    Notes: Methacryloyl chloride was polymerized in a number of solvents, and the polyacid chloride was converted into the methyl ester. The yield and viscometric molecular weight are given for each sample, whereas the osmometric molecular weight is given for selected samples. The choice of solvent controls the degree of polymerization between the limits of 500 and 8,800. Polymer yields were about 20% and the degree of polymerization was 1500 when dioxane was used as a solvent, whereas the yield increased to 45% and the DP was very high when such solvents as cyclohexane and n-hexane were used. Polymerization in solvent mixtures gave intermediate molecular weight values. Infrared analysis showed the presence of anhydride.

    Notes: A water immiscible liquid is encapsulated in a thin skin of an insoluble copolymer by dissolving a difunctional acid chloride in said liquid and spraying the solution in fine droplets into a dilute aqueous solution of excess glycol or diamine. The polymer, which is not hygroscopic, forms as an insoluble film on the periphery of each drop. The microcapsules may be separated from the aqueous phase and dried without coalescing. Dyes, pigments, stabilizers, and other materials may be incorporated within the capsules.

    Notes: The movement of coloured markers (polydimethyl siloxane and low density polythene) placed near the die entry before extrusion or injected through the side of the die during extrusion has been measured both above and below the onset of instability. During stable flow the observed velocity distributions are similar to those predicted by assuming that polymer at the die wall is stationary, but when melt fracture occurs there is movement at the wall. The possibility of a stick-slip mechanism being responsible for unstable flow is examined and evidence in support of this hypothesis is presented. Available evidence suggests that for a given molecular weight, the critical shear stress increases with the polarity of the polymer, and the material of which the die is made has been found to affect the instability.

    Notes: The effect of polymers with conjugated bonds, namely polyphenylvinylene, phenylacetylenep-diethinylbenzene copolymer and thermally dehydrochlorinated polyvinylchloride, upon the stability of polyvinylchloride suspensions has been investigated. It has been shown that polymers with conjugated double bonds are efficient inhibitors of the thermal dehydrochlorination of polyvinylchloride. The most active inhibitor, polyphenylvinylene, is not light stabilized.

    Notes: Attempts to operate plastic processing equipment at higher production rates is often accompanied by erratic flow behavior of the polymer melts. In order to study this problem, a constant-rate, capillary rheometer was used to study the melt flow over a wide range of shear rates. These data demonstrate that many high-density polyethylenes exhibit a discontinuous shear-stress shear-rate curve; that is, under certain conditions the observed load oscillates “between two values differing by about 15%”. This region of unstable melt flow is not necessarily allied with melt fracture although it is dependent on molecular weight and melt temperature. Unstable melt flow occurs at lower shear rates with a decrease in the melt index (increase in molecular weight) or melt temperature.

    Notes: It is demonstrated that the bonding characteristics of commercial liquid adhesive primers made from nitrile rubbers and phenol-formaldehyde polymers, cured by means of hexamethylenetetramine can be quite sensitive to the presence of small amounts of excess water and ammonia. The added water partially decomposes the hexamethylenetetramine to formaldehyde and ammonia, the concentration of the latter thus increasing to such a point as to promote a lower temperature curing of the phenolic resin. Restoration of the original adhesive properties can be attained by the addition of reagents which remove either the water or the ammonia, or both, from the adhesive. Examples of such reagents are certain organic acids, anhydrides and peroxides. The addition of these reagents has been found to reactivate and increase greatly the useful lives of nitrile-phenolic liquid adhesive primers containing hexamethylenetetramine.

In both free radical and ionic copolymerizations involving similar monomers, plots of the logarithms of the relative reactivities of several substituted styrenes toward the styrene and methyl methacrylate radicals versus the differences existing between the exaltations of substituted styrenes and the exaltation of styrene were strikingly similar to those in which Hammett's sigma values for the styrene substituents were plotted against the relative reactivities of the substituted styrenes.

    Notes: The size of the crystallites in nylon 66, polytetrafluoroethylene, and polyethylene correlates with the degree of crystallinity. This supports the suggestion that the amorphous content of partially crystallized polymers can be attributed to chain folds and other regions directly related to the crystalline structure, at least down to a level of 40-60% crystallinity. In all cases, the term “percent crystallinity” is a useful unifying concept expressing the over-all degree of order including mesomorphic structures, the relative amounts of ordered and disordered material, and the quality of the ordered regions.

    Notes: Films of polytetrafluoroethylene, irradiated in halogen atmospheres, decrease in ultimate tensile strength and elongation at break almost as rapidly as in oxygen. The rate of degradation is approximately independent of chlorine pressure down to about 1 mm Hg. The phenomena can be explained semi-quantitatively in terms of first order, cage reactions of scission, recombination, and reaction with chlorine, complicated by diffusion at low pressures. However, all reaction and diffusion parameters must shift with gradual increase of crystallinity.For a given decrease of elongation at break, polytetrafluoroethylene irradiated in a vacuum retains a relatively higher ultimate tensile strength. In the copolymer of tetrafluoroethylene and hexafluoropropylene, the changes in ultimate tensile strength and elongation at break are much slower under all conditions. Both polytetrafluoroethylene and the copolymer retain appreciable tensile strength but little elongation up to a dose of 2 x 108r.

    Notes: This study summarizes the flow behavior of concentrated and melt systems of linear amorphous polymer. For low molecular weight polymer, viscosity increases regularly with a lower power dependence on molecular weight. This dependence decreases towards first power with increasing temperature but is independent of shear, with viscosities being constant up to the highest attainable steady state stresses, about 106 dynes/cm2. At a higher molecular weight, 2,000-40,000, depending on polymer type, the viscosity dependence on molecular weight changes abruptly to a high power which is generally about 3.4. Above the transition, this high power function is independent of temperature but depends markedly on shear. Non-Newtonian flow sets in abruptly and predictably above the transitional molecular weight.

    Notes: A simplified creep apparatus was used to evaluate the creep and recovery of polyethylene and polypropylene. The creep resistance of polypropylene containing as much as 9% n-heptane (boiling) soluble material is superior to that of linear polyethylene. The lower the boiling n-heptane soluble content, the better the resistance to creep. The creep resistance of polypropylene is also improved by annealing.

    Notes: The diameter increase of a molten thermoplastic extrudate is discussed in terms of the viscoelasticity of the resin. A swelling index, the ratio of the extrudate diameter to the orifice diameter, is defined. It was observed that swelling data taken at different temperatures can be correlated with stress in the die. An unexpected influence on the swelling for polyethylenes is the length of the die where short dies show larger swelling than long dies. Some suggestions for reducing swelling tendencies are made based on a simple model for flow in a capillary.

    Notes: Based on the simplified, flat-plate, isothermal, Newtonian flow model of the screw pump, an expression for pressure as a function of time at a fixed point on the extruder barrel has been derived. This expression predicts a periodic saw tooth pressure cycle under most flow conditions. It also establishes the flow conditions under which an inversion of the saw tooth cycle will occur. Pressure transducer data, in conjunction with extrusion rate data, can be used to calculate the average fluid viscosity in a region near the transducer.

    Notes: The melt extrusion of a commercial polypropylene resin was studied with a screw extruder and a series of capillary dies of length:radius ratios varying from 0 to 65. Three temperatures and the range of shear rates encountered in industrial processing were covered. The temperature dependence of the apparent melt viscosity of polypropylene was intermediate between linear and branched polyethylene. Extrapolated zero shear activation energy for viscous flow was 9 ± 1 Kcal/mole. Shear dependence of the apparent viscosity of polypropylene melt was greater than for branched polyethylene in turn, greater for linear.

    Notes: The sorption of nitrogen vapors on polyethylene, polytetrafluoroethylene, saran, polyvinylalcohol, polyvinylacetate, polystyrene, cellulose and ethylcellulose at the nitrogen boiling point has been investigated. It has been shown that polyethylene, polytetrafluoroethylene, saran, polyvinylalcohol and polyvinylacetate are non-porous sorbents with dense packing of the macromolecules. It has been shown that the specific surface area of cellulose, ethylcellulose and the total pore volume of polystyrene calculated from nitrogen vapor sorption is less than the corresponding value determined from the sorption of organic substances. Nitrogen vapor sorption at the nitrogen boiling point cannot be used for estimating the porosity of loosely packed polymers.

    Notes: Resin structures available from chlorinated polyethylene have been determined by studies of the infrared absorption spectra, and have been related to the glass transition temperatures and degree of crystallinity of the products. The relationships between chlorine content, molecular weight, glass transition temperature and a number of mechanical and rheological properties are described. Data are presented on the effect of blends of various resins in this series on glass transition phenomena. The stability of the resins to dehydrochlorination is discussed.

    Notes: The influence of channel curvature on flow through screw pumps and melt extruders is derived for infinite channel aspect ratios. Flow rate and horsepower relations for flow of Newtonian liquids are given as functions of design parameters. The results of this cylindrical theory are compared with those of conventional theories. Curvature factors converting the equations of the flat plate theory to those of the cylindrical theory are shown. Pressure distributions in screw pump channel are analyzed.

    Notes: Mixing of viscous fluids in an annular channel is analyzed theoretically. Four basic mixing patterns are described which can be generated by altering the relative rotational velocities of the annular boundaries. Only one aspect of the mixing problem is isolated, i.e., reduction in striation thickness. Problems of front to back mixing to iron out variations of feed stream composition and mixing of solid particles are not considered. The theory was verified experimentally using molten polystyrene.

    Notes: The permeability of films to O2, CO2, and H2O vapor is reduced by drawing processes. The permeability data are compatible with the hypothesis that crystallinity reduces permeability because of (1) a decrease in the volume fraction of penetrable amorphous polymer; (2) a lengthening of the diffusion path by impenetrable barriers, presumably crystallites; and (3) a reduction in segmental motion within the amorphous volume. Analysis of the effect of molecular diameter on diffusion rate shows that the segmental motion within amorphous polypropylene regions is less than that in natural rubber.

    Notes: This investigation studies the behavior of several cylindrical specimens of a Buna-N elastomer subjected to compression impact by a finite mass. Initial strain rates fall in the range between 40/sec and 400/sec, with maximum nominal strains up to 0.4. It is shown that values of nominal stress, true and nominal strain, energy dissipation and impact duration can be correlated with elastomer geometry and initial kinetic energy by means of two parameters derived from an analysis of a linear viscoelastic model. Other sources of variability of elastomer behavior are discussed.

    Notes: The application of a free vibration technique to the study of the cross-linking of an epoxy resin by an amine is demonstrated. Curing curves are presented showing the changes in dynamic mechanical flexural modulus and damping during reaction. At room temperature, the glass transition point is reached after a conversion of about 65% of the epoxide groups. Comparisons of the dynamic mechanical and infrared curing curves indicate a lowering of the reaction rate after this point is reached. Finally, an activation energy for the epoxy-amine cross-linking reaction is calculated.

    Notes: The polymer was prepared by pyrolyzing the sodium salt of 3,5,7,8,-tetrachloroperfluoro-octanoic acid and subsequent dechlorination of the resulting heptene with activated zinc. The heptadiene was separated from other dechlorination products and brought to a high degree of purity through use of vapor phase chromatography and polymerized in bulk utilizing gamma rays at room temperature, 100°C and 150°C, and utilizing ultraviolet light at 150°C. The properties of these brittle and glossy polymers were determined. Also the purified 4-chloroperfluoroheptadiene-1,6 was telomerized with dibromodifluoromethane yielding a cycle, clear, colorless, liquid telomer.

    Notes: The influence of bulk physical properties on the bond strength of an adhesive was studied. A conventional epoxy resin when cured with various mixtures of phthalic acid and anhydride exhibits maximum bond strength when the curing agent is 75% acid and 25% anhydride. This effect is explained on the basis of physical properties of the unfilled resin systems. Most significant of these were the heat distortion temperature and the compressive break and yield strengths.

    Notes: Seventeen different samples of styrene polymers and copolymers were examined in a capillary extrusion rheometer over broad ranges of temperatures and shears. They were injection molded in a spiral mold over the same temperature range. If viscosities at constant shear stress are obtained from the rheometric data, they cannot be correlated with spiral flow results. However, if viscosities are calculated at a proper “high” shear rate, capillary data correlate will with spiral flow results at all temperatures and for all materials.

    Notes: The problems of unsteady-state heat transfer associated with the melting and cooling of crystalline plastics such as polyacetals, high density polyethylene and polypropylene, are complex because of the latent heat of fusion that must be dealt with in problems where the temperature range being considered extends across the melting range. In the case of polyacetals, the use of a computer is shown in the literature to provide solutions to those problems. For many molding shops, such aids are not available. Numerical methods for the solution of such problems have been investigated, and their results are compared with those obtained using a computer and published for polyacetals. Cooling curves derived from numerical calculations, for slabs and rods made from high density polyethylene and polypropylene, are presented. Applications of the method are indicated.

    Notes: Dilatometric measurements show that both isotactic and atactic polypropylenes have two distinct glass transition temperatures. Because of the two transitions, three values for the cubical coefficient of thermal expansion are necessary to accurately define the volume expansion of polypropylene from -80° to +90°C. A method is outlined for calculating the content of amorphous atactic, amorphous isotactic, and crystalline isotactic polymer in a given polypropylene sample.

    Notes: The chemical changes taking place in polyethylene in the process of flame spraying have been investigated qualitatively and quantitatively by a spectroscopic method. It has been shown that oxidation occurs during the process with the formation of acidic, ketonic, ester carbonyls, associated hydroxyls and -C-O- bonds. The degree of unsaturation also increases due to growth in the number of vinyl and trans-vinylene groups. A small increase in the number of CH3 groups is also observed.

    Notes: The radiation-induced polymerization of n-tetradecafluoroheptene-1 was investigated at pressures of 8100, 11,900 and 17,100 atmospheres and temperatures between 90 and 267°C. The data are interpreted in terms of a free radical mechanism and the polymer has a low ceiling temperature. As temperature is increased, the depropagation step becomes important and reduces the rate of polymerization. Under some conditions transfer seems to be a significant part of the mechanism. Several radiation-induced polymerizations at high pressure were carried out with 1,1,2-trifluorovinylphenyl ether as well as one with 1,2,3,4,5-pentafluorophenyl 1,1,2-trifluorovinyl ether. The results suggest that transfer limits the molecular weight of the polymers.

    Notes: The effect of resin mass on the degree of crosslinking during the initial polymerization of three typical polymer compounds was determined by electrical resistivity techniques. Activation energies for the electrical conduction process (Ec) were calculated and used as an index of the extent of additional crosslinking occurring during sixteen months of storage. Test data show continued polymerization at slow rates for periods of time ranging from three days to over sixteen months before the polymerization process is completed. The final degree of crosslinking is shown to be dependent on the extent of initial crosslinking.

    Notes: Analysis in liquid or solid state and in various conditions of cure, have been developed. The analysis is carried out by spectroscopy in the near-infrared region. Epoxy values are determined preferably at 2.205 microns, or else at 1.159 microns. The required path length in the specimen is between 1 and 10 mm, solid resins were examined either in disposable glass cells or in the form of cast sheet. A direct method of evaluation is used when the path length is precisely known, a relative method based on internal calibration when this is not the case. Similarly, a near-infrared method for the determination of hydroxyl groups in epoxy resins was developed. The use of the isosbestic principle led to an accurate method for the determination of hydroxy values in uncured epoxy resins.

    Notes: An attempt has been made to categorize the various materials which have been utilized in polyester compositions to impart or enhance the fire-retardancy of these compositions. Examples of halogen-containing and phosphorus-containing compounds are classified according to where they may be thought to occur in the complex chemical structures which are polyesters. The classifications are basically designed to take advantage of the known chemical reactions which lead to polyesters; the fire-retardant component may be incorporated into the final composition in the diacid, diol, crosslinking agent or may be merely mechanically added as a diluent.

    Notes: Variations were made in the concentration of tin and amine catalysts used in a typical one-short polyether flexible foam system. Differences in foaming characteristics and foam stability were correlated with the relative rates of the chemical reactions which are believed to occur during the foaming process. Physical properties measured on the samples showed in many cases a dependence on the balance between the tin and amine catalysts.

Polytactic polymers are taken into consideration for the most part.     Notes: To date, quantitative stereochemical data are available for a few polymers only. A convenient and graphic representation of this data has been developed. Analysis of this data yields two parameters characterizing a polymer, one representing the tacticity and the other representing the nature of the tactic sequence distribution. The statistical parameters obtained may be related to the polymerization conditions and, hence, to the mechanism of stereoregulation. Furthermore they may be related to a number of physical properties, such as melting point, equilibrium crystallinity, and melting curve. Thus, the proper analysis of the NMR data supplies a link between polymerization conditions and the properties of the resulting polymer.

    Notes: 1,2,2,-Trifluorovinyl phenyl ether and 2,3,4,5,6,-pentafluorophenyl 1,2,2-trifluorovinyl ethers have been prepared, and the nucleophilic reactions of the phenoxide and 2,3,4,5-pentafluorophenoxide salts with tetrafluoroethylene have been investigated. In general, it is some-what difficult to control the reaction so as to produce the desired olefinic monomer. Under truly anhydrous conditions and when other sources of active protons are avoided, the olefin is the chief product if excess tetrafluoroethylene is used. The monomer produced is very reactive with phenoxide ions, and the diphenoxyolefins are the chief secondary products. Both monomers failed to polymerize under normal, free radical conditions. With boron trifluoride an oily material was obtained from the trifluorovinyl phenyl ether. High pressure and gamma rays converted both monomers into polymers.

    Notes: These compounds were synthesized by starting with the addition of the Grignard reagent of bromopentafluorobenzene to trifluoroacetaldehyde and chlorodifluoroacetaldehyde, respectively. The new alcohols, 2,3,4,5,6-pentafluoro-α-(trifluoromethyl) benzyl alcohol and 2,3,4,5,6-pentafluoro-α-(chlorodifluoromethyl) benzyl alcohol, are formed in good yield and undergo smooth conversion to (1,2,2,2-tetrafluoroethyl) perfluorobenzene and (2-chloro-1,2,2-trifluoroethyl) perfluorobenzene, respectively, in excellent yield when treated with sulfur tetrafluoride using n-pentane as diluent. The major pyrolysis product was identified as (2,2-difluorovinyl) perfluorobenzene (α-hydroheptafluorostyrene) and the minor one as perfluorostyrene. Perfluorostyrene was also synthesized in excellent yield by bromination of (1,2,2,2-tetrafluoroethyl) perfluorobenzene and subsequent pyrolysis of the brominated derivative. The dehydrochlorination of (2-chloro-1,2,2-trifluoroethyl) perfluorobenzene provided another good synthesis of perfluorostyrene.

    Notes: These experiments show that ABS can be adsorbed and desorbed from microbial cell surfaces. The adsorption equilibria between the free and adsorbed phases are primarily influenced by the pH of the system. The effects of ABS upon the biochemical activities of bacteria depended upon the amount of ABS adsorbed and the type of bacteria present. The behaviour of ABS is essentially the same whether in the presence of pure or mixed bacterial cultures. Two desorption experiments with soil from a reclaimed water spreading basin which has received sewage over a six month period showed that at least 82% of the ABS “loss” was recoverable from the soil. In all the experiments carried out in this study no evidence was obtained for the biochemical degradation of ABS.

    Notes: Because branched-chain alkyl benzene sulfonate (ABS) is well known to create water pollution problems in surface water and ground water, a study was made on the biodegradation of this surfactant along with other anionic and nonionic surfactants in detergents by water bacteria. Biodegradation was evaluated by the conventional river die-away test employing the methylene-blue method or surface-tension techniques as analytical tools. Under anaerobic conditions, sugar detergents (sucrose esters) seem to be an ultimate solution to water problems created by branched-chain ABS; in aerated water, straight-chain ABS, sulfo tallow methyl esters, and nonionic surfactants are preferable to branched-chain ABS to improve water problems.

    Notes: Shock pressure waves with a frequency of 50/sec. and a maximal pressure of 60-70 kg./cm.2 greatly reduced the viability of E. coli. A similar effect was obtained also on staphylococci, enterococci, Salmonella Zanzibar, and coliphages T2. On the contrary no effect on spores of B. subtilis was found. The effect was practically independent of the maximal pressure within the range from 30-80 kg./cm.2, the highest pressure tested. The effect decreased with increasing cell density of the bacterial suspension. Bacteria surviving shock treatment died considerably more rapidly when stored at 4°C. than bacteria treated in the same way except for exposure to pressure waves. Morphological examinations with the aid of ordinary light microscopy as well as electron microscopy suggest that the effect was due to mechanical action on the bacterial cell walls.

    Notes: Sphaerotilus is usually present in biological waste treatment processes and is considered to be a nuisance organism because it may cause bulking of activated sludge. However, it may play an important role in the decomposition of putrescible organic matter in properly operated biological waste treatment processes. The research described herein is a study of the variety of types of organic compounds which may be decomposed by Sphaerotilus and the effect of some environmental factors upon the rate of decomposition. Although there are some types of compounds which cannot be decomposed by Sphaerotilus, it is shown that this organism is able to assimilate and oxidize a large variety of organic compounds and that it is tolerant of an extremely wide range of environmental conditions. The role of Sphaerotilus in biological waste treatment is discussed in the light of the data presented.

    Notes: It has been found that a bacterium (isolant C12B) grew poorly on 6 and 8 carbon alcohols, well on 10 and 12 carbon alcohols, and slowly but well on the 14 to 18 carbon compounds. Unexpectedly, resting cells oxidized the 6 and 8 carbon compounds most readily and cell-free extracts oxidized 6, 8, and 10 carbon alcohols. Extracts from cells adapted to sodium lauryl sulfate oxidized the 12 carbon alcohol as well, if small concentrations of substrate were used. The bacterium grew on benzene sulfonate and adapted resting cells oxidized that compound. Pure culture degradation studies revealed that isolant C12B used only 11% of tetrapropylene ABS supplied as a sole carbon source, whereas 66% of sodium lauryl sulfate was degraded. Intermediate quantities of kerosene-based alkyl benzene sulfonate and a tallow-derived detergent were utilized. Extracts of another bacterium, TEG-5, grown on tetraethylene glycol, contained enzymes which oxidized polyethylene glycols up to an average molecular weight of 600, but not any larger polymers. This oxidation was visualized by coupling to reduction of ferricyanide.

    Notes: Ferrocyanide-treated beet molasses was fermented in fully baffled mechanically agitated, 2.5-liter, tank-type fermenters using pellets of Aspergillus niger grown in shake flasks as an inoculum. The object of the study was to adapt the fermentation method previously developed in an unstirred tower-type fermenter to a conventional tank-type fermenter. Under selected conditions of stirring (agitator speed 400-700 rpm) and sparging (medium porosity sintered disk), high yields of citric acid (8.6% citric acid, 70% conversion of available sugar) were obtained in 140 hr. of fermentation. There was little relation between citric acid yield and aeration conditions in the fermenter as judged by mash viscosity measurements and sulfite oxidation studies.

    Notes: A method of inoculating nutrient surfaces uniformly with bacterial cells has been developed. The method is based on controlled exposure of samples in a humidified chamber to a spray produced from a bacterial suspension. Factors affecting uniformity of inoculation (ionic strength of suspension, force of spray, distance between spray nozzle and sample, and exposure time) were studied in detail. Under optimum condition using agar medium in Petri plates, differences between total counts for six plates inoculated simultaneously, or between counts for individual 2 cm.2 areas taken at any location on any of the plates, were not statistically significant at the 5% level. The coefficient of variability for counts on 2 cm.2 areas renged from 8 to 12% between tests of 216 counts each.

    Notes: By growing Aspergillus parasiticus (C.M.I. 15957) under controlled conditions on sterilized peanuts an average of 265 mg. of aflatoxin per kilogram of peanuts has been produced and subsequently isolated. Other strains from the A. flavus-oryzae group gave lower yields of aflatoxin and one such strain gave aflatoxin from which certain normal components were absent. The aflatoxin produced on sterilized peanuts by any particular strain of A. flavus-oryzae was shown by thin-layer chromatography to contain the same major components as were produced by that strain on unsterilized whole peanuts.

    Notes: A method by which a paramagnetic oxygen analyzer is used to determine oxygen consumption and carbon dioxide production during aerobic fermentation of Torulopsis utilis is described. The volume of carbon dioxide produced is not necessarily related to the volume of oxygen consumed by the microorganisms. For this reason, all carbon dioxide is removed from the effluent gas before it enters the analyzer when determining oxygen consumption. A method for calibrating the analyzer to correct for this removal is outlined. When determining carbon dioxide, effluent gas bypasses the carbon dioxide absorption tower before entering the analyzer. The carbon dioxide which it contains dilutes the oxygen remaining in the gas. The magnitude of this dilution can be used to calculate the percentage of carbon dioxide produced in terms of the air flow to the fermentor. Equipment required to alternately measure and record oxygen consumption and carbon dioxide production is described.

    Notes: Cobamide production by Butyribacterium rettgeri Barker and Haas (ATCC 10825) was studied on a laboratory scale. Yields of the order of 5 μg./ml. were obtained when this anaerobe was grown for 6 days at 37°C. in a medium containing glucose, cornsteep liquor, cobalt nitrate, calcium carbonate, and sufficient ammonium hydroxide to keep the pH between 6.0 and 7.2. All of the vitamin was found associated with the bacterial cells, and examination of extracts of these cells showed the presence of 5,6-dimethylbenzimidazolylcobamide coenzyme in large quantities. Supplementation of the fermentation medium with a number of benzimidazoles resulted in formation of traces of “new” cobamides with these bases in the nucleotide protion of the cobamide. Supplementation with a number of purines did not result in incorporation of these moieties in the cobamide portion of the molecule.

    Notes: Removal of the pungent factor, allyl isothiocyanate, will partly determine whether mustard seed can become a commerical source for oil and meal in the United States. In processing studies at the Northern Laboratory, the mustard glucoside was converted enzymatically and the pungent oil was removed. This process has now been extended to pilot-plant scale by using filtration-extraction equipment at the Southern Laboratory. After desolventization and further steam stripping, the extracted meal had a residual content of 2.9% crude fat and 0.004% allyl isothiocyanate.

    Notes: A number of factors was found to affect the cup plate assay for lysine with Streptococcus faecalis ATCC 6057. Hydroxylysine, aspartic acid, and Tween 80 were found to be among the essential components of the assay medium. Removal of any of these three components produced diffuse growth zones or otherwise unsatisfactory plates. The inclusion of asparagine and additional dipotassium phosphate to the medium was found to be advantageous but not an absolute requirement. It was important not to autoclave the hydroxylysine and the additional dipotassium phosphate together with the rest of the assay medium, but to add these components to the sterilized medium immediately before pouring the plates. With certain concentrations of hydroxylysine in the medium, the addition of L-glutamine was found to potentiate the effect of hydroxylysine and thereby improve the lysine plates. The effect of graded amounts of the above mentioned medium components and of the pH of the assay medium was also studied. Increasing the sterilization time of the medium from 5 to 10 min. at 120°C. improved the readability of the lysine plates and led to more easily reproducible results. Factors influencing the preparation of the inoculum for the lysine plates were also studied: the age of the stab culture, the amount of inoculum used per plate, and the influence of the nature of the suspension medium used for washing the inoculum.

    Notes: The metabolic pathway of degradation of naphthalene to salicylic acid has been studied by several investigators. The rates of conversion and of cell growth are the basis for the present study. In the early phase of the fermentation, the salicylic acid accumulation followed an exponential curve, as did the bacterial cell accumulation. In the second phase, most of the salicylic acid was produced according to a linear or zero reaction, at rates of 0.3 to 0.5 g./l./hr. A linear accumulation of microbial cell weight accompanied the linear salicylic acid formation. Despite the correlation of growth and salicylic acid production, the type or amount of nitrogen supplied for growth had only a small effect on yield over broad limits. The optimum pH for salicylic acid accumulation was approximately 6.5. Below this value a slower rate of production and lower final yield was seen. Above pH 6.5, a rapid degradation of salicylic acid was observed. A temperature range from 25 to 30°C. was optimum. At 35°C., no growth occurred, while at 20°C. very slow salicylic acid accumulation resulted.

    Notes: Stabilization ponds have been widely accepted in recent years, following studies that established deeper understanding of the phenomena associated with the stabilization process. A compilation of information on the use of both sewage and industrial ponds is presented showing types and geographical location in the United States. A total of 1,304 stabilization ponds currently are used as the principal method of sewage treatment serving a population of 2,138,085. Ponds are used for tertiary treatment serving a population of 2,146,951, in 343 instances. Use of 827 industrial ponds by 31 industries in 44 States is also indicated. Indiana ranks first in usage with 159 ponds and Callfornia second with 140. Ponds were classed as effective by 66 percent of the 455 users who reported on this item. Of 149 reports on odors, 73% indicated nuisances. Insect problems were reported in only a small number of instances. Data are presented on design considerations and the mechanism of waste stabilization ponds. Aerobic ponds are given greatest emphasis. Photosynthesis and its dependence upon the algal mass, suitable temperature, incident light penetration, nutrient supply, and induced vertical mixing by wind are of prime importance in the stabilization mechanism. Odors are associated with prolonged anaerobic conditions, and these may persist up to 4 weeks following extended ice cover in cold climates, if BOD loadings are 25 lbs. per acre per day or greater. Nitrogen and carbon may be limiting factors in the development of an algal mass. Coliform, BOD, and nutrient removals are presented for various physical, chemical, and climatic conditions. Although fecal streptococci removal averaged 97% in a study pond, they were still discharged in the effluent at a rate of 700,000 per second in winter and 70,000 per second in summer. A striking similarity exists generally among the algal speciation in stabilization ponds, regardless of geographic location. The algal mass is, however, dependent upon unique pond conditions and location, and may vary upwards to nearly 5 million algal cells per milliliter, 34,000 p.p.m. by volume, or 30-35 tons per acre per year. The problem of blue-green algal mats and resultant pigpen odors in extremely localized areas is duscussed. Midges and midge larvae may have an impact on the nutrient cycle in a stabilization pond, and may pose a potential localized nuisance in some areas. Mosquito problems have usually been associated with growth of aquatic vegetation. Suggestions for minimizing the potential problem are discussed.

    Notes: A sensitive cup plate assay method for lysine on large plates using S. faecalis ATCC 6057 as the test organism is described in detail. Well-defined zones of dense growth were obtained in a rectilinear dose - response range from 5 to 3000 μg. of L-lysine/ml. Comparative assays on twelve different samples were carried out using the turbidimetric tube assay method with Leuc. mesenteroides ATCC 8042 and the cup plate method with S. faecalis ATCC 6057. Good agreement between the two assay methods was obtained for five samples. For the remaining seven samples the tube assay values were appreciably higher than those obtained by the plate method. This discrepancy is discussed. Pure lysine added to various sample extracts could be quantitatively recovered. The specificity of the lysine requirement of S. faecalis ATCC 6057 in the plate assay was also investigated. Under certain experimental conditions, a weak and variable response was obtained with D-lysine and the polyamines cadaverine, spermine, and spermidine. There was generally no difficulty in distinguishing the D-lysine and polyamine growth zones from the L-lysine growth zones. This growth effect of D-lysine and polyamines could be diminished and even completely abolished by increasing the DL-hydroxylysine hydrochloride level in the plate medium. The lysine peptides glycyl-L-lysine and L-lysylglycine gave excellent growth zones of the same appearance as those produced by L-lysine. The activity of these two peptides in cup tests were on a molar basis about 70% of that of L-lysine.

    Notes: A process has been developed giving over-all yields of about 50%: which could be increased to about 80% with higher material costs and processing time. The preferred method of recovering nucleotides from the cytoplasm has been thermal extraction by means of a heat exchanger followed by filtration, protein precipitation, and carbon adsorption. The concentrated eluate from the carbon is absorbed onto Dowex 1 ion-exchange resin, and groups of nucleotides are removed by pH-defined eluents and recovered by a second carbon adsorption. Finally, standard ion-exchange resolutions have been used to recover individual nucleotides. Other biochemicals in the extract have been investigated, and preliminary work has been done on the combination of nucleotide extraction with the recovery of nucleic acids from the residual cells.

    Notes: In the present paper the authors described laboratory fermentors used in the Antibiotics Research Institute since 1958 for research concerning the technology of the biosynthesis of antibiotics and amino acids and other aerobic fermentations. The lids of the fermentors are made from stainless steel, the vessels are made from glass. The fermentors are placed, always four each, in eight water baths with automatic temperature regulation within a range of ±0.5°C. Details regarding construction, auxiliary equipment, and working procedures used for the sterilization of fermentors and media, inoculation, injection of additives, and defoaming are given. The set-up for one-stage and multi-stage, batch and continuous cultivation are discussed.

Germ-free conditions are maintained during operation by means of a positive pressure of sterile air.     Notes: Microbial polysaccharides which have many potential industrial applications are receiving more and more attention. One of the economic obstacles to the commercialization of these polysaccharides has been the cost of isolating them from fermented broth. To reduce this cost a recycling process was developed. The polyanionic polysaccharide synthesized from glucose by the activity of the bacterium Xanthomonas campestris NRRL B-1459 is precipitated from the fermented broth with a quaternary ammonium compound (QAC).QAC is removed from the precipitate with methanol leaving the water-soluble polysaccharide. Evaporation of the methanol wash removed the methanol and substantially all the QAC was recovered in the concentrate. This was recycled for subsequent precipitations. Chemical assay and the viscosity of water solutions of the isolated polysaccharide indicate no adverse effects on it from recycled QAC. The cost to make this polysaccharide by the described procedure is estimated to be $1.14/lb. This cost estimate includes land, buildings, raw materials, equipment, labor and supervision, utilities, factory supplies, working capital, and plant overhead.

    Notes: Bacillus subtilis NRRL B-2612, isolated at this laboratory, was used to produce polyglutamic acid from devitalized wheat gluten. Conditions for production of the polypeptide were investigated. When NRRL B-2612 was grown on wheat gluten in shaken flasks with phsophate buffer and salts at pH 6.5, yields of polymer reflected a 25-40% incorporation of glutamic acid into polyglutamic acid. Repeated alcoholic precipitation yielded peptides containing between 45 and 90% D-glutamic acid. Physical and chemical properties of a typical purified polyglutamic acid indicated that the product was a gamma-linked polypeptide composed of 75% D-glutamic acid and 25% L-glutamic acid with a molecular weight of approximately 200,000.

    Notes: The distribution of dissolved oxygen in agitated, sparger-aerated aqueous solutions has been investigated by polarographic techniques. The results of the investigations are discussed in relation to earlier studies in bubble distribution in such systems. Liquid-phase mixing is good at agitation intensities well below that necessary to disperse bubbles throughout the liquid.Suspended solids interfere with the transfer of oxygen unless they are freely suspended. Discrete fibers (paper-pulp) are much easier to suspend than molds in either filamentous or pellet forms. Mass-transfer at the cell-liquid interface appears to be unaffected, in both bacterial and mold suspensions, by the intensity of agitation, provided that the organisms are freely suspended.

These mixtures include ternary, quaternary, and quinary systems consisting of normal and isoparaffins, olefins, acetylenes, naphthenes, and aromatics. The method, based upon the mole fraction of the low-boiling component in the mixture, graphically presents the ratio of the actual critical pressure to the pseudocritical pressure as a function of a boiling parameter. For mixtures of more than two components the pseudocritical pressure is based on the critical pressure of the pure low-boiling component and on the actual critical pressure of the mixture consisting of all the remaining higher-boiling components. From an analysis of twenty-seven binary systems (139 compositions) the average deviation of calculated values from reported values is 1.27%. For thirty-four mixtures containing from three to seven components the average deviation is 1.64%.Based upon a similar approach graphical relationships are presented for the estimation of the critical molar density of binary hydrocarbon mixtures of known composition, which also may contain normal and isoparaffins, olefins, nuphthenes, and aromatics. The same boiling point parameter is used to correlate the ratio of the critical density to the reciprocal of the molar average critical volume for nonmethane systems and the product of the critical pressure and the critical volume for methane systems. From an analysis of fifteen binary systems (eighty compositions) the average deviation of calculated values form reported values is 1.54%.

    Notes: Simultaneous mass, heat, and momentum transfer was experimentally investigated for the flow of air through packed and distended beds of spheres saturated with water. The porous spheres were approximately ⅝ in. in diameter. The void fraction of the packed bed was ∊ = 0.416, and the distended beds, whose spheres were positioned in a body-centered cubic arrangement, had respective void fractions of ∊ = 0.576 and 0.778. The distended beds were formed by separating the spheres and connecting them with short lengths of fine rigid wire.Similar relationships for the packed and the distended fixed beds were obtained when the transfer factors were related to the modified Reynolds number. The relationship obtained for mass transfer in packed and distended beds was found to apply to data reported for fluidized beds of spheres.Pressure drop measurements were made which were not exacting but which indicate that the Ergun relationship for packed beds can be used to relate the friction factor to the modified Reynolds number for both the packed and distended beds investigated.

    Notes: Pressure drop measurements were made across packed and distended beds having five layers of smooth plastic spheres, 1.23 in. in diamter, arranged in cubic, body-centered cubic, and face-centered cubic orientations of void fractions varying from 0.354 to 0.882. The distended beds were prepared by separating the spheres with permanently attached short lengths of fine rigid wire. Friction factors were calculated from the overall pressure drop measurements with the Ergun equation and were plotted against the corresponding modified Reynolds number which ranged from 2,550 to 64,900. A single relationship resulted for both types of beds which is independent of the geometric orientation and void fraction of the spheres of the bed.To eliminate entrance and exit effects of the air flowing through the bed pressure drop measurements were also made across the middle layer of each distended bed. Again a single relationship between the corresponding friction factors and modified Reynolds number was obtained which is independent of the geometric orientation and void fraction of the bed.From the two relationships between friction factors and modified Reynolds number the ratio Pk/fk for a packed or distended bed having five layers of spheres is 1.13. This ratio should decrease with increasing number of layers of spheres and approach the limiting value of 1 when the number of layers becomes very large.

    Notes: An elution gas that might be either a single component or a binary mixture, but which in any case was appreciably soluble in the fixed liquid phase on the column matrix material, was used to form a multicomponent liquid phase within the chromatographic column. Vapor-liquid equilibrium was studied both with the solute of interest at essentially infinite dilution conditions and with the solute present at some finite concentration in both phases. From the elution data vapor-liquid equilibrium ratios, or K values, were calculated by expressions relating the solute retention volume to the solute K value in the vapor-liquid system maintained within the chromatographic column.Data were taken for the solutes ethane, propane, and n-butane at infinite dilution in the methane-n-decane system at 160°, 70°, 40°, 0°, and -20°F from 20 to 2,000 lb./sq. in.; for propane at infinite dilution in methane-n-hexadecane at 70°F and 20 to 200 lb./sq. in.; and propane in the system methane-propane-n-decane at 40°F from 20 to 460 lb./sq. in. The univariant gas-liquid-solid locus was experimentally determined for the methane-n-decane binary system.The chromatographically determined K values for n-butane at infinite dilution in methane-n-decane were compared with published static equilibrium values and found to be in substantial agreement. Activity coefficients calculated from the data for all the solute isotherms were compared at atmospheric pressure with the results of the Brönsted and Koefoed relation for estimating activity coefficients of hydrocarbons, and close agreement was again found.

    Notes: The present paper concerns the mechanism of mass transfer accompanied by a first-order irreversible chemical reaction between two phases. Based on the film-penetration concept a general mathematical model to describe the physico-chemical behavior at the interface has been formulated. Mass transfer mechanism may be analyzed and evaluated in terms of the dimensionless groups appearing in the derived equations.For limiting conditions the derived general equations can be reduced to those based on the simple postulations such as the film theory, the penetration theory, and the surface renewal theory. For nonlimiting cases the film-penetration concept provides information which cannot be obtained by either the film theory or the surface renewal theory alone.Experimental results appearing in literature show that the physical mass transfer coefficient is proportional to the molecular diffusivity to the v-th power and that v varies widely between 0.15 and 1.0. The film-penetration concept theoretically predicts this v-variation, whereas, in accordance with the film theory or the surface renewal theory, v has to be a certain fixed value.It is shown that if an accurate physical mass transfer coefficient is available, the film-penetration concept, the film theory, and the surface renewal theory all predict practically the same effect of chemical reaction on the mass transfer rate. However if the chemical mass transfer coefficient is to be predicted without an accurate physical transfer coefficient, the choice of the theory or the mechanism may become important.

    Notes: When drops in a two-liquid phase chemical reactor are able to mix with one another by coalescences and redispersions, any spread of concentration among the drops tends to be averaged out. This phenomenon can affect average reaction rate and selectivity in non first-order reactions or mass transfer rate controlled reactions in the dispersed phase. It was found that for mass transfer rate controlled reactions, or the equivalent zero-order reaction, quite large dispersed phase mixing rates are required to bring conversions close to the level obtained with complete mixing.
