    Notes: Summary Two min of intraluminal perfusion of the rat proximal tubules with autologous serum induced marked ultrastructural alterations including extensive cytoplasmic vesiculation due to swelling of rough endoplasmic reticulum cisternae and occasional extrusion of nuclei and cytoplasm into the lumen. Within 4 min pronounced vesiculation of mitochondria was observed. These findings are consistent with the notion that serum-induced inhibition of proximal tubular fluid absorption is due to cell lysis, presumably mediated by complement activation.

    Notes: Summary To investigate early immunological disturbances at the onset of diabetes, lymphocyte function and islet cell surface antibodies were studied in streptozotocin-treated C57BL/10 and B10.BR mice. In C57BL/10 mice, streptozotocin given in multiple low doses depressed lymphoblastic transformation to phytohaemagglutinin and pokeweed mitogen, but not to concanavalin A on day 6 after the first administration. On day 20, the transformation remained suppressed with phytohaemagglutinin, but recovered to the control level with pokeweed mitogen. In the early phase after treatment, the islet cell surface antibodies were elevated and then declined. Single high dose administration depressed responses to phytohaemagglutinin with no detectable islet cell surface antibodies. In B10.BR mice transformations to pokeweed mitogen and concanavalin A were suppressed in the early phase. The strain of mice may be a factor to be considered. Thus, it was suggested that the deterioration of immunological function with the formation of islet cell surface antibodies preceded the onset of hyperglycaemia in mice treated with multiple low doses of streptozotocin.

    Notes: Summary The hypotensive effect of acute and long-term, intravenous and oral administration of the calcium antagonist, diltiazem, was investigated in 8 normotensive volunteers and 55 patients with essential hypertension. Diltiazem i.v. infusion of 45 mg/h (0.5 mg/min, then 1.0 mg/min, each for 30 min rapidly decreased both blood pressure (BP) from 164±22/98±8 to 144±15/86±9 mmHg (mean±SD) and total peripheral resistance from 32.6±8.4 to 25.3±5.4 mmHg/l/min (p〈0.001), and increased stroke volume from 58.2±9.5 to 64.2±8.6 ml/beat (p〈0.05). It altered neither heart rate nor cardiac output in the hypertensives (n=10). Oral diltiazem 60 mg rapidly decreased BP from 155±10/103±6 to 142±12/90±8 mmHg after 3 hours (p〈0.01/p〈0.001) in hypertensives (n=8), but not in normotensives (n=8). Diltiazem 90 mg p.o. decreased BP from 157±15/102±9 to 129±13/83±8 mmHg (p〈0.01) in hypertensives (n=15), and reduced the heart rate from 71±8 to 65±8 beats/min (p〈0.01). The drug did not change plasma renin activity either in normotensives or hypertensives. The fall in diastolic BP was correlated with the plasma diltiazem concentration (r=0.910, n=6, p〈0.05). Long-term treatment with diltiazem 30mg t.d.s. decreased BP from 163±12/104±8 to 145±9/88±9 mmHg (p〈0.001, n=13), and 60mg t.d.s. decreased BP from 169±15/102±6 to 148±13/87±8 mmHg (p〈0.001, n=8), and significantly reduced the heart rate (p〈0.01) in hypertensives. Thus, the hypotensive action of diltiazem, which is due to arterial dilatation, is effective, either on intravenous or oral administration, during acute and long-term treatment of essential hypertension.

    Notes: Summary The acute and long-term hypotensive effects of low doses of nifedipine, and the correlation between the fall in the blood pressure (BP) and the plasma nifedipine concentration, were investigated in patients with essential hypertension. The oral administration of nifedipine 5 mg rapidly decreased BP from 163±22/101±10 to 127±12/82±9 mmHg (mean±SD; p〈0.001), and increased heart rate from 72±8 to 76±6 beats/min (p〈0.05), plasma renin activity rose from 1.2±0.6 to 1.4±0.8 ng/ml/h (p〈0.05), and plasma nifedipine concentration was 75.6±22.0 ng/ml 30 min after administration (n=7). The nifedipine concentration was significantly correlated both with the fall in BP (r=0.410, p〈0.02, n=31) and the rise in the heart rate (r=0.412, p〈0.02, n=31). Treatment with nifedipine 5 mg t.d.s. alone or in combination either with propranolol 10 mg t.d.s., or thiazide 1 tablet daily, or propranolol and thiazide, controlled BP in 36 patients during the 22 week study period. During the long-term nifedipine therapy, the plasma nifedipine level was significantly correlated with the fall in systolic (r=0.577, p〈0.01, n=20) and diastolic (r=0.595, p〈0.01, n=20) BP. It was concluded that the plasma nifedipine concentration could be correlated with the fall in BP, and that low doses of nifedipine, either as monotherapy or in combination, were effective in the acute and long-term treatment of patients with essential hypertension.

    Description / Table of Contents: Zusammenfassung Es wird eine einfache und genaue Methode zur Bestimmung von Eisen im Blut unter Verwendung von Säureaufschluß im Heizblock beschrieben. Der Aufschluß benötigt wenig Mühe und erlaubt die Verarbeitung einer großen Zahl von Proben. Der verdünnte Aufschluß wird zur kolorimetrischen Eisenbestimmung mittels o-Phenanthrolin benutzt und die entsprechende Hämoglobinkonzentration im Blut berechnet. Es zeigte sich eine gute Korrelation der Ergebnisse mit der beschriebenen und der Cyanmethämoglobin-Methode anhand von 21 Standardproben, die 4,9 bis 26,8 g/dl Hämoglobin enthielten.
    Notes: Summary A simple and accurate determination of iron in blood involving acid digestion in a heating block is described. The digestion requires little attention and permits the handling of large numbers of samples. The diluted digest is used for the colorimetric determination of iron using o-phenanthroline, and the equivalent hemoglobin in blood is calculated. There was a good correlation between the results of the present method and the cyanmethemoglobin method with 21 standard blood samples containing 4.9–26.8 g/dl of hemoglobin.

    Notes: Summary Neutralizing (NT) and hemagglutination-inhibiting (HI) antibodies to calf diarrhea coronavirus (CDCV) and hemagglutinating encephalomyelitis virus of swine (HEV) (strain 67 N) were detected in high proportions of normal adult cattle and pigs in Japan. Since comparison of NT and HI titers in the serum samples suggested an antigenic difference between the viruses, cross NT and HI tests of these viruses were carried out with antisera raised in rabbits. The homologous NT titers were markedly higher than the heterologous titers. In HI tests essentially the same results were obtained. These findings indicate the presence of a marked difference in antigenic make-up between CDCV and HEV. NT and HI tests can clearly differentiate the viruses, although there is some cross reaction.

    Notes: Summary Three cytopathic rotavirus strains were isolated in MA104 cells from faecal specimens of pediatric patients with acute gastroenteritis. Pre-treatment of virus with trypsin and incorporation of a small amount of trypsin in maintenance medium were important for establishment of the strains in these cells. The isolates were antigenically closely related with strain Wa of human rotavirus and had some antigenic relationship with strain Lincoln of bovine rotavirus.

    Notes: Summary A causative agent, provisionally designated as CARS, was isolated from the enlarged submaxillary gland of rat which was characterized as a sialoadenitis, using mouse-derived Balb/c3T3 clone A31 (3T3) cell culture. The virus could be propagated in 3T3 cell culture where it produced multinucleated giant cells and formed clear plaques. It was identified as a member of the coronavirus group from the following results: RNA content was suggested by the lack of the effect of cytosine arabinoside, the infectivity was sensitive to lipid solvents and inactivated at 56° C for 5 minutes, the viral particle showed typical coronavirus morphology which was approximately 100 nm in diameter. Serologically, although CARS and sialodacryoadenitis virus (SDAV) actually belonged to the rat-coronavirus group, some antigenic variations existed between these two agents in the results of both neutralization and complement-fixation tests using monovalent antisera. When inoculated intranasally into susceptible rats, CARS caused clinically and histologically overt sialoadenitis as observed in the natural outbreak, and retained its virulence for rats in several passages of mouse brain and even when cell culture was used, while rats which were inoculated with SDAV were asymptomatic.

    Notes: Summary A reversed passive hemagglutination (RPHA) method was developed for the detection of bovine coronavirus in fecal specimens. Sheep erythrocytes fixed with glutaraldehyde, and then treated with tannic acid were coated with anti-bovine coronavirus rabbit antibodies purified by affinity chromatography using bovine coronavirus linked to Sepharose 4B. The RPHA test was carried out by a microtiter method. Erythrocytes coated with purified specific antibodies were agglutinated by bovine coronavirus, but not by bovine rotavirus or enterovirus. The reaction was inhibited by antiserum to bovine coronavirus, confirming the specificity of the reaction. The RPHA test detected bovine coronavirus in 13 of 22 fecal specimens (59 per cent), from natural cases of diarrhea, while the positive rates were only 14 per cent (3/22) and 22 per cent (5/22) for immunofluorescent staining of primary cultures of calf kidney cells infected with the specimens, and immune electron microscopy respectively. The advantages of the RPHA method are its simplicity, high sensitivity and rapidity.
