    Notes: Abstract Background: Irinotecan (CPT-11) is metabolized by esterase to form a SN-38, which is further conjugated by UGT1A1. Genetic polymorphism has been shown in a promoter region of UGT1A1 and is related to its activity. We investigated whether there might be an inter-individual difference in pharmacokinetics of SN-38 and its glucuronide, depending on the genotypes of UGT1A1. Patients and methods: Nine male patients with lung cancer were treated with irinotecan (50 mg/m2) and carboplatin. Pharmacokinetic parameters were calculated with full sampling plasma data. Genotypes were determined by analyzing the sequence of TATA box of UGT1A1 of genomic DNA from the patients. Results: The genotyping analysis revealed one heterozygote (6/7) and one homozygote (7/7) for (TA)7TAA allele (UGT1A1*28). The remaining seven patients were homozygote for (TA)6TAA allele (6/6, wild type). The metabolic ratios (SN-38/SN-38 glucuronide) in the patient with 7/7 genotype were uncharacteristically higher than those in the patients with other genotypes (6/6 and 6/7). Biliary index was 6980 versus 2180 ± 1110 (range 840–3730) in patients with 7/7 versus 6/6 genotypes, respectively. Conclusion: These results support the idea that the patient with 7/7 genotype has an impaired capacity for glucuronidation of SN-38.

    Notes: Abstract The interaction of an anticancer drug, 5-fluorouracil with sodium carboxymethylcellulose in aqueous solution was studied with a spectral method and viscosity measurement. From the binding data, the standard molar change in enthalpy, entropy and the number of binding sites on polymer were calculated. The standard molar change of enthalpy of 5-fluorouracil is about — 7 Kcal/mol with sodium carboxymethylcellulose. The enthalpy change is a considerably greater.

    Notes: Abstract The measurement of the diffusion coefficients of anticancer drugs and compounds having a similar structure in aqueous solution at 30 °C are carried out and the effect of some groups affecting the diffusion coefficients is investigated. The measurements of viscosity are also carried out in order to obtain the viscosity coefficient. The radius of molecule from the diffusion coefficients is in agreement with the value from the viscosity measurements. Under the concentration range studied, it seem that these compounds have a same state, for example, monomer or dimer. However, from Van der Waals radius of the results obtained, these solutes are monomer state. In spite of a large size, the diffusion coefficient of Uridine-5′-monophosphoric acid indicates a high value because of the potential gradient occurs in diffusion. From the result, 5-fluoro-2′-deoxy-5′-monophosphoric acid diffuses quickly in human body in spite of the large molecular weight.

    Notes: Abstract The present study was undertaken to explore the acute effect of hyperosmolality on the response of cultured rat inner medullary collecting duct (IMCD) cells to atrial natriuretic peptide (ANP). In contrast to the stimulatory effect of chronic incubation (12 h) in hypertonic medium, it was found that short-term incubation (〈 2 h) reversibly suppressed the ANP-dependent cyclic guanosine monophosphate (cGMP) production. Urea, NaCl and mannitol were equi-potent as the osmolyte in suppressing the ANP-dependent cGMP production. Receptor binding assay revealed that hyperosmolality induced a rapid and marked reduction of the maximum binding (B max) of ANP without a significant change of the dissociation constant (K d). Pretreatment with protein kinase C inhibitors (calphostin-C, staurosporin) or with cytoskeleton modulators (cytochalasin-B, colchicine) did not affect the inhibitory effect of hyperosmolality. In conclusion, acute hypertonicity inhibited the ANP-induced cGMP production in contrast to chronic hypertonicity, and reduction of the number of ANP binding sites was considered to be a mechanism responsible for the inhibitory effect of hypertonicity.

    Notes: Abstract We used the hydrostatic pressure P 〈1 GPa as a tool to change the c-axis critical current (interlayer coupling) of Bi-2201 and Bi-2212 single crystals. The c-axis critical current drastically increases with pressure (up to 270 % GPa −1 ). The superconducting critical temperature, on the contrary, only slightly increases with a rate of ∼ 2-6 % GPa −1 . This implies that the CuO-interlayer coupling has little effect on T c in Bi-compounds.
