology and Environmental Aspects of Inhalant Allergens of the European Academy of Allergology and Clinical Immunology.This trial was undertaken to study, in several geographically spread European countries, the prevalence of skin prick test (SPT) positivity to Alternaria (A) and Cladosporium (C) in subjects with nasal and/or bronchial symptoms of suspected allergic cause. Each patient completed an anamnestic questionnaire and underwent SPT with a panel of common inhalant allergens and also A and C supplied by three different laboratories, to allow for manufacturer bias. Specific scrum IgE determination was carried out only in subjects with SPT positivity to A and/or C with an immunoassay system. In nine European allergology centers, a total of 877 subjects was enrolled in the trial; 83 of them showed SPT positivity to A and/or C; only nine patients showed monosensitization to A, and none to C The highest percentage of positive subjects was found in Spain (20%); the lowest in Portugal (3%). In the other seven centers, the variation was 7–10%. The age range of mold-positive subjects was 5–60 years. Rhinitis was by far the most common symptom, whether associated or not with asthma and/or conjunctivitis.

    Notizen: Abstract  In a cell line from human pleomorphic adenoma (AP2 cells) we studied the response of these cells to basement membrane proteins. The culture was characterized as myoepithelial-like by transmission electron microscopy and immunocytochemistry. AP2 cells were grown in contact with a reconstituted basement membrane (Matrigel). Cells grown on Matrigel showed conspicuous phenotypic alterations, depending on how the substrate was applied. Cells grown on the top of Matrigel developed a dendritic phenotype, exhibiting thin, long and intercommunicating cytoplasmic extensions resembling normal myoepithelial cells. Cells grown inside Matrigel formed multi-layered clusters. Light, confocal and transmission electron microscopy showed that these clusters were formed by double-layered epithelioid cells delimiting luminal spaces. The cells facing the lumen were cuboidal, showing microvilli at the apical plasmalemmal and junctional complexes. The spatial arrangement of basement membrane is a key modulator of morphogenetic changes and cytodifferentiation of tumour myoepithelial cell lineage in culture.

    Notizen: Abstract  The identification of tumor-associated antigens recognized by cellular or humoral effectors of the immune system has opened new perspectives for cancer therapy. Different groups of cancer-associated antigens have been described as targets for cytotoxic T lymphocytes (CTLs) in vitro and in vivo: 1) cancer-testis (CT) antigens, which are expressed in different tumors and normal testis; 2) melanocyte differentiation antigens; 3) point mutations of normal genes; 4) antigens that are overexpressed in malignant tissues; and 5) viral antigens. Clinical studies with peptides derived from these antigens have been initiated to induce specific CTL responses in vivo. Immunological and clinical parameters for the assessment of peptide-specific reactions have been defined, i.e., delayed-type hypersensitivity (DTH), CTL, autoimmmune, and tumor regression responses. Preliminary results demonstrate that tumor-associated peptides alone elicit specific DTH and CTL responses leading to tumor regression after intradermal injection. Granulocyte-macrophage colony-stimulating factor (GM-CSF) was proven effective in enhancing peptide-specific immune reactions by amplification of dermal peptide-presenting dendritic cells. Long-lasting complete tumor regressions have been observed after induction of peptide-specific CTLs. However, in single cases with disease progression after an initial tumor response, either a loss of the respective tumor antigen targeted by CTLs or of the presenting major histocompatibility complex (MHC) class I allele was detected as a mechanism of immune escape under immunization. Based on these observations, cytokines to enhance antigen and MHC class I expression in vivo are being evaluated to prevent immunoselection. Recently, a strategy utilizing spontaneous antibody responses to tumor-associated antigens (SEREX) has led to the identification of a new CT antigen, NY-ESO-1, which is regarded as one of the most immunogenic antigens known today inducing spontaneous immune responses in 50% of patients with NY-ESO-1-expressing cancers. Clinical studies involving antigenic constructs that induce both antibody and CTL responses will show whether these are more effective for immunotherapy of cancer.

    Notizen: Zusammenfassung Die Verfasser wollen den Blick auf die Oberflächengestalt der Haut des tätigen Menschen hinlenken. Sie versuchen zu zeigen, daß der rauhen oder glatten Oberfläche der Haut eine große Bedeutung zukommt bei allen Fragen, die Schmutz, Waschen, Speichern hautschädigender Arbeitsstoffe und andere Beziehungen betreffen, die zwischen der Haut des tätigen Menschen und seiner Arbeitsumgebung bestehen. Die hautschädigenden Stoffe wirken dort stärker, wo sie längere Zeit und in größeren Mengen in der Haut gespeichert werden. Nach Einsatz der Fluorescenzanalyse, einer Raumbildmikromethode und einer fluorescenzmikroskopischen Methode unter Verwendung der Ultrabeleuchtung im Auflicht gelang eine genaue Betrachtung und Beschreibung der Hautoberfläche. Die Mikromethoden sind in erster Linie für das wissenschaftliche Laboratorium und für die Klinik bestimmt, aber die Betrachtung der Haut im filtrierten (unsichtbaren) Ultraviolettlicht und die Methode der mikroskopischen Raumbildbetrachtung (Mikrostereomethode) sollen vor allem im praktischen Betrieb, beim Fabrikarzt, Gewerbearzt, Vertrauensarzt und beim Sicherheitsingenieur angewandt werden. Mit diesen Hilfsmitteln kann die Rauheit der Haut, die Speicherwirkung für den hautschädigenden Schmutz, der Erfolg des Waschens und die Brauchbarkeit der einzelnen Waschmittel untersucht und beobachtet werden. Die Methoden sollen ein Hilfsmittel sein, wo es zu entscheiden gilt, wann ein hautgefährdetes Gefolgschaftsmitglied vorbeugend behandelt werden muß oder wann ein geheilter Ekzematiker an seinen Arbeitsplatz zurückkehren darf. Es wird in der Arbeit untersucht, an welchen Stellen der Hautoberfläche der Schmutz sitzt, und es wird festgestellt, daßeinem Schmutz eine andere Bedeutung zukommt, je nachdem er an verschiedenen mikroskopisch festgelegten Hautstellen liegt. Im Text ist an einigen Beispielen die praktische Bedeutung der Untersuchungsergebnisse klar gelegt worden. Die Methodik, vor allem die Technik der Auflichtmikroskopie bei starken Vergrößerungen wird eingehend geschildert.

    Notizen: Summary The histochemical fluorescence method of Falck and Hillarp was used to locate monoamine-containing neurones in the nervous system ofStrophocheilus, a Brazilian land snail. Green-fluorescing (catecholamines) cell bodies were found only in cerebral ganglia. Neuropiles of all ganglia contained many green-fluorescing fibers most of which appeared to enter from the periphery. Giant (up to 500 μ) non-fluorescing cells were seen in most ganglia of the suboesophageal mass. Green-fluorescing fibers surrounded, and apparently ended on, their axon hillocks and proximal segments of axons. Green-fluorescing sensory cells were seen in the eyeless tentacles. Cells fluorescing yellow (5-hydroxytryptamine) or yellow fading to green on exposure to ultraviolet light were found in cerebral, visceral and right palliai ganglia. Ligature of the cardiac nerve for 24 hrs resulted in an accumulation of yellow or “yellow green”-fluorescing masses in axons proximal to the constriction. The distribution of monoamine-containing neurones inStrophocheilus is rather similar to that found in other pulmonate molluscs such asHelix.
