    Notes: Summary This study was undertaken to evaluate the efficiency of flow cytometry in the detection of the serological H-Y antigen, and to survey expression of HY in the normal human population. Peripheral blood leukocytes (granulocytes) were reacted with monoclonal H-Y antibody, gw-16, and with FITC-conjugated goat anti-mouse IgG, and then were scored for fluorescence in the flow cytometer. Assigned H-Y phenotype was correlated accurately with sex phenotype in 33 out of 38 women and 62 out of 65 men. Data were evaluated by exploratory data analysis (EDA).

    Notes: Summary The expression of theHer2/neu gene product p 185 was retrospectively analyzed in 58 patients with gastric carcinoma. The results were correlated to various clinicopathological and prognostic factors. Positive membrane staining for p185 could be detected in 38% of the patients (22/58). Membrane staining was significantly greater in well and moderately differentiated tumors of the intestinal type when compared with poorly differentiated lesions and carcinomas of the diffuse type (P〈0.01). Positive membrane staining did not correlate with site and tumor stage, but T1 lesions had less membrane staining than more advanced primary tumors. Overall survival showed no difference between p185-positive and negative cases. Multivariate analysis defined a subgroup of curatively resected patients with stage III and IV disease that had a statistically significant poorer survival when p185 was overexpressed (P=0.005). Overexpression of theHer2/neu product p185 appears to be associated with intestinal-type gastric carcinoma and may sociated with intestinal-type gastric carcinoma and may help in identifying a subset of patients at increased risk for shorter survival.

    Notes: Zusammenfassung 1. Von der Sachverständigen Kommission des Office International des Fabricants du Cacao et du Chocolat wurde ein Einheitsverfahren zur Ermittlung von Lactose und Saccharose in Kakaoerzeugnissen aufgestellt, das die chemische Bestimmung der Zucker mit der polarimetrischen verbindet. Es erschien zweckmäßig zu prüfen, wie weit die damit erhaltenen Ergebnisse von den Inhaltsstoffen der Kakaokerne beeinflußt und angewendete bekannte Zuckermengen wiedergefunden werden. 2. Die Menge der reduzierenden Stoffe der Kakaokerne beträgt, ermittelt nach dem Einheitsverfahren, zwischen 0,56 und 1,97%, berechnet als Lactose. 3. Die Ermittlung der Lactose kann nach dem Einheitsverfahren mit großer Genauigkeit durchgeführt werden. 4. Bei der Ermittlung der Saccharose nach dem Einheitsverfahren treten Fehler auf, die, wie auch Th. von Fellenberg gefunden hat, auf Adsorption des Rohrzuckers durch den zum Klären verwendeten Niederschlag von Bleiphosphat zurückzuführen sind. 5. Die von H. Fincke vorgeschlagene Abänderung des Verfahrens zur Verbesserung der Ablesegenauigkeit beim Polarisieren ist sehr zweckmäßig und liefert bessere Werte als das ursprüngliche Verfahren. 6. Durch Ansäuern mit Essigsäure an Stelle des Klärens mit Bleiacetat und Natriumphosphat werden unter Befolgung der von Fincke vorgeschlagenen Abänderung bessere Saccharosewerte als nach der Arbeitsweise des ursprünglichen Einheitsverfahrens erhalten.

Bei der Rohfaserbestimmung besteht bei Verwendung von Glasfiltertiegeln die Gefahr, daß ein Teil der Mineralstoffe mit den am Glas haftenden Resten der Rohfaser im Tiegel zurückbleibt, bei der Veraschung nicht erfaßt wird und so zu unrichtigen Werten führt. 2. Die bei der Rohfaserbestimmung nach K. Scharrer und K. Kürschner verwendete Aufschlußflüssigkeit wirkt in der Kälte auf Filtrierpapier nicht ein. 3. Die Verwendung von Papierfiltern bei der Rohfaserbestimmung nach Scharrer und Kürschner bietet in mancher Hinsicht Vorteile, da sowohl der durch unvollständige Erfassung der Mineralstoffe verursachte Fehler vermieden wie auch das Filtrieren und Auswaschen erleichtert und damit die Arbeitszeit verkürzt wird. 4. Zum Auswaschen der Rohfaser nach Scharrer und Kürschner wird statt Alkohol zweckmäßig Aceton verwendet.

    Notes: Abstract Despite the importance of symptom control in the cancer population, few studies have systematically assessed the prevalence and characteristics of symptoms or the interactions between various symptom characteristics and other factors related to quality of life (QOL). As part of a validation study of a new symptom assessment instrument, inpatients and out-patients with prostate, colon, breast or ovarian cancer were evaluated using the Memorial Symptom Assessment Scale and other measures of psychological condition, performance status, symptom distress and overall quality of life. The mean age of the 243 avaluable patients was 55.5 years (range 23–86 years); over 60% were women and almost two-thirds had metastatic disease. The Karnofsky Performance Status (KPS) score was≤80 in 49.8% and 123 were inpatients at the time of assessment. Across tumour types, 40–80% experienced lack of energy, pain, feeling drowsy, dry mouth, insomnia, or symptoms indicative of psychological distress. Although symptom characteristics were variable, the proportion of patients who described a symptom as relatively intense or frequent always exceeded the proportion who reported it as highly distressing. The mean (±SD, range) number of symptoms per patient was 11.5±6.0 (0–25); inpatients had more symptoms than outpatients (13.5±5.4 vs. 9.7±6.0, p〈0.002) and those with KPS≤80 had more symptoms than those with KPS〉80 (14.8±5.5 vs. 9.2±4.9, p〈0.0001). The number of symptoms was highly associated with heightened psychological distress and poorer quality of life (for example, r=0.67 for the relationship with the Functional Living Index-Cancer, a QOL measure). These data clarify the prevalence and characteristics of cancer-related symptoms and suggest that the number of symptoms per patient may be a useful quality of life indicator.

    Notes: Summary Our previous investigations correlated the degree of cytogenetic and immunophenotypic heterogeneity of cultured normal glia, astrocytomas and malignant gliomas. The possible significance was suggested by the statistical correlation of individual antigens with diagnosis and patient survival [1]. The present study has established the patterns of covariation of titers of monoclonal antibody reactivity with a panel of cell surface antigens among normal glia (8), astrocytomas (4), anaplastic astrocytomas (12), mixed malignant gliomas (8) and glioblastomas (21). A mean aggregate titer across 43 antigens was computed for each culture and then subtracted from the observed individual titers. Factor analysis was performed to determine a small number of Factors, derived as the weighted average of the 43 mean-adjusted antigens, which accounted for a significant proportion of the covariation of immunophenotypic expression in the sample of 53 cultures. Clusters of antigens were found to independently segregate in their deviation from the aggregate phenotype. Adjusting for age and diagnosis, Factors 1 and 4 correlated with patient survival among recurrent and primary neoplasms, respectively. Factor 2 additionally discriminated between primary and recurrent gliomas. Factor 3 was associated with age at diagnosis. Factors 1 and 2 correlated with the histopathologic grade of glial tumor. Scatter plots of Factor 1 vs. 2 revealed the minimal immunophenotypic diversity of the normal glia. Astrocytomas were similar but not identical. Progressive divergence was evident between the immunophenotypes of anaplastic astrocytomas, mixed gliomas and glioblastomas. These data suggest that qualitative and quantitative differences in antigenic heterogeneity may identify stages in glial tumor progression. Furthermore, the aggregate immunophenotype may be a useful prognostic indicator, which is independent of established clinical variables such as age at diagnosis and pathologic grade of neoplasm. Factor analysis has been shown to be an efficient means of reducing the dimensionality of the data without sacrificing its informative character.
