    Notizen: Abstract An algebraic multigrid method is developed which can be used as a preconditioner for the solution of linear systems of equations with postitive definite matrices. The method is directed to equations which arise from the discretization of elliptic equations of second order, but only the matrix is the source for the information used by the algorithm. One has only to know whether the matrix stems from a 2-dimensional or 3-dimensional problem and whether the elliptic equations are scalar equations or belong to a system.

    Beschreibung / Inhaltsverzeichnis: Summary Preventive strategies aim to reduce gastric acidity (H2ms (sucralfate, antacids, pirenzepine) and to normalize gastric mucosal microcirculation (sucralfate, pirenzepine). Thus, the most important prophylactic measure is an optimized emergency and ICU regime aiming to improve oxygenation and microcirculation. All specific drugs used for stress ulcer prophylaxis have been shown to be effective in prospective controlled studies. Furthermore, pirenzepine has been found to be superior to H2-antagonists, at least in neurosurgical patients. Insufficient or no data exist to support the use of prostaglandins or omeprazole for stress ulcer prophylaxis. The most important adverse effect of stress ulcer prophylaxis is nosocomial pneumonia due to gastric alkalinization. This may occur in long-term ventilated patients with a gastric pH〉4 and may account for up to 50% of all nosocomial pneumonias in certain groups of patients. Mortality is not influenced by antacids or H2-antagonists, while sucralfate has been shown to reduce mortality, most probably by inhibition of bacterial translocation.
    Notizen: Zusammenfassung Die Ursachen akuter oberer Gastrointestinalblutungen bei Intensivpatienten sind vielfältig: Ulkuskrankheit, Medikamenteneffekte, Sondenläsionen, akutes Nieren- oder Leberversagen und Streßläsionen. Die letzteren manifestieren sich typischerweise als multiple oberflächliche Mukosaerosionen, während Ulzerationen meist bei Patienten mit Schädelhirntrauma oder nach neurochirurgischen Eingriffen oder Verbrennungstrauma beobachtet werden. Nur bei Patienten mit intrakraniellen Läsionen kommt es regelmäßig zur Hyperazidität. Neuere Erkenntnisse haben deutlich gemacht, daß ein aktiv säuresezernierender Magen resistenter gegen Noxen ist als ein Magen mit nur basaler Säuresekretion. Aktive Säuresekretion erfordert ausreichende Sauerstoffzufuhr und ausreichenden ATP-Gehalt in der Magenmukosa. Hypotension und Schock führen zur Mukosaischämie und sind die bedeutsamsten Risikofaktoren der Streßblutung. Prophylaktische Maßnahmen zielen darauf ab, die Magensäure zu vermindern und insbesondere die protektiven Mechanismen zu stärken. Die wichtigste prophylaktische Maßnahme ist deshalb eine frühzeitige optimale Schocktherapie. Alle derzeit in Deutschland zugelassenen spezifischen medikamentösen Prophylaxeregimes haben sich in kontrollierten Studien als wirksam erwiesen. Für neurochirurgische Patienten konnte darüber hinaus eine überlegene Wirksamkeit von Pirenzepin gegenüber H2g;blutungsprophylaxe. Die bedeutsamste Nebenwirkung ist eine erhöhte Pneumonierate unter säuresupprimierender Streßblutungsprophylaxe. Diese Nebenwirkung beobachtet man bei langzeitbeatmeten Intensivpatienten mit einem Magensaft pH 〉4. Bei speziellen Risikogruppen werden bis zu 50% aller nosokomialen Pneumonien durch diese Nebenwirkung verursacht. Die bei längerfristig beatmeten Patienten beobachtete geringere Mortalität unter Sucralfat im Vergleich zu H2-Antagonisten/Antazida kann wahrscheinlich durch die verminderte Translokation von Bakterien aus dem Gastrointestinaltrakt erklärt werden.

    Notizen: Abstract Flavonoids are plant phenolic compounds involved in leguminous plant-microbe interactions. Genes implied in the central branch (chalcone synthase (CHS), chalcone isomerase (CHI)) or in the isoflavonoid branch of the flavonoid biosynthesis pathway have been characterized in Medicago sativa. No information is available to date, however, on genes whose products are involved in the synthesis of other types of flavonoids. In this paper we present the genomic organization as well as the nucleotide sequence of one flavanone-3-hydroxylase (F3H) encoding gene of M. sativa, containing two introns and exhibiting 82–89% similarity at the amino acid level to other F3H proteins. This is the first report on the gennomic organization of a f3h gene so far. We present also the sequence of a partial dihydroflavonol-4-reductase (DFR) M. sativa cDNA clone. Southern blot experiments indicated that f3h and dfr genes are each represented by a single gene within the tetraploid genome of M. sativa. By a combination of Northern blot and RT-PCR analysis, we showed that both f3h and dfr genes are expressed in flowers, nodules and roots, with a pattern distinct from chs expression. Finally, we show that dfr is expressed in M. sativa leaves whereas f3h is not. The role played by these two genes in organs other than flowers remains to be determined.

    Notizen: Summary A field trial was carried out with oil seed rape plants that had been genetically modified to contain genes coding for marker characters (β-glucuronidase, kanamycin resistance and asulam resistance). The aim of the experiment was to examine a method of studying gene dispersal in an agricultural environment. The central area of the field plot comprised 150 genetically modified plants and 450 plants not expressing the marker genes. These were surrounded by additional non-expressing plants. The plants were allowed to freely cross pollinate and a sample of the resultant progeny from non-expressing plants were screened for expression of the marker characters and for the presence of the introduced genes (by PCR). Limited gene dispersal was detected and the frequency of modified seedlings amongst the progeny of a plant appeared to have been strongly influenced by the genotype of the immediately adjacent plants.

    Notizen: Abstract Logistic problems of large-scale reforestation necessitate freezer-storage of conifer seedlings. Frozen stock is typically thawed slowly at low temperatures for up to several weeks before shipping to the plantation site, but the necessity of this practice is questionable. Experiments were conducted to study effects of different thawing regimes on photosynthetic recovery, frost hardiness, water relations and growth initiation in “interior spruce” (white spruce (Picea glauca (Moench) Voss) and Engelmann spruce (Picea engelmannii Parry) hybrid complex). One year-old container-grown seedlings were planted after 9 days post-storage thawing at 5–15 °C or still frozen, directly from the freezer. During a 29 day observation period after planting, both groups showed changes in xylem water potential (Ψw), carbon fixation (A), stomatal conductance (g s ), chlorophyll a fluorescence and xanthophyll cycle pigments. Treatment differences in fluorescence and pigments peaked within one hour after planting. All differences in Ψw, A, g s , ratio of internal to external CO2 concentration (Ci/Ca), fluorescence, pigments and root number disappeared after 5 to 8 days. Terminal bud burst occurred 2.6 days earlier in the pre-thawed seedlings. When seedlings were rapidly thawed in the dark at 21 °C they achieved maximum Ψw (−0.2 MPa) in 3–4 hour. When evaluated 45 min after planting, A, g s , Ci/Ca and fluorescence values of rapidly thawed seedlings were intermediate between those for seedlings planted frozen or after 9 days slow thawing, showing that the recovery process was well underway a few hours after removal from the freezer. These results suggested that a suitable on-site operational protocol for rapid thawing might be to lay frozen bundles on the ground at ambient temperature overnight. In field trials of this method, rapidly thawed seedlings broke bud 3.3 days later than slowly thawed stock and also had greater frost hardiness at time of planting. Height, shoot and root mass did not differ after 3 months growth.

    Notizen: Involvement of tyrosine phosphorylation in osteoclastic bone resorption was examined using osteoclast-like multinucleated cells prepared from co-cultures of mouse osteoblastic cells and bone marrow cells in the presence of 1α,25-dihydroxyvitamin D3. When osteoclast-like cells were plated on culture dishes in the presence of 10% fetal bovine serum, they were sharply stained in their peripheral region by anti-phosphotyrosine antibody. Western blot analysis revealed that 115-to 130-kD proteins were tyrosine-phosphorylated in osteoclast-like cells. Using immunoprecipitation and immunoblotting, one of the proteins with 115-130 kD was identified as focal adhesion kinase (p125FAK), a tyrosine kinase, which is localized in focal adhesions. Immunostaining with anti-p 125FAK antibody revealed that p125FAK was mainly localized at the periphery of osteoclast-like cells. Herbimycin A, a tyrosine kinase inhibitor, not only suppressed tyrosine phosphorylation of p125FAK but also changed the intracellular localization of p125FAK and disrupted a ringed structure of F-actin-containing podosomes in osteoclast-like cells. Antisense oligodeoxynucleotides to p125FAK inhibited dentine resorption by osteoclast-like cells, whereas sense oligodeoxynucleotides did not. These results suggest that p125FAK is involved in osteoclastic bone resorption and that tyrosine phosphorylation of p125FAK is critical for regulating osteoclast function.

    Notizen: The protective effect of melatonin on lipopolysaccharide (LPS)-induced oxidative damage in phenobarbital-treated rats was measured using the following parameters: changes in total glutathione (tGSH) concentration, levels of oxidized glutathione (GSSG), the activity of the antioxidant enzyme glutathione peroxidase (GSH-PX) in both brain and liver, and the content of cytochrome P450 reductase in liver. Melatonin was injected intraperitoneally (ip, 4mg/kg BW) every hour for 4 h after LPS administration; control animals received 4 injections of diluent. LPS was given (ip, 4 mg/kg) 6 h before the animals were killed. Prior to the LPS injection, animals were pretreated with phenobarbital (PB), a stimulator of cytochrome P450 reductase, at a dose 80 mg/kg BW ip for 3 consecutive days. One group of animals received LPS together with Nw-nitro-L-arginine methyl ester (L-NAME), a blocker of nitric oxide synthase (NOS) (for 4 days given in drinking water at a concentration of 50 mM). In liver, PB, in all groups, increased significantly both the concentration of tGSH and the activity of GSH-PX. When the animals were injected with LPS the levels of tGSH and GSSG were significantly higher compared with other groups while melatonin and L-NAME significantly enhanced tGSH when compared with that in the LPS-treated rats. Melatonin alone reduced GSSG levels and enhanced the activity of GSH-PX in LPS-treated animals. Additionally, LPS diminished the content of cytochrome P450 reductase with this effect being largely prevented by L-NAME administration. Melatonin did not change the content of P450 either in PB- or LPS-treated animals. In brain, melatonin and L-NAME increased both tGSH levels and the activity of GSH-PX in LPS-treated animals. The results suggest that melatonin protects against LPS-induced oxidative toxicity in PB-treated animals in both liver and brain, and the findings are consistent with previously published observations related to the antioxidant activity of the pineal hormone.

    Notizen: Three distinct genes encode an identical calmodulin protein in mammalian cells. In addition, multiple mRNA transcripts, with approximate sizes of 1.6 kb and 4.4 kb, are visualized on Northern blots hybridized to calmodulin-I cDNA probes. To elucidate the mechanism generating multiple calmodulin mRNAs, the complete sequence of the 4194 base human calmodulin-I mRNA was determined from cDNA clones and 3′ rapid amplification of complementary ends (3′ RACE). The 5′ untranslated region of calmodulin-I mRNA contains a GC-rich domain containing multiple repeats of GGC interrupted by a GCA sequence, as well as a tandem repeat sequence of eight GCA triplets. The 3′ untranslated region of calmodulin-I mRNA contains two canonical and one aberrant (ATTAAA) polyadenylation signal, consistent with the sizes of 1.6 kb and 4.4 kb mRNAs visualized on Northern blots, and a potential minor 4.2 kb mRNA detected by 3′ RACE. Hybridization experiments using specific probes upstream and downstream of the polyadenylation signals demonstrated that alternate use of polyadenylation signals is the molecular mechanism for multiple calmodulin-I mRNA transcripts in human cells. Thirteen adenine rich elements with the motif AUUUA were detected in the 3′ untranslated region. Three such motifs are embedded in regions that are conserved with the rat 3′ untranslated region of calmodulin-I mRNA. One of these is surrounded by an adenine-uridine rich region that can form an 11-base pair stem structure. We propose that sequences in the 3′ untranslated region of calmodulin-I mRNA may play a role in the regulation of calmodulin expression.

    Notizen: Garlic is known as a potent spice and a medicine with broad therapeutic properties ranging from antibacterial to anticancer, antidiabetic, and anticoagulant. Two major proteins of 40 KD and 14 KD constituting approximately 96% of total garlic proteins have been recently purified at our Institute. This immunocytochemical and ultrastructural study revealed that the 40 KD protein was localized in the parenchyma sheath cells (PSC) of garlic bulbs, whereas the 14 KD protein was present in the cortical cells (CC). Immunogold electron microscopy study indicated that the 40 KD protein was specifically localized in the globular granules of the cytoplasmic area of PSC. Each globular granule was amorphous and homogenous with membrane limiting its outermost layer. The yellowish color of PSC in freshly cut slices of garlic bulb suggested that PSC may have sulfur-containing compounds such as allicin, the primary contributor of the pungency and medicinal properties of garlic. Ellman's reagent test quantitatively revealed that there were 17.8 n moles sulfhydryl (SH)/ml of 40 KD garlic protein. Microtubule tubulin in mitotic figures from PHA-stimulated human short-term whole blood cultures reacted strongly with antitubulin antibody but reacted negatively with anti-40 KD garlic protein antibodies and therefore was not related to the 40 KD garlic protein immunocytochemically.
