    Notes: In the present study, we assayed mannan-specific IgE and IgG antibodies in samples of serum isolated from blood collected from adult patients with bronchial asthma, using a liquid-phase method with a polysaccharide, mannan (Mn), purified from Candida albicans (C. alb), and investigated the relationships of allergenicity among a crude extract of C alb. purified Mn. and acid protease (AP). The correlations between the titers of anti-Mn A and anti-Mn B IgE and IgG were very strong, and the levels of inhibition of anti-Mn A IgE and IgG reactions by Mn A and Mn B were almost identical. Although no common allergenicity was observed between Mn A and AP because there was no correlation between the titers of anti-Mn A and anti-AP IgE, and no inhibition of the anti-Mn A IgE reaction by AP, both antigens were found to exist in crude C alb. The level of inhibition of anti-crude C. alb IgG reaction by Mn A or Mn B was about 60%. Approximately 70% inhibition of the anti-Mn A IgE reaction was observed for eight different fungal allergen extracts, but no inhibition was observed for 11 of the other fungal allergen extracts tested. The above results indicate that common antigenicity was observed between Mn A and Mn B in the human IgE and IgG antibody production system, and the cross-allergenicity observed among some fungi was considered to be the result of the common antigenicity of Mn isoforms.

    Notes: Background Very little is known about the mechanistns underlying adverse reactions to radiocontrast medium. On the basis of the clinical features of the adverse reactions, il has generally been considered that an IgE-dependent mechanism is not involved in these adverse reactions, and only a few studies have demonstrated the presence of IgE antibody to radiocontrast medium iti patient sera. Methods We assayed for IgE antibody to ioxaglie acid (Hexabrix®), a representative radiocontrast medium, in the sera of patients who had developed adverse reactions to ioxaglie acid. A conjugate was prepared by coupling succinyl ioxaglie acid to human serum albumin. Enzyme immunoassay and radioimmunoassay for the detection of IgE antibody to ioxaglie acid in patient sera were constructed by physical adsorption or by eovalent eoupling of the conjugate on solid supports. Results When the radioactivity or the absorbance exceeding the mean plus 3 SD for normal healthy subjects was regarded as positive, IgE antibody was detected in the sera of 47.1% of the patients who had a past history of adverse reactions to ioxaglie acid and 16.2% of those who had experienced adverse reactions to ioxaglie acid within 24 h before the blood collection, although the IgE antibody levels were low. On the other hand, IgE antibody to ioxaglie acid was not detected in the sera of patients with no history of adverse reaetions to ioxaglie acid. Inhibition experiments revealed the presenee of IgE antibody speeific to ioxaglie acid in the serum defined as positive for IgE antibody to ioxaglie acid. The presenee of IgE antibody to ioxaghe acid did not always eorrelate with the activation of mast eells due t o the occurrence of adverse reaelions to ioxaglie acid. Conclusions A small amount of IgE antibody to ioxaglie acid was detected n i the sera of some patients with a history of adverse reactions to ioxaglie acid, and these adverse reactions may be partly explained by the presence of IgE antibody in the serum of at least some patients.

    Notes: Multilayer supermirrors stacked with three sets of Pt/C combinations have been fabricated on a flat float-glass and conical replica foil mirror using a magnetron DC sputtering system, and applied to X-ray optical systems in the hard X-ray region. The design of the supermirror is optimized to obtain the highest integrated reflectivity in the energy band and at the grazing angle concerned. X-ray reflectivities of 30% in the 25–35 keV band at an incidence angle of 0.3° were obtained.

    Notes: Summary In diabetic nephropathy leukocytes, mainly composed of monocytes/macrophages, which accumulate in the glomeruli and the interstitium, play an important part in the progression of glomerulosclerosis. The infiltration of leukocytes into inflammatory tissues or atherosclerotic lesions is mediated by adhesion molecules, which are expressed on the vascular endothelial cells, although little is known about the mechanism of leukocyte infiltration into diabetic renal tissues. P- and E-selectin are leukocyte adhesion molecules, which are expressed on the vascular endothelial cells and promote the adhesion of leukocytes to the endothelium. We investigated the expression of P- and E-selectin in the kidney tissue of patients with diabetic nephropathy and compared it with that of patients with other glomerular diseases (minimal change nephrotic syndrome, membranous nephropathy, IgA nephropathy, mesangioproliferative glomerulonephritis, and lupus nephritis). Expression of P- and E-selectin were both significantly increased in the glomeruli and the interstitium of patients with diabetic nephropathy as compared with those with other glomerular diseases. P- and E-selectin were both expressed along the glomerular capillaries and the peritubular capillaries in the interstitium. Neither P- nor E-selectin were correlated with the number of infiltrated leukocytes in the glomeruli, however, interestingly the E-selectin expression on peritubular capillaries was correlated with the number of infiltrated CD14 positive cells in the interstitium. These results suggest that E-selectin may play a key role in leukocyte infiltration into the renal interstitium in patients with diabetic nephropathy. [Diabetologia (1998) 41: 185–192]
