Notes: Two different electron density reconstruction methods for 8-keV neutral lithium beam probing have been developed for the Compact Helical System (CHS). Density dependences on emission and ionization processes are included by using effective rate coefficients obtained from the collisional radiative model. Since the two methods differ in the way the local beam density in the plasma is determined, the methods have different applicable electron densities. The beam attenuation is calculated by iteration from the electron density profile in method I. In method II, the beam remainder at the observation point z is determined by integrating the Li i emission intensity from z toward the position of emission tail-off. At the emission tail-off, the fast lithium beam is completely attenuated. Selecting an appropriate method enables us to obtain edge electron density profile well inside the last closed flux surface for various ranges of plasma densities (1012–5×1013 cm−3). The electron density profiles reconstructed by these two different methods are in good agreement with each other and are consistent with results from ruby laser Thomson scattering.

Notes: A plasma source based on an inductive breakdown of a supersonic gas puff is described. The source was developed to provide an anode plasma for an annular, extraction geometry, magnetically insulated ion diode. In this source, plasmas with densities of 1013 cm−3 were generated and accelerated to velocities of 20–30 cm/μs; plasma fluxes of 10–40 A/cm2 were obtained. Operating the source under the diode insulating field effect, plasma fluxes above 100 A/cm2 were observed. When the plasma source was used in conjunction with a magnetically insulated diode gap, intense ion beams with proton fluxes of more than 100 A/cm2, energies of 100 keV, and beam pulses longer than 1 μs were extracted.

Notes: The localization of DNA replicating cells, epidermal growth factor (EGF) receptor-expressing cells and ras oncogene product p21 (p-21ras) positive cells were examined in various skin tumours to elucidate the role of EGF receptor and p21ras in the epidermis. Normal skin, keratoacanthoma (KA), solar keratosis (SK), Bowen's disease (BD), squamous cell carcinoma (SCC), basal cell carcinoma (BCC) and extramammary Paget's disease (PD) were studied. EGF receptors were seen in proliferating layers, where DNA replicating cells localize, but p21ras was found in the more differentiated layers. We conclude that EGF receptor expression is closely associated with cellular proliferation, but p21ras may play a role in the differentiation of cells in various skin tumours.

Notes: Summary It has been proposed that basic fibroblast growth factor (bFGF) is an autocrine growth factor of melanoma cells, in contrast with normal melanocytes where bFGF acts as a paracrine growth factor. As this notion is mostly based on the different requirements for bFGF in cultures of benign and malignant pigment cells in vitro, we performed an immunohistochemical study to examine bFGF expression in vivo using paraffin sections from naevus cell naevi (NCN) and malignant melanoma (MM). All the NCN (n=7) showed strong and homogeneous expression of bFGF protein, whereas the primary MMs (n=5) showed heterogeneous expression, with a population of negative cells. Metastatic MMs (n=5) also showed heterogeneous expression, and had a greater population of negative cells. These results suggest that bFGF has some, as yet unidentitied, role in the growth of benign NCN, and that overexpression of bFGF is neither a prerequisite for melanoma genesis nor for progression to metastatic MM.

Notes: Summary A solution is obtained for the second mixed boundary value problem with two boundaries loaded with external forces, and the other two boundaries simply supported. A rational mapping function formed as a sum of fractional expressions and complex stress functions is used for the analysis. A closed form solution can be obtained under uniform tensile and shearing loadings. These solutions are applicable to crack problems in linear elastic fracture mechanics. In order to illustrate the general solution, a double connected region is considered, in which interaction between a square hole with a crack extension and another crack is investigated. Stress distributions and stress intensity factors are obtained.

Notes: Summary The temperature-sensitive mutant ts-1 of influenza virus A/WSN/33 carries mutations in the gene encoding RNA polymerase PB2 subunit. Effect of temperature on various steps of viral RNA synthesis was examined using disrupted virions of ts-1 mutant. The initiation of RNA synthesis with dinucleotide ApG primer was not affected by elevated temperature, whereas that with primer RNA containing 5′-terminal cap-1 structure was temperature-sensitive. The result supports the previous notion deduced from the UV-crosslinking experiments, that PB2 is involved in the cap-1 dependent initiation of RNA synthesis. In addition, the ts-1 mutant showed a defect in RNA chain elongation. Nucleotide sequence analysis of RNA segment 1 of ts-1 mutant revealed that the amino acid number 417 is essential for the recognition of cap-1 structures and/or the interaction with catalytic unit of the RNA polymerase.

Notes: Summary After feeding various diets we studied the effects of dietary calcium, magnesium and phosphorus on the formation of struvite stones in rats with urinary tract infections, and also studied the effects of the administration of vitamin D3 and aluminium gel on stone formation. A low-magnesium diet decreased urinary magnesium and prevented stone formation, but a medium-calcium diet did not significantly decrease stone weight. A high-calcium diet decreased urinary phosphorus and inhibited stone formation. A high-calcium and high-phosphorus diet decreased urinary excretion of magnesium and inhibited stone formation. Although the administration of vitamin D3 did not inhibit stone formation, aluminium gel decreased the urinary level of phosphorus and prevented stone formation. A marked decrease in urinary magnesium and/or phosphorus may prevent struvite stone formation in rats with urinary tract infections.

Notes: Summary In order to predict the material temperature throughout the hot rolling process of aluminium, a numerical model is proposed in a theoretical as well as simple way. Using the model, the possibility of the improvement in productivity by reforming the finishing mill arrangement is examined under aspect of the control of material temperature. The results so obtained show the positive effect of increasing the number of finishing mills on the productivity.

Notes: Summary Immunohistochemical and immunochemical analyses were performed on a monoclonal antibody designated 1-2B7B which was derived from immunizing mice with human prostate epithelial tissue. The 1-2B7B antigen was expressed not only along the acinous basement membrane zone (BMZ) of the prostate and testis, but also along the BMZ of the epithelia of several other organs including the skin, oesophagus, urinary bladder, ureter, stomach, intestine and bile duct. The antigenic epitope was not expressed in these tissues of lower mammals. Immunoelectron microscopic studies on normal human skin revealed that the 1-2B7B antigen was localized mainly just beneath the hemidesmosomes of basal keratinocytes, but not beneath melanocytes. Indirect immunofluorescence and immunoelectron microscopic studies on 1 M NaCl-split skin confirmed that this antigen was not separated from the cytoplasmic membrane of basal cells after salt treatment. SDS polyacrylamide gel electrophoresis of immunochemically purified protein from the epidermis demonstrated the molecular weight of the antigen to be 120 kDa. 1-2B7B monoclonal antibody should be a useful probe for studying the pathomechanism of some blistering diseases, as well as the assembly and function of the epidermal-dermal junction.
