    Notizen: With the aid of particle simulation, we discuss efficiency enhancement in a single-pass Cherenkov laser by varying the permittivity of a dielectric sheet loaded on a parallel plate waveguide. For the analysis of the problem, a two-dimensional model for the Cherenkov laser is considered which is composed of a planar relativistic electron beam and a parallel plate waveguide, one plate of which is loaded with a dielectric sheet. In order to maintain the synchronism between an electron beam and an electromagnetic wave, the permittivity of the dielectric is properly varied in accordance with the variation in the average velocity of the electron beam. The result of numerical simulation shows that the efficiency of energy transfer from the electron beam to the electromagnetic wave is greatly enhanced by properly varying the permittivity of the dielectric sheet. © 1997 American Institute of Physics.

    Notizen: On the basis of the fluid model of the electron beam, amplification characteristics of a two-dimensional plasma-filled Cherenkov laser at 100 GHz bands are analyzed. By injecting a dense plasma into the background of a Cherenkov laser, the coupling between a relativistic electron beam and an electromagnetic wave at millimeter wavelengths has been improved, and the weak beam–wave interaction problem has become less serious. In particular, we show that due to electron–ion collisions in the background plasma, proper choices of parameters are essential for good operation in this regime. The numerical results indicate that a Cherenkov laser with background plasma can generate higher output power with a greatly enhanced growth rate at a more reasonable interaction length. The plasma injection, in addition, enables the device to gain more output power with a higher beam current. © 2000 American Institute of Physics.

    Notizen: Melatonin is the pineal hormone that acts via a pertussis toxin-sensitive G-protein to inhibit adenylate cyclase. However, the intracellular signalling effects of melatonin are not completely understood. Melatonin receptors are mainly present in the suprachiasmatic nucleus (SCN) and pars tuberalis of both humans and rats. The SCN directly controls, amongst other mechanisms, the circadian rhythm of plasma glucose concentration. In this study, using immunoprecipitation and immunoblotting, we show that melatonin induces rapid tyrosine phosphorylation and activation of the insulin receptor β-subunit tyrosine kinase (IR) in the rat hypothalamic suprachiasmatic region. Upon IR activation, tyrosine phosphorylation of IRS-1 was detected. In addition, melatonin induced IRS-1/PI(3)-kinase and IRS-1/SHP-2 associations and downstream AKT serine phosphorylation and MAPK (mitogen-activated protein kinase) phosphorylation, respectively. These results not only indicate a new signal transduction pathway for melatonin, but also a potential cross-talk between melatonin and insulin.

    Notizen: Background There are no reports of respiratory epithelial damage induced by immuiioglobulin-stimulated eosinophils.Objective We tried to induce damage to respiratory epithelium by guinea-pig (GP) eosinophils stimulated with guinea-pig IgG (GP-lgG)-coated Sepharose 4B beads. Methods GP tracheal epithelium was cultured together with GP eosinophils that had been collected from the peritoneal cavity, purified on a Percoll gradient, and stimulated with GP-lgG-coated beads (GP eosinophils:beads = 20:1). Damage to the epithelium was observed with an inverted microscope.Results After 24 h of culture with three 106 eosinophils. irregularity of the surface of the epithelium, desquamation. shedding of cilia, and abnormal beating of cilia were observed. This damage was first observed after 12 h of incubation, and was more severe at 24 h. No damage was found when beads coated with human serum albumin (HSA) were used. GP eosinophils stimulated with GP-IgG released significantly more EPO than those stimulated with HSA. at 6 and 24 h. Heparin and eatalase partially inhibited the epithelial damage. O2-production by eosinophils was also enhanced with GP-IgG-coated beads. Conclusion Both granule basic proteins and reactive oxygen radicals may be responsible for epithelial damage, probably via an EPO + H2O2+ halide system. These results confirmed that stimulated eosinophils can damage respiratory epithelium.

    Notizen: Background We have shown that interleukin-5 (IL-5) and granulocyte-macrophage colony-stimulating factor (GM-CSF) are present in sputum from patients experiencing acute asthma attacks, by eosinophil survival assay. The viability of guinea-pig eosinophils was significantly increased in the presence of such sputum extracts after 3 days' culture, and it was inhibited by the addition of anti-IL-5 and anti-GM-CSF antibodies. However, the contribution of IL-5 to the increase in eosinophil viabihty was less than expected from the values of IL-5 measured by enzyme-linked immunosorbent assay (ELISA). Therefore, we speculated that something in sputum inhibited the function of IL-5.Objective Tratnsforming growth factor-β (TGF-β) was the only cytokine we tested that inhibited the prolongation of survival of guinea-pig eosinophils induced by IL-5. The objective of this study is to detect TGF-β in the same sputum. Methods Guinea-pig eosinophils were cultured with or without anti-TGF-β antibody in the presence of sputum extracts, and the eosinophil viability was counted after 3 days. Measurement of TGF-βl in sputum was performed by ELISA. Results Eosinophil viabilities with and without anti-TGF-β antibody were 79.7 ± 2.9% and 69.0 ± 2.7%, respectively, and the difference between them was statistically significant (P 〈 0.05, n = 9). The concentration of TGF-β1 in the sputum was 21.7 ± 3.3 ng/mL (n = 9).Conclusion These observations suggest that TGF-β is present in sputum from patients with bronchial asthma.

In the present study, using the euglycaemic hyperinsulinaemic glucose clamp technique, we investigated the effects of hyperinsulinaemia on sodium-water metabolism and the pressor system in obesity, both of which have been reported to be closely associated with insulin resistance and/or hyperinsulinaemia.2. Sixteen obese young subjects and 24 non-obese young subjects who were all normotensives, participated in this study. The 2h euglycaemic hyperinsulinaemic glucose clamp was performed in a fasting state. The mean glucose infusion rate needed to maintain a fasting blood sugar level (FBS) during the last 30 min of the clamp was used as an indicator of insulin sensitivity (M-value). Before and after the clamp, the following parameters were measured: creatinine clearance (Ccr); urinary excretion of sodium (UNaV); fractional excretion of sodium (FENa); plasma renin activity (PRA); plasma aldosterone concentration (PAC) and plasma noradrenaline concentration (PNA).3. The M-value was significantly lower in obese subjects compared with non-obese subjects, although FBS and fasting immunoreactive insulin levels were similar in both groups. UNaV and FENa fell only in obese subjects during the clamp, while Ccr showed no significant change in either group. PNA and PRA increased significantly and PAC tended to increase in both groups.4. These results suggest that obese subjects have insulin resistance with respect to glucose metabolism, but that urinary sodium excretion and the pressor system remain insulin-sensitive; the sensitivity of the sodium retaining action to hyperinsulinaemia was actually higher in obese subjects than in non-obese subjects. Therefore, if compensatory endogenous hyperinsulinaemia was raised by insulin resistance, these two factors may lead to chronic sodium retention and pressor system stimulation and, in turn, to hypertension in obesity.
