Summary Acid mucopolysaccharides (MPS) are first demonstrable in the mast cells (MZ) in rats on the 16th–17th day of pregnancy. One to two days before this, the mast cells give a diffuse, positive PAS reaction. In the first stages of development the acid MPS are diffusely distributed in the cytoplasm; later they localize in increasing amounts at the granules. Parallel to this, in studies in which the number of mast cells staining with toluidine blue in solutions of graduated p H are counted, in the early stage of development only few mast cells show a strong acid MPS reaction. The number of mast cells staining at higher p H rapidly increases before birth. Shortly after birth the increase is only slight. The mast cells show an increased content of acid MPS. The formation and maturation of the acid MPS parallels the deposition of histamine, as found byParrat andWest in mast-cell-rich tissues. These results represent the conditions for an intracellular salt-like heparin-histamine union. In adult animals all of the histochemically demonstrable enzymes in the mast cells appear with the formation of granules.

Zusammenfassung Am 16.–17. Schwangerschaftstag werden bei Ratten in den Mastzellen (MZ) erstmals saure Mucopolysaccharide (MPS) nachweisbar, 1–2 Tage vorher zeigen die MZ bereits eine diffuse positive PAS-Reaktion. Die sauren MPS sind in den ersten Entwicklungsstadien diffus im Cytoplasma lokalisiert, später findet sich zunehmend eine Bindung an die Granula. Parallel hierzu zeigt die Auszählung der mit Toluidinblau in abgestuften p H -Lösungen gefärbten MZ, daß im Frühstadium der Entwicklung nur wenige MZ stark saure MPS aufweisen. Die Zahl der bei höherem p H färbbaren MZ nimmt vor der Geburt rasch zu. Kurz nach der Geburt ist die Zunahme nur mehr gering. Die MZ zeigen also bereits einen hohen Gehalt an sauren MPS. Die Bildung und Reifung der sauren MPS geht der vonParrat undWest gefundenen Histamineinlagerung in MZ-reichen Geweben parallel. Dieser Befund stellt die Voraussetzung für eine intracelluläre salzartige Heparin-Histaminbindung dar. Alle bei erwachsenen Tieren in den MZ histochemisch nachweisbaren Fermente treten bei der MZ-Entwicklung bereits mit der Bildung der Granula in Erscheinung.
