Notizen: Abstract The magnetostriction (MS) of REBa2Cu3O7−δ (RE=Y, Nd, Eu, Gd, Ho, Er, Yb) is reported as a function of magnetic field and temperature forH〈5 T and 1.5〈T〈40 K. With nonmagnetic rare earths the MS is small and a linear function of the field atH〉1 T. The MS is much larger when the rare earth carries a magnetic moment, as usual, but the volume MS observed with moment carrying REBa2Cu3O7−δ is larger than that of nonsuperconducting intermetalics. This effect is explained by the shielding currents which are induced by the 4f magnetic moments in the superconducting CuO-lattice. The thermal expansion of GdBa2Cu3O7−δ shows a large peak at the ordering temperature of 2.2 K in both the superconducting and the nonsuperconducting samples.

Notizen: The reaction of electronically excited Na(3P) atoms with HCl has been studied in a crossed molecular beams experiment. At collision energies slightly above the endoergicity of the reaction, Na(3P) shows a dramatic enhancement of reactivity over ground state Na(3S). Detailed measurements of the laboratory angular and velocity distributions of the reactively scattered NaCl product at 5.4 kcal/mol collision energy have allowed determination of the product center-of-mass translational and angular distributions. These experimental results are compared to the DIPR model of electron transfer reactions. The broad translational energy distribution is in qualitative agreement with the DIPR model, but the angular distribution exhibits reduced intensity for scattering perpendicular to the relative velocity vector which cannot be reproduced by the DIPR model. The preferred transition state configuration, Na–Cl–H, is consistent with what would be predicted by a diffuse 3P orbital where the Na atom appears ion-like. This configuration is opposite to that given by the dominant term in the long range multipolar expansion of the neutral reactant potential.

Notizen: Using histochemical methods, morphofunctional aspects of the alimentary tract of larval coregonids were investigated. Larvae of Coregonus lavaretus were reared for 34 days with either zooplankton or one of two dry diets. Ontogeny, localization and diet-related modifications of the following enzymes were examined: trypsin (luminal digestion), aminopeptidase, maltase, alkaline phosphatase (brush border-bound digestion) and unspecific esterase (intracellular nutrient processing). All of the enzymes studied were present in 13-day-old larvae. Except for the intracellularly located unspecific esterase, there was an ontogenetic enhancement of enzyme staining intensities accompanied by a significant increase in the volume of the intestinal mucosa. Enzyme activities differed within and between intestinal regions. This finding suggests that a spatial gradient of nutrient breakdown and absorption already exists in the morphologically and physiologically incompletely developed digestive system of larval coregonids. Digestive enzyme activities were modified in response to the dietary regimen. There was no obvious correlation between enzymic response and growth performance of the larvae.

Notizen: Abstract We isolated from Candida maltosa mutants lacking saccharopine reductase (lys9) and saccharopine dehydrogenase (lys1). They accumulated α-aminoadipate-δ-semialdehyde (AASA) in the cell and excreted it into the culture medium. In the presence of 15 g glucose/l, 1.25 g NH4H2PO4/l and 50 mg l-lysine/l in a minimal salt medium C. maltosa G285 (lys1) produced about 80–90 mg AASA/l within 48 h. It is the first report of lysine-requiring yeast mutants that accumulate and excrete AASA. In contrast, Pichia guilliermondii lys9 mutants lacked this AASA overproduction. The AASA accumulation by C. maltosa mutants may be explained by the low feedback regulation of their homocitrate synthase and the equilibrium of the enzyme reactions involved in the lysine biosynthesis.

Notizen: Abstract A novel aminotransferase catalysing the first step of lysine catabolism, the oxidative transamination of the ?-group of L-lysine, was found and characterised in the yeast Pichia guilliermondii. The enzyme, L-lysine : pyruvate aminotransferase (Lys-AT), is strongly derepressed in cells grown on L-lysine as sole nitrogen source and its activity is highly specific for both L-lysine and pyruvate. We could successfully isolate a regulatory mutant which is unable to use lysine as sole nitrogen source based on its inability to derepress the Lys-AT.

Notizen: Summary X-irradiation of the head of adult mice leads to DNA repair synthesis (unscheduled DNA synthesis, UDS) in non-proliferating cells of the brain as shown autoradiographically after injection of3H-thymidine and subsequent irradiation. The extent of UDS induced by one and the same X-ray dose varies between different cell types and also between different brain areas. Within the range of X-ray doses studied (2 to 100 Gy) a linear dose effect relationship was observed. No evidence of a saturation effect was found. The slopes of the regression lines for the dose effect relationship differ considerably for the different cell types. Two interesting correlations were found, if the present results were compared with other data in the literature: (i) There seems to be a correlation between the extent of UDS and radiosensitivity of the different cell types, the cells with low DNA repair synthetic rates being more radiosensitive. (ii) The extent of UDS of the different cell types correlates well with the extent of protein synthesis of the corresponding cell types. Apart from radiation induced UDS, spontaneous UDS was found to occur in sham-irradiated animals. The extent of spontaneous UDS also differs considerably between different cell types as well as between different brain areas. The increase of spontaneous UDS with increasing duration of immobilization of the animals during sham irradiation suggests a relationship between spontaneous UDS and stress.

Notizen: Summary Single, slow muscle fibers fromRana temporaria were equilibrated in normal Ringer's. 95 mmol/liter K1-solution containing various concentrations of Ca2+, Ni2+, Mn2 or Mg2+ was applied, and the ensuing contractures were recorded isometrically. While peak tension (F max) was little affected, maintained tension (measured 1 min after onset of contracture) strongly depended on the concentration and species of divalent cations. Tension was maintained at its peak value in the presence of all species of divalent cations provided their concentrations were adequately increased. Dose-response curves were hyperbolic: Lineweaver-Burk plots revealed straight lines with different slopes intersecting near 1/F max, and indicating the following order of efficiency: Ni2+〉Ca2+〉Mn2+〉〉Mg2+. Hill plots for these cations resulted in straight lines with slopes near 1. Qualitatively similar relationships were obtained with contracture solutions containing D6000 (3–12 μmol/liter). However, under these conditions higher concentrations of Ca2+ or Ni2+ were required in order to fully maintain tension. After a step concentration change in the medium during contracture, the effects of Ca2+ or D600 were detectable only after a delay of 9 and 18 sec, respectively. It is concluded that divalent cations and D600 compete for the same binding site according to a 1:1 reaction. This site is presumably located inside the transverse tubular system and controls inactivation of the contractile force.

Notizen: Summary K contractures of single slow muscle fibers ofRana temporaria were measured isometrically in the presence of normal, reduced, and increased Ca2+ concentrations at 18 to 20°C. At normal Ca2+ concentration (1.8mm) contracture tension decreased from its peak value of 35.4±8.2 N/cm2 to 59.4±23.9% within one minute, and to 48.3±27% within two minutes (30 fibers). Peak tension was virtually unaffected by changes of the Ca2+ concentration, but maintenance of tension was impaired by low (0.2mm), and improved by high (10mm) Ca2+ concentrations. When Ca2+ was added during the K contracture, there was practically no restoration of lost tension. Effects similar to those of Ca2+ were observed upon addition of foreign divalent cations to the medium. Co2+, Ni2+, and Cd2+ were slightly more effective than Ca2+ and Mn2+; the smallest effects were obtained with Mg2+, Sr2+, and Ba2+. The effects of foreign divalent cations were independent of the presence of Ca2+. It is concluded that in slow fibers ofRana temporaria maintenance of contracture tension is not due to an influx of Ca2+ ions. Instead, binding of divalent cations to superficial sites seems to be essential.
