    Notes: Quantification of the degree of acclimation of a mixed culture is demonstrated. An industrial landfill leachate was used as a model mixed substrate. The biodegradable fraction, the specific rate of substrate uptake, and the apparent reaction order were measured. The method of acclimation did not appear to affect the biodegradable fraction. The apparent reaction order is shown to be related to the difference between the rates of uptake of the various leachate components. This difference is defined as the differential in the rate. The effects of a supple mental carbon source and different inocula were investigated. Addition of glucose tends to diminish the differential but also decreases the specific rate of substrate uptake. The outcome of an acclimation procedure is dependent on the quality of the inoculum; however, a poor inoculum does not preclude an acclimation that yields a superior culture. A normalized rate coefficient is shown to have merit as an indicator of the degree of acclimation. Two microorganisms were isolated, Acinetobacter calcoaceticus var. Iwoffi and Moraxella sp., that could grow axenically with the leachate as their sole source of carbon. Comparisons are made between pure and mixed cultures with regard to biodegradable fraction, specific rate of substrate uptake, and apparent reaction order.

    Notes: Using commercial immobilized glucose isomerase (SWETASE®, Nagase Co.), the effect of substrate protection on enzyme deactivation has been studied in a batch manner. The data analysis was carried out based on Briggs-Haldane kinetics in which enzyme deactivation accompanying the protection of substrates was also considered. The protection factor was proposed to elucidate the dependence of the degree of substrate protection. The existence of the protection of glucose isomerase by the substrates has been verified experimentally. Also, the enzyme-substrate complex deactivates with a decay constant which is one-half that of the free enzyme. Theoretical analysis of enzyme deactivation with substrate protection offers an effective understanding which is essential for enzyme replacement and process optimization.

    Notes: A tube rheometer system has been constructed for aseptic study of the rheology and fundamental flow properties of mycelial fermentation fluids. The rheometer consists of a U-formed tube circuit starting and ending inside the fermentor. The mash is pumped through the tubes with a lobe rotor pump. The flow is measured by an electromagnetic flow meter. Pressure drops have been measured with a system of differential membrane transducers for different flow rates. The rheometer system was tested with Newtonian and non-Newtonian fluids.

    Notes: Cellulase (Cellulosin AC-8) was immobilized on poly-L-glutamic acid. This immobilized cellulase (IC) is water soluble in the neutral and alkaline solutions, where IC has the activity, while IC can be made insoluble by lowering the pH so that it can be recovered from the reaction mixture with its activity. The optimum pH and temperature were determined to be 5.5 and 55°C, respectively. The stability of IC against change in the pH and temperature was improved by the immobilization. Solvolysis of 3N-NaOH-treated cellulose, with IC under the optimum conditions found here, led to the production of low-molecular-weight compounds.

    Notes: Biodehalogenation of 10-5 M concentrations of bromotrichloromethane (BTM) and 1,2-dibromo-3-chloropropane (DBCP) was studied in static cultures of Pseudomonas putida PpG-786. The experimental cultures were prepared by growing P. putida on camphor, which is known to induce the synthesis of high concentrations of cytochrome P-450 in this bacterium. Measurements of bromide ion release were found to be approximately consistent with the amounts of halocarbon degraded. Gas chromatography/elctron capture detection (GC/ECD) measurements of hydrocarbon degradation products as a function of incubation time showed the transitory appearance of chloroform and bromodichloromethane from BTM and the transitory appearance of lower boiling but unidentified products from DBCP. The degradation of BTM to trihalomethanes and the halide ion is consistent with the enzymatic reductive dehalogenation by cytochrome P-450 reported by others. The dependence of initial conversion rates on halocarbon concentration (0.1-2 ppm) and cell mass concentration (1-28 g cell/L) was determined by measuring the decline of parent halocarbon in stirred batch cell suspensions. The rate of DBCP conversion was up to 10-fold higher than the rate of BTM conversion. When the intracellular, enzyme-catalyzed conversion BTM is analyzed by the effectiveness factor of heterogeneous catalysis, the initial conversion rates measured suggest that intrinsic enzyme kinetics, rather than halocarbon permeation of the cell membrane or other diffusive processes, is rate limiting.

    Notes: There are many scaling formulas that predict the oxygen mass transfer coefficient as kL·a = constant·(Hp/V)αVsβ Exponents α and β frequently are scale dependent themselves. A general formula has been derived from the work of Calderbank,1 Miller,2 and Tilton,3 resulting in kL·a = C1 φ + C2 log (Pm/V) φ where φ equals the gas-holdup fraction and Pm/V equals the effective mechanical power input per unit of volume. This formula is consistent with the formula of Westerterp4 modified by Miller.2 Gas holdup can be predicted in several ways. Gas-sparged isothermal expansion power input, used for predicting φ, demonstrates that scaling can be done by using either superficial air velocity or volume per volume per minute for aeration.The importance of mixing in replenishing oxygen at the boundary layers of microorganisms will be assessed and compared with the kL·a as the oxygen transfer ratelimiting step.

    Notes: Since a stable alcohol oxidase with a high specific activity is not commercially available, we propose to produce and purify this enzyme from a strain of the yeast Hansenula polymorpha. This alcohol oxidase was immobilized into a gelatin matrix and its activity was estimated by a pO2 sensor. The enzyme electrode obtained was then used in a continuous flow system to measure methanol or ethanol concentrations. The sample oxygen content dependence of the signal was minimized by the support properties. Measuring time for each sample were less than two minutes including response data treatment and rinsing step. The enzyme electrode response was set for ethanol from 0.5mM to 15mM and for methanol from 10mM to 300mM. On repeated use, the electrode signal for 10mM of ethanol was stable for at least 500 assays. Analysis have been performed in different beverages such as wine and beer, and the results compared to those obtained with classical methods of analysis.

    Notes: The soil yeast Trichosporon cutaneum was grown in continuous culture with phenol as the sole carbon source. The cultures were operated as carbon-limited chemostats or as steady-state continuous cultures without carbon limitation. Selected comparative runs were also conducted on glucose or acetate as carbon source. In addition to growth parameters, the activities of several intracellular enzymes were determined, comprising those directly involved in the degradation of phenol as well as auxiliary enzymes required for the generation of reducing power. All enzymes were assayed in detergent-permea-bilized cells. Phenol was found to serve as an excellent carbon source, comparable to glucose or acetate. The utilization of phenol in T. cutaneum is very efficient as indicated by a low maintenance requirement (0.01 g phenol/g cells·h). The cell yields obtained were on the order of 0.8 g cells/g phenol. Although the phenol-limited chemostats were run with fully phenol-induced cells, a further increase in the activities of isocitrate DH(NADP+), maleate DH and the phenol-degrading enzymes occurred after transition to nonlimiting condition. Enzyme activities increased in parallel with increasing phenol levels in the effluent, as well as with increasing toxicity. The significance of this phenomenon is discussed.     Notes: The development of Penicillium roqueforti on buckwheat seeds proceeds roughly into four steps, involving a lag phase and three growth phases. First, it appears as a spore germination and external colonization of the grains by the mycelium. Then, mainly external sporulation and internal colonization of the seeds occur and finally internal sporulation takes place. The Stoichiometry of the growth and the sporulation is established. Kinetic experiments performed in a fixed bed reactor show that the growth of the microorganism (biomass production) may be estimated by the protein content of the medium. This growth occurs with a very low μmax value close to 0.030 h-1. The chitin content of the medium is an indicator of the sporulation, just as the metabolic liquor (mainly water) produced during the course of a cultivation. The values of the observed respiratory quotient are close to those predicted by stoichiometry.

    Notes: Rapid liquid-liquid extraction of lactate dehydrogenase from muscle by using a low-cost aqueous bipolymer two-phase system was achieved by using a centrifugal separator. Extraction of the target enzyme into the upper phase was enhanced by including the dye Procion yellow HE-3G (bound to polyethylene glycol). The dye acted as an affinity ligand for the enzyme. The isolation of the enzyme was carried out either by using a cell extract or by homogenizing the muscle directly in the system. The latter approach reduced the preparation time with a factor of 0.25. The two methods gave, respectively, 310 and 360 kU lactate dehydrogenase/kg muscle (measured at 22°C). By using a small centrifugal separator, Alfa Laval LAPX 202, 3-5 kg muscle could be processed/h in a 30-L, two-phase system.

    Notes: In this article, a two-phase system for the digestion of wastes with a high solid content is simulated. The solids are charged to the hydrolyzer and then leachate recirculation is activated until biodegradation is nearly complete. Several parameters are tested, namely moisture, leachate recirculation flow rate, and hydrolyzer-methanizer volume ratio. The results show that recirculation rate is an important parameter subject to optimization, with optimal values corresponding to hydrolyzer hydraulic retention times below 1 day. The quantity of recirculating water must be the highest possible. As a consequence, the organic load to the methanizer is reduced, making thus possible the use of a smaller methanizer volume.

    Notes: O-Acetylserine sulfhydrase in the form of a crude extract from Salmonella typhimurium LT2 was used for the production of L-cysteine from L-O-acetylserine and sodium hydrosulfide at pH 7.0 and 25°C. The two substrates have quite different pH stability relationships. O-Acetylserine readily rearranges to N-acetylserine and the rate of this O → N acyl transfer reaction increases at higher pH, temperature, and concentration of O-acetylserine. On the other hand, sodium hydrosulfide is more soluble at a higher pH. A stirred-tank bioreactor with a continuous substrate feed was employed to overcome this problem. The O-acetylserine feed was stored at its saturation level (2.05M) at pH 5.0, and the sodium hydrosulfide feed was dissolved at 2.05-2.3M without pH adjustment (pH ≥ 11.5). Both substrates were simultaneously introduced into the bioreactor. The performance of the bioreactor was optimized by employing an automatic feedback control system to regulate the concentration of O-acetylserine in the bioreactor. This feedback control system was based on the fact that as the bioconversion proceeds, protons are produced along with cysteine. A pH controller thus detected the decrease in pH and activated the substrate pumps. After mixing in the bioreactor, these two substrate solutions behaved as a base due to the high alkalinity of sodium hydrosulfide. Thus, substrate infusion started when the pH was lower than the set point, i.e., the reaction pH, and stopped when the pH was raised higher than the set point. The amount of substrate introduced was determined by the alkalinity of the mixture of the two substrates, which in turn was controlled by the concentration of sodium hydrosulfide. After optimizing the sodium hydrosulfide concentration and the substrate feed rate, the bioconversion gave a productivity of 3.6 g L-cysteine/h/g dry cell weight S. typhimurium, an L-cysteine titer of 83 g/L and a molar yield based on O-acetylserine of 94%.

    Notes: The growth dynamics of Escherichia coli with the temperature-sensitive plasmid, pOU140, were examined. Recombinant cells exhibited nearly identical kinetic behavior to host cells at low culture temperatures and low copy numbers. However, at higher temperatures, in which the copy number was significantly increased, the recombinant cells showed decreased stability along with lower growth rates and substrate yields as compared to the host. Furthermore, the production of a constitutive cloned-gene protein was shown to increase with temperature in an Arrhenius fashion when culture temperature was varied between 38 and 42°C. These results suggest that temperature can be used to quantitatively control the production of a desirable plasmid-coded gene product.

    Notes: During unsteady-state continuous culture of Saccharomyces cerevisiae on sugarcane blackstrap molasses, the invertase activity of the intact yeast cells oscillated. Disturbances were produced by changing medium composition, air rate, impeller speed, and dilution rate. The influence of the oxygen supply rate and of the dilution rate on the invertase activity depend on the medium composition. The highest invertase activity was obtained when, after a steady-state attained using unsupplemented culture medium, nutrients were added to the feeding mash. A Monod-like equation seems to be the best representation of the correlation between the specific rate of reducing sugars consumption and the specific rate of nonreducing sugar hydrolysis by the yeast cells.

    Notes: Reaction rates of hydrophobic enzymes, aminopeptidase, and alkaline phosphatase, in microsomes prepared from the porcine brush border membrane and in vesicles pre pared from microsomes and phospholipids were measured at various temperatures. Interactions between the hydrophobic enzymes and the phospholipid layers are discussed as well as the effects of fluidity change of phospholipid layers on enzyme activity. Further, reaction properties and stabilities of the immobilized vesicles containing microsomal enzymes were studied.

    Notes: The relationship between cell mass and cell number dynamics for bacteria such as Escherichia coli depends on the cell cycle parameters C and D. Effects of plasmid copy number on these cell cycle parameters have been studied for Escherichia coli HB101 containing pMB1 plasmids propagated at different copy numbers ranging from 12 to 122. Determination of cell cycle and cell size parameters was accomplished using flow cytometry data on single-cell light scattering and DNA content frequency functions in conjunction with a mathematical model of cell population statistics. Two independent methods for estimating C and D intervals based on flow cytometry were developed and applied with essentially identical results. The presence of plasmids decreases the C and D periods, mean cell sizes, and initiation masses for chromosome replication by 14, 24, 38, and 18%, respectively, relative to corresponding values for plasmid-free host cells. Plasmid copy number has a negligible influence on these parameters, suggesting that host-plasmid inter actions which determine these properties are centered on the single plasmid selected for replication according to the random selection model established for ColE1-type plasmids.

    Notes: Both kLa and kL measurements were carried out by an unsteady state technique at impeller speeds ranging from 1.6 to 5.8 s-1 in a mechanically agitated animal cell culture vessel of working volume 1.5 L. Checks were made that the time constant of the oxygen electrode was negligible compared to the time for aeration and that the oxygen electrode reading was not a function of agitator speed in the range employed. The kL values by surface aeration of (1.18-3.54) × 10-5 m/s and kLa values by sparged aeration of (2.8-8.5) × 10-4 s-1 were found. The former are in reasonable agreement with published experimental values and the latter in accord with values estimated from published correlations based on agitator power input and aeration rate. The fluids used were water, basal medium, and basal medium supplemented with 5% (v/v) foetal calf serum; for each of these, kL and kLa values were similar. However, the addition of silicone antifoam (6 PPM) reduced the kLa value by ca. 50%.

    Notes: The equations of a chemostat in which one microbial population grows on multiple rate-limiting nutrients are formulated. The dynamics of a chemostat involving growth on complementary nutrients is studied through stability analysis of the system of equations. Some conditions are derived that relate the dynamic behavior of the chemostat to its operating conditions and can be applied to any model for the specific growth rate of the population. It is shown that, if maintenance of the population is neglected, the system exhibits no sustained or damped oscillations. If maintenance of the population is considered, damped oscillations are observed for some operating conditions.

    Notes: The effects of diffusion on a reaction taking place in a permeable catalyst are examined theoretically for the case where the reaction has two substrates supplied from opposite sides of a catalytic slab. The solutions of the reaction-diffusion equation for combinations of zeroth-and first-order kinetics are given in terms of an effectiveness factor and a parameter describing the position in the layer where the reaction occurs. In these terms, the results vary only weakly with reaction order. The use of the exact solutions for a reaction that is zeroth order in both sub strates is proposed as a general rule for estimating the reaction rate and the reaction position.

    Notes: The effect of acoustic conditioning on the particle size distribution of isoelectric and calcium-ion-precipitated soya protein has been examined in low-residence-time chambers. In a previous study a beat frequency of 5 Hz obtained using a dual-source system of opposing vibrators was determined as giving optimal improvement in particle-settling characteristics for isoelectric soya protein precipitate. In this study the effect of amplitude of vibration, a measure of acoustic power input, and residence time of acoustic conditioning has been examined.Acoustic power input changed the flow pattern in the conditioning chamber from laminar streamline flow to a well-mixed, turbulent pattern. Such a mixing effect promoted the rapid aggregation of fine particles, a process that was modeled on the basis of orthokinetically controlled collisions. The rate of removal of fine particles due to acoustic conditioning was shown to be proportional to a mixing effect that was releated to the acoustic power dissipated per unit volume.The consequences of fine-particle aggregation on the centrifugal recovery of the precipitate are discussed.

    Notes: Changes of pH and sulfate concentration in high-sulfur coal refuse slurries are used as measurements of microbial pyrite oxidation in the laboratory. Sodium lauryl sulfate (SLS), alkylbenzene sulfonate (ABS), benzoic acid (BZ) and combinations of SLS plus BZ and ABS plus BZ effectively inhibited formation of sulfate and acid when added in concentrations greater than 50 mg/L to inoculated 20 or 30% coal refuse slurries. Here 25 mg/L concentrations of SLS, ABS, and ABS + BZ stimulated acid production. Formic, hexanoic, oxalic, propionic, and pyruvic acids at 0.1% concentrations were also effective inhibitors. Four different lignin sulfonates were only slightly effective inhibitors at 0.1% concentrations. It was concluded that acid formation resulting from microbial oxidation in high-sulfur coal refuse can be inhibited.

    Notes: Invertase from baker's yeast (Saccharomyces cerevisiae) was covalently bound via benzoquinone and glutaraldehyde to a macroporous polystyrene anion exchanger. The behavior of the invertase-polystyrene complexes in batch and packed-bed reactors was characterized kinetically. In addition to kinetic studies on sucrose hydrolysis at low initial substrate concentrations, the dependence of conversion degree on flow rate at high, industrially used substrate concentrations was determined. The described invertase-polystyrene complexes are suitable for technical application in the production of glucose-fructose mixtures because of their high specific and relative activities, as well as the good hydrodynamical and mechanical properties of the polystyrene matrix.

    Notes: High methanol concentrations have a negative effect on the growth rate and the biomass yield of growth transients induced by methanol pulses in continuous cultures of Methylomonas L3. The physiological basis of this effect is investigated by measuring the effect of the methanol pulse on the cell energy charge (EC) and ATP, ADP, and AMP concentrations, and by comparing the results of the pulse transients against an unstructured model. The methanol pulse is shown to lead to increased values of the cell EC and ATP concentration, and thus, inhibition and reduced availability of biosynthetic energy are excluded as causes of inhibition. When the biomass and methanol profiles of the transient experiments are compared in phase-plane diagrams against computer simulations based on the model, satisfactory agreement between experimental data and model predictions is found in single-substrate, high-dilution-rate experiments. Conversely, poor agreement between experimental data and simulation results indicates a more severe growth inhibition than the model predicts at low dilution rates and a less severe one in mixed-substrate experiments. Based on these findings and other relevant physiological information, we propose that the large variations in the negative effect of methanol on growth result from the fact that cells accumulate methanol to widely different concentrations depending on their physiological state. In their effort to detoxify from the high intracellular methanol and formaldehyde concentrations, cells oxidize considerably more methanol than they can incorporate into biomass. This leads to a useless ATP surplus, which the cells must hydrolyze without doing any useful biosynthetic work, and this results in lower biomass yields.

    Notes: The cellulase activity in cell-free broths from the thermophilic, ethanol-producing anaerobic bacterium Clostridium thermocellum is examined on both dilute-acid-pretreated mixed hardwood (90% maple, 10% birch) and Avicel. Experiments were conducted in vitro in order to distinguish properties of the cellulase from properties of the organism and to evaluate the effectiveness of C. thermocellum cellulase in the hydrolysis of a naturally occurring, lignin-containing substrate. The results obtained establish that essentially quantitative hydrolysis of cellulose from pretreated mixed hardwood is possible using this enzyme system. Pretreatment with 1% H2SO4 and a 9-s residence time at 220, 210, 200, and 180°C allowed yields after enzymatic hydrolysis (percentage of glucan solubilized/ glucan potentially solubilized) of 97.8, 86.1, 82.0, and 34.6%, respectively. Enzymatic hydrolysis of mixed hardwood with no pretreatment resulted in a yield of 10.1%. Hydrolysis yields of 〉95% were obtained from ∼0.6 g/L mixed hardwood pretreated at 220°C in 7 h at broth strengths of 60 and 80% (v/v) and in approximately 48 h with 33% broth. Hydrolysis of pretreated mixed hardwood is compared to hydrolysis of Avicel, a pure microcrystalline cellulose studied previously. The initial rate of Avicel hydrolysis saturates with respect to enzyme, whereas the initial rate of hydrolysis of pretreated wood is proportional to the amount of enzyme present. Initial hydrolysis rates for pretreated wood and Avicel at 0.6 g/L are greater for wood at low broth dilutions (1.25: 1 to 5 :1) by up to 2.7-fold and greater for Avicel at high broth dilutions (5 : 1 to 50 : 1) by up to 4.3-fold. Maximum rates of hydrolysis are achieved at 〈2 g substrate/L for both pretreated wood and Avicel. The substrate concentration at one-half the maximum observed rate for C. thermocellum broths is smaller for pretreated mixed hardwood than for Avicel and decreases with increasing broth dilution for both substrates. An initial activity per volume broth of approximately 11 μmol soluble glucose equivalent produced/L broth/min is observed for mixed hardwood pretreated at 220°C and for Avicel at high broth dilutions; the initial activity per volume broth for Avicel is lower at low broth dilutions. The results indicate that pretreated wood is hydrolyzed at rates comparable to Avicel under many conditions and at rates significantly faster than Avicel under several conditions.

    Notes: A recursive estimation scheme, the Extended Kalman Filter (EKF) technique, was applied to study enzymatic deactivation in the enzymatic hydrolysis of pretreated cellulose using a model previously developed by the authors. When no deactivation model was assumed, the results showed no variation with time for all the model parameters except for the maximum rate of cellobiose-to-glucose conversion (r′m).The r′m variation occurred in two zones with a grace period. A new model of enzymatic hydrolysis of pretreated cellulose deactivation was proposed and validated showing better behavior than the old deactivation model. This approach allows one to study enzyme deactivation without additional experiments and within operational conditions.

    Notes: A mathematical model which describes ethanol production in a packed bed fermenter containing. Zymomonas mobilis entrapped in small spheres of calcium alginate within a packed bed fermenter has been developed. The equations combine simultaneous diffusion and reaction as well as a complex flow pattern to calculate glucose and ethanol profiles in the column type reactor. As part of the study, diffusivity values for glucose and ethanol in cell-loaded calcium alginate were determined. Also a freecell kinetic expression for Z. mobilis at 33°C and ph 6.0 was developed. Comparison of the model with actual experimental results were made showing average deviations of ca. 30-40%.

    Notes: There is good evidence that the specific feeding and growth rates of a population of suspension-feeding ciliates on a population of bacteria is described by the classic Monod model or something very like it. However, use of this model combined with the assumption that feeding of the ciliates is the only interaction between the populations leads to predictions grossly at variance with experimental observations. A second interaction between the populations, growth of the bacteria on products of lysis, and perhaps metabolism of the ciliates is known to occur and might be an important factor in determining the dynamics of ciliate-bacterial systems. Whether or not this is the case is examined in the present article. It is concluded that accounting for the second interaction does not remove the discrepancies between model predictions and experimental observations. It is concluded also that the second interaction is likely not important except in batchtype situations where the supply of available energy for supporting biological activity is not replenished.

    Notes: The technique of autohydrolysis steam explosion was examined as a means for pretreatment of sugarcane bagasse. Treatment conditions were optimized so that following enzymatic hydrolysis, pretreated bagasse would give 65.1 g sugars/100 g starting bagasse. Released sugars comprised 38.9 g glucose, 0.6 g cellobiose, 22.1 g xylose, and 3.5 g arabinose, and were equivalent to 83% of the anhydroglucan and 84% of the anhydroxylan content of untreated bagasse. Optimum conditions were treatment for 30 S with saturated steam at 220°C with a water-to-solids ratio of 2 and the addition of 1 g H2SO4/100 g dry bagasse. Bagasse treated in this manner was not inhibitory to fermentation by Saccharomyces uvarum except at low inoculum levels when fermentation time was extended by up to 24 h. Pretreated saccharified bagasse was inhibitory to Pachysolen tannophilus and this was attributed to the formation of acetate from the hydrolysis of acetyl groups present in hemicellulose. The major advantage of the pretreatment is the achievement of high total sugar yield with moderate enzyme requirement and only minor losses due to sugar decomposition.

    Notes: In order to carry out an effective enzyme reaction, the preparation of soluble-insoluble immobilized enzyme was investigated. Proteases were selected as model enzymes, and their immobilization was carried out by using an enteric coating polymer as a carrier. Among the polymers tested, methacrylic acid-methylacrylate-methylmethacrylate copolymer (MPM-06) gave the most active soluble-insoluble immobilized papain. This immobilized papain showed insoluble from below pH 4.8 and soluble form above pH 5.8; it was also soluble in water-miscible organic solvent. It was reusable and more stable with heat and water-miscible organic solvents than native proteases. Furthermore, various proteases could be immobilized by using MPM-06 with high activity. Chymotrypsin immobilized by this method catalyzed the effective peptide synthesis in a heterogeneous reaction system containing water-miscible organic solvent.

    Notes: Static and dynamic characteristics of continuous cultures of recombinant methylotrophs, which are designed to improve the selectivity of plasmid-bearing cells and the plasmid stability, are investigated in detail. Operational regions in which coexistence (survival of plasmid-bearing and plasmid-free cells) operation is feasible have been identified in the entire space of kinetic parameters and operating variables. The stability characteristics of each steady state are examined. The existence of oscillatory states around the coexistence steady state is investigated using the dynamic (Hopf) bifurcation analysis. For proper startup of the continuous culture operation, it is critical to identify the sets of initial conditions, if any, which lead to transients that ultimately result in washout of plasmid-bearing cells and avoid such conditions. For the numerical illustrations presented, the coexistence steady state happens to be locally stable over much of its region of existence, particular for the operating conditions corresponding to maximum productivity.

    Notes: The effect of operating parameters like pH, protein concentration, column geometry, and gas flow rate on the separation efficiency of proteolytic enzymes from crude human placental homogenate has been studied in a batch foam column. Purification has been found to be optimum at pH 8.0, close to the isoelectric pH, at which the surface adsorption of the protein on the foam bubbles is maximum. Both purification and recovery varied significantly with total protein concentration. Stable bubble formation was hindered at lower protein concentrations, while extraneous proteins rather than the protease were preferentially adsorbed at higher protein concentrations, decreasing the purification efficiency. Column diameter and column height should be optimized for any specific feed protein concentration and gas flow rate. However, the enrichment ratio was found to decrease with the increase in flow rate. The results indicate that foam fractionation is an effective separation process for recovering valuable biochemicals from biological materials.

    Notes: An artisanal static process for protein enrichment of cassava by solid-state fermentation, developed in laboratory and tested on pilot units in Burundi (Central Africa), provides enriched cassava containing 10.7% of dry matter protein versus 1% before fermentation. Cassava chips, processed into granules of 2-4-mm diameter, are moistened (40% water content) and steamed. After cooling to 40°C, cassava is mixed with a nutritive solution containing the inoculum (Rhizopus oryzae, strain MUCL 28627) and providing the following per 100 g dry matter: 3.4 g urea, 1.5 g KH2PO4, 0.8 g MgSO4·7H2O, and 22.7 g citric acid. For the fermentation, cassava, with ca. 60% moisture content, is spread in a thin layer (2-3 cm thick) on perforated trays and slid into an aerated humidified enclosure. The incubation lasts ± 65 h. The production of protein enriched cassava is 3.26 kg dry matter/m2 tray. The effects of the variation of the nutritive solution composition and the inoculum conservation period on the protein production are equally discussed.

    Notes: An optimal time of 45 min was established for extraction of rennin (chymosin) from abomasa using ultrasound. Effect of swelling of raw material and increased temperature of extraction medium (45°C) was established. As rennin is resistant to ultrasound treatment and enhanced temperatures, ultrasound extraction results in a significant increase in the yield and activity. However, purified rennins were sensitive to ultrasound action. There were found changes of the electrophoretic properties and amino acid composition. Physicochemical properties were different and quality characteristics were higher for ultrasonic extracted rennin than for the control, particularly specific clotting activity, proteolytic activity, and rheological properties of curd. However, nonspecific proteolysis, coefficient of rennin effectivity, appearance, odor, solubility, moisture, protein, and NaCI content were similar. Ultrasonic extraction caused a decrease in the total viable counts of microorganisms of rennin, because of the short time of extraction.

    Notes: A simple and convenient method for measuring KLa in large-scale fermentors was proposed. This method was based on the measurement of the dissolved oxygen concentration under steady state conditions established by an equivalency of the sulfite ion feed and chemical oxidation rates. This method had the following advantages: It was a steady state method, and so it was not necessary to consider the response lag of a dissolved oxygen probe and the response lag due to gas phase mixing in fermentors. The oxygen content of the effluent gas in this measuring system was nearly the same as that of the sparged air. Therefore, it was possible to use the oxygen partial pressure of the sparged air for the calculation of the driving force of oxygen transfer. The detailed information on the kinetics of sulfite oxidation was not necessary, because the dissolved oxygen concentration in steady state was not influenced by sulfite oxidation rates. The KLa measurement was finished in as short a period as 150 s, even in a fermentor with a volume of 10 m3. Since the amount of Na2SO4 accumulation in the test fermentors was very small because of the quick measurement, the KLa values obtained by this method were applicable to the electrolyte-free system. Furthermore, we could discharge the used liquid from the fermentors into a drain without any pretreatment due to the low salt concentration.

    Notes: Interactions between two bacterial species which exhibit opposite substrate preferences in response to the same environmental conditions were investigated. Klebsiella oxytoca preferentially utilizes glucose over citrate as a substrate while Pseudomonas aeruginosa utilizes citrate over glucose. Both species were grown on single as well as mixed substrates, namely, D-glucose and citrate, with various ratios of inocula and substrates. Competition is observed between the two species with citrate alone while amensalism is observed with glucose alone. In mixed cultures-mixed substrates experiments, amensalism of K. oxytoca on P. aeruginosa prevailed until the exhaustion of glucose.

    Notes: Acinetobacter has been found to be the major species responsible for mediating biological phosphate removal. The growth kinetics and phosphate uptake were investigated for an isolated Acinetobacter strain growing in a defined medium. The phosphate uptake is dependent on growth rate, temperature, and pH. Polyphosphate granules occurred in a balanced growth stage. The maximum phosphorus content in cells was 4.8% at the dilution rate of 12 day-1. The specific phosphate uptake rate was found to be a quadratic polynomial function of the dilution rate. Increased calcium (up to 36 mg/L) and magnesium (up to 15 mg/L), and the addition of yeast extract (100 mg/L), primary effluent (20%), and fluoride (10 mg/L) did not affect phosphate uptake. Anaerobic conditioning (N2 stripping), low pH (CO2 stripping), and addition of sodium acetate under anaerobic conditions failed to stimulate immediate phosphate release. Nevertheless, After 21-24 h, the phosphate release was ca. 3, 5, and 15 mg P/g cell, respectively, for N2 purging, the addition of acetate, and CO2 purging. For two-stage completely stirred reactor operation, there was negligible phosphate overplus at the second reactor when phosphate was added, when the first reactor was subjected to phosphate limitation. When both phosphate and carbon limited the growth in the first reactor, there was slight phosphate accumulation under endogenous respiration conditions in the second reactor.

    Notes: Toluene was oxidized by a mutant strain of Pseudomonas putida (strain NG1) to toluene Cis-Glycol (TCG). Product was accumulated in fed-batch cultures to concentrations (18-24 g/L) higher than hitherto achieved. In vitro activities of toluene dioxygenase from P. Putida NG1 were fivefold lower than that from the toluene-grown wild-type organism, whereas comparable activities of both catechol 2,3- and catechol 1,2-oxygenase were obtained; irreversible inhibition of toluene dioxygenase activity by TCG was shown in vitro. Ammonia deprivation during the production phase limited the growth of revertant organisms but had little effect on either the duration (25h) of the process or the final concentration of TCG achieved. The rate of glucose utilization decreased throughout the biotransformation and cell death accompanied the cessation of TCG accumulation in cultures. These changes were a consequence of TCG formation and a cooperative toxic effect was demonstrated for toluene and TCG. Adenylate energy charge values decreased from ca. 0.8 to 0.2 over the course of the biotransformation but were maintained above 0.5 in the absence of TCG. Similarly, cellular AMP levels increased dramatically during biotransformation, presumably as a consequence of RNA degradation, but were maintained at low levels in the absence of TCG. The results suggest that TCG is the mediate of a gradual deterioration in the state of the culture which leads to a loss of both in vivo and in vitro toluence dioxygenase activity and a marked decrease in culture viability.

    Notes: A process has been developed for the bulk purification of cellulase-free β-1,4-D-xylanase from the fungus Trichoderma harzianum E58. The process involved the primary step of ultrafiltering the culture filtrate via a 10,000-molecular-weight cut-off membrane to separate the cellulase (retentate) and xylanase (permeate) fractions. The cellulase component was concentrated by 40- to 60-fold, resulting in an enzyme complex that could effectively hydrolyze high concentrations of cellulose and xylan to glucose and xylose. The xylanase was concentrated and solvent exchanged by adsorption to a cationic exchanger, SP-ZetaPrep 250, followed by elution with a pH change in the buffer to give a purified and concentrated xylanase complex dissolved in a low-salt buffer. The resultant xylanase system was pure by the criteria of sodium dodecyl sulfate polyacrylamide electrophoresis, had a very high specific activity of 2400 IU/mg protein, was virtually free of filter paper activity, and had a ratio of contaminating filter paper activity of 2 × 10-6 (0.009% endoglucanase activity). Approximately 3.3 g protein, which contained in excess of 7 × 106 IU xylanase activity, was obtained from 17 L original culture filtrate. The process scheme was designed to facilitate scale-up to an industrial level of production.

Through comparison of corresponding intrinsic and non-intrinsic models in dimensionless form, constraints for non-intrinsic model usage in terms of biokinetic and fermenter operating parameters can be identified a priori. Analysis of a simple product-inhibition model indicates that the non-intrinsic approach is suitable only when the attainable biomass volume fraction in the fermentation broth is less than about 0.10. Inappropriate application of a non-intrinsic model can lead to gross errors in calculated substrate and product concentrations, substrate conversion, and volumetric productivity.

    Notes: Ethanol fermentation by yeast was carried out in a cell filtration recycle system with a hollow-fiber membrane filter. Maximum biomass concentrations up to 210 g dry wt/L were obtained, but in normal operation concentrations they were between 100 and 150 g/L. The ethanol productivity using 14% glucose feed was 85 g/L h, with an ethanol concentration of 65 g/L and an ethanol yield of over 90%. The ethanol productivity and yeast growth rate decreased as the cell concentration increased beyond a certain level. The cell mass in the reactor was maintained by a proper manipulation of diluticn rate and bleed ratio depending on the growth rate.

    Notes: The experiment of bacteria adhesion onto starch granules is conducted. It is found that anaerobic saccharolytic bacteria have the highest adhesion ability in their growth and initial stage of stationary phase. Starch granules with a low crystallinity, low bulk density, and high water-holding capacity have a high adhesion capacity. The optimum temperature for both bacterial growth and their adhesion is 30°C. The optimum pH for the bacterial adhesion range from 5.0 to 6.5. Anaerobic conditions cause an appreciable decrease in percentage of adhesion. The percentage of adhesion is not sensitive to the type of soluble saccharide on which bacteria were grown.

    Notes: Ethanolic fermentation of spent sulphite liquor with ordinary bakers' yeast is incomplete because this yeast cannot ferment the pentose sugars in the liquor. This results in poor alcohol yields, and a residual effluent problem By using the yeast Candida shehatae (R) for fermentation of the spent sulphite liquor from a large Canadian alcohol-producing sulphite pulp and paper mill, pentoses as well as hexoses were fermented nearly completely, alcohol yields were raised by 33%, and sugar removal increased by 46%. Inhibitors were removed prior to fermentation by steam stripping. Major benefits were obtained by careful recycling of this yeast, which was shown to be tolerant both of high sugar concentrations and high alcohol concentrations. When sugar concentrations over 250 g/L (glucose: xylose 70:30) were fermented, ethanol became an inhibitor when its concentration reached 90 g/L. However, when the ethanol was removed by low-temperature vacuum distillation, fermentation continued and resulted in a yield of 0.50 g ethanol/g sugar consumed. Further improvement was achieved by combining enzyme saccharification of sugar oligomers with fermentation. This yeast is able to ferment both hexoses and pentoses simultaneously, efficiently, and rapidly. Present indications are that it is well suited to industrial operations wherever hexoses and pentoses are both to be fermented to ethanol, for example, in wood hydrolysates.

    Notes: The effect of surface coverage of biomass on biofilm kinetics in an expanded-bed, granular activated carbon an aerobic reactor was evaluated. Acetate was used as the sole organic carbon source. An assumption of 10% surface coverage of biofilm was examined and compared to 100% coverage. Best estimated values of ka and Ksa did not differ significantly from one case to the other. The confidence region analysis also showed that the biofilm was fully penetrated in the expanded-bed reactor for the case of 10% coverage, as well as when 100% coverage was assumed. Because the biofilm was fully penetrated, a model having no internal diffusion resistance and using the best estimates of ka and Ksa obtained from the 10 and 100% coverage assumptions was capable of giving good predictions of effluent acetate concentrations for an in dependent experiment having a reduced liquid detention time. Consideration of biofilm surface-loading criteria demonstrated how the results can be applied to other reactors for the purpose of predicting when the extent of surface coverage and internal diffusion resistance are not significant factors in biofilm modeling.

Flow pattern behind the board consists of large vortex loops causing the fluid to circulate from bottom to top and producing a thorough mixing effect. The drag board seems to have several advantages over conventional paddle wheels for the mixing and stirring of algal cultures in shallow ponds.

    Notes: The Luedeking-Piret equation was used to fit the kinetic data of pullulan fermentations from peat hydrolyzate substrate. In batch mode, the kinetic parameters m, n, α, and β varied as a function of fermentation conditions: aeration rate, agitation speed, and temperature. In constant-feed fed-batch mode, the parameters Varied according to the feed rates. In peat hydrolyzate medium, the polysaccharide synthesis was strongly growth associated in batch and continuous fermentations but entirely growth associated in fedbatch fermentations. The fed-batch mode of fermentation with an appropriate feed rate is more advantageous with respect to batch and continuous fermentations. Therefore, if the fermentation is started batchwise and then followed by fed-batch mode at a constant feed rate, the overall polysaccharide productivity (g pullulan/L h) is significantly higher than those obtained with batch or continuous fermentations using the same total medium volume.

    Notes: Volumetric mass-transfer coefficients were measured in an 11-L external-loop airlift fermenter with deionized water, a fermentation medium, and during a fermentation. Both a Mackareth oxygen electrode and a novel rapid-response probe were used. When the conventional step-change dynamic method was used for water, the long, nonlinear response time of the Mackareth electrode made correction of its readings difficult; this problem did not occur when the rapid-response probe was used. A comparison was made with a method of mass-transfer coefficient determination which does not involve any assumptions about the gas residence time distribution. However, this method requires that the liquid phase is well-mixed and this requirement was not met in the airlift fermenter. Comparison of the present results with other KL a determinations for airlift fermenters showed that KL a in water depends on the active gas holdup, the value of KL a/∊ at 20°C being ca. 0.37 s-1. Although higher gas holdups were obtained with the fermentation medium than for water, the values of KL a/∊ were lower, ca. 0.22 s-1 at 20°C.

    Notes: A theoretical relationship has been developed to allow the effect of free ligand concentration on the capacity of an affinity Chromatography matrix to be determined where the protein adsorbed shows co-operative binding. Computer simulations using literature values for association constants show that under optimal conditions resin capacity can be increased significantly in the presence of a small but finite concentration of free ligand. The model also allows prediction of the soluble ligand concentration required for biospecific elution. The results obtained suggest the possibility of a new elution technique, “reverse biospecific elution,” that reduces the amount of free ligand required to effect elution.

    Notes: A start-up routine for a novel type of anaerobic gas-lift reactor using sand as support particles for Bacterial adhesion and involving a dilution rate shift-up is shown to result in rapid formation of mixed-culture aggregates from freely suspended cells. Formation of aggregates changed the general metabolism from acetate-butyrate production to acetate-propionate production. This change is attributed to a selection by washout, favoring propionate-producing bacteria with superior adhesive properties. Sand is shown to be essential in establishing, but not in sustaining, elevated holdup ratios. The importance of maintenance processes and cellular lysis in deeper parts of aggregates are manifest from a reduced effluent biomass concentration and a pronounced production of valeric acid.

    Notes: Five strains each of Gibberella fujikuroi and Fusarium monoliforme were screened to select G. fujikuroi P-3, a strain capable of giving consistent production of gibberellic acid (GA3) by solid state fermentation (SSF). The comparative production of GA3 by SSF and submerged fermentation (SmF) indicated better productivity with the former technique. The accumulation of GA3 was 1.626 times higher in the case of SSF. On the basis of available carbohydrates in the media, the percent conversions were 0.096 and 0.156 in SmF and SSF, respectively. The use of coarse wheat bran of the particle size of 0.3-0.4 cm resulted in an increase of 2.5 times in the yield of GA3. The enrichment of commercial wheat bran with soluble starch gave enhanced accumulation to an extent of 3.5 times. The relation between GA3 production and cell growth in SSF was similar to that encountered in SmF. The consistent and improved yields to a tune of 1.22 g GA3 per kilogram dry moldy bran (DMB) establish the potential and feasibility of SSF for the production of GA3 by G. fujikuroi P-3. On preliminary cost analysis, a net savings of about 60% and 50% on fermentation medium cost and the expenditure on down-stream processing, respectively, as compared to the presently employed SmF technique was evident.

    Notes: An adaptive steady-state optimization algorithm is presented and applied to the problem of optimizing the production of biomass in continuous fermentation processes. The algorithm requires no modeling information but is based on an on-line identified linear model, locates the optimum dilution rate, and maintains the chemostat at its optimum operating condition at all times. The behavior of the algorithm is tested against a dynamic model of a chemostat that incorporates metabolic time delay, and it is shown that large disturbances in the subtrate feed concentration and the specific growth rate, causing a shift in the optimum, are handled well. The developed algorithm is also used to drive a methylotroph single-cell production process to its optimum.

    Notes: The changes in growth kinetics in aerobic batch cultures of Klebsiella pneumoniae were followed by measurements of extracellular metabolites, rates of gas exchange, dissolved oxygen tension, pH, and carbon balance at all stages of growth. When the initial growth-limiting glucose concentration in media without pH control was increased from 1.0 g carbon L-1 to 2.2 g carbon L-1, the number of different, mainly acidic, extracellular metabolites of glucose at the end of exponential growth increased, while the proportion of acetate decreased. During the postexponential growth phase, the extracellular metabolites were oxidized, resulting in an increasing complexity of changes in pH, gas exchange, and dissolved oxygen tension with increasing initial substrate concentration. All these parameters showed concomitant stepwise changes. This pattern was independent of the dissolved oxygen tension in the range 30-200 μM. When pH was kept constant, the number, slope, and relative magnitude of the steps in gas exchange and dissolved oxygen tension were pH-dependent, being most complex at low pH. Detailed carbon balances showed that 20% of the initial glucose was converted into extracellular metabolites at the end of exponential growth at neutral pH. In the postexponential phase, pyruvate (2%) was reoxidized first followed by acetate (13%). The observed molar growth yield coefficient (YATP) was 8.4 if the transitory occurrence of pyruvate and acetate was accounted for, and 6.4 if it was neglected. The corrected observed molar growth yield coefficient (Y′ATP) was 9.4 and compared well with the true molar growth yield coefficient (YMaxATP), which was found to be 11.0. Specific in situ respiration rates of the exponential growth phase of cultures grown at different controlled pH values compared well with in situ values for energy-limited chemostat grown cells at the same growth rates, suggesting that growth in the batch culture was energy-limited throughout the exponential growth phase. This view was supported by low levels of intracellular glycogen and exopolysaccharides of all cultures, by the value of Y′ATP of 9.4, and by a constant specific production rate of the extracellular metabolites throughout exponential growth. It was concluded that even under strictly aerobic conditions, control of pH is as important as control of dissolved oxygen tension during growth of enterobacteriaceae in batch cultures.

    Notes: Experimental and modeling studies were conducted to analyze the dynamic response behavior of a phenol-oxidizing fixed film using a differential, fluidized-bed bioreactor in a recycle loop with a well-mixed reservoir. With the bioreactor at steady state, a pulse of phenol was added to perturb the system, and the phenol concentration was monitored continuously until steady state was again achieved.The experimental dynamics were compared with a dynamic mathematical model based on diffusion and reaction within the biofilm, liquid mixing, and biofilm growth. Constant-pH experiments could be adequately described using an unstructured, double-Monod kinetic expression with substrate inhibition by phenol.However, in dynamic experiments without pH control, the pH of the liquid phase dropped, and damped oscillations were observed in the phenol concentration and reaction rate trajectories. Oscillatory solutions could not be induced in the model, even when product inhibition was included, and a linear stability analysis did not reveal tendencies toward instability. The cause of the experimental oscillations remains unknown.

    Notes: The adhesion of three microorganisms (Saccharomyces cerevisiae, Acetobacter aceti, and Moniliella pollinis) to different materials has been studied using various supports (glass, metals, plastics), some of which were treated by an Fe(III) solution. The surface properties of the cells were characterized by the zeta potential and an index of hydrophobicity; characterization of the supports involved surface chemical analysis (XPS) and contact angle measurements. Cell suspensions in pure water at a given pH were left to settle on plates; the latter were then rinsed and examined microscopically, Saccharomyces cerevisiae and A. aceti adhere to metals under certain pH conditions but do not adhere to any of the other materials tested unless it is previously treated by ferric ions; adhesion of these hydrophilic cells is essentially controlled by electrostatic interactions. Moniliella pollinis adhere spontaneously to glass and to polymeric materials, but its attachment is also influenced by cell-cell or cell-support electrostatic repulsions; near the cell isoelectric point, cell flocculation is competing with adhesion to a support.

    Notes: The optimal periodic operation of the biological reactor in which the metabolites belonging to Type I or II in Gaden's classification are produced was investigated from the viewpoint of the multiobjective programming problem. In addition to the productivity of the desired product, its concentration and the conversion of the substrate which have a large influence on the performance of the separation process following the fermentation process were adopted as the objective functions. The growth of cells was assumed to be inhibited both by the substrate and the product, and the Luedeking-Piret model was employed for the specific production rate. The optimal periodic operation was determined by use of the optimization method due to Miele. It was clarified that the noninferior set for the periodic operation was generally composed of the repeated batch portion and the repeated fed-batch one.

    Notes: An identifiedEnterobacter aerogenesutilizing methanol as a sole carbon source was studied for the optimization of biomass production and the reduction of its nucleic acid content. Results indicated that the highest yield and conversion were obtained at 0.5% methanol. The addition of seawater as a source of trace elements has an adverse effect. However, the addition of urea as source of nitrogen enhanced the growth of E. aerogenes. Heat shock at 60°C for 1 min followed by incubation at 50°C for 2 h caused 72.6% reduction in the nucleic acid.

    Notes: L-Phenylalanine was produced continuously from L-as-partate and phenylpyruvate by transaminase from a newly screened Pseudomonas putida strain. The process was carried out with an isolated enzyme in homogeneous phase in an enzyme membrane reactor and with immobilized whole cells in a stirred tank reactor, respectively. Due to the difference in transport resistance, the productivity of the free enzyme in homogeneous phase (72 mmol/L h) was about 3 times higher than the productivity achieved using immobilized cells. However, a better stability of the biocatalyst was observed with immobilized cells.

    Notes: Biological denitrification of drinking water was studied in a fluidized sand bed reactor using a mixed culture. Hydrogen gas was used as the reaction partner. The reaction kinetics were calculated with a double Monod saturation function. The Ks value for hydrogen was below 0.1% of saturation. No appreciable biofilm diffusion effects were detected. Reactor performance was a function of the culture's past history. Batch experiments always exhibited an accumulation of NO2-, but continuous experiments with a sufficiently long residence time always resulted in complete nitrogen removal. Rates of up to 23 mg N/L h, 25 mg N/g DW h, and 7.9 mg H2/L h were achieved. Residence times of 4.5 h would be required for complete denitrification of water containing 25 mg NO3--N/L or approximately 1 h for every 5 mg/L.

    Notes: A dual hollow fiber bioreactor, consisting of an outer silicone membrane for oxygen supply and an inner polyamide membrane for substrate permeation, was used as an immobilized enzyme reactor to carry out enzymatic glucose oxidation. Attaching a silicone tube oxygenator to provide an additional oxygen supply improved the conversion in glucose oxidation when the oxygen supply was rate-limiting. The reactor was operated in both diffusion and ultrafiltration modes. In the latter case, the conversion was much higher, but the stability of the immobilized enzyme was better maintained in the diffusion mode. As the inlet glucose concentration increased from 10mM to 500mM, the conversion decreased from 70 to 20%.

    Notes: A kinetic model for the agglomeration of milk micelles following κ-casein hydrolysis is described. The key features of the model are: (1) the surface potential of casein micelles is sufficient to explain the colloidal stability of the milk system; (2) the reduction in surface potential following κ-casein hydrolysis explains the loss of stability; (3) partial hydrolysis leads to limited agglomeration; and (4) the kinetics of agglomeration are compatible with the theory that completely hydrolyzed micelles have only a limited number of interaction sites. The model accurately predicts solution turbidity increase assuming that micelles have only circa 1.2 interaction sites on the average under the experimental conditions of this study.

    Notes: A mechanistic kinetic model of gel firmness development during milk gel formation is presented. The model correctly accounts for the influence of enzymatic κ-casein hydrolysis on the rate of firmness development in renneted milk gels. The model used is based on two first-order reactions occurring in series. The first reaction is enzymatically controlled and corresponds to the formation of gel crosslink sites by κ-casein hydrolysis. The second reaction is nonenzymatic and corresponds to the process of crosslink formation and depletion of active sites. The model successfully predicts gel firmness development in the temperature range 31-45°C for a variety of initial enzyme concentrations.

    Notes: Methanogenesis was studied in downflow stationary fixed-film bioreactors. The support materials in this study included ceramic Raschig rings, hardwood chips, and sized charcoal. The performances of these support materials have been compared using both synthetic acid mixture and acid products obtained from paper mill sludge. Woodchips appeared to be the most promising support material: The maximum methane productivity of 3.56 L/L day at a nominal retention time of 0.78 day was obtained using initial total acid concentrations of 9.125 g/L. Higher productivity was achieved at the cost of efficiency of the process in terms of conversion of acids. From nitrogen balances, it was deduced that ammonia supplemented methane generation by supplying hydrogen for there duction of carbon dioxide.An ionic balance was developed to ascertain the relationship between the composition and the pH of the liquid and the mole fraction of carbon dioxide in the gas phase. From these ionic balance equations, it was possible to predict the gas phase composition at various retention times. The maximum error between the computed and the experimental values was less than 13%.

    Notes: The growth of Thiobacillus ferrooxidans in a copper-containing ore suspension incubated in shake flasks was studied by determining the number of colony-forming units both in solution and attached to ore particles. The amounts of iron and copper released from the ore under experimental conditions were also determined. The total ferrous iron either released from the minerals or generated by reduction of the ferric iron in the minerals could account for the observed growth of bacteria in solution. Only a small fraction of the total colony-forming units - about 500 per mg ore - was found to be associated with the ore particles throughout the experiments. However, the rapid development of these colonies when ore particles were plated suggested that they were produced by a number of bacteria associated with each ore particle. Accordingly, when the amount of bacteria attached to ore particles was determined by monitoring the formation of ferric iron in the plates, the percentage of the total activity associated with attached bacteria was found to be between 1 and 10%.

    Notes: A tapered column type of bioreactor system packed with immobilized Saccharomyces cerevisiae was used to study the bioreactor performance as a function of design and operating variables. The performance of tapered column bioreactor system was found to be better than that of the conventional cylindrical column reactor system for the ethanol fermentation. The new bioreactor design alleviated problems associated with carbon dioxide evolution and provided a significantly better flow pattern for both liquid and gas phases in the bioreactor without local channelling. A mathematical simulation model, which takes into account of the axial convection and dispersion, interphase mass transfer, and apparent kinetic design parameters, was developed. The effect of radial concentration gradients on the bioreactor performance was found to be insignificant. For the reactor system studied, the maximum ethanol productivity obtained was 60 g ethanol/L gel/h, and the maximum glucose assimilation rate was 140 g glucose/L gel/h. One of the most important findings from this study was that the apparent kinetic parameters change at the glucose concentration of 2 g/L This change was found to be due to the changes in yeast physiology and metabolism. The values of Vm′ and Vm′ decreased from 0.8 to 0.39 g ethanol/g cell/h and from 97mM to 11mM, respectively. The substrate inhibition constant was estimated as 0.76M and the product inhibition constant was determined as 113 g ethanol/L The degree of product inhibition showed practically a linear relationship with an increasing ethanol concentration. Based on the hydro-dynamic analysis of the bioreactor system, it was found that the Peclet number, NPe was not a strong function of the flow velocity at low flow rates through the bioreactor system, but its value decreased somewhat at an interstitial velocity greater than 0.03 cm/s. The tapered column bioreactor system showed a much better flow pattern of gas and liquid phases within the reactor, thereby providing a more homogeneous distribution of gas-liquid-solid phases in the reactor without any phase separation.

    Notes: A new immobilized cell system providing protection against toxic solvents was investigated so that normal fermentations could be carried out in a medium containing toxic solvents. The system consists of immobilized growing cells in Ca-alginate gel beads to which vegetable oils, which are inexpensive absorbents of solvents, had been added. The ethanol fermentation of Saccharomyces cerevisiae ATCC 26603 was used as a model fermentation to study the protection afforded by the system against solvent toxicities. The fermentation was inhibited by solvents such as 2-octanol, benzene, toluene, and phenol. Ethanol production of one batch was not finished even after 35 h using immobilized growing yeast cells in conventional Ca-alginate gel beads in an ethanol production medium (5% glucose) containing 0.1% 2-octanol, which is used as a solvent for liquid-liquid extraction and is one of the most toxic solvents in our experiments. With the new immobilized growing cell system using vegetable oils, however, four repeated batch fermentations were completed in 35 h. Castor oil provided even more protection than soy bean, olive, and tung oils, and it was possible to complete six repeated batches in 35 h. The immobilized cell system with vegetable oils also provided protection against other toxic solvents such as benzene and toluene. A possible mechanism for the protective function of the new immobilized cell system is discussed.

    Notes: Polymers produced by horseradish-peroxidase-catalyzed coupling of phenols have been explored as potential substitutes for phenol-formaldehyde resins. To overcome low substrate solubilities and product molecular weights in water, enzymatic polymerizations in aqueous-organic mixtures have been examined. Peroxidase vigorously polymerizes a number of phenols in mixtures of water with water-miscible solvents such as dioxane, acetone, di-methylformamide, and methyl formate with the solvent content up to 95%. As a result, various phenolic polymers with average molecular weights from 400 to 2.6 × 104 D were obtained depending on the reaction medium composition and the nature of the phenol. Peroxidase-catalyzed copolymerization of different phenols in 85% dioxane was demonstrated. Poly(p-phenylphenol) and poly(p-cresol) were enzymatically prepared on a gram scale. They had much higher melting points, and in addition, poly(p-phenylphenol) was found to have a much higher electrical conductivity than phenol-formaldehyde resins.

    Notes: General rules for the optimization of different biocatalytic systems in various types of media containing organic solvents are derived by combining data from the literature, and the logarithm of the partition coefficient, log P, as a quantitative measure of solvent polarity. (1) Biocatalysis in organic solvents is low in polar solvents having a log P 〈 2, is moderate in solvents having a log P between 2 and 4, and is high in a polar solvents having a log P 〉 4. It was found that this correlation between polarity and activity parallels the ability of organic solvents to distort the essential water layer that stabilizes the biocatalysts. (2) Further optimization of biocatalysis in organic solvents is achieved when the polarity of the microenvironment of the biocatalyst (log Pi) and the continuous organic phase (log Pcph) is tuned to the polarities of both the substrate (log Ps) and the product (log Pp) according to the following rules: |log Pi - log Ps| and |log Pcph - log Pp| should be minimal and |log Pcph - log Ps| and |log Pi - log Pp| should be maximal, with the exception that in the case of substrate inhibition log Pi, should be optimized with respect to log Ps In addition to these simple optimization rules, the future developments of biocatalysis in organic solvents are discussed.

    Notes: The influence of microheterogeneity on enzyme inactivation kinetics is presented. Examples of different enzymes are given where microheterogeneity has been detected by different techniques. The different statistical models are presented which include the influence of microheterogeneity on enzyme inactivation kinetics and stability. As the microheterogeneity of the enzyme increases, there is a sharper decline in the normalized activity during the initial stages of the deactivation but a greater stability and activity, compared to similar homogeneous enzyme, as the deactivation proceeds. Microheterogeneity makes the deactivation reaction have a higher apparent order of reaction. The implications of microheterogeneity on enzyme inactivations are high lighted by different examples. The analysis provides fresh physical insights into the chemistry, subpopulations, structure, and function of enzymes.
