Notes: Residue mixtures of pesticides in foods which were difficult to separate by packed-column gas chromatography were determined by capillary gas chromatography and electron capture detection. Manual injections of sample and reference standard were made with a 10 μL syringe on a septum-enclosed, split/ splitless inlet system. Optimal peak height repeatability (± 3%( was achieved with three-μL injections which were made within time periods of four hours or less. The average linearity coefficient (slope( obtained for detector response as a function of solution concentration was 1.1.

Notes: A procedure for packing 15 cm × 1 mm id reverse-phase microbore columns with 3 μm silicas obtained from different manufacturers is described. The speed of analysis and detection limits are compared to those obtained with a 50 cm × 1 mm id column packed with 10 μm ODS. The effect of detector time constant on the system, and flow rates on column efficiency are also examined.

Notes: The volatiles of fresh cherimoya (Annona cherimolia, Mill.) fruit pulp were separated by combined high-vacuum distillationliquid/liquid extraction (pentane/dichloromethane 2+1). After preseparation by liquid chromatography on silica gel three fractions (I-III) were obtained by eluting with pentane (I), pentane/diethyl ether (9+1) (II), and diethyl ether (III), and fractions II and III were subjected to HRGC/FT-IR analysis. 26 volatile compounds comprising nearly all the main components (range, 10-300 μg/kg fruit pulp) were identified by this technique for the first time in cherimoya fruit.

Notes: A method is described for the qualitative and quantitative determination of configurational isomers of zeaxanthin (=3,3′ -dihydroxy-β, β -carotene) and lutein (=3,3′ -dihydroxy-α -cartotene). It is based on the reaction of these zeaxathin and lutein isomers with (S)-(+)-α-(1-naphthyl) ethyl isocyanate to afford diastereomeric dicarbamates, which are analyzed by HPLC.

Notes: A comparative study is reported on separation of series of mono-, di-, and trisubstituted methyl 5β-cholanates, which differ only in the position and stereochemistry of hydroxyl or keto groups at position and stereochemistry of hydroxyl or keto groups at positions C-3, C-7, and/or C-12, by reversed-phase [with chemically-bonded (C-18) silica gel] and normal-phase (silica gel) high-performance thin-layer chromatography (HPTLC). Methnol (or acetonitrile)/water systems were employed as mobile phase. Reversed-phase HPTLC found to be particulary effective for separation of the stereoisomers of di- and trisubstituted compounds whereas the less polar monosubstituted isomers are well resolved in normal-phase HPTLC.

Notes: Analyses of hydrocarbon fractions from different areas of the marine environment are described to illustrate the possibilities and limitations of high resolution gas chromatography (HRGC) in the analysis of environmental samples. Examples are given of dissolved, particulate, and sedimentary hydrocarbons and organochlorine compounds; the importance of an adequate sampling of the marine environment is stressed. HR chromatographic profiles obtained in two columns of low and high polarity (SE-52 and PEG 20M) permit the sources and transport pathways of both natural and anthropogenic hydrocarbons to be traced. Analysis of tissues of marine mammals, which metabolize or excrete many of the biogenic and petrogenic hydrocarbons present in other areas of the marine environment, may provide an opportunity to obtain relatively clean profiles of many anthropogenic compounds of interest. The plotting of mass chromatograms from data compiled by COM-GC-MS remains the most appropriate method for the conclusive indentification of these compounds.

Notes: Pyrolysis gas chromatography (P/GC) has been successful in differentiating coal tar pitches which are used in the manufacture of high temperature graphite materials [1,2]. In the present study, coal tar and petroleum pitches received from five suppliers were readily differentiated using three distinct methods of polymer analysis: dynamic headspace and pyrolysis gas chromatography mass spectrometry (DH/GC/MS and P/GC/MS, [3]), and pyrolysis mass spectrometry (P/MS, [4,5]). All of these methods are appropriate for the analysis and pattern recognition study of coal and petroleum products; however, methods which incorporate GC usually necessitate long analysis times and are subject to problems associated with gas chromatographic reproducibility (e.g. active sites [6], and nonconstant retention times). As a fingerprint analysis method, P/MS is rapid and ideally suited to computerized pattern recognition study of pitches due to the simple format of P/MS data (computer tabulation of normalized intensities versus masses); whereas, in the present study, the normalized peak area versus retention time date from DH/GC/MS and P/GC/MS analyses were not suitable for direct input to a computerized pattern recognition analysis program, (due to non-constant retention times and improper data format). One limitation is that P/MS does not provide positive identification of thermally evolved products (as can DH/GC/MS and P/GC/MS) due to the complexity of the spectra of a mixture of compounds produced by P/MS. Within the limits of the commercial instrumentation and computer programs used in this study, a combination of all three techniques provided the greatest capability for the differentiation of the five coal tar and petroleum based pitches studied.

Notes: A new design for an electron capture detector cell is described. The cell is compatible with the requirements of high resolution capillary columns and is shown to be useful in applications that require high analysis speed. A unique method of sample introduction reduces the problems of sample loss by adsorption on the surfaces of the cell. Previously reported problems of sample loss by adsorption on active surfaces within the cell when using hydrogen carrier gas are shown to have been eliminated. Examples are shown demonstrating the increased speed of analysis that can be obtained when using hydrogen carrier gas and 100 micron diameter columns.

Notes: Analytical methods are described for identification and monitoring of the oxygenated metabolites of dibenzothiophene. Since such compounds are thermolabile, GC analysis is seen to be hardly feasible with conventional injectors such as all-glass moving needle or splitless injectors. Only on-column injection gives no degradation products. In addition, reversed-phase HPLC is particularly suitable for the analysis of the sulfone or sulfoxides of dibenzothiophenes.

Notes: A method is described for the determination of acetylsalicylic acid and salicylic acid in human whole blood and plasma which uses liquid chromatography with UV detection. The enzymatic degradation of acetylsalicyclic acid in blood and plasma is examined. Addition of potassium fluoride as enzyme inhibitor and deep freezing after plasma preparation allows storage of plasma for one week. A mixture of acetonitrile and diluted acetic acid as mobile phase give the best peak shape. Traces of iron lead to slight peak broadening; addition of EDTA results in excessive tailing. The detection limit for acetylsalicylic acid and salicylic acid is about 250 ng/ml for a 1.0 ml primary sample. The method is sensitive enough for the monitoring of both drugs in bioavailability studies.

Notes: Theoretical equations are given to relate resolution with development distance. Comparative experiments undertaken with quaternary ammonium compounds as test substances and using a selection of basic drugs are presented. As expected, resolution decreases with shorter development distance. However, this effect is less pronounced for compounds with small Rf values and when the spot size at the starting point is kept small. It was also shown that, for correctly assessing resolution in relation to development distance, the amounts of material spotted have to be decreased for shorter development distances.

Notes: The condition for in situ chemical modification of commercially available thin-layer plates have been varied and the influence thereof upon reversed phase plate properties has been studied. The reaction variables studied were: average silica pore size, silica water content, use of tri- or monochlorosilanes as modifiers, reaction temperature, and addition of base. Coverage and activity of residual silanol groups was studied on modified plates.Desired plate properties are high speed of development in aqueous solvents and low content of active residual silanol groups. Under the conditions tested, the lowest amount of residual silanols was achieved at the expence of the speed of development in aqueous media and vice versa.

Notes: A rapid, sensitive, and reproducible separation of purine bases, nucleosides and nucleotides by reversed-phase high-performance liquid chromatography is described. A multi-step gradient of acetonitrile in 0.1 M potassium phosphate was used to detect picomole amounts of each compound within 22 min. The high sensitivity of the method made it suitable for the analysis of purine nucleotide pools extracted from 1--2 × 106 hamster fibroblasts grown as monolayer (BHK line) and for the detection of purine bases and nucleosides leaked form the cells into the incubation media. The chromatographic procedure was applied to study the effects of hexavalent chromium (potassium dichromate) on purine metabolism. Several steps are affected by this treatment, causing a marked unbalance of both the guanylate and adenylate pool, which was seen as a strong decrease in the level of triphosphates and accumulation of their precursors.

Notes: An investigation of techniques suitable for precision cutting of glass and fused silica capillary tubing has been carried out. Accurate trimming of capillary tubing is important when precision splitters and manifolds have to be made. Techniques employed for cutting optical fibers proved to be useful, and factors affecting the suface quality of the fracture are discussed. It was found to be difficult to obtain a square cut with a flat surface in the case of fused silica tubing. The best results were obtained when the tubing was scored with the edge of a broken silicon wafer before the fracture was accomplished. This technique allows precision cutting of glass tubing with an outer diameter of 100 μm or less.

Notes: It has become common practice with capillary columns to call film thicknesses of 0.1 - 0.3 μm regular or standard, whereas 0.3 - 1.0 μm is considered to denote thick films. Consequently, films with thickness beyond 1μm (we have explored the range up to 8.0 μm) may be termed very thick. Increasing film thickness, at least to about 5 μm, brings important additionl advantages primarily in terms of loading capacity. Columns with very thick coatings may replace the traditional SCOT columns thanks to at least equivalent loading capacity combined with higher separation efficiency, and better stability characteristics. Additional advantages are increased retention, essential for the analysis of very volatile substances, and increased elution temperature, providing a basis for direct water injection. We observe that TZ (the separation number) drops to roughly 50% when the film thickness is increased by a factor of 300 from 0.025 to 8.0 μm, provided TZ is measured with a standard flow of hydrogen. The reduction is only 30% when the thick-film colums are run with individual optimization with nitrogen as a carrier gas. In contrast to standard columns, very thickly coated columns show slightly increased separation efficiency with nitrogen at drastically reduced flow rate. The HETP increases very slightly in the indicated range of flim thickness. This contradicts widespread theoretical expectations, which probably account for the fact that very thick coatings have so far been neglected. Since the preparation of very thick coatings involves some deviations from the standard procedure, complete and detailed instructions are given.

Notes: The use of sequentially coupled columns to achieve a binary liquid phase mixture has been simplified by the availability of zero dead volume fittings compatible with fused silica columns. The extent to which the velocity gradient through the coupled column affects the “apparent” liquid phase ratio can be determined by graphic interpolation; the length ratio of the coupled column segments can then be adjusted so that the “apparent” liquid phase ratio actually experienced by the solutes agrees with the targeted value. The fact that the direction of flow through the coupled column affects the chromatographic dispersion suggests that accepted generalizations on flow optimization throughout the column may not be precisely correct.

Notes: Commercially available OV-17 was partitioned into a low and a higher molecular weight fraction by supercritical fluid extraction with carbon dioxide. This higher molecular weight fraction is more thermally stable than the original OV-17 due to its more viscous and gum-like nature. Based on the growing evidence in the literature that a deactivating agent for fused silica open tubular columns should be compatible with the stationary phase, a precoat of 1,3,5-trimethyl-1,3,5-triphenylcyclotrisiloxane was applied prior to the static coating of high molecular weight OV-17. Fused silica OV-17 columns prepared in this fashion are stable up to 300°C.

Notes: Since capillary columns with well immobilized stationary phases are expected to withstand contact with supercritical fluids, we wished to study their amenability to SFC. Simultaneously, we wished to learn how far SFC can be accomplished with the ordianary tools of capillary GC.The study demonstrates that truly supercritical, not just relatively high, pressure is required to ensure the typical effects of SFC. Results obtained with sub-and supercritical pressure are compared and discussed. A comprehenshive study of the parameters permitting SFC with capillary GC equipment showed a clear preference for CO2 as a carrier, FID detection, and oncolumn sampling. While no additional equipment is required, a critical feature is the flow restrictor to be mounted on the end of the column. The production and properties of this restrictor are discussed in detail. It is reasonable to hope that SFC with 0.1 mm id capillary columns can be realized in the pressure range of 100-150 bar, where substances which cannot be eluted from a capillary colum under GC donditions are expected to be analyzed.

Notes: Solvent vapors in air may be measured with capillary gas chromatographic columns. By using large diameter columns and sample loops of approximately the same internal diameter, the column may be connected directly to the gas sampling valve. This approach eliminates the use of a splitter or cryogenic trapping and allows low levels to be measured. By operating the column at high velocity, column efficiency is sacrificed for increase in speed.

Notes: Single, long columns (1-5 m) can be prepared efficiently using reversed phase packings (3-10 μm particle diameter). 1-m columns packed with 3 and 10 μm packing provide 110 000 and 50 000 theoretical plates, respectively. Very efficient columns can resolve highly complex mixtures and difficult-to-separate compounds. Temperature gradient elution is a powerful technique for LC with microbore columns.

Notes: A 30 m, 0.25 mm ID, fused silica capillary column at temperatures from -60 to -100°C has been shown to be a quantitative trap for organic compounds with volatilities ranging from that of 1, 1 -dichloroethene that of chlorobenzene. This type of “whole column cryotrapping” provided sharp peaks (peak width approximately 4-7 seconds) for all compounds at a trapping temperature of -80°C and with high carrier gas pressures and linear velocities (30 psi and 110 cm/s, respectively). Whole column cryotrapping possesses great simplicity, chromatographic efficiency (no trapping loop connections), and a built-in indicator of quality assurance for trapping efficiency (i.e., peak shape). These advantages are extremely attractive and are indicative of the fact that the potential of this approach has not yet been fully appreciated.

Notes: There has been much effort spent in recent years developing the technology for free radical crosslinking of polysiloxane polymers to prepare capillary columns coated with thermally stable and nonextractable stationary phases. Organic peroxides have been used extensively as the free radical initiators for the in situ polymerization of the stationary phases. However, these peroxides adversely affect the phase polarity and column activity. Seven azo compounds were studied for crosslinking efficiency. Azo compounds can be used as free radical initiators to prepare nonextractable stationary phases without the adverse effects caused by peroxides.

Notes: The gas chromatographic use of flexible thin walled soft glass capillary columns coated with non-polar stationary phases is compared to similar columns made of fused silica glass. With non-polar soft glass columns, the use of surface roughening viagaseous HCI followed by a Carbowax 20 M pretreatment gave adsorptive phenomena, and thermal instability. With very polar soft glass columns where a variety of cyanopropyl silicone phases were coated directly onto the NaCI crystal matrix, adsorptive effects were again prominent and frequent break-down in film stability with time, was also observed. These undesirable effects were due to the presence of metal oxides in the soft glass. Attempts to remove these materials from the thin walled soft glass surface by means of acid leaching produced significant brittleness. This deleterious result was further increased by attempts at high temperature silylation or polysiloxane deactivation. In sharp contrast, the fused silica surface was essentially free of metal oxides and the surface silanol groups are easily neutralized by silylation or polysiloxane deactivation techniques. No brittleness was observed following these procedures. An increasing series of high molecular weight, viscous, polymeric vinyl containing non-polar and highly polar stationary phases have been produced which readily wet the surface of the fused silica and are easily crosslinked in the presence of free radical generators. These columns are essentially free of all the problems noted with flexible thin walled soft glass. When all of the parameters involved in the fabrication of a glass capillary column are assessed, it appears at this time, that the flexible fused silica glass column with cross linked phases approaches the “ideal” capillary column.

Notes: The results of ion-pair chromatography with UV-absorbing counterions are described. Hydroxynaphthalenesulfonic acids are used as ion pair formers, and alkaloids as test samples. The separations are performed on silanized silica gel from Merck and KC18F plates from Whatman. The influence of the counterion on the Rf-value is demonstrated. Some alkaloids exhibit characteristic fluorescence as ion pairs.

Notes: The effect of sample injection temperature on quantitation is examined for on-column and conventional split modes of sampling in capillary gas chromatography. Discrimination effects can be observed even with on-column injection if the injection temperature is too far above the boiling point of the solvent (or that of a major low boiling constituent(. This is attributed to higher boiling components being left behind in the syringe needle, and a set of simulation experiments are described to illustrate this effect. Various discrimination patterns using conventional split injection were observed, depending on temperature of injection. Apart from syringe needle effects, discrimination is probably due to the preferential venting of higher boiling components as liquid sample droplets, which can have a lifetime greater than the time of transit to the splitter. With such a two-phase system, involving variable droplet size, the flow distribution in the splitter will be critical to uniform sampling. The use of combination on-column/split sampling, with the appropriate temperature control to provide sample uniformity to the splitter is discussed as an advantageous alternative to conventional split sampling.

Notes: The linear relationship between the reciprocal of analyte-line intensity, and the reciprocal of concentration has been applied to the determination of ten major and minor elements in silicate rocks and minerals. The analyses were carried out on fused specimens of 11 rock standards, with and without added La2O3, and with and without added NaNO3. The method requires no absorption matrix effect corrections, and is ideally suited to the analysis of geological materials with a wide range of chemical composition. It clearly has the advantage of simplicity and speed, and can be applied to the analysis of samples when suitable standards are not available. The results obtained in all cases are in excellent agreement with the literature. However, the discs with La2O3 added yield results with better precision and accuracy than do those without La2O3. Na2O is normally determined on separately prepared rock powder pellets, but can be satisfactorily analysed by this method, using the La2O3 added discs, together with all of the other major and minor elements. Na2O analyses are more readily obtained using discs without added NaNO3. We, therefore, recommend that the analysis of geological materials be carried out on fused glass discs with La2O3 added and without NaNO3.

Notes: The exponential equation R0 = R · eR0τdescribes the counting losses in wavelength-dispersive x-ray spectroscopic (WDF) detectors up to very high counting rates. The correction error can be considerably lower than 1% for a true counting rate of 7 · 105s-1. The dead time constant τ has however to be determined very accurately for this. Methods of determining τ with the desired accuracy are also described. And finally, a formula for correcting for dead time losses for two detectors in series, where each detector has a different dead time, is deduced and verified by experiment. Here again, one can calculate the true counting rate with an accuracy of 1% at a true counting rate of about 7 · 105 s-1.

Notes: The analysis of various elements in coal has become important as coal utilization increases. The application of x-ray fluorescence (XRF) to this type of analysis is complicated by matrix-correction needs and lack of matrix-matched standards. The application of fundamental parameter mathematical corrections using easily obtainable doped graphite standards is evaluated.

Notes: Cross-sections for bremsstrahlung production in thin films may be derived theoretically only if certain approximations are made or if there is recourse to numerical techniques. In this paper, two approximate equations, one based on the Sommerfeld theory and the other on the Bethe-Heitler theory, are assessed for their accuracy in predicting cross-sections differential in photon energy and emergence angle when a thin foil target is bombarded with electrons with energies between 40 and 100 keV. Experimental data are presented from eight different target materials and these are processed initially to make comparisons between experimental and theoretical spectral shapes and subsequently to compare observed and predicted absolute cross-section values. Except for the heaviest target material (Au), the Bethe-Heitler theory provided a satisfactory description of all experimental observations. In contrast, the Sommerfeld theory provided a significantly inferior description of spectral shapes and consistently overestimated absolute cross-sections by 15-20%.

Notes: It is shown that when a narrow x-ray beam is used in energy dispersive fluorescence analysis of thick, low-Z samples large deviations from the predictions of the commonly used fundamental parameter model occur. A simple method by which the effects can be studied empirically is described. An approximate model allowing a direct calculation of the effécts is introduced. The validity of the model has been confirmed experimentally.

Notes: The stoichiometry and thickness of TiNxOy films has been determined by electron microprobe and nuclear microanalysis. The difficulty arising in the electron microprobe from the interference between NKα and TiLI lines has been overcome by using a suitable Monte Carlo computing scheme. This procedure takes into account the SiKα intensity which was found to be sensitive to composition and thickness of the overlying TiNxOy film. The good agreement between electron microprobe and nuclear microanalysis results indicate the validity of the method.

Notes: In the Japanese Industrial Standard for the x-ray fluorescence analysis of steels, the ‘estimated binary calibration curve’ method has been adopted. In this method, the calibration curve for the binary system, consisting of an analyte and the principal constituent elements is estimated and derived by using multicomponent standard samples. The measured x-ray intensity is first converted into a tentative analytical value from this calibration curve, and then the analytical result is derived from this tentative value using the correction procedure. In this paper, some methods and standard samples of making the estimated binary calibration curve are compared and discussed, also, the characteristics of the JIS method are described.
