    Notes: During early postnatal development of the kitten visual cortex the ectoenzyme 5′-nucleotidase undergoes a characteristic redistribution. Until about postnatal week 6 it is essentially confined to synaptic contacts in input layer IV and its expression is related to the use-dependent segregation of thalamic afferents into ocular dominance columns. Subsequently, 5′-nucleotidase becomes distributed uniformly throughout all layers and is then associated selectively with glial cells. Here we describe an age-dependent alteration in the expression of a carbohydrate epitope of 5′-nucleotidase which correlates with the developmental change of the enzyme's localization. We have isolated 5′-nucleotidase from the occipital cortex of kittens of varying age and from adult cats and investigated by immunoblotting the association of the HNK-1 carbohydrate epitope with the protein. 5′-Nucleotidase carries the HNK-1 epitope in kittens of 3–9 weeks but the epitope is absent from 12-week-old kittens or adult cats. Thus, the appearance of the HNK-1 epitope correlates with the transient localization of the enzyme at synapses. The HNK-1 carrying 5′-nucleotidase may be involved in synaptogenesis and use-dependent modifications of synaptic connections.

    Notes: Summary As no long-term results of a blind opening up of the pulmonary valve either by balloon valvoplasty or closed Brock valvotomy have been published, we examined the outcome of 12 patients with valvar pulmonary stenosis 17±5 (11–22) years after surgery. The Brock valvotomy had been carried out at a mean age of 3±2.8 (0.2–8.5) years. The mean pressure gradient across the right ventricular outflow tract had been 116±45 (75–97) mmHg at the catheter study or 106±43 (40–160) mmHg as measured intraoperatively. The right ventricular pressure after the Brock procedure was measured in the operating room in five patients as 46±15 (30–60) mmHg. Seven patients had been recatheterized at a mean age of 9.5±2 (7–12.7) years; at that time the gradient across the pulmonary valve had been 20±14 (10–37) mmHg. At a mean age of 21.7±3 (15–26) years these and five further patients were reexamined by echo Doppler. This time the pressure gradient across the pulmonary valve was 13±6 (7–20) mmHg. Moderate pulmonary incompetence was present in four and mild incompetence in eight patients; two had mild tricuspid insufficiency. All except one patient, who had suffered a cerebrovascular accident before surgery, were in NYHA functional class 1 and pursuing a profession. From these data we conclude that the blind opening-up of the pulmonary valve achieves excellent long-term palliation.

    Description / Table of Contents: Summary Microcallus formations are demonstrable in nearly all cancellous bones by means of suitable preparation techniques. Histologically, these structures are immature fibrous bone formed in local overloaded parts of the trabeculae. Using a preparation technique that allows combined two- and threedimensional analysis, 26 normal human spines and 11 osteoporotic spines were investigated for microcallus. Microcallus formations occur frequently in people over 45 years of age. They are mainly localized in the lower thoracic and lumbar spine and occur significantly more frequent in females than in males. The number of microcallus formations depends more on the microarchitecture of the cancellous bone than on individual trabecular parameters. In about 33 % of cases microfractures are demonstrable in the centre of the microcallus formation. In noninvasive studies the bone mass could be misinterpreted due to microcallus. Although it indicates instability of the bone structure, microcallus formation is not a purely negative mechanism. It stabilizes and regenerates the bone tissue. Furthermore, complete new trabeculae can be formed due to bridges of microcallus between residual trabeculae. Osteoporosis is not the result of an inability to form microcallus.
    Notes: Zusammenfassung Mikrokallusformationen lassen sich in nahezu allen Skelettabschnitten der Spongiosa nachweisen. Mikrokallus besteht aus Geflechtknochen, der sich an lokal überbelasteten Stellen in der Spongiosa bildet. Mit Hilfe einer speziellen Präparationstechnik wurden 26 skelettgesunde und 11 Wirbelsäulen von Fällen mit Osteoporose untersucht. Mikrokallusformationen finden sich bevorzugt bei Frauen älter als 45 Jahre in den unteren Wirbelsäulenabschnitten. Dabei hat die Mikroarchitektur der Spongiosa (TBPf) einen stärkeren Einfluß auf die Anzahl der Mikrokalli, als individuelle Trabekelparameter (Tb.N, BV/TV und Tb.Th). Nur in 33 % der Formationen lassen sich Frakturspalten nachweisen. Mikrokallusformationen können nichtinvasive Knochenmassemessungen verfälschen. Auch wenn Mikrokallusformationen Indikatoren für eine Instabilität der Spongiosa sind, tragen sie zur Knochenregeneration bei, und die Entstehung neuer Trabekel ist durch sie möglich. Die Vorstellung, daß Osteoporose das Resultat einer verminderten Mikrokallusbildung ist, trifft nicht zu.

    Notes: Summary In five of eight human transitional carcinoma cell (TCC) lines a proliferative response has been reported during exposure to interleukin-3 (IL-3), granulocyte-macrophage colony stimulating factor (GM-CSF) and granulocyte colony stimulating factor (GM-CSF). To elucidate possible growth-modulating effects of these factors combined with clinically relevant antineoplastic agents, cells of the human TCC lines EJ28 and T24 were exposed to methotrexate (MTX), vinblastine (VBL), doxorubicin (DXR) and cisplating (CDDP) with and without single or continuous exposure to IL-3, GM-CSF and G-CSF at concentrations of 1–100 ng/ml. Compared with cells exposed only to chemotherapy, significant inhibitory effects occurred as a result of continuous exposure to IL-3 or GM-CSF at the highest activities with CDDP and MTX in the T24 and EJ28 lines; continuous G-CSF administration (100 ng/ml) in combination with MTX led to significant growth inhibition in the EJ28 line. In contrast, no significant growth modulation was found on combined administration of DXR or VBL with any one of the three colony stimulating factors tested.
