    Notes: The selectivity of a column system, S, defined by equation 28, includes a variation coefficient (β) of plate number with capacity ratio which has a large influence on the peak capacity, as shown in Fig. 18. Some typical chromatograms are given. In order to predict S for a column system from Kovat's Index, equations 40 and 41 are given to calculate the constant of the carbon number rule for squalane at different temperatures or for different stationary phases. The specific retention value of heptane on squalane at different temperatures can be calculated from equation 42. The nonpolarity index, defined by equation 43, was used to calculate the retention value of heptane on various stationary phases. In liquid chromatography, the order of elution may be reversed by changing the composition of the eluent on the same chemically bonded silica (manufactured in China). The linear relations between the log retention values of different kinds of solutes or of a single solute on the silicas of different surface areas when using the same eluent are given.

    Notes: The sensitivity of an FID may change when the carrier gas flow rate changes during a chromatographic run. Sample parts which are eluted at reduced FID sensitivity produce a reduced peak area, hence are discriminated as compared to other components. Sensitivity changes were studied for hydrogen as carrier gas. For the detector tested, differences in the carrier gas flow rates of 1 ml/min shifted the FID sensitivity by 1 to 5% (depending on the fuel gas supply). Thus the stability of the sensitivity is no longer ensured as soon as the carrier gas flow rate is changed manually or by an automatic programmer during an analysis. Sensitivity drifts may also occur during temperature programmed runs with a pressure regulated carrier gas supply since the gas flow through the capillary drops with increasing temperature. Such shifts in the response became noticeable as soon as relatively high carrier gas flow rates combined with long range temperature programmes were used. The typical patterns of such discriminations are shown, closing with a discussion on the possibilities for minimizing such undesired effects.

    Notes: Fused silica is an evolutionary product of continuing efforts to improve our chromatographic capabilities. It offers the normal user new horizons in inertness, chromatographic reproducibility, ease of column installation, and possibilities in on-column trapping. Since this writing, chemically bonded phases in several film thicknesses became available on fused silica columns, and these are especially well suited to on-column injections. The use of fused silica syringe needles to inject directly onto fused silica columns seems particularly interesting. Evolution is, of course, a continuous process and it would appear that we can expect continuing developments in this exciting area.

    Notes: This paper shows how simply and yet very rapidly fatty acid esters of monohydric alcohols, but particularly partial and full fatty acid esters of fully hydric alcohols can be separated and determined by means of high-pressure liquid chromatography on LiChrosorb RP-8 with methanol/water. We have separated quantitatively the methylesters of the fatty acids C8:0 to C22:0 and C24:0, the i-propyl-, i-butyl-, n-hexyl- and i-octyl-esters of the even-numbered fatty acids C8:0 to C18:0, mono- and difatty acid esters of the 1,3-bis-(2-hydroxyethyl)-5,5-dimethyl-hydantoin, mono-, di- and triesters of the trimethylolpropane as well as the tetraesters of the penta-erythrite.

    Notes: This paper demonstrates how simply and rapidly fatty acid esters of mono- and polyhydric alcohols can be separated quantitatively in preparative quantities on Lobar® RP-8 packed columns. After the separation of unknown mixtures, the isolated esters are identified from spectroscopic data (IR/NMR) and, after saponification of the ester components (fatty acids and alcohols), from the retention times of gas and high-pressure liquid chromatographic separations. Thus, in particular, sparingly volatile or nonvolatile partial esters of polyhydric alcohols, as well as the long-chained full esters, can be determined qualitatively and quantitatively.The following fatty acid esters of mono- and polyhydric alcohols have been separated: the i-propylesters of the laurinic and myristinic acids, the i-butyl-, i-octyl- and i-octadecyl-esters of the palmitinic and stearic acids, the mono- and di-fatty acid esters of the 1,3-bis-(2-hydroxyethyl)-5,5-dimethylhydantoins, the mono-, di- and tri-esters of the trimethylalpropane and the full esters of the pentaerythrite.

    Notes: Methanol, 2-Me-propanol-2, propanol-2, benzene, toluene and xylenes in gasoline blends were determined using a valveless multi-dimensional switching system (MDSS) with two packed columns and temperature programming.Compounds of interest, interfering with paraffins and naphthenes, were in a sequence of four heart cuts transferred onto the second column. The resulting baseline separation enabled precise quantitative measurement within a short analysis time. Instrument settings, times of switching and integration parameters were stored in an instrument memory file (user table) and thus high precision in the measurements was achieved.

    Notes: Stable silica gel sorbents with aliphatic or aromatic groups are formed by chemical modifications of the silanol groups with special reactive silanes. Various lipophilic surface modifications on silica gels with varying pore structures are tested with regard to their chemical and physico-chemical characteristics, their wettability and their chromatographic retention data. The main problem in TLC is the preparation of abrasion-resistant layers on glass or on foils which meet the usual high standard of quality and are also suited for quantitative determinations. Thin-layer chromatography on reversed-phase layers can only be performed if the complete wettability of the lipophilic stationary phase in dry form is guaranteed by the mobile phase. Adsorption-chromatographic separations with lipophilic eluents and reversed-phase partition-chromatographic separations with hydrophilic eluents are performed, for example, with dyes, with polycyclic aromatic hydrocarbons and with lipids. The great differences in selectivity caused by the various modifications of the sorbent and the varying eluent composition are remarkable. Ready-for-use layers with lipophilic surface modifications complement the existing range of pre-coated layers and thus widen the application of TLC and HPTLC considerably.

    Notes: A gas-chromatographic separation of underivatized steroids is described, in which the liquid crystal N,Ń-bis(p-phenylbenzylidene)α,α′-bi-p-toluidine (BPhBT) was used isothermally at 265°C for the stationary phase. The nematic range of this crystal is 247 to 403°C. Use of the BPhBT crystal allowed operations in a temperature range where steroids are volatile and unlikely to display broad elution peaks and long retention times.With columns precoated with 2.5 weight percent of BPhBT, steroids containing minor structural differences in the ring system were readily differentiated. For example, 5-androsten-17α-ethinyl-3β, 17β-diol was found to elute more rapidly than its 3-keto analog, ethisterone. Ethisterone was distinguished readily also from its 19-nor analog, norethisterone. Additionally, differences in side-chain functionality and resultant molecular dimensions facilitated the separation of steroids of identical ring structure, as in the comparison of cholesterol with methyl 3β-hydroxy-5-cholenate and 25-hydroxycholesterol.An enhancement of separation according to the molecular length-to-breadth ratio was observed; in general, the larger this length-to-breadth ratio, the greater the retention period on the column.

    Notes: High performance thin-layer chromatography was used for the separation of nine therapeutically important and structurally similar β-adrenoceptor blocking drugs on silica gel plates. In situ scanning of the plate in the reflectance mode gave detection limits in the low nanogram range depending on the individual drug and the wavelength selected for its determination. Scanning the HPTLC plate at several wavelengths can be used to aid the identification of partially resolved components. Continuous development and continuous multiple development techniques are compared for the simultaneous separation of the drug mixture.

    Notes: The promising technique of controlling chromatographic selectivity by the adjustment of individual column temperatures in systems of series-coupled columns is investigated by means of a general model incorporating the effects of temperature and mobile phase compressibility. Expressions are derived for the linear flow velocity, the effective partition coefficient and the retention time for a system of n columns assuming an ideal mobile phase gas, under conditions of constant overall pressure drop and neglect of the temperature dependence of the mobile phase viscosity. The results indicate the importance of thermodynamic parameters, relative to parameters influencing the linear flow velocity, in determining the effect of temperature on the chromatographic retention time. Numerical results are illustrated graphically for two-column systems which are discussed in greater detail. Switching of columns is also discussed and it is shown that even if thermodynamic contributions remain unchanged, non-thermodynamic contributions have a notice-able effect.

    Notes: “Leaching” or “etching” by strong mineral acids seems to be a necessary pretreatment step for the most commonly used deactivation procedures of glass capillaries by reaction with either polyethylene glycol or silylation reagents. The acidic sites which are formed on the surface during this acid treatment cannot be completely removed by the subsequent deactivation process. This drawback can be overcome by performing the leaching with water vapour, resulting in an accumulation of cations at the surface and a decrease in the number of silanol groups. Capillaries of this type show excellent properties for the chromatography of strongly basic compounds. After the wash-out of the alkaline surface layer, the acidity of the support is suited for the chromatography of strongly basic as well as strongly acidic compounds. Due to a lack of reactive acidic sites, special deactivation procedures have to be applied to capillaries produced in this way.

    Notes: A new surface pretreatment for the preparation of wall-coated open tubular (WCOT) glass capillary columns has been evaluated. This technique involves the application of a non-extractable layer of Superox™-4, a 4,000,000 MW polyethylene glycol, to the glass surface as a pretreatment and deactivation agent. Unlike other polyethylene glycols, Superox-4 is stable at high temperatures (〉 300°C) in the absence of oxygen, coats smoothly onto a bare glass surface and resists droplet formation. WCOT columns (SE-54, Carbowax 20M, and SP-2250) prepared using this technique are compared to columns prepared using a modified Grob [2,3] BaCO3 procedure. The Superox-4 pretreated columns were equal or superior in quality to the BaCO3 pretreated columns, based on the appearance of an activity standard and the effective theoretical plates (Neff) per meter. Chromatograms showing practical application of the WCOT columns prepared using both methods is given.

    Notes: Glass capillary columns are ideal for analysis of urinary steroid profiles (i.e. the total steroid neutral fraction without prior separation into sub-groups).In this paper performance of both SCOT and wide-bore WCOT columns has been compared, resulting in no significant quantitative difference on urine extracts run as trimethylsilyl ether derivatives. However, because of increased efficiency the WB-WCOT column exhibited more sensitivity in measurement of small amounts of 11-ox-aetiocholanolone. Sensitivity of other steroids was greatly enhanced by use of either SCOT or WB-WCOT, leading to an almost tenfold increase over conventional packed columns (e.g. androsterone and aetiocholanolone 〈 40 μg per 24 h urine sample). A simple splitless injection system (based on a design of Dr. W. Greenaway, Oxford University) is presented.Experiments were also carried out using a SCOT column in conjunction with an FM-electron capture detector. Whilst efficiency was impaired due to the design of the detector, use of He (carrier) and N2 (make up gas) showed that several steroid derivatives could be easily measured at well below the 10-10 g level with much more rapid retention times than with a 1½m packed column. Highly successful separation of 19 amino acids (as fluoro-acyl derivatives) was achieved in 35 minutes using a SCOT (30 m) SP2100 column.

    Notes: A method for the HPLC separation of phosphatidylglycerol (PG), phosphatidylinositol (PI), phosphatidylcholine (PC), and sphingomyelin (SPH) was achieved using five in-series columns packed with LiChrosorb, Partisil, and μ-Porasil adsorbents, a solvent mixture of chloroform/methanol/ammonium hydroxide (50 : 36 : 6.7, by volume), and a Pye LCM2 Moving Wire (FID) detector. The same phospholipid mixture was also separated using four μ-Porasil columns with the same eluent and detector. The latter conditions were found to be suitable for the analysis of phospholipids obtained after centrifuging, extraction, and precipitation of surface-active lipid components of patient amniotic fluid collected at amniocentesis section. The lecithin/sphingomyelin (L/S) ratios, determined by the HPLC method, correlated well with those determined by the TLC technique in four normal pregnancies, whereas results of shake tests did not correlate too well with L/S ratios determined by the above two chromatographic methods. Besides the lecithin/sphingomyelin ratio, the present method was able to supply additional information: the concentrations of phosphatidylglycerol and phosphatidylinositol, for prediction of fetal lung maturity, and the palmitic acid content of amniotic fluid phosphatidylcholine.

    Notes: The use of HPTLC in the analysis of some sixteen-membered ring macrolide antibiotices was examined. In the case of Spiramycins, instrumentalized HPTLC proved to be very efficient for the separation and determination of these antibiotics. With the use of an internal standard together with the datapair technique in sampling and evaluation of the HPTLC plates, a coefficient of variation less than 1.5% could be achieved when determining the different Spiramycins. Other sixteen-membered macrolides, such as Tylosins, Turimycins and 9-Propionylmaridomycins can be separated with sufficient resolution for quantitative work, in spite of their extremely simular structures and large molecular weights. Detection is always at wavelengths which agree with the intrinsic absorption maximum of the chromophors of the components (e. g. 282 nm for Tylosins, 232 nm for Spiramycins and Turimycins, 195 nm for 9-Propionylmaridomycins).

    Notes: Combination of various GC detectors by using a Varian effluent splitter with glass capillary columns has been found to be a rapid procedure for profiling organics extracted from sludges and river sediments. The selectivity and the increased sensitivity of the thermionic nitrogen-phosphorus detector (TSD), the electron capture detector (ECD), and the flame photometric detector (FPD) over the flame ionization detectors (FID) or mass spectrometers allow the detection of compounds present at trace levels without need for extensive sample cleanup. Furthermore, the combination of two selective detectors may supplement the information with regard to the chemical functionalities required for structure elucidation.

    Notes: The point of our published papers since 1957 is reviewed. The relations between the required value of peak resolution, K1 (or R), and peak separation, K3 (eqn 9); K1 and relative accuracy of a peak height quantitative method, Ph (eqn. 10); K1 and relative accuracy of a peak area method, Pa, (eqn. 12) at different concentration ratios, φ, are derived. The final result in Table 2 shows a large influence of φ on the required value of K1.The approximately linear relation between peak width and retention value (eqn. 18) exists not only in GC. but also in HPLC. Plate height values H1 and H∞ for a solute with capacity ratio, k′, equal to unity or approaching infinity, respectively, are used to evaluate the column efficiency (eqn. 20). The measuring methods (eqn. 21,22,23) and parameters effecting on H1 and H∞ are given for GC packed column (eqn. 24), GC open tubular column (eqn. 25) and HPLC (eqn. 26). In the light of this, columns of high efficiency were developed. Some typical chromatograms for high speed analysis and separation of complex mixtures are given.

    Notes: Chlorimipramine and other tricyclic antidepressant drugs are widely used for the treatment of depressive disorders. We analysed chlorimipramine and its major metabolite desmethylchlorimipramine after a two step extraction from plasma with diethylether. The underivatized samples were separated on a short (40 cm × 2 mm) silanized glass column packed with 3% OV 17 on Chromosorb HPW-DMCS, 80-100 mesh, which was directly coupled to the mass spectrometer. Detection of the tricyclic antidepressant drugs and their respective deuterium labelled internal standards was performed by chemical ionization using methanol vapour as reactant gas. The method is simple and sufficiently sensitive to permit the routine analysis of therapeutic plasma levels of tricyclic antidepressant drugs in extracts originating from 1 ml plasma.

    Notes: An assay method has been developed to determine the free base of Org 6001 (3α-amino-5α-androstan-2β-ol-17-one. HCI) in biological samples. The assay procedure consists of protein denaturation, ethyl acetate extraction, derivatization with tert-butyldimethylchlorosilane in dimethylformamide followed by n-hexane extraction and selected ion monitoring using trideuterated Org 6001 as an internal standard. For quantitation, the signals were monitored corresponding to the masses of the [M - 57]+ ions of 2-O-tert-butyldimethylsilyl, N-formyl derivatives of Org 6001 and the internal standard at m/z 390 and m/z 393, respectively. The assay method was applied to plasma samples from the human and rat, and to rat tissues including the target organ, the heart. The relationship between the Org 6001 plasma level and clinical effect in the human, the distribution of Org 6001 in rat tissues, and the correlation between the plasma level and the amount of Org 6001 in total heart tissue of the rat were determined.

    Notes: A sensitive and specific method has been devised for the determination of carpronium chloride, a parasympathomimetic agent, in plasma. The method is based on selected ion monitoring combined with the pyrolysis reaction of (3-isopropoxycarbopropyl)trimethylammonium chloride derived by transesterification of the drug. Carpronium chloride-[2H9] is used as an internal standard. The drug and the internal standard are separated from deproteinized plasma by ion pair extraction, and converted into isopropyl ester derivatives. The derivatives are pyrolysed to produce the respective isopropyl N,N-dimethyl-γ-aminobutyrates available for selected ion monitoring. Measurement is carried out in the electron impact mode by monitoring the fragment ion [M—OC3H7]+ (m/z 114, 120). The method is applicable to 5 ng ml-1 plasma with about 10% coefficient of variation, and has been utilized successfully for the pharmacokinetics of carpronium chloride in man.

    Notes: A pseudoracemic technique utilizing a stable isotope in one enantiomer was employed for the simultaneous determination of (R) and (S)-warfarin from plasma of human subjects. The assay includes high performance liquid chromatographic clean-up prior to mass spectral analysis to eliminate ion interferences from either co-administered drugs or contamination of the source. The assay is reliable, accurate and precise to within 5% at the submicrogram level.

    Notes: The stereospecific elimination of the 3-deuterium atom from metabolites of [2H]-analogues of Δ1-tetrahydrocannabinol (Δ1-THC), Δ6-THC and Δ7-THC has been investigated as a possible method for determining the stereochemistry of metabolites substituted with hydroxy or acid groups in the terpene ring. Elimination of HCOOTMS was found to involve the 3-hydrogen of the axial but not the equatorial isomer of hexahydrocannabinol-7-oic acid, a metabolite of all three cannabinoids. Similar stereospecific eliminations were observed during the loss of TMSOH from the TMS derivatives of 5α-hydroxy-Δ6-THC, 6α-hydroxy-Δ1-THC and 1α,2β-dihydroxy-Δ1-THC. Loss of TMSOH from 1α,7- and 1β,7-dihydroxy-HHC involved the 3-hydrogen in both cases but the isomers could be distinguished as their alkane-boronate derivatives; only the derivative of 1α,7-dihydroxy-HHC lost the boronate ring with stereospecific removal of the 3-hydrogen. The stereochemistry of the four isomers of 1,6-dihydroxy-HHC could not be determined in this way as the [M-TMSOH]+· ions from all four compounds had lost the 3-hydrogen, presumably as the result of 1,6-bond cleavage.

    Notes: As a result of the increasing concern over the environmental exposure of humans to carcinogenic alkylating agents, assay methods are being developed in order to quantitate the extent of in vivo alkylating reactions. Such methods will be of value also in connection with the use of alkylating agents as anti-cancer drugs. The reactions carried out by these compounds include the alkylation of cysteine in proteins, and a procedure has now been developed for the estimation of S-methylcysteine in haemoglobin following exposure of rats to the carcinogen methyl methanesulphonate. Haemoglobin is particularly appropriate for the analysis of alkylating agent reactions as this protein is readily available from blood and has a long in vivo half-life. The analytical method involved isolation of globin from blood, hydrolysis in 6 M hydrochloric acid, partial purification of S-methylcysteine by chromatography on Dowex 50 ion exchange resin, and subsequent analysis by gas chromatography mass spectrometry. The amino acid mixtures were derivatized by n-butyl ester formation and N-heptafluorobutyrylation, and were chromatographed on a capillary column coated with OV-101 or Chirasil-Val using a solid injection system. For the mass spectrometric determination [2H3]-S-methylcysteine was employed as an internal standard. Monitoring of the [MH]+ ion of derivatized S-methylcysteine in chemical ionization (isobutane) allowed the detection of 10 pg of the compound with a signal-to-noise ratio of 20:1.

    Notes: A patient with recurrent severe hypoglycemia resembling Reye's syndrome was found to have large accumulations of ω - 1 hydroxy and keto acids in serum and urine that persisted following clinical recovery. A deficiency of mitochondrial medium chain acyl CoA dehydrogenase activity is proposed on the basis of evidence obtained using gas chromatographic mass spectrometric techniques. Analytical data is presented that will allow the recognition of this variant presenting in other patients.

    Notes: Fluorouracil in human plasma at a concentration as low as 2 ng ml-1 can be assayed with the use of gas-liquid chromatography and a multiple ion detector. Fluorouracil is isolated from plasma by anion exchange extraction. The extract is butylated and the derivative obtained extracted into a mixture of cyclohexane + dichloromethane (95:5). An aliquot is submitted to combined gas-liquid chromatography mass spectrometry. The use of chlorouracil or oxygen-18 labelled flurouracil as internal standard is discussed. Linearity is proven from 100 μg to 5 ng of fluorouracil per ml plasma. The coefficients of variation are 7.2 and 8.4% for within-run and between-run precision, respectively, at the 50 ng ml-1 level. The biological applicability of our procedure is demonstrated by analysing plasma samples obtained at different time intervals from three patients given 1 g of fluorouracil once intravenously and once orally.

    Notes: Indicine-N-oxide was analyzed quantitatively in biological samples using a direct partial purification method involving acetonitrile precipitation or methanol precipitation followed by ion exchange chromatography. Trimethylsilyl derivatization of the resultant provided either of two derivatives, depending on the reaction conditions used, both of which had good gas chromatographic qualities. Heliotrine-N-oxide was used as the internal standard for this work. Data are presented to show that this is a reliable and useful internal standard based on its behavior in the partial purification method and on the gas chromatographic characteristics of its two derivatives. In addition, both low and high resolution mass spectral data indicate that heliotrine-N-oxide produces two trimethylsilyl derivatives analogous to those produced by indicine-N-oxide under the same conditions. Application of this procedure to urine and blood samples from cancer patients in clinical trials indicates that over 95% of the drug is removed from the circulation and excreted in the urine over the course of 48 h.

    Notes: The urinary excretion in man of pethidine and five of its metabolites (norpethidine, pethidinic acid, norpethidinic acid, p-hydroxypethidine and pethidine N-oxide) after a single intramuscular dose has been investigated using analytical procedures based on selected ion monitoring. New methods for the determination of p-hydroxypethidine and pethidine N-oxide have been developed, and the selectivity and sensitivity of the procedures are discussed. Only some 5% of the administered dose was excreted as unchanged pethidine. The mean urinary excretion of norpethidine was 16.4%. Pethidinic acid and norpethidinic acid constituted the major fraction of the dose excreted in urine. The total excretion of pethidinic acid and norpethidinic acid was 41.7 and 23.2%, respectively. Pethidine N-oxide constituted less than 0.5% of the excreted amount, and p-hydroxypethidine could only be detected in the urine of two of the patients. A mean total recovery of about 90% of the dose was found in urine as pethidine and the five metabolites.
