    Notes: Varicocele occurs in approximately 15% of the male population. It affects the left side of the scrotum in 90% of subjects, is bilateral in 9%, and right sided in only less than 1%. Its etiology remains controversial. Hypotheses include: the longer length of the left testicular vein, its right-angle entry into the left renal vein, the “nut-cracker” pressure of the superior mesenteric vein, pressure of a loaded sigmoid colon, failure of the fasciomuscular venous pump of the spermatic cord, and absence of valves in the left vein.We have investigated the last hypothesis in a study of 31 male dissecting room cadavers and 15 male autopsy subjects. On the left side, valves were absent throughout the course of the main testicular vein in 20 subjects, and on the right they were absent in eight. However, on the left side six subjects also had an accessory testicular vein; in five of these, valves were absent. On the right, two subjects had an accessory vein, but both had valves present. Thus a valveless venous pathway of drainage was present in 25 of the subjects on the left and eight on the right. This difference was statistically significant to P 〈 0.001.This correlates with the experimental production of varicocele in the dog 3 months after testicular valve destruction (Saypol et al. J Clin Invest 68:39, 1981).

    Notes: The phenomenon of limb regeneration is briefly described, and factors empirically known to enhance regeneration are reviewed. Regeneration is compared to the initial development of the limb, with which it shares many similarities. Recent advances in understanding of the control mechanisms underlying regeneration are described; in particular, the role of retinoids, and their relationship with homeobox-containing genes are considered. The long-term prospects of inducing regeneration in human amputees are considered, and a cautious “thumbs up” is given to this notion, at least in a limited form.

    Notes: The ligaments of the lumbar neural foramina have not been systematically described. We sectioned the lumbar spines of 15 cadavers in the sagittal plane with a cryomicrotome and identified the ligaments in the neural foramina. Fibrous ligaments were identified in every lumbar neural foramen. The most constant ligament was a band of fibrous tissue behind the anulus fibrosus and distinct from it, originating in one vertebral body and inserting in the next. Ligaments connecting the posterior disc margin and the superior articular process were found in 48% of the neural foramina. A band of fibrous tissue originating from the anulus fibrosus and inserting along the pedicle and superior articular process was found in 44% of the foramina. Three other distinct types of ligament were found less commonly. Ligaments, which some investigators believe may contribute to lateral spinal stenosis, are commonly present in the neural foramina.

    Notes: Postal questionnaires were sent to anatomy demonstrators who had been in that post at the University of Manchester, England, between 1975 and 1986. Replies received from 83 former staff (faculty) (72% response rate), of whom 79 were medical graduates, indicated that the posts were thought of very highly for learning to teach, for studying for the Primary Examinations for Fellowship of the Royal Colleges of Surgeons (FRCS), and for gaining subsequent employment. The 93% success rate for those who took the Primary FRCS Examination, usually while in the post, is remarkable. The results of the survey and the future for demonstrator posts are discussed in the light of changes in the regulations for the FRCS Examinations, changes in the university environment for biological subjects, and changes in the financial circumstances of both the universities and the National Health Service.

    Notes: Magnetic resonance imaging (MRI) is an excellent modality for tomographically visualizing abdominal structures, particularly the liver, In an effort to simulate the natural color scheme of the living abdomen using MRI, we have generated color composites from sets of gray tone MR images obtained at corresponding anatomic slice positions from two healthy individuals. Pulse sequences used for our image sets included T1-, T2-, and proton density-weighted spin echo sequences as well as an angiographic FISP gradient echo sequence. A PC/AT-compatible computer with 24-bit graphics display capbilites, along with commerical and customized image-processing software, was used for composite generation. The applied colors were selected based on quantitative characteristics tissue intensity patterns so that tissue contrast could be optimized in the final image. The generated composites were correlated with cadaver sections to evaluate the color scheme of these false-colored images. With our composite generation techniques, it was possible to generated near-natural appearing color images of the abdomen. Color composites may be useful for teaching human cross-sectional anatomy and may also have diagnostic applications in abdominal MRI studies. © 1992 Wiley-Liss, Inc.

    Notes: We describe a three-dimensional computer-generated model of the human mediastinum. Contours were manually traced from photographs of a serially sectioned, unfixed frozen cadaver block, Using software developed in-house, the contours were digitized, automatically assembled and placed in registration, and displayed on a high performance graphics workstation as surfaced images. The model thus produced has advantages over traditional representations of anatomical structures; it is realistic, accurate, efficient, and flexible. Each anatomical component of the mediastinum is stored as a separate file in the computer and can be selectively accessed, displayed, and manipulated, allowing the model to be disassembled, reassembled, dissected, and viewed in any orientation. The model can be divided into a series of transversely cut “slabs,” the surfaces of which correlated with the anatomy displayed in the original cryosections and with radiological scans, thus permitting 2-D sectional anatomy to be viewed in the context of the 3-D model. Animation of the model can be accomplished by recording pre-planned scripts onto videodiscs. The videodiscs serve as image archives that can be viewed in an interactive, self-paced manner or accessed from a computer-based multimedia system that is being developed by the Digital Anatomist Program. Anatomical reconstructions based on datasets of x-, y-, z-coordinates have potential clinical applications as models that can be displayed to match the sectional planes of radiological images. The mediastinum model we describe is part of a large image database being developed by the Digital Anatomist Program that will form the spatial component of a knowledge base in structural biology. © 1992 Wiley-Liss, Inc.

    Notes: Rib-vertebra angles (RVAs) were measured (T1-12) on the chest radiographs of 412 children aged 0-17 years attending the hospital with minimal disorders or diseases (boys 193, girls 219). A new method for measuring RVAs is used and the asymmetry of rib-vertebra angles is calculated as rib-vertebra angle differences (RVADs). The data are analysed in three age groups - infancy, childhood, and puberty, after the classification of Karlberg (1989).The findings show the following:(1) RVAs decrease from T1-12, especially so between (T8 and T12.2) Between infancy and childhood, RVAs of the upper ribs incrase, more so in boys than girls. (3) Between childhood and adolescence there is a further elevation of ribs, involving more ribs in boy than girls (boy T1-10, girls T1-8). (4) Between infancy and childhood, the lower ribs droop more in girls than boys (boys T9-10/12, girls T7/8-12). There is no change in the RVAs of the lower ribs between childhood and puberty in either boys or girls. (5) The hypothesis is suggested that RVAs are influenced by the central nervous system through its influence on trunk muscle activity. (6) Ribvertebra angle asymmetries (or differences, RVADs) are related to age and sex; their pattern reflects the common age, sex, and laterality patterns of idiopathic scoliosis. Extremes of such asymmetries may be an etiological factor for both infantile and adolescent idiopathic scoliosis. © 1992 Wiley-Liss, Inc.

    Notes: The lateral ulnar collateral ligament (LUCL) of the elbow has been illustrated variably in anatomy texts. The purpose of this investigations was to determine the percentage of specimens in which this structure is present, and to describe its anatomy and function. The LUCL was identified as part of the lateral capsulo-ligamentous complex in 17 of 17 fresh frozen cadaver elbows (P 〈.0001). The LUCL originates on the lateral epicondyle, blends with the fibers of the annular ligament as it arches superficial to it, then curves to insert on the tubercle of the supinator crest of the ulna. It is distinct at its insertion, but not at its origin where its fibers blend with those of the common extensor origin. The insertion is exposed in the interval created between (the deep surface of) the fascia of the supinator and its muscle fibers. The insertion can be palpated by applying a varus stress to the elbow. Cutting the ulnar fibers permitted posterolateral rotatory subluxation of the ulno-humeral joint and varus subluxation of the elbow. © 1992 Wiley-Liss, Inc.

    Notes: No published studies on the reactions of British medical students to human dissection have been found. A class of 167 first year British preclinical students completed a questionnaire covering reactions to the dissecting room (DR) and other causes of stress for medical students, 6 Weeks after starting dissection.Five percent of students were very upset by the DR during the first few sessions while 36% were slightly upset, and 58% were unaffected. Six weeks later none were very upset and 78% were not at all upset by the DR. Those upset by the DR had higher stress levels by a standard measure than those not disturbed. Four percent of students had been so disturbed that their concentration was disrupted during the first DR session. The DR was a relatively unimportant cause of stress to students compared with work-related stressors, such as too much work and exam anxiety. The majority of students felt themselves mentally prepared for the DR. About half had seen a dead body before and these students were signigicantly more likely to feel mentally prepared.Some students are, understandably, upset by their first experiences of the DR but most adapt fairly quickly. It appears that a small number continue to be seriously disturbed. The possiblity of using students'reactions to the DR as an educational exercise for their medical career could be explored further. © 1992 Wiley-Liss, Inc.

    Notes: The present paper proposes a selective antermodial approach to the third cervical vertebra, based on the topographic and stratigraphic anatomy of the carotid regions. The study was performed on 12 subjects and involved 20 observations. The investigations showed the existence of an “access window” limited in one direction by the external carotid artery and pharynx, and in the other direction, by the superior thyroid artery and the hypoglossal nerve.     Notes: This paper provides a decription and a mini-atlas (with bold labeled anatomic drawings and magnetic resonance images) (MRIs) of the pulmonary arterial system and surrounding structures for the purpose of interpreint MRIs of heart patients examined in sagittal planes. The pulmonary arterial system (right outflow tract) was studied by examination of cadaver sections made in sagittal planes and by examination of corresponding MRIs from two living normal adult males. We propose a nontraditional format for describing the pulmonary arterial system by dividing in into four parts, rather than the customary three parts, adding a Pulmonary valve region between the ending of the conus arteriosus and the beginning of the pulmonary trunk. This region contains the entire pulmonary valve. It is approximately 1.5 cm long and is defined or outlined by the triple-scalloped fibrous annulus of the pulmonary valve as decribed by Zimmerman (Ann R Coll Eng 39:348, 1966). Defining this region as a separate anatomic entity provides for a more precise description of the relationships of the left coronary and anteior interventricular arteries, the left sinus of the aortic valve, and left auricle to the outflow tract than that found in conventional descriptive anatomy. This paper also defines the ending of the pulmonary trunk, and the orifices of the left and right pulmonary arteries in relationship to the carina found in the upper posterior wall of the pulmonary trunk. An explanation for preferential enlargement of the left pulmonary artery in pulmonary valve stenosis is correlated to our study. © 1992 Wiley-Liss, Inc.

    Notes: “Late” incisional hernias are those which appear more than 1 year after surgery. However, they are not associated with the same causal factors as “early” incisional hernias. In this study, scanning (SEM) and transmission electron microscopy (TEM) were used to compare the collagen of 12 samples of intact linea alba with 8 samples of scar tissue, aged between 6 weeks and 10 years, taken from patients who had undergone previous midline abdominal incisions. No differences of collagen organization were observed using qualitative SEM. TEM, however, showed that intact tissue has collagen fibrils with larger mean diameters and differs from scar tissue in the shape of its fibril diameter distribution. Collagen fibrils from scar tissue in elderly subjects resembled collagen fibrils from intact tissue of young subjects. Neither TEM or SEM revealed any differences between scars of different maturity. © 1992 Wiley-Liss, Inc.

    Notes: The high recurrence rate of inguinal hernias following primary repair has prompted us to re-examine the anatomy of the inguinal region with particular emphasis on the iliopubic tract (IPT). The IPT is described as an aponeurotic band forming the inferior margin of the transversus abdominis lamina. We documented the presence and degree of development of the IPT in dissections of 151 embalmed inguinal regions and in serial sagittal sections of four body halves. The iliopubic tract was identified in all specimens. It stretched between the anterior superior iliac spine laterally and the pubic tubercle and the pubic tubercle and pectineal line medially. The intervening arch formed a discrete structure of variable thickness and was substantial in 63 specimens. Histological sections demonstrated that the IPT is connected to the inguinal ligament, fascia lata, and anterior femoral sheath and is composed primarily of collagenous fibers with a minor elastic component. These data indicate that the iliopubic tract is a consistent and easily identified structure in the inguinal regions. These results suggest that dissection of the iliopubic tract, like the inguinal ligament, should become an integral part of the assessment of groin anatomy during hernia repair. © 1992 Wiley-Liss, Inc.

    Notes: The gross anatomy course at St. Louis University was designed to utilize peer teaching. Eight students are assigned to each cadaver and they work in teams of two, so that every student does one out of every four dissections. During one class period, the lecture for a unit is given to all class members, followed by a prosection demonstration to only the dissectors of that unit (one-fourth of the class). At the beginning of the following class period, the dissectors give a prosection demonstration of their dissection to the other six student assigned to their cadaver. The lecture and prosection demonstrations for the next unit are given to the next team of dissectors as above. The students anonymously evaluate the course each year. Passing Part I of the National Board Examinations is required. For 12 years, the National Board Unit Examination of gross anatomy was given as the final examination for the course.The course is liked by the students, can be taught with a minimal number of faculty, utilizes only half the cadavers of a traditional course, has less contact hours than the national average, yet students do better than the national average on the anatomy part of the National Board Examination. © 1992 Wiley-Liss, Inc.

    Notes: Morphologic changes in the walls (stratum granulosa and theca interna and externa) of preovulatory follicles were studied in 15 clinically normal women. Ovulation was visualized in 11 of 15 (73%) women. Images were recorded using high-resolution transvaginal ultrasonography. Mean follicular diameter immediately before ovulation was 20.7 ± 0.6 mm. Echoes representing the cumulus oophorus/oocyte complex were observed in 10 of 11 (91%) follicles for which ovulation was ascertained. Thickness of the deep internal wall increased (P 〈 0.02) beginning from 4 to 5 h before ovulation. The apical wall thinned (P 〈 0.0001) to nondetectable limits immediately before follicular rupture. Four women (27%) did not ovulate during the imaging period [34-41 h post-human chorionic gonadotropin hCG]. The four follicles that did not ovulate remained of uniform thickness; stigma formation was not observed. The data supported the hypothesis that ultrasonographically detectable alterations in preovulatory follicles precede ovulation. Clinical applications include prediction of the potential for, and interval to, rupture of the preovulatory follicle and timing of ova retrieval in women undergoing ovarian stimulation for amelioration of infertility. © 1992 Wiley-Liss, Inc.

    Notes: This paper reports a segmental analysis of the lateral spinal radiographs of 37 children referred to hospital in a school screening study of 4,890 school children aged 12-13 years. On each lateral radiograph a line was drawn along the posterior surface of each vertebral body from T5-L5 and the angle of this line from the vertical was recorded.After measuring the scoliosis curve (Cobb) angle and using conventional criteria for diagnosis, three groups of patients are defined, namely, 1) a control group (average scoliosis curve angle (Cobb) of 5°, n = 14), 2) a group with lumbar curves (average Cobb angle 21°, n = 7), and 3) a group with thoracic curves (average Cobb angle 19 degrees, n = 10). Individual lateral spinal profiles are also analyzed.The findings show: (1) In the control group, there are different degrees of vertical backward tilt (declivity) from T7-L3, with a maximum tilt at T12 (mean 26°). The most vertical vertebrae are T6 and L4, with forward tilting (proclivity) at each of (T5 and L5.2) In the lumbar curve group, the segmental sagittal tilt is not significantly different from that in the control group. The mean declivity at T12 is 25°. (3) In the thoracic curve group, the segmental sagittal tilt is significantly less than that in the control group at each of T10-L1. The mean declivity at T12 is 17°. A more vertical T12 is associated with a larger Cobb angle. (4) The individual sagittal spinal profiles of the thoracic curve group (but no other group) show lordosis in the region of the lateral spinal curve (scoliosis) and a kyphotic angulation at an average of three vertebrae below the apical vertebrae of the scoliosis curve. (5) It is suggested that as a thoracic lordoscoliosis develops, the appearance of a kyphotic angulation in the thoracolumbar and upper lumbar spine is determined by the compressibility of each disc in relation to the length of the articular processes at the corresponding level. Where the combination of disc weakness to facet length is most adverse, forward flexion occurs, as in a spinal fracture, to produce the kyphotic angulation. © 1992 Wiley-Liss, Inc.

    Notes: The patterns of the tendons of extensor digiti minimi, extensor digitorum, and extensor indicis have been studied on the dorsum of the hands of 25 embalmed cadavers.Each of the tendons of extensor digitorum (ED) is usually single, but those of the middle and ring fingers are occasionally multiple. Intertendinous connections are of variable pattern. The ED tendon of the little finger lies closely to that of the ring finger, with which it may be fused, or it may be absent. It is suggested that it may be undergoing evolutionary reduction.Extensor digiti minimi (EDM) usually has two tendons, but may have three or four. In addition to EDM, the little finger also receives either a separate tendon, or a tendinous slip, from ED.Extensor indicis (EI) varies in the number of its tendons, in its position relative to the ED tendon to index finger, and in its connection to extensor pollicis longus tendon.Examples of supernumerary muscles (extensor digitorum brevis manus, extensor indicis brevis, and extensor pollicis et indicis communis) were also found. © 1992 Wiley-Liss, Inc.

    Notes: An anatomic study focused on the proximal cervical spinal nerves has not been reported. Therefore, we dissected the cervical spinal nerves of three cadavers and examined stained sections in parasagittal and axial planes through the proximal cervical spinal nerves and root sheaths. These studies documented distinct segments of the spinal nerve which had not been completely described in the previous anatomic studies. The sheath originates from the dural sac as a common sleeve, divides into two sleeves, one containing the ventral root and one the dorsal root, and then distal to the dorsal root ganglion fuses again into one sleeve. A space, the interradicular cleft, separates the dorsal and ventral portions of the sleeve. The proximal segment of the spinal nerve proper distal to the dorsal root ganglion is composed of multiple small fascicles surrounded by a dense epineurium. The presence of an interradicular cleft in the cervical nerve root sheath and of fascicles in the cervical spinal nerve has significance for imaging of the cervical spinal nerves and for the pathogenesis of symptoms in cases of partial compression. © 1992 Wiley-Liss, Inc.

    Notes: The purpose of this study was to review venous collateralization resulting from inferior vena cava obstruction. The elements responsible for the extent and distribution of venous collaterals in inferior vena cava obstruction and whether or not the obstructive lesion involves tributaries of the inferior vena cava. Common etiologies of inferior vena cava obstruction include extensions of iliofemoral vein thrombi, thrombosis from intraluminal tumors or following trauma, and external compression. The anatomy of the venous collateral systems may be divided into deep and superficial networks, each of which is composed of systems of primary or secondary clinical significance, as defined by the degree of restoration of adequate venous return and the extent of visceral venous decompression.The most common obstructions of the inferior vena cava involve the lower third of the vessel. The azygos-hemiazygos and vertebral venous plexus systems play the most significant roles, while the superficial systems are less prominently involved. In upper level inferior vena cava obstruction, reestablishment of venous circulation is less developed, which usually leads to a poorer clinical outcome. © 1992 Wiley-Liss, Inc.

    Notes: This study reports a retrospective analysis of the details of the anatomy of the renal arteries of 403 kidney donors and the arterial complications related to 513 subsequent kidney transplants in our transplant unit. Multiple renal arteries occurred bilaterally in 10.2% of donors and unilaterally in 20.8%, a total incidence of 31%. There was a higher incidence of vascular-related complications following transplantation of kidneys with multiple renal arteries. Attention is drawn to the need for careful technique in identification of multiple renal vessels, especially aberrant vessels, at the time of donor nephrectomy and also to the different techniques available for anastomosis of multiple renal arteries in kidney transplant recipients. © 1992 Wiley-Liss, Inc.

    Notes: A reassessment is presented of the functional anatomy of the spine and associated musculature. Recent detailed anatomical studies of the erector spinae muscle group have shown a complex organization which is not well explained by traditional models of the mechanical behavior of the spine. In an arch-like model for the spine this complexity is required to control the curvature and the compressive thrust along the spine to ensure mechanical stability. This model also assigns a biomechanical function to the erector spinae muscles that is consistent with their anatomical description, unlike previous lever models. Implications are that posture, the many-curved configuration of the spine adopted during maneuvers, is important when assessing spinal function and that the spine forms a dynamic structure in which all the muscles, ligaments, discs, and vertebrae must be considered synergistically. © 1992 Wiley-Liss, Inc.

    Notes: The anatomy of the anterior spinal artery in pigs was studied to determine whether or not the artery was continuous, as it is humans, or discontinuous. Six porcine carcasses were embalmed, and 24 hr later injected with pigmented latex through the thoracic aorta. The spinal column was removed, and the spinal cord was dissected with analysis of the anterior spinal artery. In all cases, the artery was continuous, and perfused by 18 ± 3 medullary arteries. The pig would thus be suitable as a model for the study of human diseases in which the anatomy of the anterior spinal artery is a factor. © 1992 Wiley-Liss, Inc.

    Notes: The precise morphology of the human sphincter of Oddi was investigated. Anatomic samples, including the terminal portion of the bile duct and main pancreatic ducts, the major duodenal papilla, and the periampullary duodenal wall, were removed from 22 cadavers of various ages of patients who had died without any history of biliary or pancreatic disease.The specimens were fixed and processed by routine techniques and examined by light microscopy; serial sections were stained with haematoxylin-eosin, Masson's trichrome, and Van Gieson methods.The muscular bundles of the human sphincter of Oddi were divided into the following tracts: The X-Y tract, the distance from the papillary opening to the first muscular thickening that showed a circular trend; the Y-Z tract, the thickest portion of the circular muscular bundles (the true sphincter of the hepatopancreatic ampulla( (HPA) (Oddi), reinforced by fibres arising from the duodenal wall and the Z-W tract, the terminal thinner portion of the circular bundles that pass from the duodenal wall to the bile duct. The X′, Y′, Z′, and W′ points were fixed by the same criteria along the wall of the main pancreatic duct.The single tracts were measured, and the data derived therefrom subjected to statistical analysis.

    Notes: In spite of its clinical importance in cleft palate, there are few detailed accounts of the blood supply of the soft palate and palatal muscles. A series of dissections was carried out to investigate the supply of the levator and tensor veli palatini muscles. Ten adult cadavers and 12 fetuses were used, all of which had been previously injected with a variety of media to outline the vascular tree. Conventional dissection was performed on the cadavers, and microsurgical instruments and a dissection microscope were utilized for fetal dissections.In 70% of dissections the m. levator veli palatini had a dual arterial supply, from the ascending palatine and ascending pharyngeal arteries. In the remainining cases the muscle was supplied by a single artery, either the ascending palatine or ascending pharyngeal. In 79% of dissections the tensor muscle had a dual supply from the accessory meningeal artery along with either the ascending palatine, ascending pharyngeal, or, in one case, the lesser palatine arteries.The results suggest that in careful surgical closure of the cleft soft palate the normal dual arterial supply would protect the tensor and levator muscles from vascular damage. In the minority of patients where only a single vessel supplies the levator, a radical intrapalatal dissection may jeopardize its vascularity.

    Notes: Vein valve transplantation is a recently described operation to treat the sequelae of lower extremity venous hypertension resulting from valvular dysfunction. The operation involves harvesting a segment of an arm vein contianing a competent valve and transplanting it into a lower extremity vein. To determine the optimal incisions for obtaining a suitable valve-containing segment of vein, 20 cadaver arms were dissected. Cephalic, basilic, and brachial veins were examined for size, distribution of valves, and presence of tandem valves (defined as valves separated by 2 cm or less). Because patency rates are low in vascular grafts 〈 4 mm in diameter, only veins of this size or greater were considered adequate for transplantation. Cephalic veinswere found to have a consistently smaller diameter than basilic or brachial veins (P 〈 0.0001). Incisions for exposure of the basilic and brachial veins were evaluated. When exposure was limited to the middle third of the arm, an adequate vein, either the basilic or one of the brachial veins, was found in all cases. When either the distal or proximal third of the arm was exposed, however, an adequate vein was found in only 70% of the cases (P 〈 0.02). Tandem valves, although potentially useful, were present in only 5%, 25%, and 20%, respectively, of distal, middle, and proximal arm segments. Based on the results of this study, a medial incision along the middle third of the arm, exposing a basilic or brachial vein, is recommended as the optimal approach for vein harvesting in vein valve transplantation.

    Notes: This study was conducted on X-ray films of the shaft of normal femur and second metacarpal bones of adult male and female Nigerians to compare the percentage cortical thickness relative to the subperiosteal diameter (cortical index) of these bones. Age was correlated with cortical index (CI) using Pearson's product of moment correlation coefficient. Confidence limits of CI were calculated for clinical use. Results obtained were analysed for each sex and compared at P〈0.05 level of significane. CI of the second metacarpal is significantly greater than of the femur in both the male and female. Age maintained significant inverse correlation with CI of the femur and nonsignificant correlation with the second metacarpal in the female. In the male, age had nonsignificant correlation with the femur or with the second metacarpal bone.It is concluded that (i) the amount of percentage relative cortical bone to subperiosteal diameter of the second metacarpal, a nonweight-bearing bone, is greater than that of the femur, a weight-bearing bone, (ii) age of the adult has significant effect on the amount of cortical bone of the femur in the female but not in the male, and (iii) age of the adult does not have significant effect on the amount of cortical bone of the second metacarpal in both sexes.

    Notes: In the academic year 1989-1990, the Department of Surgery at the University of Kentucky College of Medicine introduced a modified third-year surgery clerkship. Its goals were to eliminate the lecture method of instruction, to involve students actively in the educational process, and to emphasize decision-making and problem-solving skills in the third year of medical school. Hands-on training in patient care procedures would be offered in a fourth-year acting internship after students had learned both problem-solving and life-long learning skills while concurrently developing a medical knowledge base. Students were removed from the traditional surgical service teams, since it was decided that team duties might be detrimental to educational goals. Problem-based tutorial sessions were designed, and a block of unscheduled time was provided for students to use in obtaining, exploring, and reflecting on medical information. Informal comparisons of students in the previous clerkships with those in the modified clerkship have noted positive changes in the students' intellectual maturity, motivation, enthusiasm, and time-management skills. Faculty and students have responded positively to the programs which are part of this innovative clerkship.

    Notes: Following the application of a topical anesthetic agent to the tympanic membrane, a number of patients described a sensation of numbness in the mouth. On examination these patients were found to have a significant area of anesthesia on the inferior surface of the ipsilateral soft palate.As a result of auditory tube dysfunction many of the tympanic membranes were atrophic and retracted. In these cases the pars tensa was closely related to the medial wall of the tympanic cavity, and local anesthetic agents applied to the tympanic membrane could easily diffuse through to the promontory and related structures. If the bone over the tympanic plexus or that over the facial nerve were dehiscent these nerves would be exposed to the effect of the anesthetic. We feel that our observation suggests that in some individuals at least, the pathway of sensory innervation of the soft palate may involve either the facial nerve or the tympanic branch of the glossopharyngeal nerve.

    Notes: To examine the microvascular changes of villi after transplantation, the small intestine was transplanted in 36 Wistar/Shi rats (16 syngeneic, 20 allogeneic; donor Lewis). At days 1, 4, 8, and 10, four rats of the syngeneic group and five of the allogeneic group were killed. Corrosion casts of the vessels of the graft were prepared and examined by scanning electron microscopy. In the grafts of each group at 1 day, the villous microvascular casts were reduced in height, and the capillary plexus was much less dense, with signs of degeneration. By 4 days, however, the villi of syngeneic grafts had been restored to normal, both in their dimensions and in the richness of the capillary plexus. On the other hand, the microvascular casts of allografts at the same stage showed the villi still to be greatly reduced in height, although the density of the capillary net had been restored essentially to normal. At 8 and 10 days, the allogeneic grafts again showed short villi, but with greatly reduced capillary networks, Our results suggest that the regeneration of intestinal villi following ischemic damage at operation is rapid in syngeneic grafts and that in allogeneic grafts the processes of rejection had started by 4 days. © 1992 Wiley-Liss, Inc.

    Notes: John C. Boileau Grant was one of the most outstanding teachers of anatomy in the 20th century. His life's work is honored by the thirty editions to which, collectively, his three textbooks have run; by the J.C.B. Grant Museum in the Anatomy Department of the University of Toronto; and by the establishment by the Faculty of Medicine of that university of a John C.B. Grant Memorial Award and Lecture. The present article is an abridgement of the first lecture in this new eponymous series. An account and an appraisal are given of Grant's life and contributions. Reference is made to the deleterious impact on anatomy of overspecialization - not of research methods and goals, but of the minds of anatomists. The catalytic and revitalizing effect that an array of new imaging techniques is exerting on the interest in, and emphasis upon, gross human morphology is stressed and the author underlines R. Ger's claim that, thereby, all gross anatomy has become clinical anatomy. © 1992 Wiley-Liss, Inc.

    Notes: The enumerations of the modern 12 pairs of cranial nerves by a number of anatomists are reviewed in a continuous historical perspective using original texts and modern translations. The history of the numeration of the nerves is traced from Galen's classification into seven pairs (second century A.D.), through to Willis' nine pairs (1664) and Soemmerring's 12 pairs (1778). Authors describing individual structures (e.g., trochlear and olfactory nerves) for the first time are particularly noted.The contributions made by medieval anatomists such as Achillini, Berengario da Carpi, and Massa are emphasized, as also are the political and religious constraints placed upon the practical activities of anatomists up until the 16th century. It is shown that although the classification of the nerves into 12 pairs was accepted after 1778 by European authors, British texts continued to classify them as nine pairs until the end of the 19th century. © 1992 Wiley-Liss, Inc.

    Notes: The incidence and distribution of landmarks relating to the coronary sinus (c.s.) were evaluated in 240 human hearts. Special attention was directed to the myocardial coat of the c.s., the distribution and pattern of the cardiac veins, and their dimensions. In all specimens the myocardial coat of the c.s. also covered the adjacent 2 to 11 mm of the great cardiac vein. In 15% of cases this myocardial layer was thickened in a sphincter-like fashion, the edge of which was formed obliquely, in a crescent-like manner, or diffusely without a sharp border.In 3% of the hearts the myocardial cover of the c.s. extended over the terminal 10 mm of the middle cardiac vein as a strong fiber cord. In 9%, single isolated belts of fibers fixated the terminal parts of the adjoining cardiac veins to the posterior wall of the left atrium, and in 8% two or three myocardial cords, embedded in the fatty tissue of the left coronary sulcus, did the same.Because the myocardial cover extended leftwards to variable distances over the c.s., the left edge of the myocardial covering cannot serve to define the beginning of the coronary sinus. The location of the ostial valve of the great cardiac vein (valve of Vieussens) was variable as well, being found on the average 2.5 mm proximal to the opening of the oblique vein of the left atrium. Moreover, the valve of the great cardiac vein was found in only 87% of cases; therefore it is inappropriate for defining the beginning of the coronary sinus. Finally, the dot-like ostium of the oblique vein was most constant, and from the viewpoint of embryologic development, it is the logical landmark for determining the beginning of the coronary sinus, a necessary presupposition for cardiologic procedures like reperfusion of cardiac veins.

    Notes: The thickness of the fibrous connective tissue sheath around the corpus cavernosum was measured in a population of 41 men aged 30-90 years. Specimens were obtained from embalmed cadavers and compared with tissue from fresh cadavers. Sections of tissue were stained with hematoxylin and eosin, Masson's trichrome, or VerHoeff-Van Gieson's to show the connective tissue in the best manner. The thickness was correlated with the presence of inflammation and/ or fibrosis and showed an age-dependent thinning after the sixth decade of life. The results are related to the age occurrence of Peyronie's disease and the possibility of men with chronic thickening of the tunica albuginea having a predisposition toward developing the disease.

    Notes: The organization of chromatin and cytoplasmic microtubules changes abruptly at M-phase entry in both mitotic and meiotic cell cycles. To determine whether the early nuclear and cytoplasmic events associated with meiotic resumption are dependent on protein synthesis, cumulus-enclosed hamster oocytes were cultured in the presence of 100 μg/ml puromycin or cycloheximide for 5 hr. Both control (untreated) and treated oocytes were analyzed by fluorescence microscopy after staining with Hoechst 33258 and tubulin antibodies. Freshly isolated oocytes exhibit prominent nucleoli and diffuse chromatin within the germinal vesicle as well as an interphase network of cytoplasmic microtubules. After 4-4.5 hr in culture, most oocytes were in prometaphase I of meiosis as characterized by a prominent spindle with fully condensed chromosomes and numerous cytoplasmic asters. After 5-5.5 hr in culture, microtubule asters are no longer detected in most cells and the spindle is the only tubulin-positive structure. Incubation for 5 hr in the presence of inhibitors does not impair germinal vesicle breakdown, chromatin condensation, kinetochore microtubule assembly, or cytoplasmic aster formation in the majority of oocytes examined; however, under these conditions, a population of oocytes retains a germinal vesicle, exhibiting variable degrees of chromatin condensation and cytoplasmic aster formation. Meiotic spindle formation in inhibited in all oocytes. These effects are fully reversible upon culture of treated oocytes in drug-free medium for 5 hr. The data indicate that meiotic spindle assembly is dependent on ongoing protein synthesis in the cumulus-enclosed hamster oocyte; in contrast, chromatin condensation and aster formation are not as sensitive to protein synthesis inhibitors during meiotic resumption. © 1992 Wiley-Liss, Inc.

    Notes: Insulin-like growth factor-II (IGF-II) mRNA exists as multiple transcript size classes, such as 6.0, 5.3, 4.9, 3.2, and 2.2 kb mRNAs in various human tissues. Three different promoters, 2 different polyadenylation sites, and alternative splicing are involved in producing these multiple transcripts. Initiation of transcription at the 3 different promoters results in multiple mRNAs which contain identical coding regions but different 5′-untranslated regions (5′-UTRs). The first promoter is thought to direct expression of 5.3 kb mRNA in adult human liver. The second promoter region directs expression of 6.0, 3.2, and 2.2 kb mRNAs in human fetal tissues and several adult nonliver tissues. The third promoter specifies transcription of a 4.9 kb mRNA in various tissues. We isolated and sequenced a cDNA clone (pIGF-II-1-70) from a human plcental cDNA library, which contains the IGF-II coding region and the 5′-UTR associated with the third promoter. By using a 5′-UTR-specific probe from the clone, we found that this third 5′-UTR is contained in the IGF-II mRNA of 2.2 kb and is absent in the 3.2 kb IGF-II mRNA. We also found an 0.9 kb transcript expressed in placenta, which hybridized strongly to the third 5′-UTR specific probe but not to IGF-II coding region probes. This finding might indicate the existence of an mRNA encoding an IGF-II-associated peptide. © 1992 Wiley-Liss, Inc.

    Notes: Ten embryonic stem (ES) cell lines from mink blastocysts were isolated and characterized. All the lines had a normal diploid karyotype; of the ten lines studied, five had the XX and five had the XY constitution. Testing of the pluripotency of the ES-like cells demonstrated that (1) among four lines of genotype XX, an X was late-replicating in three; both Xs were active in about one-third of cells of line MES8, and analysis of glucose-6-phosphate dehydrogenase revealed no dosage compensation for the X-linked gene; (2) when cultured in suspension, the majority of lines were capable of forming “simple” embryoid bodies (EB), and two only showed the capacity for forming “cystic” multilayer EBs. However, formation of ectoderm or foci of yolk sac hematopoiesis, a feature of mouse ES ceils, was not observed in the “cystic” EB; (3) when cultured as a monolayer without feeder, the ES cells differentiated into either vimentin-positive fibroblast-like cells or cytokeratin-positive epithelial-like cells (less frequently); neural cells appeared in two lines; (4) when injected into athymic mice, only one of the four tested lines gave rise to tumors. These were fibrosarcomas composed of fibrobalst-like cells, with an admixture of smooth muscular elements and stray islets of epithelial tissue; (5) when the ES cells of line MES1 were injected into 102 blastocyst cavities and subsequently transplanted into foster mathers, we obtained 30 offspring. Analysis of the biochemical markers and coat color did not demonstrate the presence of chimaeras among offspring. Thus the cell lines derived from mink blastocysts are true ES cells. However, their pluripotential capacities are restricted. © 1992 Wiley-Liss, Inc.

    Notes: High molecular weight proteins in Rattus norvegicus that are immunoreactive with an antiprotamine 2 specific antibody but not with an anti-protamine 1 specific antibody are described. These proteins were detected by coupling high-performance liquid chromatography (HPLC) with an enzyme-linked immunosorbent assay (ELISA). Briefly, following HPLC separation of rat sperm nuclear proteins, the HPLC fractions were probed with the antibodies. We estimate that the antibody probes are 100-1000 times more sensitive than UV absorbance measurements. Immunoblot analysis following acid-urea electrophoretic separation of rat sperm nuclear proteins, and of the HPLC fractions, also detected putatie protamine 2 precursor proteins. The proteins reactive with the anti-protamine 2 antibody are most likely not mature protamine 2, since they were detected in a region of the chromatogram where we would not expect protamine 2 to migrate based on the chromatographic locations of human and mouse protamine 2. Likewise, the immunoblotting experiments demonstrated that the anti-protamine 2 antibody recognized proteins with slower electrophoretic mobilities than would be expected for a mature protamine 2. An anti-protamine 1 monoclonal antibody, Hup1 N, that binds rat protamine 1 is also described. Hup 1 N allowed for identification of the HPlc fractions that contained rat protamine 1. Finally, we demonstrated that Hup 1 N binds protamine 1 from a large number of species, suggesting a conserved epitope for Hup 1 N. © 1992 Wiley-Liss, Inc.

    Notes: The objectives of this study were to compare the fertilization rate of bovine in vitro matured oocytes by in vitro fertilization (IVF) and by microinjection of a single spermatozoon (MI) and to relate these rates with fertility reported for these bulls in artificial breeding. Bull A (Holstein) had a nonreturn rate of 75%. Semen from this bull is routinely used in our standard IVF procedure. Bull B (Ayrshire), used regularly in artificial breeding and related to bull D, had a nonreturn rate of 69.2%. Bull C (Brown Swiss), with a chromosomal translocation and trisomy, achieved a nonreturn rate of 42%. Bull D (Ayrshire) produced nonmotile spermatozoa (SPZ) and had an abnormality described as “tail stump defect.” No pregnancies sired by bull D have been reported. Oocytes were either fertilized in vitro by capacitated SPZ or by microinjection of a single immobilized SPZ into the ooplasma. SPZ were treated with 0.1 μM A23187 and used for IVF. For microinjection SPZ were cocultured for 5 h with bovine oviduct epithelial cells (BOEC) and then immobilized by freezing and thawing twice without cryoprotectant. A single batch of killed SPZ (stored at - 25°C) was used for all microinjections. All oocytes were cultured in Medium 199 for 22 h at 39°C and subsequently fixed, stained, and examined for evidence of fertilization (i.e., female and male pronucleus formation, SPZ decondensation). Fertilization rates following IVF with semen from bulls A, B, C, and D were 80%, 54%, 1%, and 2%, and following microinjection were 39%, 22%, 21%, and 34%, respectively. © 1992 Wiley-Liss, Inc.

    Notes: Preimplantation development encompasses the “free”-living period of mammalian embryo-genesis, which culminates in the formation of a fluid-filled structure, the blastocyst. Cavitation (blastocyst formation) is accompanied by the expression of a novel set of gene products that contribute directly to the attainment of cell polarity with the trophectoderm, which is both the first epithelium of development and the outer cell layer encircling the inner cell mass of the blastocyst. Several of these gene products have been identified and include the tight junction (ZO-1), Na/K-ATPase (α and β subunits), uvomorulin, gap junction (connexin43), and growth factors such as transforming growth factor-α (TGF-α) and epidermal growth factor (EGF). This review will examine the role(s) of each of these gene products during the onset and progression of blastocyst formation. The trophectodermal tight junctional permeability seal regulates the leakage of blastocoel fluid and also assists in the maintenance of a polarized Na/K-ATPase distribution to the basolateral plasma membrane domain of the mural trophectoderm. The polarized distribution of the Na/K-ATPase plays an integral role in the establishment of a trans-trophectoderm Na+ gradient, which drives the osmotic accumulation of water across the epithelium into the nascent blastocoelic cavity. The cell adhesion provided by uvomorulin is necessary for the establishment of the tight junctional seal, as well as the maintenance of the polarized Na/K-ATPase distribution. Growth factors such as TGF-α and EGF stimulate an increase in the rate of blastocoel expansion, which could, in part, be mediated by secondary messengers that result in an increase in Na/K-ATPase activity. Insight into the mechanism of cavitation has, therefore, directly linked blastocyst formation to trophectoderm cell differentiation, which arises through fundamental cell biological processes that are directly involved in the attainment of epithelial cell polarity. © 1992 Wiley-Liss, Inc.

    Notes: A homogeneous preparation of casein kinase 2 has been isolated from the phytopathogenic fungus Verticillium dahliae (the parasite of cotton). The enzyme consists of three subunits with molecular masses of 53, 41, and 38 kDa. Highly specific immune serum against casein kinase 2 has been obtained. By means of immunoblotting, enzyme-linked immunosorbent assay (ELISA), and immunochemical isolation on protein A-Sepharose, it is shown that the amount of casein kinase 2 increases under heat shock conditions (at least in part due to the synthesis de novo), while the synthesis of the majority of other proteins falls. The activity of casein kinase 2 is supressed during heat shock and so does not correlate with its content. The results give an evidence for the two-step model of casein kinase 2 regulation during heat shock.

    Notes: The present study was carried out to examine the effect of maturation media on male pronucleus formation of pig oocyte matured and fertilized in vitro. Follicular oocytes collected from prepubertal gilts at a local slaughter house were cultured (36 h) in three different media (mTCM-199, Waymouth MB 752/1, and mTLP-PVA), fertilized in vitro, and assessed for nuclear maturation and male pronucleus formation. The addition of 10% (v/v) pig follicular fluid (pFF) to maturation media significantly increased the rate of nuclear maturation of pig oocytes (P 〈 0.01), whereas the rate of nuclear maturation of pig oocytes among three different media did not differ. However, the rate of male pronucleus formation of pig oocytes was significantly higher in pig oocytes matured in Waymouth MB 752/1 with or without pFF than in oocytes matured in the other two media (P 〈 0.01). In experiment 2, the addition of cysteine (the same concentration as in Waymouth medium, 0.57 mM), to mTLP-PVA significantly increased the rate of male pronucleus formation of pig oocytes compared with the control (P 〈 0.01). The results indicate that the composition of maturation medium affects the ability of pig oocytes to form male pronuclei following sperm penetration; media containing a high concentration of cysteine (possibly as a substrate of glutathione), such as Waymouth MB 752/1, can remarkably promote this ability.

    Notes: The sensitive technique of mRNA phenotyping with the reverse transcription-polymerase chain reaction was employed to determine the patterns of gene expression for several growth factor ligand and receptor genes during bovine preimplantation development. Several thousand bovine embryos encompassing a developmental series from one-cell zygotes to hatched blastocysts were produced by the application of in vitro maturation, fertilization, and oviductal epithelial cell embryo coculture methods. Transcripts for transforming growth factor (TGF-α) and platelet-derived growth factor (PDGF-A) are detectable in all preimplantation bovine stages as observed in the mouse. Transcripts for TGF-β2 and insulin-like growth factor (IGF-II) and the receptors for PDGF-α, insulin, IGF-I, and IGF-II are also detectable throughout bovine preimplantation development, suggesting that these mRNAs are products of both the maternal and the embryonic genomes in the cow, whereas in the mouse they are present only following the activation of the embryonic genome at the two-cell stage. In contrast to the mouse embryo, IGF-I mRNA was detected within preimplantation bovine embryos. Basic fibroblast growth factor (bFGF) is a maternal message in the bovine embryo, since it is only detectable up until the eight-cell embryo stage. Bovine trophoblast protein (bTP) mRNA was detectable within day 8 bovine blastocysts. As was observed in the mouse, the transcripts for insulin, epidermal growth factor (EGF), or nerve growth factor (NGF) were not detectable in any bovine embryo stage. Analyses of this type should aid the development of a completely defined culture medium for the more efficient production of preimplantation bovine embryos.

    Notes: The ability of the rabbit blastocyst to reduce nitroblue tetrazolium (NBT) to formazan in the presence of cyanide was assayed as an indicator of extramitochondrial oxidase activity capable of generating the superoxide radical. A cytochemical method initially developed for the detection and localization of hydrogen peroxide production at the ultrastructural level in phagocytosing leukocytes (Briggs et al.: J Cell Biol 67:566, 1975) was also applied to the blastocyst. The results demonstrate that the rabbit blastocyst acquires the ability to reduce NBT by a cyanide-insensitive process and to generate hydrogen peroxide between the fourth and fifth days postcoitum. The enzymatic activity responsible is apparently an NAD(P)Hdependent oxidase in the outer, microvillous plasma membrane of the trophoblast.

    Notes: Extracellular matrices (ECM) present around unfertilized and fertilized mammalian oocytes were studied ultrastructurally in samples prepared in the presence of ruthenium red to facilitate stabilization of extracellular materials. Unfertilized mouse, hamster, and human oocytes have an ECM comprising granules and filaments in their perivitelline spaces (PVS). This matrix is more abundant in the human than in hamsters and mice. The granule/filament matrix appears identical to the matrix seen between cumulus and corona radiata cells following ruthenium red processing and previously shown to comprise protein and hyaluronic acid. By including ruthenium red during fixation, it is possible to demonstrate the existence of cortical granule exudate in the PVS of fertilized oocytes from hamsters, mice, and humans. Much of the cortical granule exudate is trapped in the PVS and forms a new coat around the fertilized oocyte. This material is particulate when stained with ruthenium red and appears to be uniformly dispersed around the entire oocyte surface. We refer to this new coat as the cortical granule envelope. This envelope is observed in the PVS of all developmental stages up to and including blastocysts in all three species. Following hatching of mouse and hamster blastocysts, the cortical granule envelope is no longer present. Possible functions of this envelope are discussed.

    Notes: The quantitative changes in the mRNAs for ribosomal proteins L7a, L18a, and S15 were assayed in slot hybridization experiments using labeled cRNA probes with total RNA from late growth-phase oocytes, ovulated eggs, and early embryos through the blastocyst stage. All three mRNAs showed a similar developmental pattern of prevalence, but their copy numbers per oocyte or embryo fluctuated according to developmental stage. There are on an average about 17,000 copies of each mRNA in the late growth-phase oocyte; this number drops to one-fifth to one-tenth in the ovulated egg and two-cell embryo but increases rapidly during cleavage to about 25,000 in the eight-cell embryo and about 42,000 in the blastocyst. A comparison of the levels of these mRNAs with the reported rates of ribosomal protein synthesis (LaMarca and Wassarman, 1979) suggests that, in late growth-phase oocytes, ribosomal protein synthesis is regulated primarily at the translational level and is kept low by some factor limiting mRNA utilization. On the other hand, the high rate of ribosome biosynthesis during early embryogenesis from the two-cell stage onward appears to involve the coordinate activation and transcription of ribosomal RNA and ribosomal protein genes coupled with the immediate translational utilization of ribosomal protein mRNAs.

    Notes: Because recent studies have particularly implicated the insulin growth factor family in early development, the effects of insulin-like growth factor (IGF-1) on the development of mouse embryos in vitro were investigated in detail. When added to the medium for culture of two-cell embryos, IGF-1 stimulated the number of cells in the resultant blastocysts after 54 hr, entirely by increasing the number of cells in the inner cell mass (ICM) (16.0 ± 0.5 vs. 12.6 ± 0.5 cells/ICM). This stimulation was also achieved when ICMs were isolated from blastocysts prior to culture for 24 hr with IGF-1 (22.3 ± 1.0 vs. 17.5 ± 0.8 cells/ICM). There was no effect of IGF-1 on trophectoderm (TE) cell proliferation. In morphology studies, IGF-1 also increased the proportion of blastocysts (62% ± 3% vs. 49% ± 4%) while decreasing the number of embryos remaining as morulae (32% ± 3% vs. 38% ± 2%) or in the early cleavage stages (7% ± 3% vs. 13% ± 3%) after 54 hr culture from the two-cell stage. All these effects were achieved with EC50s of approximately 60 pM IGF-1, which is in the range for IGF-1 receptor mediation; however, cross reaction with insulin, IGF-2, or other unknown receptors is not excluded. Nonetheless, the results show that physiological concentrations of IGF-1 (17-170 pM, 0.1-1 ng/ml), which have been observed in the reproductive tract, affect the early embryo, suggesting a normal role for this factor in the regulation of growth of the developing conceptus before implantation.

    Notes: The Arg-Gly-Asp peptide (RGD), contained in several extracellular matrix proteins such as fibronectin, laminin, vitronectin, and collagen, is a tripeptide that plays a role as a recognition sequence in many cell-to-cell and cell-to-matrix adhesion mechanisms, through its interaction with several receptors of the integrin family. We previously described the ability of the oolemma of hamster oocytes to bind GRGDTP coupled to the surface of activated immunobeads and demonstrated that RGD-containing oligopeptides inhibit the adhesion of human and hamster spermatozoa to zona-free hamster oocytes and their subsequent penetration. In the present experiments, we show, utilizing immunobeads coated with an RGD-containing peptide (PepTiteTM 2000), that the oolemma of unfertilized human eggs is also able to recognize this adhesion sequence. The binding of PepTiteTM 2000-coated immunobeads to the oolemma was inhibited by the oligopeptide GRGDTP as well as by fibronectin and laminin. When immunobeads were prepared with a PepTiteTM concentration of 10 μg/ml, GRGDTP 150 μg/ml, laminin 80 μg/ml, and fibronectin 60 μg/ml inhibited bead rosetting on the egg surface. These data suggest that a specific binding moiety for RGD is present on the human egg surface. The binding of fibronectin to the oolemma was also demonstrated by the rosetting of immunobeads coupled with antifibronectin antibody to human oocytes after their exposure to 1 mg/ml free fibronectin. Such binding of fibronectin to the oolemma could be inhibited by coincubation with a monoclonal antibody directed against the cell adhesion fragment of fibronectin. In addition, oolemmal rosetting of immunobeads coupled with a monoclonal antibody directed against the α5 subunit, usually part of the fibronectin receptor VLA 5 (α5β1), provided additional evidence that a putative fibronectin receptor is present on the oolemma of human eggs.

    Notes: Epididymal sperm cells, incubated with plasmid DNA, showed a spontaneous tendency to interact with the exogenous nucleic acid. We have investigated the molecular basis of such interaction. Exogenous DNA is taken up by sperm cells over a 15- to 20-min period and is specifically localized on the nuclear area of the sperm head. DNA was reversibly bound to spermatozoa since it can be competed out by excess of cold competitor DNA or by other polyanions as heparin and dextran sulphate. By contrast, poly-L-lysine, a polycation, favours the uptake. DNA molecules of large size (7 kb) were preferentially taken up as compared to smaller ones (150-750 bp). Acidic proteins were also taken up and concentrated, as for DNA, at the nuclear level. These data strongly suggested that ionic interactions may occur between foreign molecules and a substrate located in the sperm head. On the basis of Southwestern analysis, a sperm head protein(s) of 30-35 KD is identified as potential substrate for exogenous DNA binding. Moreover, we have found that seminal plasma contains factor(s) which abolish sperm permeability, exerting a powerful inhibitor effect on DNA uptake. The presence of a specific binding protein for the DNA and of a factor inhibiting such interaction support the existence of a mechanism controlling, through specific factors, the sperm-DNA interaction.

    Notes: A new medium, SOM, produced by simplex optimization, has been used to study the joint effects of NaCl, glutamine, and glucose on the development of outbred CF1 mouse zygotes to the blastocyst stage. Contrary to previous reports, glucose has no significant inhibiting effect on development to the blastocyst stage in this medium. Even in the presence of 5 mM glucose, 70% of the embryos develop to at least four cells, and 60% reach the blastocyst stage. Raising the concentration of NaCl from 75 to 125 mM, in the absence of glutamine, progressively inhibits development. Moreover, the response to glutamine depends on the concentration of NaCl in the medium. When the NaCl concentration is low, glutamine inhibits development. In contrast, when the NaCl concentration is high, glutamine protects against the inhibitory effect of the salt. We propose that glutamine protects against high concentrations of NaCl in the medium by acting as an organic osmolyte.

    Notes: A simple and successful method of microinjection of a single spermatozoon under the zona pellucida of a mouse oocyte has been developed. A characteristic of this method is that the tip of the sperm injection needle pierces the zona pellucida without touching the ooplasmic membrane. All the ova (277) used for this series of experiments had normal morphology after the injection procedure. Spermatozoa preincubated in culture medium for capacitation and those treated with ionophore A23187 for induction of acrosome reaction were used. In combination with some of these injections, a manipulation assisting the adhesion of the sperm head onto the ooplasmic membrane was employed. The fertilization rate (67.3%) of the ova injected with the ionophore-treated sperm using the sperm-adhesion treatment was significantly higher (P〈0.005) than that obtained by the injection of the preincubated sperm without applying the adhesion treatment (23.6%). All three of the recipients that received the 24 fertilized ova became pregnant and gave birth to 11 off-spring (45.8%). The inseminations performed with the sperm-adhesion treatment using the immotile sperm from the preincubated population and/or those from the ionophore-treated population did not result in fertilization in any case. These results suggest that the fertilization rate of subzonal insemination with motile ionophore-treated sperm can be improved by applying the sperm-adhesion treatment and that sperm motility might be involved in the establishment of fertilization, even after the adhesion of the sperm head with the mouse ovum membrane.

    Notes: The classical concept of sex determination in mammals is that a Y chromosomal gene controls the development of the indifferent gonad into a testis. Subsequent divergence of sexual phenotypes is secondary to this gonadal determination. The most likely candidate gene is SRY (sex-determining region Y) in humans, and Sry in mouse. However, several lines of evidence indicate that sexual dimorphism occurs even before the indifferent gonad appears. Here we present evidence that bovine male embryos generally develop to more advanced stages than do females during the first 8 days after insemination in vitro. Corresponding relationships between both cell numbers and mitotic indices and sex were also seen. Although it is not clear whether this phenomenon involves factors originating before or after fertilization, these findings suggest that sex-related gene expression affects the development of embryos soon after activation of the embryonic genome and well before gonadal differentiation.

    Notes: The use of fluorescein-conjugated Pisum sativum agglutinin (FITC-PSA) was evaluated for its ability to distinguish acrosome-intact from acrosome-damaged stallion spermatozoa. Incubation of fresh (acrosomeintact) and frozen-thawed (acrosome-damaged) spermatozoa with FITC-PSA resulted in acrosome-intact spermatozoa that exhibited no fluorescence, while acrosome-damaged spermatozoa exhibited fluorescent staining over the rostral portion of the head and equatorial segment. Experiments using mixtures of various ratios of acrosome-intact and acrosome-damaged spermatozoa determined the precision (intrasample coefficient of variation), and linearity (increased percentage of spermatozoa with PSA binding, with increased percentage of frozenthawed spermatozoa in a sample) of FITC-PSA binding. The binding of FITC-PSA increased in samples as the portion of frozen-thawed (acrosome-damaged) to fresh (acrosomeintact) spermatozoa increased. A positive correlation existed (r = 0.98, P 〈 0.05) between the percentage of FITC-PSA bound sperm and the proportion of damaged spermatozoa added to a sample. Location of PSA lectin binding on acrosome-damaged spermatozoa, determined by electron microscopy using gold-conjugated PSA, was to components of the outer acrosomal membrane and acrosomal matrix. These results demonstrate that FITC-PSA binding may be useful in determining acrosomal integrity of fresh and frozen-thawed stallion spermatozoa. © 1992 Wiley-Liss, Inc.

    Notes: The acrosome reaction induced by the zona pellucida in mouse sperm has been shown to proceed in two stages experimentally distinguishable by the fluorescent probe chlortetracycline. Entry into the first stage of sperm bound to isolated, structurally intact zonae pellucidae is blocked by the compound 3-quinuclidinyl benzilate. In this study, we show, utilizing the fluorescent Ca2+ indicator fluo-3, that the first stage of the zona-induced acrosome reaction is characterized by an increase in intracellular Ca2+, followed by a decrease as the acrosome reaction proceeds. This calcium transient is completely suppressed by 3-quinuclidinyl benzilate. We conclude that the Ca2+ transient is induced by the zona pellucida and is required for the zona-induced acrosome reaction. Blockage of this sperm intracellular Ca2+ transient provides a mechanism for the inhibitory action of 3-quinuclidinyl benzilate on the zona-induced acrosome reaction in mouse sperm. © 1992 Wiley-Liss, Inc.

    Notes: This study examines the evolution of mitochondria in the follicular cells during the development of the ovarian follicle in the teleostean fish Epiplatys spilargyreus. The mitochondria are few in number until the end of previtellogenesis; their matrix is dense, and their cristae are well developed. They proliferate during vitellogenesis and then are modified by deterioration of their matrix. Multilamellar structures are organized in the vacuolized mitochondria. During postvitellogenesis, these modifications become more advanced. The mitochondria degenerate, leaving vacuoles that contain heterogeneous structures, which will be released into the intercellular spaces. At the end of these mitochondrial transformations, the follicular cells degenerate. They release the elements which will participate in forming the secondary envelope. © 1992 Wiley-Liss, Inc.
