    Notes: Summary The plasma concentration profile of oxprenolol after intravenous bolus injection, during intravenous infusion and following sustained oral administration was studied in a total of 106 patients with coronary heart disease. Speed of onset of pharmacodynamic activity, as measured by suppression of isoprenaline tachycardia, was discernible within a few seconds of central injection and complete within 5 min in all patients; variability in response was small. Following both i.v. bolus and intravenous infusion, plasma oxprenolol concentrations showed considerable between patient variability The plasma concentration/time profile observed in 16 patients following single intravenous oxprenolol bolus therapy was substantially higher, particularly during the early distribution phase, than observed and predicted volunteer data. Higher plasma oxprenolol concentrations were also attained during the more extended time sampling of the infusion studies; these findings would be compatible with reduced oxprenolol clearance in patients with ischaemic heart disease. During chronic oral therapy there was a many-fold between-subject variability in plasma concentration achieved following a given ingested dose. Correlation of antagonism of exercise tachycardia inhibition with plasma oxprenolol concentration in 15 male volunteers demonstrated near complete blockade of exercise stimulation of chronotropic beta-adrenoceptors at an average plasma oxprenolol concentration of 150 ng/ml. In coronary heart disease, such plasma concentrations can most conveniently be achieved by a 4 mg oxprenolol intravenous bolus with simultaneous infusion of 0.05 mg/kg/h; however, these studies provide sufficient information to allow alternative regimens to be derived should lesser plasma concentrations be considered desirable.

    Notes: Summary The pharmacokinetic profile of Sobrerol, a mucolytic drug, has been studied in patients with acute exacerbations of chronic bronchitis and dense sputum. In addition to measurement of serum and urine levels, the concentration in bronchial mucus was also examined, and their correlation was calculated. Mass fragmentographic analysis was used to assay free sobrerol and its principal urinary metabolites hydrated carvone and glucuronidated sobrerol. After the doses and administration routes used, there appeared to be accumulation of sobrerol in bronchial mucus. This is a feature of great interest and value for a drug which has the specific action of liquefying mucus.

    Notes: Summary An HPLC analytical method was applied to the determination of plasma concentrations of 5,6-dihydro-5-azacytidine (NSC 264880, DHAC) in two foxhounds after a rapid intravenous infusion of 300 mg/kg DHAC. The dose employed is the mouse equivalent LD10 dose which results in mild reversible toxicity in the dog. The decline in DHAC plasma concentrations was greater than three log decades after dosing. The plasma concentration-time data was computer-fitted by NONLIN to a three compartment open model with first-order elimination using the equations for a short intravenous infusion. The half-lives corresponding to the three exponential phases were: t1/2α =5.78 min, t1/2β =1.57 h and t1/2γ =22.0 h in dog 1 and t1/2α =7.41 min, t1/2β =2.25 h and t1/2γ =21.6 h in dog 2. The terminal phase of the plasma concentration time profiles represented a minor contribution (2.2–3.2%) to the total area under the curve. The plasma concentration time data for the first 12 h after dosing was computer fitted to a two compartment open model. The initial and terminal half-lives determined from the two compartment fits were similar to the t1/2α and t1/2β values of the fits to the three compartment open model. Similar total body clearance values were calculated from the areas under the plasma concentration time curves from time zero to infinity for the computer fits to the three and two compartment models, respectively. Thus, for practical purposes, it appears feasible to define adequately the total body clearance of DHAC by analysis of the plasma concentration time data during the time interval in which the plasma concentration is described as a bi-exponential equation. Renal excretion of the parent drug is the principal route of excretion of DHAC from the dog.

    Notes: Abstract The pharmacokinetics of methotrexate (MTX) and 7-hydroxy-methotrexate (7-OH-MTX), a major metabolite, were investigated in rabbits after intravenous bolus injection and infusion using a specific HPLC assay. The arterial sampling (from the carotid artery) was used in all the studies since peculiar and significant arterial-venous differences in the plasma concentration of MTX and 7-OH-MTX were found following bolus administration of the drug. The disposition kinetics of MTX appeared polyexponential with a small terminal phase having a half-life of 10.2–27.5 hr. Extensive formation of 7-OH-MTX occurred at the two dose levels (15 and 50 mg/kg). Nonlinear disposition of MTX was reflected in several aspects of data analysis. A disproportionate increase in the AUC with dose was observed. An increase in dose not only reduced the mean total body clearance (7.49 vs. 4.26 ml/min/kg) and renal clearance (4.89 vs. 2.76 ml/min/kg), but also prolonged the mean residence time (26.2 vs. 43.3 min). The steady-state volume of distribution (Vss) of MTX was estimated to range from 0.16 to 0.25 L/kg. More than 90% of the dose was excreted as MTX and 7-OH-MTX within 8 hr after dosing. Renal clearances decreased with the increasing plasma levels, suggesting active tubular secretion as one of the excretion mechanisms. A similar pattern for renal clearance of 7-OH-MTX was obtained. Infusion studies of 7-OH-MTX revealed that this metabolite had a longer residence time and a larger Vss as compared with MTX, which were in accordance with its physicochemical properties. Essentially complete doses of 7-OH-MTX could be recovered in the rabbit urine.

    Notes: Abstract This study was conducted to evaluate the influence of route of administration upon the bioavailability and pharmacokinetics of methylprednisolone sodium succinate. Fourteen healthy adult male volunteers received 40 mg doses of methylprednisolone as the following treatments after an overnight fast in a 4-way crossover design: (a) as a 1 ml i.v. bolus;(b) as a 1 ml i.m. injection;(c) administered as an oral solution;and (d) as 5×8 mg oral tablets. Both the ester and free methylprednisolone were determined in plasma and urine. Study results indicate that the ester is rapidly and extensively converted to free methylprednisolone after all routes. The extent of methylprednisolone absorption was equivalent after i.v. and i.m. administration. Both orally administered treatments resulted in a lower extent of absorption attributed to a first-pass effect. Although a slightly lower extent of absorption was demonstrated following the oral administration of the methylprednisolone sodium succinate solution relative to the methylprednisolone oral tablets, this average difference of 9% would probably be of minimal therapeutic importance.

    Notes: Abstract The present study was conducted to develop, test, and apply a simple exponential time-dependent volume of distribution function to describe the pharmacokinetics of compounds following instantaneous, zero-order and first order input. Simulations were used to show the applicability and flexibility of the equations. Experimental data from the literature were fitted using the equations developed in the present study. In addition, the results of these fitted curves were compared to the results of the original fitting procedures to compare and contrast the methods. In the present analysis, the change from an initial volume (V1)to the total volume (VT)is perceived as a simple exponential function. Therefore, a single exponential term to describe elimination and a single exponential term to describe the change from V1 to VT were used to describe the entire blood concentration profile. The results from present simulations and fitted data indicate that the transition from V1 to VT is a more continuous process than observed with classical methods and is consistent with results obtained from physiologic flow-limited models. This observation suggests that the present curve-fitting technique may be more akin to physiologic reality, in that it depicts the change from the initial volume of distribution to the total volume of distribution as a continuous exponential function which reflects the establishment of an equilibrium.

    Notes: Abstract The pharmacokinetics and pharmacodynamics of intravenous bumetanide (0.250 mg/kg), alone (treatment I) and after probenecid pretreatment (treatment II), were studied in four mongrel dogs. Lactated Ringer's solution was administered by vein throughout both treatments at a flow rate of 2 ml/min to avoid fluid and electrolyte depletion. Bumetanide and probenecid concentrations were analyzed by HPLC, sodium by flame photometry, and creatinine by colorimetry. Although the probenecid markedly reduced the plasma and renal clearances of bumetanide, as well as the fraction excreted unchanged in the urine, there was no significant difference between treatments I and II in the 4-hr natriuretic and diuretic responses. However, analysis of the dose-response curves between treatments I and II showed that sodium, excretion was better correlated with bumetanide urinary excretion rate than with plasma concentration. The reasons for a poor correlation between treatments during the early time periods are discussed.

    Notes: Abstract After bolus intravenous dosing of d-tubocurarine (d-TC) to rats, the twitch heights of the tibialis anterior muscle indirectly stimulated were followed, and its decrease was defined as pharmacologic response of d-TC. The relation between dose and response intensity was found to be well described with Hill's equation. According to a theory proposed by Smolen, Hill's equation was also applicable to the biophase d-TC concentration-response relation; the time courses of the relative biophase d-TC concentration indicated linear kinetics with dose levels ≤0.15 mg/kg and the occurrence of dose-dependent disposition with 0.30 mg/kg. After bolus i.v. dosing of3H-d-TC, plasma d-TC concentration obeyed a dose-independent two compartment model with doses ≤0.15mg/kg, but not with 0.30 mg/kg. This finding matched the above estimated with pharmacologic data. The active metabolite was not found in plasma and urine. The extent of d-TC plasma protein binding was independent of the wide range of plasma levels and its mean (±SD) value was 30.5 (±3.8). Plasma d-TC levels and pharmacologie response intensity were well correlated by Hill's equation and a three compartment model (the general two and the biophase compartments) in the dose range ≤0.15 mg/kg.

    Notes: Abstract Naltrexone release rates from a controlled release delivery system have been quantitated over a time period greater than one month in the monkey. The method requires calibration of the pharmacokinetic parameters of each monkey utilizing an intravenous bolus dose and assay of unchanged naltrexone levels in plasma as a function of time after dosing. Also required are periodic plasma levels of unchanged naltrexone obtained subsequent to administration of the delivery system. Release rates are then calculated as well as the total amount released. Application of the methodology to a biodegradable copolymer naltrexone delivery system in three monkeys showed an initial release rate of 3– 8% of the dose per day over the first 3– 5 days followed by a slow, rather constant release rate of 1– 3% per day from day 5 to the time of the last measurable plasma sample (36– 43 days). Comparison of alternative calculation methods using both experimental and simulated plasma naltrexone data verified the accuracy of the release rate calculations. The sum of the calculated total amount of naltrexone released plus the assayed amount remaining in the delivery system after removal from the animal accounted for 91– 94% of the administered dose in the two monkeys in which complete data were obtained.

    Notes: Summary Using a paired ion exchange high pressure liquid Chromatographic assay, pharmacokinetic evaluation of methyl glyoxal bis guanylhydrazone (methyl-GAG) was performed in nine male New Zealand albino rabbits following administration of a single intravenous bolus dose of 50 mg/kg B.W (550 mg/m2 BSA). Blood samples were collected before and at intervals of 5, 10, 15, 30 min and 1, 2, 3, 4, 6, 8, 12, 18, and 24 h after administration of the drug. The analysis of experimental data indicates a three compartment open model with first order elimination from the central compartment described by the equation C pt =A.e−αt +B.e−βt +C.e−γt , where A, B, C, are 107.985, 4.785, and 0.763 μg/ml, respectively, α, β,γ, are 5.466, 0.487, and 0.030 h−1, respectively, and $$T_{\frac{1}{2}\alpha ,\beta ,\gamma } $$ are 7.6, 85.3 min and 23.1 h, respectively. The mean volume of distribution in the central compartment V c was 0.44 liters (l)/kg, volume of distribution Vd area 30.326 l/kg, and the total body clearance 0.9097 l/kg/h. The existence of a long terminal plasma half life of methyl-GAG reported previously in human studies was also confirmed in experimental animals and may explain the cumulative toxicity of this drug.

    Notes: The chromatographic behavior of phenols in reversed-phase mode liquid chromatography differs from that of non-ionic compounds such as alkyl alcohols, alkylbenzenes, halogenated benzenes, polyaromatic hydrocarbons, and aromatic acids. Therefore, the retention times of 61 phenols were measured in a system of an octadecyl bonded silica gel and acetonitrile/water mixtures. The logarithm of the capacity ratio (log k') was found to be a linear function of the hydrophobicity (log P) in acidic acetonitrile/water mixtures. This result was applied to a different octadecyl bonded silica gel. Eight phenols were selected as standard compounds, and their log k' values were measured in 0.05 M phosphoric acid in 10 to 90% acetonitrile/water mixtures. An empirical polynomial relation was obtained between the concentration of acetonitrile and the slope of the log k' vs log P curve. Finally the capacity ratio of all phenols were calculated in given eluents by the equations derived from the measurements of standard compounds and the calculated log P values. The difference between predicted capacity ratios and measured ones was within 10%.

    Notes: Silane-coupling agents, commonly used for fiberglass reinforced plastics (FRP), were applied for deactivation of silanol sites in glass capillary columns prior to coating with mediumpolarity phases such as Carbowax and Superox. The columns, prepared in a two-stage process in the case of glass (acidic leaching and drying, dynamic cold silanization followed by static coating with the phase) or a one stage process in the case of fused silica, gave the best results when deactivation and hence wettability were induced by glycidoxypropyltrimethoxy silane.

    Notes: A test mixture containing 28 compounds plus their stable isotopically labeled analogs was analyzed on a daily basis for one month during a series of routine fused silica capillary column GC/MS analyses in order to establish the precision with which retention times and responses were produced. These long-term precision data were evaluated to determine how to best predict retention times and how to best reproduce quantification. Results clearly indicate that relative retention times should be calculated using the reference compound eluted most closely to the target compound and that quantification should be based on relative response using a chemically “similar” compound.

    Notes: A method is described for the determination of fenvalerate (cyano(3-phenoxyphenyl)methyl 4-chloro-α-(1-methylethyl)-benzeneacetate) residues on woolen cloth. The fenvalerate was Soxhlet-extracted from the treated fabric with acetone for four hours at a reflux rate of six solvent exchanges per hour. The fenvalerate residue was determined by gas-liquid chromatography with a flame ionization detector. The results were quantitated by comparing integrator counts obtained from extracts of treated samples with those obtained from known standards. The method was sensitive to 0.00054% fenvalerate by weight. Recoveries from fabric treated with 0.00054% to 0.215% averaged 94.8%. The response of the detector was linear over a 400-fold range of fenvalerate concentrations.

    Notes: Five commercial varieties of uncoated fused silica capillary tubing used in high resolution gas chromatography were subjected to cobalt-60 gamma radiation. The advantages and superior performance of fused silica open tubular columns coated with OV-1 and subsequently immobilized or crosslinked in situ by radiation is reported in a previous paper in this Journal; however, in this investigation a loss in flexibility of irradiated capillaries was also noted. Quantitative measurements on flexibility were performed on specimens of irradiated fused silica capillaries, indicating that changes do indeed occur upon irradiation. Scanning electron micrographs also illustrate slight deterioration of the outer protective polyimide coating. Most surprisingly, it was also established that a variation does exist in the flexibility of the commercially available raw material studied.

    Notes: An ion mobility detector that has been specifically developed for interfacing with capillary gas chromatography is investigated in the negative ion mode. Like the electron capture detector, to which this instrument is closely related, the ion mobility detector shows an enhanced response to low molecular weight halogenated compounds when a small quantity of oxygen is doped into the make-up gas flow. Under O2 doping conditions, the device can operate in a reactant ion monitoring mode responding universally to compounds capable of capturing thermal electrons and in a tunable selective product ion mode providing increased selectivity over that achieved by the ECD. At an oxygen concentration of 0.5%, minimum detectable amounts as low as 600 femtograms have been realized for carbon tetrachloride. Selectivity of chloro- versus bromo- compounds is demonstrated using a mixture of p-dichlorobenzene and p-dibromobenzene.

    Notes: Interest in monitoring halogenated organics in drinking water and natural surface and ground water in the low ppb range continues to grow. There is a tendency to include still more volatile halocarbons, the trace determination of which is known to be rather demanding. This prompted us to re-examine the feasibility of large-volume direct aqueous injection onto capillary columns, coupled with ECD. A primary problem was to avoid simultaneous elution of water with halocarbons, since water suppresses the ground current of the ECD. The following measures contributed to the solution of this problem.Apolar, extremely inert, columns are required to elute water completely, and even before very light halocarbons. Their coatings have to be far thicker (≍ 5 mUm) than commonly employed thick films since they must permit isothermal analysis at a column temperature around 100°C in order to ensure rapid and complete elution of water. Finally, it is essential that sampling be carried out on-column for two reasons: diffusion of water vapor in the injector, resulting in delayed elution, is then eliminated, and peak distortion during splitless injection is avoided.Although we now know that persilylated columns with immobilized coatings withstand routine water injections, more longterm experience is needed to provide detailed recommendations for the handling of these columns.

    Notes: Four para-substituted cholesterol cinnamates were synthesized and evaluated for utility as gas chromatograph (GC) liquid crystal stationary phases. The capability of the phases to separate olefinic geometrical isomers was found to be dependent on the position of the olefinic bond. When unsaturation occurred at positions four carbons or greater from the terminal methyl of the compounds investigated, the relative ability of the phases to separate geometrical isomers was p-Cl 〉 p-Me 〉 cholesterol cinnamate 〉 p-MeO derivative 〉 p-NO2. When unsaturation occurred at positions four carbons or less from the terminal methyl the relative ability of the para-derivatives of cholesterol cinnamate to separate geometrical isomers was reversed.

    Notes: Conditions for maximizing resolution and signal-to-noise ratios in fluorescence scanning densitometry of separations obtained by high performance thin-layer chromatography are described. Resolution was found to be independent of most instrument parameters with a small decrease observed at wide slit widths. Signal was found to be markedly dependent on experimental parameters such as slit width, slit height, sensitivity setting, and the time constant of electronic filters. For recording chromatograms a slit width of 1.00 mm, a slit height slightly greater than the spot diameter, a low sensitivity setting (50 mV), electronic filtering, and a scan rate of 24 or 48 mm min-1 are commensurate with minimum recording time and maximum sample detectability without incurring a significant decrease in resolution.

    Notes: A study has been undertaken on the effect of temperature on retention characteristics in thin-layer chromatography (TLC) with low-volatility mobile phases (MP). It is shown that temperature variations in TLC in melts bring about variations in both the relative retention values and, in some cases, in the order of migration of the chromatographic zones across the layer. The variation in the capacity factor k' with temperature agrees well with Martin's equation. To explain the temperature dependence of Rf one must, in general, take into account the variations with temperature in both the partition coefficient and the phase ratio. To describe the variation in Rf with temperature in TLC with low-volatile MP one can use an approximate equation in which In Rf is a linear function of 1/T. The experiments indicate that temperature is a major factor in TLC in melts.

    Notes: It is well known that removal of metal ions from the glass surface greatly improves both the thermal stability and the adsorption characteristics of glass capillary columns. Usually metal ions are removed from a glass surface by acid leaching; in particular, the static leaching procedure with hydrochloric acid is used very often for the preparation of glass capillary columns. This paper discusses the results of an investigation into dynamic leaching. Special attention is paid to those factors which affect the adsorption properties of the leached surface. The importance of the non-specific adsorptivity of the capillary surface due to changes of surface properties induced by the leaching process is pointed out.

    Notes: Fused silica capillaries deactivated with D4 and coated with OV-1 were subjected to cobalt-60 gamma radiation in order to elucidate the effect of incident radiation on column performance. The chromatographic performance of these open tubular columns in which OV-1 was polymerized in situ was found to be dependent on the dosage of radiation and was evaluated before and after the irradiated columns were rinsed with solvent. Of the radiation dosages employed, 3 MRad produced superior chromatographic performance coupled with very favorable residual surface activity.

    Notes: Two-dimensional gas chromatography with two capillary columns of different polarity (SP 2100 and OV 225) is used for pesticide residue analysis in food samples. By means of “live chromatography” applying pneumatic switching technique according to Deans, unequivocal identification of 57 organophosphorus pesticides in food samples is achieved at trace concentrations. The instrument is equipped with only one injection port and one flame photometric detector. On-line data processing is very helpful, especially in calibration and checking the system's reliability with the multitude of test compounds. The complete pesticide residue analysis including clean-up of about six food samples can be completed by one person in 8 hours.

    Notes: The usefulness of pump or loop injectors in preparative liquid chromatography was evaluated. The concentration distribution of samples flowing through a UV detector cell, directly connected to the injector, was recorded at various flow velocities, at various diameters of loop tubing, and using various methods of injection [injection with sample volumes equal to a fraction or total loop volume, etc.]. The most advantageous methods were found to be either use of a loop injector to inject only a fraction of its total volume or use of a pump. Both of these methods ensure a almost rectangular concentration distribution.

    Notes: A comparison of the effects on peak shape is made between a 350 and 100 microliter displaced coaxial and a 350 microliter concentric cylinder electron capture cell. Peaks as fast as W1/2 = 0.5 second were detected using electronics operating in the constant current mode. The measured peak shapes are compared to those obtained by computer simulation assuming either complete mixing or plug-like flow within ECD cell. The advantage of controlling the flow pattern through the cell to obtain plug-like flow is demonstrated. Differences in peak shape between the displaced coaxial and concentric cylinder cells of the same volume are explained in terms of the electrostatic fields within the cells.

    Notes: A novel solute focusing technique for on-column injection of liquid samples onto capillary GC columns is described. The focusing technique allows injection of 8.0 microliters or more of sample without producing the peak distortion or splitting observed under conventional on-column injection conditions. The experimentally determined performance of the technique is given for a wide volatility range sample. Solute focusing is useful in situations where on-column injection of 1.0 microliter or greater is required.

    Notes: A comparison is made between air and carbon dioxide as secondary cooling gases in cold on-column injection into capillary columns. Because CO2 is a more effective coolant than air, the injection zone can be kept at a colder temperature, thus preventing loss of sample vapor through the open injector even as the initial oven temperatures because greater than the boiling point of the solvent. Sample loss at these elevated temperatures is shown as a reduction in peak area as functions of column temperature during injection and different injection zone temperatures. The maximum allowable oven temperature during injection is significantly higher when CO2 is used as secondary coolant rather than air.

    Notes: Recent developments in the use of negative ion detection in GC-MS have indicated the potential for highly sensitive analyses for certain compounds. This paper describes the application of the techniques to the assay of Nitroglycerin and its metabolites yielding sensitivity to two femtograms. Because of the low levels of nitroglycerin in biological media such sensitivity is highly desirable if not essential for meaningful measurements. In addition to sensitivity the method is shown to yield extremely good selectivity contributing further to the credibility of quantitative results.

    Notes: A high-performance thin-layer chromatographic (HPTLC) method is described for the determination of tributyltin compounds (bis(tri-n-butyltin) oxide, TBTO, and tri-n-butyltin naphthenate, TBTN) and their degradation products (dibutyltin and monobutyltin compounds). The organotin compounds are extracted from wood with ethanol containing 0.5% (v/v) of hydrochloric acid and the separation of the defferent kinds of organotin compounds is achieved by thin-layer chromatography. The sample spots are measured using a scanning densitometer after decomposing the organotin compounds to inorganic tin by ultraviolet irradiation and visualization of the spots with pyrocatechol violet.Applications of the method to detection and quantification of organotin compounds in preservative solutions, in recently impregnated wood, and in wood samples from five-year-old window frames are described.
