    Notes: Vacuum ultraviolet emission and electrical characteristics of a simple discharge configuration consisting of two planar cylindrical electrodes operated with a dc voltage have been measured over a wide range of He/Xe mixtures and discharge pressures. Breakdown characteristics are consistent with those found in the literature, however current–voltage characteristics and the inferred discharge resistivity suggest the presence of a complex process controlling electron emission at the cathode. Ultraviolet vacuum emission maps of atomic and molecular xenon at 147, 150, and 173 nm, respectively, have been measured as a function of pressure, from 60 to 500 Torr, and gas mixture, from pure Xe to 5% Xe in He. The calibrated ratios of each emission map help to visualize the zones of strongest ultraviolet emission over a wide range of operating conditions. One-dimensional simulations of the breakdown voltage and current discharge have been performed using the commercially available discharge-modeling package SIGLO. Good agreement with experimental results is found in the case of pure helium and xenon, however, in the case of pure xenon, the gas temperature was adjusted (elevated) in order to reproduce the measured current–voltage characteristics. Modeling of the electron number density distribution indicates that the discharge is principally composed of a thick ion sheath near the cathode. © 2001 American Institute of Physics.

    Notes: 5-Hydroxytryptamine-1A (5-HT1A) receptor agonists, including flesinoxan, reduce anxiety and activate the hypothalamus-pituitary-adrenal (HPA) axis under basal conditions. In order to investigate the underlying neural mechanisms we investigated immunoreactivity for the immediate early gene protein product Fos (Fos-ir) in rat brains 1 h after flesinoxan treatment (0.0, 0.3 or 3.0 mg/kg p.0.). Typically, 5-HT1A receptor-containing brain areas, such as the dorsal raphe nuclei, hippocampus, septum, diagonal band and the cortical and basomedial amygdala, do not show Fos-ir. Apparently, binding of flesinoxan at the 5-HT1A receptor does not directly lead to activation of c-fos in the cell, probably due to its negative coupling to adenylate cyclase. However, in typically non-5HTlA receptor-containing brain areas Fos-ir is increased due to flesinoxan treatment, as in the paraventricular nucleus of the hypothalamus (PVN), the dorsolateral part of the bed nucleus of the stria terminalis (BNSTd1) and the central amygdala (CeA). Flesinoxan-treated rats also exhibited higher plasma corticosterone levels than vehicle-treated animals, which suggests the involvement of corticotropin-releasing hormone (CRH) or vasopressin in the hypothalamus. After double immunolabelling (Fos/CRH or Fos/vasopressin), every CRH neuron detected in the PVN also contained Fos. Moreover, a significant correlation existed between the number of Fos-ir neurons in the PVN and the plasma corticosterone level. Hardly any Fos/ vasopressin double labelling was visible in the PVN. Accordingly, flesinoxan exerts its activating effects on the HPA axis via CRH neurons in the PVN. These effects are transsynaptically mediated by other brain areas, such as the CeA and BNSTdl, which also show increased Fos-ir.

    Notes: Vacuum emission, breakdown, and current–voltage characteristics of a large surface area-to-volume ratio planar dc microdischarge have been measured for different He–Xe mixtures in the range of 10%–100% Xe and pressures in the range of 60–500 Torr. The electrical measurements show that the Paschen breakdown curves and the current–voltage characteristics are primarily controlled by the xenon concentration in the mixture. A study of the vacuum ultraviolet emission of xenon atoms at 147 nm and of xenon dimers at 150 nm normalized to the discharge power reveals the presence of local maxima in the emitted intensity as a function of discharge pressure and xenon concentration, indicative of complex excited-state atomic and excimer kinetic processes. © 2000 American Institute of Physics.

    Notes: Abstract The fear-potentiated startle (PSR) paradigm is a putative behavioral model for the determination of anxiolytic properties of drugs. The present study further investigated the predictive validity of the model. Predictive validity is high, when only drugs clinically used as anxiolytics attenuate PSR dose dependently. Results showed that startle potentiation decreased dose dependently after the administration of the anxiolytics CDP (2.5–10 mg/kg, IP) and alprazolam (1–3 mg/kg, IP). After administration of the clinically non-anxiolytic drugs amitriptyline (2.5–10 mg/kg, IP), carbamazepine (5–20 mg/kg, IP), fentanyl (0.0025–0.04 mg/kg, SC), naloxone (2.5–10 mg/kg, IP), nicotine (0.4–1.6 mg/kg, IP), alcohol (500–2000 mg/kg, IP), andd-amphetamine (0.6–2.4 mg/kg, IP), a dose-dependent decrease in startle potentiation was not found. The PSR correctly discriminated most of the drugs tested in clinically anxiolytic and clinically non-anxiolytic drugs. However, haloperidol behaved as a false positive, and results of nicotine and alcohol were at variance with results reported by others.

    Notes: Abstract The fear-potentiated startle response paradigm is used to investigate anxiolytic properties of drugs. The first objective of the present study was to further investigate the predictive validity of this paradigm. The anxiolytics chlordiazepoxide (2.5–10 mg/kg IP) and oxazepam (1–10 mg/kg PO) and the putative anxiolytic flesinoxan (1–10 mg/kg PO) decreased startle potentiation dose-dependently, indicating an anxiolytic effect. The antidepressant fluvoxamine (5–20 mg/kg PO) did not affect startle potentiation. Ideally, anxiolytic drugs attenuate startle potentiation without affecting control startle levels, although some studies report altered control startle amplitudes. The second objective was to investigate whether different effects on control startle amplitudes are related to different startle devices. Therefore, the drugs were tested in two laboratories. Results showed no significant differences between laboratories, indicating that equipment is not a critical factor in the drug-induced alteration of control startle levels. In an additional experiment, it was shown that flesinoxan (10 mg/kg PO) did not affect strychnine-induced startle potentiation, supporting the idea that the attenuating effect of flesinoxan on the fear-potentiated startle response is due to its anxiolytic properties. Thus, the fear-potentiated startle response paradigm appears a valid and reliable model for anxiolytic properties of drugs.

    Notes: Abstract The effects of several types of antidepressants in a recently developed “behavioural despair” model, the tail-suspension test, are described. Drug effects on the automatically recorded duration of immobility and power of movements were measured in three strains of mice. Only in one strain (NMRI) did almost all antidepressants tested showed the expected reduction in duration of immobility. Tranquillizing drugs, but not stimulants, could be distinguished from antidepressants. The power of movements could not definitively be related to the pharmacological profile of the drugs tested. The use of the tail-suspension test as a rapid and highly predictive behavioural primary screen for antidepressant drugs is suggested.
