    Notes: Summary A peculiar ultrastructural change of the cytoplasmic membrane is described. It concerns selective destruction of the outer leaflet of the luminal side of the capillary endothelium in the white matter of the rat brain. 30 min and 3h after intracerebral injection of arachidonic acid (AA). The cristae of the mitochondria disappeared in the endothelium of many capillaries. These changes were observed mainly in the central part of brain edema, where the extracellular space was markedly widened. The possible mechanism whereby these changes developed is discussed.

    Notes: Summary The content of norepinephrine (NE) in the ventricular, basal cisternal and lumbar cerebrospinal fluid (CSF) was determined in 19 patients with ruptured cerebral aneurysms. The cisternal CSF in patients with vasospasm contained a significantly higher level of NE (0.246 ± 0.049 ng/ml) compared with those without vasospasm (0.075 ± 0.001 ng/ml) (p 〈 0.001). However, this increase is not considered to be high enough to constrict cerebral arteries, unless there is an increased NE-sensitivity in subarachnoid haemorrhage. Vascular responses to NE in vivo were then studied after reversing blood-induced prolonged vasospasm of the rabbit's basilar artery through a transclival approach as well as a newly developed basal cisternal irrigation model. However, NE in molar concentrations between 1 × 10−10 and 10−2 failed to produce further contraction of the artery. In conclusion, the increase in NE with vasospasm might be only a secondary phenomenon, and not a causative factor of vasospasm. Early removal of subarachnoid blood clots seemed to prevent the development of vasospasm.

    Notes: Summary Intracellular lectin (Con A)-binding sites of human pituitary adenoma were examined by electron microscopy using the horseradish-peroxidase (HRP)-labelling technique. In this study were used 16 cases of human pituitary adenomas operated on in our clinics between 1977 and 1981: they concluded 4 each of PRL-, GH-, ACTH-producing and hormonal non-functioning adenomas. In parallel with the detection of lectin-binding sites, basal levels of their secreting hormones were determined by the radioimmunoassay technique, and their producing hormones were characterized light microscopically by the immunocytochemical HRP-labelling technique. In the present study, for hormonal functioning adenoma cells, mature or large granules of each specific type of adenoma cells had no definite Con A-binding sites. On the other hand, immature or small secretory granules of RPL- or GH-producing adenoma cells showed a positive reaction with Con A. ACTH-producing adenomas so far examined revealed no definite binding sites. Some variable results were obtained concerning non-functioning adenomas. Lectins have been and will be very useful in the detection of different subtypes of adenomas in each specific group.

    Notes: Summary Lectin-binding sites of the human pituitary adenoma cells were examined by electron microscopy in correlation with their functional states. In this study used were 37 cases of the human pituitary adenomas which had been operated in our clinics from 1977 to 1979. They were divided into four groups: 13 cases of PRL-producing adenomas, nine of hGH-producing adenomas, three of ACTH-producing adenomas, and 12 of hormonal non-functioning adenomas. In parallel with the detection of the lectinbinding sites by means of the ferritin-labeling method, the basal levels of their secreting hormones were determined by the radioimmunoassay technique, and their producing hormones were characterized light microscopically by the immunocytochemical HRP-labeling technique. The present study clearly shows that the ferritinlabeled, lectin-binding sites of the human pituitary adenoma cells prefixed with aldehyde are different in number between PRL- or hGH-producing adenoma cells and non-functioning ones. Morphologically resemblin, non-functioning adenoma cells and mixed somatotrophic and lactotrophic adenoma cells are definitely distinguished from each other by the findings of the ferritin-labeled RCA-binding sites' distribution pattern. Lectins have been proved very useful to evaluate the membrane characteristics of the human pituitary adenoma cells in association with their functional states.

    Notes: Summary A case of anterior sacral meningocele which was clearly visualized on whole body computerized tomography is presented. Posterior surgical approach with ligation of the narrow communication between the sacral dural sac and the meningocele alleviated the subjective complaints of a feeling of abdominal distention and of dysuria. For the purpose of postoperative follow-up study, examination by whole body computerized tomography proved to be the best method of evaluating the size of the meningocele and also the state of neighbouring structures.

    New approach to brain tumour chemoradiotherapy with cellular synchronization by colcemid (1976)

    Notes: Summary Cellular synchronization using Colcemid as pretreatment for combined chemoradiotherapy was investigated. C6 rat brain tumour was cultured in RPMI medium containing 10−5-10−7 Mol. of Colcemid for 24 hours. The basic cell kinetics were analysed with a Pulse Cytophotometer, which facilitated the analysis of tumour cell cycle phase distribution according to the DNA content. The effect of Colcemid depended on the concentration, and the minimal concentration showing continuous blocking during 48 hours after removal of the drug was 10−6 Mol. G1 fraction of 2 C DNA content was reduced from 74% to 36%. G2-M phase of 4 C DNA content increased from 9% to 28%. S phase cells increased from 17% to 31%. Polyploid cells in the Tetraploid cell cycle could be recognized. The remaining 36% of cells within the G0+G1 peak of 2 C DNA content were considered to be diploid G0 cells.

    Notes: Summary The degradation of poly(l-glutamic acid) and poly(l-lysine) by X-rays and ultraviolet radiation has been studied at variouspH values, including the helix-coil transition region. It was found that X-rays cause maximum degradation in the transition region for both polypeptides, indicating that the efficiency of degradation is affected not only by the secondary structure but also by some other factors related to the transition. On the contrary, the degradation by monochromatic ultraviolet 2537 Å radiation showed a strong dependence on helix content of PLGA, suggesting that degradation occurs more effectively in the helical form than in the coil form. Theoretical calculations were made to approximate thepH dependence of degradation, assuming different cross sections for the peptide bonds in helix, coil, and junctions between helical and coil regions.
