    Notizen: Spleens from newborn mice less than 6–7 days of age contain ‘naturally occurring’ suppressor cell populations that are able to inhibit nonspecifically immune responses of third-party adult spleen cells and alloreactivity in the newborn spleen per se. Isolation of the effectors of this suppressor activity reveals that they are not classical T lymphocytes but, instead, a mixed population of cells of the monocytc series (monoblast/promonocyte/ monocyte) plus mast cells. This mixed population apparently can elicit its suppressor activity in part through secretion of soluble, in vitro culture-stable material, which in turn initiates activation of the suppressorcell/limb of the immune response. These activities, nevertheless, modulate most strongly the early activation events of T-cell responses and can result in complete suppression of alloreactive helper and cytotoxic T lymphocytedevelopment. Suppressor activity by mast cells can be demonstrated by degranulation, whereas suppressor activity by monocytes appears inherent in the newborn population. Thus, there are now at least three cell populations in the neonatc spleen—mast cells, monocytes. and T lymphocytes—which caneffect suppressor activity.

    Notizen: An anti-idiotypic (α Id) serum was induced against anti-tetanus toxoid (α TET)-reactivc F(ab')2 fragments of a single donor. After appropriate adsorption the serum was shown to react only with α TET F(ab')2 fragments and not with F(ab')2 fragments deplcted of α TET reactivity or anti-diphtheria toxoid (DIP) or anti-purified protein derivative (α PPD) F(ab')2 fragments of this donor. Inhibition studies with soluble TET indicated that the anti-idiotypic serum partly reacts with determinants close to or within the antigcnic binding site. When tested on a small panel of α TET F(ab')2 fragments derived from unrelated donors, one out of four donors tested showed cross-reactivity. When tested on α TET F(ab')2 fragments derived from related family members, the donor's father and two sisters were found to cross-react, whereas the mother and two other sisters did not. The observed cross-reactivity was independent of the litres of α TET antibodies in the serum. Furthermore, two-dimensional gel electrophoretic studies carried oui on α TET IgG of Id-positive or Id-negative family members indicated that the expression of idiotypic determinants is linked to a particular heavy chain. Immunofluorescent staining with α Id and cytofluoro-graphic analysis of donor T-lymphoblast proliferation in response to TET showed that the T cells did not express Id determinants found on his α TET antibodies.

    Notizen: Spleen effector cells for IgG- and IgM-induced antibody-dependent cellular cytotoxicity (ADCC) were characterized with respect to density and cell surface markers by using sheep erythrocytes (SRBC) coated with hybridoma-derived monoclonal anti-SRBC IgG or anti-SRBC IgM antibodies as targets. While basically the same effector celts are cytolytic for IgG and IgM antibody-coated SRBC, they differ with respect to their relative killing capacity for IgG- versus IgM-coated target cells. On the basis of physical and biochemical properties three populations with cytolytic capacity could be separated: (I) A light fraction of large cells had high cytolytic potential for both IgG- and IgM-coated SRBC. The cells were negative for the Fc receptor for IgG (Fcy-R−) and the C3-receptor (C3-R−), they carried the receptor for Helix pomatia A agglutinin (HP-A+), and reactivity was strongly reduced after treatment with anti-Thy-1 and complement (C). (II) High activity was also observed with a medium-dense fraction, preferably lysing IgG antibody-coated cells. The cells were Fcy-R+, partly C3-R+, mostly HP-A −, and only a minor portion of the cells were Thy-1+. (Ill) A dense fraction, displaying on a percell basis low cytolytic potential, was more active in IgM than IgG ADCC. The cells were Fcy-R+, HP-A+ and Thy-1+. All three effector cell populations were non-adherent, non-phagocytic, and surface immunoglobulin-negative (s-Ig−).

    Notizen: A subpopulation of murine synovial lining cells is shown to express Ia antigens coded for by both I-A and I-E/C loci of the major histocompatibility complex and to be derived from precursors in the bone marrow. This is demonstrated by use of biotinylated mouse monoclonal antibodies bound to frozen sections of tissue from normal and bone-marrow-reconstituted mice that are detected by a sensitive avidin/biotinylated peroxidase complex (‘ABC’) technique.

    Notizen: Natural killer (NK) cells constitute a cell type with an as yet undefined lineage, although certain similarities with T lymphocytes have been found in the mouse. Our present results show that NK cells have a significant difference compared with T and B cells in their capacity to traverse the blood-lymph barrier. Thoracic duct lymphocytes from mice or rats are thus devoid of NK activity, when at the same time potential, cytotoxic T lymphocyte (CTL) activity or antibody-dependent cellular cytotoxicity function can be demonstrated. Chronic thoracic duct drainage in the rat also leads to an increase in NK activity per unit cell number in the other lymphoid organs. Thoracic duct lymphocytes from mice and rats may thus serve as convenient sources of CTL and/or killer cells in situations in which it is important to minimize NK cell involvement.

    Notizen: The mouse primed lymphocyte typing (mPLT) assay, based on the sequential reactivation of specific immunocompetent, alloantigen-reactive T blast cells in secondary mixed leucocyte culture (MLC), has been utilized to define the major histocompatibility complex (MHC) Class I-associated lymphocyte-stimulating (LS) determinants. The test panel of secondary stimulating cells has been expanded to include 25 B10.W lines (mouse strains carrying MHC regions derived from wild mice), thus permitting examination of more than thirty ‘independently derived’ MHC haplotypes. Data obtained using mPLT cells generated in primary MLC between H-2 K- and/or DL-disparate strain combinations indicate that each allelic Class I product expresses a unique set of LS determinants recognized by alloreactive T lymphocytes. In addition, we have observed that Class I products, which are apparently serologically identical but which are encoded for by genes of independently derived haplotypes, can express mutually distinct LS determinants. These data suggest that Tlymphocytes possess the capacity to distinguish between Class I antigens that are serologically similar (or even identical) and support the concept that LS determinants recognized by T cells are distinct from the majority of specificities recognized by B cells. This concept has marked importance in interpretation of data in the human system, for which there is now an attempt to correlate serology and PLT.

    Notizen: The expression of lymphokine genes during infection of virulent (Tulahuén) or mild (CA-I) strains of T. cruzi was studied in mice lacking CD4 and/or CD8 molecules. The increased susceptibility of CD4− and CD4−CD8− mice to infection with CA-I or Tuiahuen was parallelled by diminished IFN-γmRNA levels. Nitric oxide release and inducible nitric oxide synthase mRNA accumulation by cells from Tulahuen infected CD4− mice was also diminished. CD8− (but not CD4−CD8− mice) showed an increased IL-4 and IL-10mRNA accumulation upon infection with both strains of T. cruzi. A Th2-like’ response (higher IL-4 and IL-10mRNA to IFN-γmRNA ratio), was also observed when cells from non-infected CD8- mice were stimulated with T cell mitogens.

    Notizen: The frequency of γδ+ T cells in the peripheral blood of 26 Turkish, 24 Swedish, 35 Japanese and 14 ‘Asian’ (non-Japanese) healthy blood donors and healthy volunteers were investigated by flow cytometry. In the Turkish group, 9.3% (median value) of the CD3+ peripheral blood T cells expressed the γδ T cell receptor. A similar level of γδ+ T cells was found in the non-Japanese ‘Asian’ healthy volunteers (9.2%), while significantly lower values were detected in the Swedish (4.2%) and Japanese (4.5%) groups. These dramatic differences in normally occurring γδ+ T cells in different groups of healthy individuals were further reflected by a low incidence of 〉10% γδ+ T cells in the Swedish (0/24) and Japanese (6/35) groups compared to the Turkish (12/26) and ‘Asian’ (5/14) groups. The described γδ+ T cell differences between distinct ethnic groups are thus likely to be a consequence of environmental factors, but additional genetic influences cannot be ruled out. The present study demonstrates the potential importance of the ethnic origin and environmental history of subjects examined in studies of γδ+ T cells–disease relations.

    Notizen: The surface characteristics of the mouse spleen cells mediating antibody-dependent cytotoxicity (ADCC) against antigen-coated chicken erythrocytes have been studied by several different column fractionation methods The major effector cells in this system were shown to be surface-adherent and could be depleted from spleen cells by passage through glass-bead ovalbumin/anti-OA immune complex columns (Fc receptor-binding), glass-bead immunoglobulin/anti-mouse Ig columns (Fc receptor and surface immunoglobulin-binding), and glass-bead mouse Ig/rabbit (Fab')2-anti-mouse Ig or Sephadex G-200/rabbit anti-mouse Ig columns (surface immunoglobulin binding) The concentration of EDTA in the medium used to fractionate the cells played a significant role in determining whether surface immunoglobulin could be demonstrated on the ADCC effector cells. From these results, the conclusion was drawn that ADCC on the part of mouse spleen cells could be mediated by surface-adherent, Fc receptor-positive cells bearing surface immunoglobulin of unknown origin. The possibility that ADCC can be mediated by a heterogenous population of cells in the mouse spleen is discussed.

    Notizen: To determine the extent of V-gene heterogeneity of blood T lymphocytes in patients suffering from Myasthenia Gravis (MG), we used eight recently available monoclonal antibodies (MoAb), directed against different Vα and Vβ gene products of the variable part of the T-cell receptor (TCR), covering approximately 25% of the α/β T cells in normal peripheral blood (PBL) of healthy individuals. Using a two-colour immunofluorescence method, we could calculate the expression of α/β V segments within the two major T-cell subsets, CD4-/CD8+ and CD4+/CD8- lymphocytes. Twenty-seven per cent (4/15) of the MG patients had T cells showing signs of abnormal expansion. Furthermore, among these expanded T cells, a restricted Vβ12 gene expansion could be seen, in three out of four patients. No correlation between TCR V-gene usage and HLA haplotypes (HLA-A, -B, -DR and -DQ) could be seen. Our data suggest that the majority of MG patients have abnormally expanded T-cell clones. The relevance of these findings is discussed.
