    Notes: A cell and gas-handling system of the supercritical material for x-ray absorption measurement are designed. The cell was made of stainless steel with a pair of Be windows of 3 mm thickness. The x-ray path can be controlled by changing the distance between two Be windows. Electric cartridge heaters are embedded in the cell to control the temperature. With this system we have studied the supercritical state of CF3Br, whose critical point is 67 °C and 39.1 atm. The results of x-ray absorption fine structure measurements for Br K edge for gaseous, liquid, and supercritical states of this material are presented together with the performance of the gas-handling system. © 1995 American Institute of Physics.

    Notes: The influence of seed quality on seedling growth of white clover cv Makibashiro plants was examined in a controlled environment. The average dry matter (DM) yield of 70-day-old seedlings from a poor-quality seed lot was significantly less than that from a high-quality seed lot. In both seed lots, a linear relationship between the DM yield and time from sowing was observed. The slope of regression of DM yield against time from sowing for the poor-quality seed lot was less than the equivalent slope of the high-quality seed lot, suggesting that possible avenues for increased DM accumulation in a high-quality seed lot are somewhat indirect (time of emergence, uniform emergence etc.); a direct effect of seed quality on the photosynthetic ability of plant tissue seems less likely to occur. The root: shoot ratios declined in both groups of plants with increasing age. Seedlings from the high-quality seed lot reached a stable root:shoot ratio (equilibrium state) much earlier than those from the poor quality seed lot. The seed quality had no influence on the proportion of DM partitioned to various organs. The results of this study are discussed in relation to field establishment.

    Notes: Changes in quantity and composition of soil organic matter (SOM) in pasture receiving annual superphosphate (SP) applications for 41 years at 0 (control), 188 and 376 kg SP ha−1a−1 were investigated in soil samples collected from 0–75 and 75–150 mm depths by determining total carbon (TC), total nitrogen (TN), biomass C (BC), biomass N (BN) and subjecting the soils to sequential extraction using cold water, hot water, a mixture of hydrochloric (0.1 M HC1) and hydrofluoric (0.3 M HF) acids (HCl/HF), and sodium pyrophosphate (Na4P2O7) followed by sodium hydroxide (NaOH) for extracting labile and stable SOM fractions. There were significant differences in some SOM fractions between control (0) and SP treatments (188 and 376), especially in the topsoil (0–75 mm) but these were not observed between the two SP treatments. Soil TN (0–75 mm), BN and BN: TN ratio (0–75 and 75–150 mm depths) and the proportion of hot–water–extractable C (HC) in soil TC (HC:TC) (0–75 mm) were significantly greater in the SP treatments than in the control. HC1/HF extractable C and the proportions of soil TC as HC1/HF extractable C (HC1/HF extractable C: TC) were smaller in the topsoil of SP treatments than in the control. Similar results were observed in humin N: TN ratio and the proportions of soil TC as cold–water–extractable carbohydrate (CWcC: TC) and of soil HC as hot–water–extractable carbohydrate (HWcC : HC). Increases in the proportion of labile fraction in SOM were reflected in values of BN, BC: BN, BN: TN, HWcC : HC and HC: TC whereas decreases in the proportion of stable fraction in SOM were found in humin N: TN and HCl/HF–extractable C: TC ratios. Increases in labile SOM (BN and N–containing compounds such as amino acids and amino sugars, which were extractable by hot water but were not present as carbohydrate) and decreases in stable SOM (HC1/ HF–extractable C and humin fraction) in soils under pastures treated with annual SP applications compared with the control were attributed to pasture improvement and the amelioration of P and S deficiency, resulting in a greater return of plant residues and animal excreta and also an increase in clover growth and associated biological N2 fixation. The additional labile SOM in SP treatments compared with that of the control was not associated with the soil mineral Al and Fe components.

    Notes: Radiation-induced defects in a new second harmonic generation material, lithium vanadate single crystal, have been studied by electron paramagnetic resonance (EPR) and optical absorption measurements. EPR detects two intense signals due to intrinsic O− and impurity-associated CO−3 trapped-hole centers at 77 K, but no such signal in the case of trapped-electron centers. The EPR signal of impurity Fe3+ ions is suppressed by 77 K irradiation. Low-temperature irradiation produces three optical absorption bands peaking at 3.14, 2.0, and 1.56 eV. It has been concluded that some of the free electrons produced by irradiation are trapped by Fe3+ ions at low temperatures. After annealing at room temperature all the optical absorption bands disappear, although the EPR signals of CO−3 trapped-hole centers and unidentified electron-type centers at g=1.96 remain stable up to 330 K. © 1995 American Institute of Physics.

    Notes: The UVSOR is a 750-MeV dedicated electron storage ring in the Institute for Molecular Science at Okazaki, Japan. The ring was constructed mainly for the use of VUV photons. However, the intensity of the soft x-ray region around 10 A(ring) region is not negligible. The ring is also equipped with a 4 T superconducting three-pole horizontal wiggler. We have constructed a beamline equipped with a monochromator in order to utilize the soft and normal x-ray photons from the ring. In order to separate the vacuum of the ring from that of the monochromator, a light tight Be foil of 10 μm in thickness is placed in the upstream of the monochromator in the beamline. The monochromator constructed is a high vacuum compatible with double crystal which provides constant offset of the output beam. The constant offset during the rotation of crystal plane was realized by a simple linkage of linear motions of the two crystals with two linear guides placed on both input and output beam levels. The mechanical movement was found to be very smooth through a full rotation of 60° (between 15° and 75° of Bragg angle). Monochromator crystals such as beryl (101¯0), quartz (101¯0), InSb(111), Ge(111), and Ge(220) whose 2d value is 15.965, 8.512, 7.481, 6.532, and 4.00 A(ring), respectively, are being used with a reasonable intensity and signal-to-noise ratio of output signal. The range of photon energy which can be used with this line is from 850 eV to 4 keV in the bending magnet section, and from 850 eV to 6 keV with a wiggler. Because of the comparatively low energy of 750 MeV of the electron, the heat load is not a severe problem, and no radiation damage was observed for the first beryl crystal during an exposure to the beam for several months.We have measured the Na K-edge XANES spectrum of a thin film of NaCl which was evaporated in situ on a collodion film in the sample chamber at room temperature by using beryl as monochromator crystal. From the doublet separation of the spectrum, we can estimate the resolution better than 1 eV, which is sufficient for this energy range. With Ge(220) as monochromator crystal, K edge of potassium has been measured with a reasonable signal-to-noise ratio. Therefore, the practical limit of the energy attainable in this beamline of bending magnet section is about 4 keV.

    Notes: A vacuum-compatible double-crystal monochromator with constant exit-beam height has been installed and operated for several years at beamline BL7A at the UVSOR facility at the Institute for Molecular Science, Okazaki, Japan, The beamline is used both for the photons from a normal bending-magnet section and those from a 4 T superconducting three-pole horizontal wiggler. Various pairs of monochromator crystals are being used with sufficient intensity and signal to noise ratio of the output signal. Basic structure of the beamline, the mechanism of the monochromator, and some typical spectra of materials with absorption edges between 850 eV and 4 keV are reported.

    Notes: Viral diseases of sweet potato are very prevalent and often seriously damaging to the plants. In particular, the severe strain of the sweet potato feathery mottle virus (SPFMV-S) causes ‘obizyo-sohi’ disease in Japan. In order to confer viral resistance against SPFMV using current biotechnology, a transgenic sweet potato has been produced, introducing hygromycin-resistant (hpt) and SPFMV-S coat protein (CP) genes, which have shown a significant resistance to SPFMV-S. In the breeding programme, it is important to confirm that the viral resistance conferred in T0 plants can be inherited by their progeny. In the present study, progeny were obtained from crosses between the transgenic T0 and a non-transgenic variety of sweet potato. The results showed that the CP gene was inherited by the next generation and that the stability of viral resistance was also confirmed. Thus, this production system for the virus-resistant transgenic sweet potato is useful in practical breeding.

    Notes: Melanin-concentrating hormone (MCH) is believed to be an important orexigenic peptide mainly localized in the lateral hypothalamic area. Its involvement in the hyperphagia induced by hypothalamic lesions and lactation remains unclear. In this study, we investigated MCH immunoreactivity in the hypothalamus using immunohistochemistry and MCH concentration in the peripheral circulation using an enzyme immunoassay in rats with a lesion in the ventromedial hypothalamus or the paraventricular nucleus, and in lactating rats. Bilateral lesions of the ventromedial or paraventricular nuclei were performed using an electrolytic method. Quantification of immunoreactivity by image analysis revealed that the number and mean staining intensity of MCH-immunoreactive neurones in the lateral hypothalamic area and the zona incerta were significantly decreased by both types of lesions compared to sham controls, whereas circulating MCH concentration was not significantly different on day 7 postlesion. By contrast, in lactating rats on days 11–12 postpartum, the expression of MCH in the lateral hypothalamic area and the zona incerta was significantly increased compared to nonlactating controls. Circulating MCH concentration was not changed in lactating rats. These results suggest that hyperphagia induced by lactation, but not hypothalamic lesion, might be induced by excessive expression of MCH in the lateral hypothalamic area and the zona incerta.

    Notes: A surface protein antigen (PAc) of Streptococcus mutans, particularly the A-region of the molecule, has been reported to interact with salivary components on the tooth surface. It might be a candidate antigen inducing the production of antibodies against the adherence of S. mutans to the tooth surface. We investigated the effects of monoclonal antibodies (MoAbs) obtained by immunization of synthetic PAc peptides that completely correspond to the amino acid sequence of part of the A-region. These MoAbs recognize several core B-cell epitopes in the sequence. Two (KH5 and SH2) of these antibodies reacted with both S. mutans and Streptococcus sobrinus, but not with Streptococcus sanguis, Streptococcus salivarius, Porphyromonas gingivalis or Lactobacillus casei. They clearly inhibited the real-time adherence of S. mutans to salivary components in a biosensor. KH5, which showed a real-time inhibition (71%), also significantly prevented the recolonization of S. mutans on the tooth surface in rats. These results suggested that the core B-cell epitope (-Y---L--Y----) recognized by KH5 was the essential sequence in the antigenic epitopes of PAc protein recognized specifically by the inhibitory antibody. Therefore, the amino acid residues were found to be important in the initial attachment of S. mutans to the tooth surface. These results provide for the mechanism of PAc molecule in the initial attachment of S. mutans on the tooth surface and more effective designs for the removal of S. mutans and S. sobrinus from the oral cavity.
